Effect of Melissa officinalis L. leaf extract on lipid accumulation by modulating specific adipogenic gene transcription factors in 3T3-L1 adipocytes

  • The objective of this study was to investigate the effects of a hypodermic injectable solution comprised of an LPM LB meso solution containing Melissa officinalis L. leaf extract (LPM) on the lipogenesis in the 3T3-L1 cells line. The lipid accumulation measured by oil red o staining in the 3T3-L1 adipocytes treated with LPM, which was reduced in a dose dependent manner and showed 91.7 to 62.9% compared to control group. Its effectiveness with a 50% solution was significantly higher than the hydroxycitric acid (positive control) treatment without showing cell cytotoxicity. In a quantitative real-time PCR, it was demonstrated that the LPM treatment appeared to upregulate the mRNA expression of the adipogenesis-related genes, which included the peroxisome proliferator-activated receptor gamma (50% concentration) while down-regulating the CCAAT-enhancer binding protein alpha (50% concentration) and the sterol regulatory element-binding protein 1c (10, 25, and 50% concentrations). The results from the current study suggest that the LPM could be useful biomaterials that can inhibit obesity in the 3T3-L1 cells, which could possibly be by regulating the specific adipogenic gene transcription factors.

후박(厚朴)과 토후박(土厚朴)의 소장운동에 미치는 영향에 대한 연구 (The Effects of Magnoliae officinalis Cortex and Machili thunbergii Cortex on Small Intestinal Motility)

  • Objectives : Magnoliae officinalis Cortex (MOC) has been used in traditional medicine for digestive diseases in Korea, China and Japan. However, Machili thunbergii Cortex (MTC) also has been used as a substitute of MOC in Korea sometimes. Thus, this study was carried out to investigate and compare the effects of MOC and MTC on intestinal motility of isolated small intestinal segments from ICR mouse. Methods : Changes in motility were recorded via isometric transducers connected to a data acquisition system and amplitude, frequency and area under the curve (AUC) of intestinal spontaneous phasic contraction were compared. Results : The MOC extracts ($1{\sim}{\mu}g/mL$) dose-dependently decreased both amplitudes and frequencies of the spontaneous phasic contraction, but not AUC. However, high concentration of MOC (100 ${\mu}g$/mL) evoked tonic contraction. And it was not inhibited by tetrodotoxin, a sodium channel blocker, and nifedipine, a L-type $Ca^{2+}$ channel antagonist. These results suggested that MOC (100 ${\mu}g$/mL)-induced tonic contraction is not mediated by nerve or L-type $Ca^{2+}$ channel. On the other hand, the MTC extracts dose-dependently inhibited amplitude and AUC, but not the frequency. Conclusions : Although both MOC and MTC affected intestinal motility, MOC is more effective on intestinal motility than MTC. And MOC has been used as a traditional medicine for a long time but not MTC. Thus, we suggested that MTC should not be used in Korea as a substitute of MOC and MOC might be useful traditional medicine for gastrointestinal disease. The mechanism of MOC is still remained to elucidate.

Neuropharmacological study of some Ayurvedic medicinal plants

  • Water extract (kwath) of six different widely used Ayurvedic medicinal plants were tested in mice for possible neuropharmacological efficacy. In the present experiments it was observed that a number of plant tested causes a significant level of Central Nervous System (CNS) depression, in that it significantly decreased the spontaneous Motor activity, and also lowered the exploratory behavior of the treated animals. Terminalia chebula (HAA), Terminalia bellerica (BHA), Emblica officinalis (AA), Piper longum 1. (PP). exhibited depressant action of on the CNS. Apart from them a mild to moderate degree of depression was evident as a consequence of administration of Zingiber officinale Rosc (SUT), Piper nigrum L. (MRC). However, none of the plant tested did not exhibit significant effects on pentobarbital induced narcosis, and this indicated that the sedating effects of the drug was not associated with the risk of fatal consequences on overdose.

Protective Effect of Sanguisorba officinalis L. Root on Amyloid ${\beta}$ Protein (25-35)-induced Neuronal Cell Damage in Cultured Rat Cortical Neuron

  • Sanguisorbae radix (SR) from Sanguisorba officinalis L. (Losaceae) is widely used in Korea and China due to its various pharmacological activity. The present study aims to investigate the effect of the methanol extract of SR on amyloid ${\beta}$ Protein(25-35) $(A{\beta}\;(25-35))$, a synthetic 25-35 amyloid peptide, -induced neurotoxicity using cultured rat cortical neurons. SR, over a concentration range of $10-50\;{\mu}g/ml$, inhibited the $A{\beta}$ (25-35) $(10\;{\mu}M)-induced$ neuronal cell death, as assessed by a 3-[4,5-dimethylthiazole-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and the number of apoptotic nuclei, evidenced by Hoechst 33342 staining. Pretreatment of SR $(50\;{\mu}g/ml)$ inhibited $10\;{\mu}M\;A{\beta}$ (25-35)-induced} elevation of cytosolic calcium concentration $([Ca^{2+}]c)$, which was measured by a fluorescent dye, fluo-4 AM. SR $(10\;and\;50\;{\mu}g/ml)$ inhibited glutamate release into medium induced by $10\;{\mu}M\;A{\beta}(25-35)$, which was measured by HPLC, and generation of reactive oxygen species. These results suggest that SR prevents $A{\beta}$ (25-35)-induced neuronal cell damage in vitro.

후박 추출물의 들깨 녹병과 잔디 녹병에 대한 방제 효과 (Disease Control Efficacy of the Extract of Magnolia officinalis against Perilla and Zoysiagrass Rusts)

  • 녹병은 Puccinales 목의 병원성 곰팡이가 일으키는 식물병으로서 다양한 식물에 발병한다. 최근 많은 연구자들은 녹병을 효과적이고 친환경적으로 방제하기 위하여 연구를 활발히 진행하고 있다. 본 연구에서는 당후박(Magjia90) 추출물로부터 제조된 유제(EC10)와 수화제(WP20)의 들깨 녹병과 잔디 녹병에 대한 방제효과를 포장에서 조사하였다. Magjia90-EC10과 Magjia90-WP20은 들깨 녹병에 대하여 47.9% 내지 69.6%의 방제효과를 보였고, 수화제는 유제보다 우수한 효과를 보였다. 또한 Magjia90-WP20은 전디 녹병에 대하여 65.7% 내지 80.5%의 높은 방제효과를 보였다. 다른 한편으로, 두 제제를 처리한 식물체 잎에서 어떠한 약해도 발견되지 않았다. 이상의 결과는 Magjia90이 녹병 방제를 위한 천연물 살균제로서 사용이 가능하다는 것을 강력하게 나타낸다.

로즈마리(Rosmarinus officinalis L.) 추출물로부터 Helicobacter pylori에 대한 항균물질 분리 및 동정 (Isolation and Identification of Antimicrobial Compounds against Helicobacter pylori from Rosemary (Rosmarinus officinalis L.) Extracts)

  • 로즈마리 추출물의 phenol 함량은 25.7 mg/g으로 매우 높게 나타났다. Helicobacter pylori 에 대한 항균효과를 측정한 결과, clear zone이 $100{sim}200{\mu}/disc$의 phenol 농도에서 각각 11, 12, 14 mm의 clear zone을 형성하여 추출물의 농도가 증가할수록 H. pylori 에 대한 저해효과가 농도 의존적으로 높아지는 것을 확인 할 수 있었다. Sephadex LH-20과 MCI-gel CHP-20 column을 이용하여 normal phase type인 $ethanol{\rightarrow}H_2O$와 reverse phase type인 $H_2O{\rightarrow}methanol$로 용액의 농도를 증가시키면서 용출한 결과 H. pylori에 대한 탁월한 저해활성을 나타낸 5종류(compound A~E)의 단일 물질로 분리하였다. 5가지 각각의 물질은 H. pylori에 대한 항균력이 미약하였으나, 2종류 이상의 다중 물질의 항균 시너지 효과에 의해 각각 10~16 mm의 clear zone을 형성하여 우수한 항균력을 나타내었으며, fast atom bombardment (FAB)-Mass, $^1H$-NMR, $^{13}C$-NMR과 IR spectrum을 사용하여 구조 동정한 결과 protocatechuic acid, coumaric acid, caffeic acid, chlorogenic acid 및 rosmarinic acid 인 것으로 확인되었다.

지유 에탄올추출물의 생체외 항산화 활성 (In vitro Antioxidant Activity of Sanguisorbae Radix Ethanol Extracts)

  • 본 연구에서는 지유 에탄올추출물의 항산화 효과를 조사하였다. Pyrogallol의 억제율을 100%로 기준하였을 때, DPPH 라디칼을 50% 억제시키는데 필요한 지유 추출물의 농도는 0.33 mg/mL으로 ${\alpha}$-tocopherol의 $IC_{50}$(0.40 mg/mL)과 유사하게 나타났다. 지유 추출물의 총항산화능은 ${\alpha}$-tocopherol에 비해 높게 나타났다. 지유 추출물의 superoxide 소거활성은 catechin에 비해 높게 나타났다. 지유 추출물의 peroxyl 라디칼 소거활성은 ascorbic acid에 비해 높게 나타났다. 지유 추출물의 구리이온 환원력은 ${\alpha}$-tocopherol에 비해 높게 나타났다. 지유 추출물은 hydroxyl 라디칼 및 peroxyl 라디칼로 유발된 supercoiled DNA strand의 절단을 억제시켰다. 지유 추출물 0.5 및 5 mg/mL의 총페놀 함량은 각각 0.50 및 3.33 mM gallic acid와 동등한 수준이었다. 또한, HepG2 세포주를 이용한 세포배양에서 지유 추출물 0.01, 0.1 및 0.5 mg/mL 농도의 첨가는 0.2 mM t-BHP로 유도된 세포독성을 각각 33.8, 79.1 및 96.9% 감소시켰다. 따라서, 본 연구 결과들은 지유 추출물의 강력한 항산화 효과와 세포독성 억제효과를 나타내며, 이러한 효능은 적어도 자유라디칼의 산화억제와 높은 총페놀 함량에 기인하는 것으로 사료된다.

  • 로즈마리(Rosmarinus officinalis)와 라벤더(Lavandura spica L.)의 잎 절편으로부터 식물생장조절물질의 첨가에 따른 캘러스 유기율, 신초 및 뿌리의 분화율을 조사한 결과와 유기된 캘러스의 항산화 활성은 다음과 같다. 로즈마리의 캘러스 유기율은 2,4-D의 모든 처리구에서 양호 하였고, 특히 배양 90일째 2,4-D 1.0mg/L처리구에서 94%의 유기율로 가장 좋은 반응을 보였다. 로즈마리의 항산화 활성은 식물생장조절물질에 따라 다른 경향을 보였고, 2,4-D 0.1mg/L를 첨가하여 배양된 캘러스에서 $43{\mu}g(RC_{50})$으로 가장 높았으며, 2,4-D의 농도가 낮을 수 록 항산화 활성이 증가됨을 알 수 있었다. 또한 NAA, BA는 2,4-D와 반대로 농도가 증가함에 따라 항산화 활성이 증가됨을 알 수 있었다. 라벤더의 캘러스 유기는 2,4-D, NAA 및 BA의 모든 처리구에서 양호 하였으며, 로즈마리와 동일하게 2,4-D 1.0mg/L 처리구에서 98%의 유기율로 가장 양호하였다. 뿌리의 형성은 NAA 1.0mg/L을 첨가하였을 때 85%의 분화율로 가장 양호 하였다. 라벤더의 항산화 활성은 잎의 경우 $69.5{\mu}g(RC_{50})$으로 낮은 항산화 활성을 보였지만 2,4-D 0.1mg/L을 첨가하여 배양된 캘러스에서 $37.6{\mu}g(RC_{50})$으로 잎 보다 2배 정도 강한 항산화 활성을 보였으며, BA의 모든 처리구에서도 잎 보다 강한 항산화 활성을 보였다.

Assessment on Antioxidant Potential and Enzyme Activity of Some Economic Resource Plants

  • The antioxidant potential and enzyme activities in Salicornia herbacea, Corylopsis coreana, Erythronium japonicum, Phragmites communis, Momordica Charantia, Nelumbo nucifera, Salvia plebeia, Portulaca oleracea, Ficus carica, Citrus junos and Cornus officinalis were determined. Their antioxidant activities were measured using DPPH radical scavenging and nitrite scavenging activity. Enzyme activities in investigated plants were evaluated as superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX). The DPPH scavenging rate from 100 to 2500 $mgL^{-1}$ was the highest in the flower of Corylopsis coreana. However, it was not detected in most of the samples at concentration below 100 $mgL^{-1}$. The nitrite scavenging activity according to each kind of resource plants was significantly higher in the stem of Corylopsis coreana and leaf of Nelumbo nucifera. The root extract of Erythronium japonicum had the highest SOD enzyme activity of 94.0% while leaf of Salvia plebeia showed the lowest SOD enzyme activity of 30.4%. The activity of CAT and APX showed higher values in the stem of Corylopsis coreana, root of Erythronium japonicum and root of Phragmites communis in comparison with other plants. The activity of POD showed significantly high values in stem of Corylopsis coreana, Momordica Charantia and pericarp of Citrus junos extracts. The antioxidant enzyme activities differ significantly in different plants. In conclusion, we showed that Corylopsis coreana, Erythronium japonicum Cornus officinalis, and Momordica Charantia had the potent biological activities. Therefore, these plant resources showing antioxidant activity could be good materials for development of source of functional healthy food.

A Randomized trial to investigate the effect of Asparagus racemosus root tablet in Relieving Postmenopausal Hot Flashes

  • Background and Objectives: As per the available data, 75% peri and postmenopausal women experience hot flashes having negative effect on their quality of life. Using herbal medicines are stepping forward to alleviate hot flashes as the available hormonal medications have been reported the presence of side effects. This study was planned with the intention to investigate the effect of Asparagus racemosus root in the treatment of postmenopausal hot flashes in comparison to the placebo. Methods: This prospective patient blinded clinical trial randomly assigned 40 postmenopausal women with symptoms of hot flashes to receive either test drug (n=20) comprising Asparagus racemosus tablet of 1g in the dosage of 2 tablet thrice in a day for the duration of 60 days continuously or to receive placebo tablet prepared from roasted wheat flour in the same dosage and duration as test drug. The efficacy was determined by reduction in the frequency and intensity of hot flashes assessed by daily diary along with the improvement in quality of life assessed through Hot Flash-Related Daily Interference Scale. Statistical analysis was accomplished by student t test and Chi-square/Fisher Exact test. Results: Test drug significantly (p <0.001) reduced the intensity and frequency of hot flashes as well as improved the quality of life without showing any side effect. Discussion & Conclusion: According to Unani scriptures, the treatment is to treat the underlying cause, such as aberrant temperament and psychological and environmental issues. Unani drugs have emmenagogue, anti-inflammatory, analgesic, and neuroprotective qualities that can help alleviate premenstrual symptoms. Unani herbs such as C. sativus, Vitex, agnus castus, P. vulgare, N. Jatamansi, M. officinalis, and Z. officinalis have been clinically demonstrated to be effective in PMS. Thus, traditional knowledge authentication and conservation are vital for future research and appreciated for application in the modern day. Furthermore, randomised controlled trials, comprehensive reviews, and meta-analyses are suggested.
