Microbial Conversion of Ginsenoside from the Extract of Korean Red Ginseng (Panax ginseng) by Lactobacillus sp.

  • Thirty-four strains of Lactobacillus species were isolated from soil and eight of these isolates (M1-4 and P1-4) were capable of growing on red ginseng agar. The M1 and P2 strains were determined to be L. plantarum and other strains (M2, M3, M4, P1, P3 and P4) were determined to be L. brevis. Fermentation of red ginseng extract (RGE) with strains M1, M2, P2 and P4 resulted in a low level of total carbohydrate content (174.3, 170.0, 158.8 and 164.8 mg/mL, respectively). RGE fermented by M3 showed a higher level of uronic acid than the control. The polyphenol levels in RGE fermented by M1, P1 and P2 (964.9, 941.7 and $965.3\;{\mu}g/mL$, respectively) were higher than the control ($936.8\;{\mu}g/mL$). Total saponin contents in fermented RGE (except M1) were higher than the control. RGE fermented by M2 and M3 had the highest levels of total ginsenosides (31.7 and 32.7 mg/mL, respectively). The levels of the ginsenoside Rg3 increased from 2.6 mg/mL (control) to 3.0 mg/mL (M2) or 3.1 mg/mL (M3). RGE fermented by M2 and M3 also had the highest levels of Rg5+Rk1 (7.7 and 8.3 mg/mL, respectively). Metabolite contents of ginsenoside (sum of CK, Rh1, Rg5, Rk1, Rg3 and Rg2) of M2 (13.0 mg/mL) and M3 (13.9 mg/mL) were also at a high level among the fermented RGE. Protopanaxadiol and protopanaxatriol content of ginsenoside of M2 (10.9 and 5.4 mg/mL, respectively) and M3 (11.0 and 5.7 mg/mL, respectively) were at higher levels than other fermented RGE.

Simulation of D-limonene Separation from Mandarine Extract in Simulated Moving Bed (SMB) (감귤 추출물로부터 D-리모넨 분리를 위한 유사 이동층 크로마토그래피(SMB) 전산모사)

  • Limonene is orange flavored natural material that is mainly contained in mandarine and lemon peels. D-limonene was extracted from cold-storaged mandarine peel by using Soxhlet extractor at $120^{\circ}C$ for 2 hours with ethanol as solvent. Henry constants of d-limonene and impurity were calculated as $H_{Lim}=8.55$ and $H_{imp}=0.223$ from the result of HPLC analysis. 4-bed SMB of limonene simulation with $0.46{\times}25cm$ columns was conducted by using Aspen chromatography program. Then effective condition for purity was found by changing $m_2$ and $m_3$ values in triangle diagram. The highest purity was 98.59% at $m_2=2.57$, $m_3=9.55$. For this case, feed, desorbent, extract, and raffinate flow rates were 1 mL/min, 1.19 mL/min, 0.857 mL/min and 1.34 mL/min, respectively. Scale-up simulation was also conducted by increasing column diameter from 0.46 cm to 1.6 cm for getting the same efficiency. The increased flow rates were 12 mL/min, 14 mL/min, 10 mL/min, and 16 mL/min for feed, desorbent, extract, and raffinate. It was possible to scale-up with maintaining same limonene purity because linear isotherms of limonene and impurity were assumed.

Impact of Application Rate of Non-ionic Surfactant Mixture on Initial Wetting and Water Movement in Root Media and Growth of Hot Pepper Plug Seedlings (비이온계 계면활성제 혼합물의 처리농도가 상토의 수분 보유 및 고추 플러그묘의 생장에 미치는 영향)

  • In developing soil wetting agent using polyoxyethylene nonylphenyl ether (PNE) and polyoxyethylene castor oil (1:1; v/v), the effect of application rates on changes in concentration of PNE, initial wetting of peatmoss + perlite (7:3) medium, and growth of hot pepper (Capsicum annuum L. 'Knockwang') plug seedlings were investigated. The elevation of application rates of wetting agent increased the amount of water retained by the root media. The treatment of 2.5 $mL{\cdot}L^{-1}$ showed similar water retention to + control ($AquaGro^L$ 3.0 $mL{\cdot}L^{-1}$). Most of the liquid wetting agent (LWA) incorporated during the medium formulation leached out in the first and second irrigation, then it decreased gradually until 10 times in irrigation. In investigation of the influence of LWA on position of water infiltrating into root media, the vertical water movements in treatments of 0.5, 1.0, and 1.5 $mL{\cdot}L^{-1}$ were much faster than those in 0.0 $mL{\cdot}L^{-1}$ (-control), but relative speed of water movement decreased by the elevation in application rate of LWA to 2.0 or 2.5 $mL{\cdot}L^{-1}$. The evaporative water loss of root media that to contained various rate of LWA and irrigated to reach container capacity was the fastest in -control among the treatments and it delayed as the application rate of LWA was elevated. The plant height of 22.2 cm in 0.5 $mL{\cdot}L^{-1}$ and stem diameter of 3.26 mm in 1.0 $mL{\cdot}L^{-1}$ were the highest among the treatments tested. The treatment of 1.0 $mL{\cdot}L^{-1}$ also had the heaviest fresh and dry weights such among treatments tested as 3.08 g and 0.861 g per plant, respectively. The elevated application rate over than 1.5 $mL{\cdot}L^{-1}$ resulted in decreased seedling growth. The results mentioned above indicate that optimum application rate of LWA is 1.0 $mL{\cdot}L^{-1}$.

  • The mutant with high productivity, X. malvacearum SNUF 560-6, was acquired from the X. malvacearum SNUF 560 with low productivity by UV-light irradiation. It was preserved is lyophilized stock culture and it was transferred to PDA slant to maintain viability fortnightly. Fermentations were started by retransfering to MY agar slant from PDA stok culture. The experiments for optimal xanthan gum production were studied in a chemically defined medium. Of the carbon and nitrogen sources tested, 0.4% sucrose medium and 10mM glutamic acid medium yielded the highest xanthan gun production respectively. The addition of 10g/l succinic acid stimulated xanthan gum production. Also 65mM $PO_4\;^{-3}\;(12.6g/l\;KH_2PO_4)$ was effective on xanthan gum production. Finally, medium 1 and medium 2 which have high xanthan gum production potencies were achieved in this stud. The components of medium 1 and medium 2 were as follows: Medium 1 : sucrose 40g/l glutamate 10mM $PO_4\;^{-3}\;54mM\;(KH_2PO_4\;12.65g/l)$ Citrate 2g/l $MgSO_4{\cdot}7H_2O\;0.2g/l$ $H_3BO_3\;0.005/l$ ZnO 0.006/l $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$ Medium 2 : $Sucrose\;40g/l\;(NH_4)_2SO_4\;2g/l$ $PO_4\;^{-3}\;65mM\;(KH_2PO_4\;12.65g/l)$ Succinate 10g/l $MgSO_4{\cdot}7H_2O\;0.02g/l$ $H_3BO_3\;0.06g/l$ ZnO 0.006g/l $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$.

Optimization of Extraction Condition on Fig (Ficus carica L.) by Response Surface Methodology (반응표면분석법에 의한 무화과 열수 추출조건의 최적화)

  • Response surface methodology (RSM) was applied for monitor the yields of desirable substances from fig (Ficus carica L) under different extraction conditions. The maximum yield was 66.46% at 22.08 mL/g of solvent to sample ratio, $90.59^{\circ}C$ extraction temperature and 148.04 min extraction time. The maximum total phenolics was $121.31{\mu}g/mL$ at 17.87 mL/g, $98.82^{\circ}C$, and 130.80 min. The maximum electron donating ability was 54.09% at $121.31{\mu}g/mL$, 18.13 mL/g, and $98.81^{\circ}C$. The maximum value of protease activity was 54.51 unit/min at 17.45 mL/g, $99.01^{\circ}C$, and 131.43 min. In addition, the maximum value of reducing sugar content was 19.14 mg/mL in 22.66 mL/g, $86.30^{\circ}C$, and 153.59 min. The optimum conditions estimated by RSM for maximal extraction of the effective components were $17{\sim}25$ mL/g of solvent to sample ratio, $80{\sim}100^{\circ}C$ of extraction temperature, and $100{\sim}170$ min of extraction time.

Magnetic Properties in Alternating Magnetic Field for the Sintered Ee-l7Cr-2M(M=Si, Nb, Mo) Alloys (Fe-l7Cr-2M(M=Si, Nb, Mo)합금 분말 소결체의 교류 자기 특성)

  • Fe-l7Cr-2M(M=Si, Nb, Mo) alloy powder was prepared by plasma electrode rotating atomizer and than the alloy powder was formed and sintered. The particle shape of the Fe-l7Cr-2M(M=Si, Nb, Mo) alloy power is spherical. The saturation magnetization of the sintered Fe-17Cr-2Mo and Fe-l7Cr-2Nb alloy are 155 emu/g. The saturation magnetization of the sintered Fe-l7Cr-2Si alloy is less than that of the sintered Fe-l7Cr-2Mo and Fe-l7Cr-2Nb alloy. The amplitude relative permeability of the sintered Fe-l7Cr-2M(M=Si, Nb, Mo) alloy has the maximum value in the range of 3∼5 Oe applied field at forming pressure 12 ton/cm$^2$, sintering temperature 1200$^{\circ}C$, and frequency 1 kHz. Power loss of the sintered Fe-l7Cr-2Nb alloy is 40 mW/cc at applied field, H$\sub$a/=5 Oe, and frequency, f=1 kHz. The power loss of the sintered Fe-l7Cr-2Nb alloy is a half of that of the sintered Fe-l7Cr-2Si and Fe-l7Cr-2Mo alloy.

Enhancement of Skin Whitening and Anti-wrinkle Activities of the Co-culture of Lactobacillus rhamnosus and Lactobacillus paracasei (Lactobacillus rhamnosus와 Lactobacillus paracasei 복합 배양의 피부 미백 및 주름 개선 활성 증진)

  • In this study, cosmeceutical activities of the co-cultures of both lactic acid bacteria isolated from the hands of women, Lactobacillus rhamnosus (L. rhamnosus) and Lactobacillus paracasei (L. paracasei) were first reported: For whitening and anti-wrinkle activities, the co-culture showed the highest tyrosinase inhibition of 20.68%, compared L. paracasei and L. rhamnosus. The co-culture also showed the highest inhibition of melanin synthesis as 63.7%. In observing the anti-wrinkle activities of the co-culture, it generated only 3726.3 pg/mL of matrixmetalloproteinase-1 (MMP-1) production when 13613.5 pg/mL and 13012.0 pg/mL of MMP-1 production were estimated from L. rhamnosus and L. paracasei. Besides these, the extract from the co-culture yielded higher collagen production as 380.7 ng/mL, compared to 323.4 ng/mL and 304.1 ng/mL from L. paracasei and L. rhamnosus. These results indicate that the co-culture of both lactic acid could improve its cosmetic activities. This hypothesis was also confirmed that the co-culture of both bacteria showed strong antioxidant activities of DPPH free radical scavenging while the extract of L. rhamnosus and L. paracasei.

Synergistic Effect of Physico-chemical Treatment and Bacteriocin Produced by Enterococcus faecium MJ-14 (Enterococcus faecium MJ-14가 생산하는 박테리오신과 물리화학적 처리의 상승효과)

  • When L. monocytogenes ($10^{5}CFU/mL$) at exponential phase cells were heated for 5 min at $65^{\circ}C$ in the presence of the bacteriocin (30 BU/mL) produced by E. faecium MJ-14, the number of viable cells was markedly reduced at p < 0.05. The bactericidal effect of bacteriocin showed synergism with combination ot organic acids (citric acid or acetic acid) or chemical preservatives (sodium benzoate, sodium lactate, sodium nitrate or potassium nitrate). For example, the number of viable cells was reduced by 4.8 log units under combination of the bacteriocin (30 BU/mL) and sodium nitrate ($100{\mu}g/mL$), while it was reduced by 1.1 log unit only under single treatment of the bacteriocin after 12 k at $37^{\circ}C$. The addition of the bacteriocin (300 BU/mL) into skim milk inoculated with L. monocytogenes ($10^{5}$CFU/mL) reduced the cells by 1.5 log unit, in case of the cell suspension stored at $4^{\circ}C$ for 24 hr. Moreover, L. monocytogenes was reduced by 2 log unit when stored at $-20^{\circ}C$ for 7 days in gound pork added with 300 BU/mL of 린e bacteriocin.

Generation of Carbon Monoxide from Saccharides (당류 중에서 일산화탄소의 발생에 관한 연구)

  • Carbon monoxide was generated when the saccharides which contained aldehyde group were added to alkali solution (NaOH solution) at $25^{\circ}C$ or $40^{\circ}C$. When the milk was added to 1.0 M NaOH solution at $25^{\circ}C$ and to 0.01 M NaOH solution at $40^{\circ}C$, carbon monoxide was generated. The carbon monoxide gas which was generated by 20 mL of 0.1 M lactose or 0.1 M xylose and 2 mL of 1.0 M NaOH solution at $50^{\circ}C$ for 2days was bubbled through 2 mL of fresh blood. The carboxyhemoglobin concentrations of both conditions were 31%. However, when the carbon monoxide gas which was generated by 30 mL of milk and 2 mL of 1.0 M NaOH at $50^{\circ}C$ for 2days or 3days was bubbled through 2 mL of fresh blood, the carboxyhemoglobin concentration was 32% or 42% respectively.

Effect of continuous pulsed electric fields treatments on quality of apple juice (사과주스의 품질에 미치는 pulsed electric field 연속 처리효과)

  • Apple juices were sterilized by continuous pulsed electric field (PEF) treatments of pulse width of 25 ${\mu}s$ at electric field intensity of 20.0 kV/cm, and with the varied pulse frequencies of 35 Hz (40 kJ/L), 55 Hz (70 kJ/L), 72 Hz (100 kJ/L) and 85 Hz (130 kJ/L). The PEF treatments of apple juice reduced the microbial counts from 5.3 log CFU/mL of initial state to 3.0 log CFU/mL after PEF treatment at energy density of 130 kJ/L. Also yeast and fungi after PEF treatments were reduced from 5.3 log CFU/mL to 3.0 log CFU/mL and Escherichia coli were from 5.3 log CFU/mL of initial state to 4.7 log CFU/mL to < $10^1$ CFU/mL. The soluble solids and free sugars did not significantly differ (p<0.05) depending on conditions of PEF treatment. The total phenolic contents and antioxidant activity such as the DPPH and ferric reducing antioxidant power (FRAP) by PEF treatments were significantly partly reduced, but the PEF-reduced value came in smaller quantities than the heat treatment at $65^{\circ}C$. The iterative PEF treatments with pulse width of 25 ${\mu}s$ and pulse frequency of 85 Hz at electric field intensity of 20.0 kV/cm showed limited in microbial reduction. Also, total phenolic contents and antioxidant activity such as DPPH and FRAP, significantly decreased depending on treatment numbers of PEF (p<0.05).
