Growth of Containerized Chamaecyparis obtusa Seedlings as Affected by Fertilizer and Container Volume (시비수준 및 용기용적에 따른 편백 용기묘의 생장 특성)

  • This study was carried out to evaluate the growth characteristics, carbon and nitrogen content of containerized 1-0 Chamaecyparis obtusa seedlings at various levels of fertilizer (2 g/L, 1 g/L, control) and three container volumes (500 mL, 400 mL, 320 mL). The growth of root collar diameter was highest in the 2 g/L (3.14 mm), followed by the 1 g/L (2.75 mm) and control (2.41 mm) treatments, while the height of seedling was significantly higher in the 1 g/L (21.88 cm) than other treatments (2 g/L: 20.92 cm; control: 19.06 cm). The growth of root collar diameter by container volume was better in the 500 mL than in the 320 mL. Dry weight of seedling was the highest in the 1 g/L ($4.41g\;seedling^{-1}$), followed by the control ($3.67g\;seedling^{-1}$) and the 2 g/L ($2.92g\;seedling^{-1}$) treatments. The dry weight of seedlings by container volume was significantly higher in the 500 mL than in the 320 mL. Nitrogen concentration in foliage was ranged from 1.51% in the control to 2.45-2.60% in the fertilizer treatments. However, carbon concentration of seedlings was not affected by the fertilizer or the container volume treatments. The growth of seedlings following planting in mountain area was better in the fertilized seedlings compared with in the unfertilized seedlings. The results indicate that the 1 g/L fertilization was an optimum rate for growth following planting of Chamaecyparis obtusa seedlings.

Antifungal Activity of the Extracts from Veratrum album L. var. grandiflorum Max. (백려노근경(白藜蘆根莖) 추출물(抽出物)의 항진균작용(抗眞菌作用))

  • Although numerous drugs are available for the treatment of superficial fungi infections of skin, the clinical effects of the majority of such drugs are not satisfactory. In the hope of searching the effective drugs for superficial fungi infections, authors studied whether Veratrum rhizoma extracts had any effect on fungi, with water extract (VRWE), ethanol extract (VREE) and methanol extract (VRME) from Veratrum album L. var. grandiflorum Max. In in vitro studies, the spores of fungi were inoculated on Sabouraud's glucose agar media which contained three extracts of Veratrun rhizoma in each concentration of $500\;{\mu}g/ml$, $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$ respectively, and the growth of the fungi were observed for 3 weeks. The species of the fungi used in these experiments were Epidermophyton floccosum, Microsporum canis, Microsporum nanum, Microsporum gypseum, Microsporum cookei, Trichophyton rubrum, Trichophyton mentagrophytes, Trichophyton tonsurans and Trichophyton verrucosum. The results of the studies were as follows: 1. The growth of M. canis, M. nanum, T mentagrophytes and T. tonsurans were slightly inhibited by VRWE $1,000\;{\mu}g/ml$, and with VRWE $5,000\;{\mu}g/ml$, the growth of E. floccosum, M. gypseum and T. rubrum were slightly inhibited, moderate inhibition on the growth of M. canis, M. nanum, M. cookei, T. mentagrophytes and T. tonsurans were showed by VRWE $5,000\;{\mu}g/ml$. 2. With $500\;{\mu}g/ml$ of VREE, the inhibition on growth of E. floccosum, M. nanum and M. gypseum were slight, however significant inhibition on the growth of M. canis, M. cookei, T. mentagrophytes, T. rubrum and T. tonsurans were observed. The growth of M. nanum and M. gypseum were moderately inhibited, and significant inhibition on the growth of E. floccosum, M. canis, M. cookei, T. mentagrophytes, T. rubrum and T. tonsurans were observed by VREE $1,000\;{\mu}g/ml$. By VREE $5,000\;{\mu}g/ml$, the growth of all tested fungi were significantly inhibited except T. verrucosuia being showed slight inhibition. 3. Significant inhibition on the growth of M. canis, T, mentagrophytes, T. rubrum and T. tonsurans were noted, and moderate inhibition of M. nanum, slight inhibition of E. floccosum and M. gypseum in growth were observed by VRME $500\;{\mu}g/ml$. The growth of E. floccosum, M. canis, M. nanum, M. cookei, T. mentsgrophytes, T. rubrum and T. tonsurans were significantly inhibited by VRME $1,000\;{\mu}g/ml$, and that of M. gypseum was moderate. With $5,000\;{\mu}g/ml$ of VRME, significant inhibition on the growth of E. floccosum, M. canis, M. nanum, M. gypseum, M. cookei, T mentagrophytes, T. rubrum and T. tonsurans were observed, and T. verrucosum was showed only slight inhibition. From the above results, it was found that the extracts of organic solvents from Veratrum rhizoma (VREE & VRME) exerted significant antifungal activity, and their effects were probably derived from the pharmacological action of steroidal alkaloids.

Settling Characteristics of Phosphorus in Nakdong Estuary Barrage (낙동강 하구호에서의 인의 침강특성)

  • A study on the characteristics of phosphorus sedimentation in Nakdong Estuary Barrage was conducted in June and July, 1989. Cylindric type sediment trap was designed for collecting of downward material. Downward flux and settling velocity of downward material were $296g/m^2/d$, 1.25m/hr in mid-layer and $955g/m^2/d$, 3.31m/hr in bottom-layer, respectively Downward flux and settling velocity of phosphorus were $0.64g/m^2/d$, 0.85m/hr in mid-layer and $1.97g/m^2/d$, 1.89m/hr in bottom-layer, respectively. Fractional composition of organic phophorus in downward material was $51.9\%$ in mid-layer water and $48.3\%$ in bottom-layer water. According to the result of this study, sedimentation of phosphorus plays an important role as a sinking effect in Nakdong Estuary Barrage.

Effects of No-tillage Rice Cover Crop Cropping Systems on Rice Root Growth (무경운 피복작물 작부체계가 벼 뿌리 생육에 미치는 영향)

  • This study was conducted to evaluate the effect of rice cover crop cropping systems on rice root growth in a rice field as affected by conventional tillage without rice straw or green manure crop treatment (CTFS, check plot), no-tillage without cover crops (NTNT), no-tillage amended with rape (NTRA), no-tillage amended with rye (NTRY), no-tillage amended with hairyvetch (NTHV), and no-tillage amended with Chinese milk vetch (NTCM). In 0-5 cm soil depth, dry weight of root in NTRS ($128g\;m^{-2}$) was significantly higher than in the other plots (p<0.05) at harvesting stage. In addition, content of active organic matter at 0-5 cm soil depth was $1,684g\;m^{-2}$ in NTCM, $1,309g\;m^{-2}$ in NTRA, $1,295g\;m^{-2}$ in NTRS, $1,072g\;m^{-2}$ in NTRY, $917g\;m^{-2}$ in NTHV, $434g\;m^{-2}$ CTFS, and $426g\;m^{-2}$ in NTNT treatment. In no-tillage rice cover crop cropping system, our findings suggest that NTRS and NTCM should be enhanced root growth and active organic matter in paddy field.

Effect of Modified Cyclone Dust Collector in Windowless Broiler Building (변형된 사이클론 먼지포집기의 이용이 무창계사의 먼지농도에 미치는 영향)

  • This study was carried out to develop the modified cyclone dust collector in windowless broiler building measuring 12 m wide, 46 m long, with a side wall height of 3 m and a capacity of 12,800 birds. Ventilation rate in windowless broiler building was $0.014{\sim}0.015\;and\;0.158{\sim}0.181cmm$ in second weeks and fifth weeks of age, respectively. Ammonia concentration was $13{\sim}16\;and\;20{\sim}24\;ppm$ in first and second weeks of age, respectively. Amount of dust collected in dust collector each week was 104.1, 274.7 and 388.6g in first, second and third weeks of age. But it was decreased from fourth weeks of age because of the increased ventilation rate. Total suspended particulate(TSP) of polluted air was $3,111.7{\sim}8,745.2{\mu}g/m^3$, but it was decreased to $530.8{\sim}2,264.4{\mu}g/m^3$ when it passed through the dust collector. Collecting efficiency was $54.2{\sim}82.9%$ in TSP. But collecting efficiency of Particulate matter smaller than $1{\mu}g$ (PM1.0) was $7.6{\sim}33.8%$, and lower than TSP. TSP concentration in control broiler house was 1,387.6 and $4,210.1{\mu}g/m^3$ in first and fourth weeks of age, respectively. But it was decreased to 876.3 and $2,535.3{\mu}g/m^3$ in broiler house operated dust collector in first and fourth weeks of age, respectively. Dust collecting efficiency for TSP was $36.8{\sim}39.8%$, and it was decreased to $11.4{\sim}39.8%$ in smaller dust size.

  • The storage stabilities of immunoliposomes incorporated with variable amounts of ganglioside $G_{M1}$ were investigated as a function of time. temperature. and composition by observing absorbance of visible light and calcein release. In the column chromatographe, the layer of unsaturated PE(dioleoylphosphatidylethanolamine : DOPE). unable to form stable liposomes at physiological temperature and pH, were formed when palmitoyl-immunoglobulin G(IgG) $(2.5{\times}10^{-4}\;mol/DOPE\;mol)$ added. The incorporation of ganglioside $G_{M1}$ into immunoliposome. enhanced the stabilities of bilayers during the extended period of storage. The turbidities of immunoliposomes coated with ganglioside $G_{M1}$ exhibited the maximum near 20 mol% $G_{M1}/DOPE$ mol. probably because of the disturbance of the bilayer characteristics, i.e., layer transition or reorientation of interaction sites. At low temperature. the higher stability was achieved than at elevated temperatures. After one week of storage. the redispersed liposomal solutions at lower temperatures maintained the original elution patterns in chromatography but broader distribution at elevated temperatures. During the storage, it is suggested the aggregation is the more dominant phenomena for liposomes kept at $5^{\circ}C$ than the fusion. while he fusion is at elevated temperatures.

Benzene Biodegradation Using the Polyurethane Biofilter Immobilized with Stenotrophomonas maltophilia T3-c

  • The benzene removal characteritics of the polyurethane (PU) biofilter immobilized with S. maltophilia T3-c, that could efficiently degrade benzene, was investigated. Maximum capacity to eliminate benzene was maintained at $100-110g{\cdot}m^-3{\cdot}h^-1$ when space velocity (SV) ranged from 100 to $300 h^-1$ -1/, however, it decreased sharply to $55 g{\cdot}m^-3{\cdot}h^-1^$ as SV increased to $400 h^-1$. The critical elimination capacities that guaranteed $90\%$ removal of inlet loading of the PU biofilter were determined to be 70,30, and $15 g{\cdot}m^-3{\cdot}h^-1$ at SV 100,200, and $300 h^-1$, respectively. Based on the result of a kinetic analysis of the PU biofilter, maximum benzene elimination velocity ($V_m$) was $125 g{\cdot}m^-3^\;of\;PU{\cdot}h^-1$ and saturation constant ($K_m$) was $0.22 g{\cdot}m^-3^$ of benzene ($65{\mu}{\cdot}I^-1$). This study suggests that the biofilter utilizing S. maltophilia T3-c and polyurethane is a very promising technology for effectively degrading benzene.

1,2-Propanediol Production by Using Saccharomyces cerevisiae M3G3 (Saccharomyces cerevisiae M3G3를 이용한 1,2-Propanediol의 생산 최적화)

  • 1,2-propanediol (1,2-PD) is a commodity chemical that is currently produced from petrochemical derivatives. Saccharomyces cerevisiae is well characterized and a successful industrial microorganism to enable the improvement of the 1,2-propanediol production by metabolic engineering. A recombinant S. cerevisiae M3G3 was used to produce 1,2-propanediol. S. cerevisiae M3G3 is the diploid strain that contains 3 copies of mgs (methylglyoxal synthase) and gldA (glycerol dehydrogenase). S. cerevisiae M3G3 was cultivated at various culture conditions by changing culture temperature, glucose concentration, and inducer concentration. Also the effect of induction time was studied to optimize the production of 1,2-propanediol. Batch and fed-batch cultivation of S. cerevisiae M3G3 was performed by using a 5 L jar fermenter. The highest concentration of 1,2-propanediol in batch cultivation was 0.86 g/L and it was further improved to 1.33 g/L in fed-batch cultivation.

PERIMETER CENTROIDS AND CIRCUMSCRIBED QUADRANGLES

  • For a quadrangle P, we consider the centroid $G_0$ of the vertices of P, the perimeter centroid $G_1$ of the edges of P and the centroid $G_2$ of the interior of P, respectively. If $G_0$ is equal to $G_1$ or $G_2$, then the quadrangle P is a parallelogram. We denote by M the intersection point of two diagonals of P. In this note, first of all, we show that if M is equal to $G_0$ or $G_2$, then the quadrangle P is a parallelogram. Next, we investigate various quadrangles whose perimeter centroid coincides with the intersection point M of diagonals. As a result, for an example, we show that among circumscribed quadrangles rhombi are the only ones whose perimeter centroid coincides with the intersection point M of diagonals.

Degradation Characteristics of Methyl Ethyl Ketone and Methyl Isobuthyl Ketone by Pseudomonas putida KT-3. (Pseudomonas putida KT-3의 Methyl Ethyl Ketone 및 Methyl Isobuthyl Ketone 분해 특성)

  • Methyl ethyl ketone (MEK) and methyl isobutyl ketone (MIBK) have been widely used as solvents in various industries. Biodegradation of MEK and MIBK by Pseudomonas putida KT-3, which could utilize MEK or MIBK as a sole carbon source, was characterized, and the cosubstrate interaction in MEK/MIBK mixture was also studied. Within the range of initial MEK concentration (from 0.5 to 5.5 mM), an increased substrate concentration increased the specific degradation rate of MEK by P putida KT-3 (from 3.15 to 10.58 mmol/g DCW$\cdot$h), but the rate sightly increased at 11.0 mM of initial MEK concentation (11.28 mmol/g DCW$\cdot$h). The similar degradation rates of MIBK (4.69-4.92 mmol/g DCW$\cdot$h) were obtained at more than 3.0 mM of initial MIBK concentation. Kinetic analysis on the degradation of MEK/MIBK mixture by P. putida KT-3 showed that MEK or MIBK acted as a competitive inhibitor. Maximum degradation rate ($V_{max}$), saturation constant ($K_{m}$) and inhibition constant ($K_{1}$) were as follows: $V_{max,MEK}$=12.94 mmol/g DCW$\cdot$h; $K_{m,MEK}$=1.72 mmol/L; $K_{l,MEK}$=1.30 mmol/L; $V_{max,MIBK}$=5.00 mmol/g-DCW$\cdot$h; $K_{m,MIBK}$=0.42 mmol/L; $K_{l,MEK}$=0.77 mmol/L.
