Screening of Antiviral Medicinal Plants against Avian Influenza Virus H1N1 for Food Safety

  • Various extracts from 30 medicinal plants were evaluated for their antiviral activity against influenza virus A/Puerto Rico/8/34 (H1N1) and cytotoxicity in MDCK cell culture. The plant material (30 g) was extracted with methanol (300 mL) at room temperature for 24 h, after which the methanolic extracts were filtered, evaporated, and subsequently lyophilized. Evaluation of the potential antiviral activity was conducted by a viral replication inhibition test. Among these medicinal plants, Tussilago farfara, Brassica juncea, Prunus armeniaca, Astragalus membranaceus, Patrinia villosa, and Citrus unshiu showed marked antiviral activity against influenza virus A/H1N1 at concentrations ranging from 0.15625 mg/mL to 1.25 mg/mL, 0.3125 mg/mL to 10 mg/mL, 5 mg/mL to 10 mg/mL, 0.625 mg/mL to 10 mg/mL, 0.625 mg/mL to 10 mg/mL, and 0.3125 mg/mL to 5 mg/mL, respectively. The extracts of Tussilago farfara showed cytotoxicity at concentrations greater than 2.5 mg/mL, whereas the other five main extracts showed no cytotoxicity at concentrations of 10 mg/mL. Taken together, the present results indicated that methanolic extracts of the six main plants might be useful for the treatment of influenza virus H1N1.

Antioxidative Activity of Rumex crispus L. Extract (소리쟁이 추출물의 항산화 효능)

  • The objective of this study was to investigate the antioxidative capacity of ethanol extracts from Rumex crispus L. The concentration of R. crispus L. extract at which DPPH radical scavenging activity was inhibited by 50% was 2.15 mg/mL, which was lower than that of ${\alpha}$-tocopherol (0.43 mg/mL), as compared to 100% by pyrogallol as a reference. Total antioxidant status was examined by total antioxidant capacity against ABTS radical reactions. Total antioxidant capacities of R. crispus L. extract at concentrations of 0.1 and 1 mg/mL were 0.47 and 2.33 mM Trolox equivalents, respectively, which were higher than those of ${\alpha}$-tocopherol. Superoxide scavenging activities of R. crispus L. extract at concentrations of 0.1 and 1 mg/mL were 21.5 and 78.9%, respectively, which were not significantly (p>0.05) different from those of catechin. Oxygen radical absorbance capacities of R. crispus L. extract at concentrations of 20 and 100 ${\mu}g/mL$ were 62.5 and 156.4 ${\mu}M$ Trolox equivalents, respectively, which were lower than those of ascorbic acid. Cupric reducing antioxidant capacities of R. crispus L. extract at concentrations of 0.1 and 1 mg/mL were 0.28 and 1.88 mM Trolox equivalents, which were similar or significantly (p<0.05) higher than those of ${\alpha}$-tocopherol, respectively. R. crispus L. extract prevented supercoiled DNA strand breakage induced by hydroxyl radical and peroxyl radical. Total phenolic contents of R. crispus L. extract at concentrations of 0.5 and 5 mg/mL were 0.58 and 3.85 mM gallic acid equivalents, respectively. R. crispus L. extract at concentration of 0.1 and 0.5 mg/mL inhibited 0.2 mM tert-butyl hydroperoxide-induced cytotoxicity by 38.5 and 63.5%, respectively, in HepG2 cell culture system. Thus, strong antioxidant and cytotoxicity-inhibiting effects of R. crispus L. extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in total phenolic contents.

Simultaneous Quantitative Analysis of Mixtures of Ixeridium dentatum (Thunb.) Tzvelev, Plantago asiatica L. and Rumex crispus L. Ethanol Extracts (씀바귀, 질경이, 소리쟁이 에탄올 추출 혼합물에 대한 주요 성분 동시 분석)

  • Recently, consume for functional cosmetics containing natural products has been greatly increased. In order to develop it as a natural cosmetic material, we selected Ixeridium dentatum (Thunb.) Tzvelev, Plantago asiatica L. and Rumex crispus L. that have antioxidant and anti-inflammatory effects. In this study, simultaneous quantitative analysis of the isolated compounds and natural product complexes (Mix.) were validated using high performance liquid chromatography (HPLC). The isolated six compounds were shown in a large linearity with a correlation coefficient (R2) of 0.99. The limit of detection (LOD) of chlorogenic acid, plantamajoside, acteoside, emodin chrysophanol and physcion were 0.36 ㎍/mL, 0.36 ㎍/mL, 0.37 ㎍/mL, 0.30 ㎍/mL, 0.22 ㎍/mL and 0.12 ㎍/mL, respectively. The limit of quantification (LOQ) of chlorogenic acid, plantamajoside, acteoside, emodin chrysophanol and physcion were 1.10 ㎍/mL, 1.08 ㎍/mL, 1.12 ㎍/mL, 0.99 ㎍/mL, 0.66 ㎍/mL and 0.35 ㎍/mL, respectively. Content analysis showed chlorogenic acid (0.19 ± 0.02%), plantamajoside (0.48 ± 0.01%), acteoside (0.65 ± 0.01%), emodin (1.15 ± 0.11%), chrysophanol (0.73 ± 0.01%) and physcion (0.69 ± 0.09%). Therefore, the results of this study may provide for basic data of standardization research natural cosmetic material development on the I. dentatum, P. asiatica and R. cripus.

  • This study was to investigate the changes of shape of the lower limb surface, the rate of the measurement of expansion and contraction and correlation coefficient between variables caused by hip joint and knee joint movements. The results of the investigation are as follows; 1. According to the development figure of shell when the leg was raised $45^{\circ}$forward($M_{2}$), total length of F.L shortened while B.L lengthened. This result is contarary to $M_{3}$raising the leg $15^{\circ}$ backward. In both $M_{2}$, $M_{3}$movements, the rate of expansion and contraction to the course direction was insignificant. When hip joint was bent $15^{\circ}$ with knee joint $120^{\circ}$bent ($M_{4}$) and hip joint was bent $30^{\circ}$ with knee joint $90^{\circ}$ bent($M_{5}$), upper section of back hip expanded while the front hip section contracted slightly. In the Movement of sitting on the chair($M_{6}$), abdomen, front hip section and upper thight section contracted to the wale direction remarkably while the back hip section expanded conspicuously. 2. According to the rate of expansion and contraction of skin (surface) by the somatometry. In $M_{2}$, C.F.L. upper and middle thight girth contracted and B.L, C.L, L.L expanded. This fact is contarary to M3. In M4, M5, C.F.L showed remarkable contraction and C.B.L expanded remarkably. In $M_{6}$, C.B.L contracted most of all the items measured and knee girth, F.L, L.L, C.B.L, hip girth expanded conspicuously. 3. According to the correlation coefficient between variables. In various movements, the correlation among girth items commonly showed a high or middle grade, the correlation among length items also commonly showed a low grade and that girth and length items showed a very low grade commonly. Waist girth, hip grith, F.L, B.L, L.L items showed that there were significant correlation.

Antioxidative Effects of Ethanol Extracts from Rhus Verniciflua Stokes (RVS) on Mouse Whole Brain Cells (옻나무 에탄올 추출물의 쥐 뇌세포에 대한 항산화효과)

  • To measure antioxidative activities, the various extracts from RVS (Rhus Verniciflua Stokes) were tried out with either DPPH or thiocyanate method. Also we used the GO (Glucose Oxidase) 20 mU/mL hydroxyl radical system in mouse whole brain cell culture. Chloroform, n-hexane or ethanol were used as extract solutions which had different polarity respectively. In DPPH and thiocyanate method, the antioxidative activities of the crude ethanol extracts were stronger than other extracts. The crude ethanol extracts were fractionated 5 peaks by glass column. Among of them, antioxidative activity of peak II $(P_{II})$ was shown stronger than other fractions, a little for peak III $(P_{III})$ and peak IV $(P_{IV})$, and none for peak I $(P_I)$ and Peak V $(P_V)$. In the antioxidative effects of crude ethanol extracts (30 mg/mL), cell viabilities were evaluated $1\;{\mu}L\;(297\;{\mu}g/mL)$, $2\;{\mu}L\;(588\;{\mu}g/mL)$ of crude ethanol extracts 59%, 68% respectively. $10\;{\mu}L\;(2,727\;{\mu}g/mL)$ addition of crude ethanol extracts had 95% cell viabilities, 0.01% significant, comparing control. In addition, the compounds related to antioxidative effect of crude ethanol extract might be glycoproteins by means of SDS-PAGE. Comparison to antioxidative effects between several antioxidants (ascorbic acid, ${\alpha}-tocopherol$, catalase) $273\;{\mu}L/mL$ addition of crude ethanol extracts corresponds to $1\;{\mu}g/mL$ catalase in antioxidative effects.

  • The purpose of this study is to know the average of hip joint range of motion and difference according to the aging for the elderly. This study consisted of elder male(n=75) and elder female(n=109). The result of assessment and analysis in hip pint range of motion are as follows : 1) The average hip flexion(knee flexed) joint range of motion in 60-69(from sixty to sixty-nine)years old are $104.26^{\circ}$(Left-Male), $101.00^{\circ}$(Right-Male), $107.05^{\circ}$(Left-Female), $107.05^{\circ}$(Right-Female). 70-79years old are $104.59^{\circ}$(L-M), $102.05^{\circ}$(R-M), $105.73^{\circ}$(L-F), $108.75^{\circ}$(R-F). 80-89years old are $101.53^{\circ}$(L-M), $101.13^{\circ}$(R-M), $96.83^{\circ}$(L-F), $97.67^{\circ}$(R-F). There was significant difference in hip flexion(knee flexed) among female group(p<.01). The average hip flexion(knee extended) joint range of motion in 60-69(from sixty to sixty-nine)years old are $73.13^{\circ}$(Left-Male), $72.04^{\circ}$(Right-Male), $77.29^{\circ}$(Left-Female), $75.97^{\circ}$(Right-Female). 70-79years old are $74.95^{\circ}$(L-M), $72.19^{\circ}$(R-M), $76.73^{\circ}$(L-F), $76.65^{\circ}$(R-F). 80-89years old are $70.83^{\circ}$(L-M), $70.37^{\circ}$(R-M), $69.00^{\circ}$(L-F), $69.00^{\circ}$(R-F). There was significant difference in left hip flexion(knee extended) among female group(p<.05). 2) The average hip extension joint range of motion in 60-69years old are $13.09^{\circ}$(L-M), $12.78^{\circ}$(R-M), $10.97^{\circ}$(L-F), $10.68^{\circ}$(R-F). 70-79years old are $8.95^{\circ}$(L-M), $8.48^{\circ}$(R-M), $11.24^{\circ}$(L-F), $10.90^{\circ}$(R-F). 80-89 years old are $8.40^{\circ}$(L-M), $8.23^{\circ}$(R-M), $7.33^{\circ}$(L-F), $7.33^{\circ}$(R-F). There was significant difference in left(p<.01) and right(p<.05) hip extension among male group(p<.05). 3) The average hip abduction joint range of motion in 60-69 years old are $33.04^{\circ}$(L-M), $33.17^{\circ}$(R-M), $33.16^{\circ}$(L-F), $33.37^{\circ}$(R-F). 70-79 years old are $31.00^{\circ}$(L-M), $30.05^{\circ}$(R-M), $32.44^{\circ}$(L-F), $32.68^{\circ}$(R-F). 80-89 years old are $29.07^{\circ}$(L-M), $27.90^{\circ}$(R-M), $28.17^{\circ}$(L-F), $28.67^{\circ}$(R-F). There was no significant difference among group. 4) The average hip adduction pint range, of motion in 60-69years old are $29.57^{\circ}$(L-M), $29.35^{\circ}$(R-M), $31.87^{\circ}$(L-F), $31.89^{\circ}$(R-F). 70-79, years old are $27.41^{\circ}$(L-M), 27.00(R-M) $30.85^{\circ}$(L-F), $31.28^{\circ}$(R-F). 80-89 years old are $26.87^{\circ}$(L-M), $26.63^{\circ}$(R-M), $24.67^{\circ}$(L-F), $24.83^{\circ}$(R-F). There was significant difference in hip abduction among female group(p<01). 5) The average hip external rotation pint range of motion in 60-69years old are $32.26^{\circ}$(L-M), $31.17^{\circ}$(R-M), $33.53^{\circ}$(L-F), $34.42^{\circ}$(R-F). 70-79 years old are $31.64^{\circ}$(L-M), $28.62^{\circ}$(R-M) $31.29^{\circ}$(L-F), $31.45^{\circ}$(R-F). 80-89 years old are $26.40^{\circ}$(L-M), $26.07^{\circ}$(R-M), $24.77^{\circ}$(L-F), $24.27^{\circ}$(R-F). There was significant difference in left(male, female p<.01) and right(female p<.0l) hip external rotation among group. 6) The average hip internal rotation joint range of motion in 60-69years old are $30.30^{\circ}$(L-M), $28.13^{\circ}$(R-M), $34.27^{\circ}$(L-F), $36.03^{\circ}$(R-F). 70-79years old are $31.24^{\circ}$(L-M), $29.57^{\circ}$(R-M), $28.51^{\circ}$(L-F), $29.10^{\circ}$(R-F). 80-89 years old are $24.63^{\circ}$(L-M), $24.40^{\circ}$(R-M), $24.27^{\circ}$(L-F), $24.27^{\circ}$(R-F). There was significant difference in left(male p<.05, female p<.01) and right(female p<.01) hip internal rotation among group.

Sleep-Inductive Effect of GABA on the Fermentation of Mono Sodium Glutamate (MSG) (Mono sodium glutamate (MSG) 발효 GABA의 수면유도 효과)

  • Relatively large amounts of GABA can be produced by the fermentation of rice bran. Therefore, this study was conducted to investigate the effects of GABA on the secretion of melatonin and serotonin for the development of a sleep inductive compound. The secretion levels of melatonin and serotonin from mice were found to be $3.425{\pm}0.182\;pg/mL$ and $5.37{\pm}0.963\;ng/mL$, respectively, in response to feeding 120 mg/mL of GABA while they were $2.607{\pm}0.41\;pg/mL$ in the control. The secretion of both melatonin and serotonin was increased up to the 13.51% and 34.99%, respectively, when compared to the negative control. However, the feeding of milk alone did not have a great effect on the melatonin and serotonin secretions. Conversely, feeding of milk with GABA enhanced the secretion of serotonin. The amounts of both melatonin and serotonin secreted increased with respect to the increase in GABA concentrations during feeding. Interestingly, the induction level of melatonin was relatively higher than that of serotonin in response to feeding 120 mg/mL of GABA. This is the first study to report that GABA has an ability to induce sleep related hormones in mice; therefore, it has the potential for use as a natural sleep aid.

Effect of Watering on Growth and Essential Oil Content of Sweet Basil (Ocimum basilicum) (관수량이 sweet basil(Ocimum basilicum)의 생장과 정유함량에 미치는 영향)

  • This experiment was carried out to investigate watering with relation to growth, quality and yield of essential oil in sweet basil. The degree of water stress was taken as amount of watering. D1 was watered with 25mL for 2 weeks and 30mL from 4 to 5 weeks after planting in Wagner pot (1/20000a, ${\phi}24cm$). As this, 75mL and 90mL, 225mL and 270mL, and 675mL and 810mL were watered in D2, D3, and D4 treatment respectively. DFT was set up as water stress was not inflicted. The growth of basil in D3 and D4 was better than that of others, in which root activity was as much twice as that of D1. Essential oil of D1 was recorded the lowest content as 0.33%. The result of proline content, peroxidase activity, photosynthesis, stomatal conductance and stomatal resistance were proved D1 to be stressed. This treatment consequently increased the content of essential oil. In consideration of growth and essential oil content, D3 treatment was highest as 47.37mg in oil production per plant. Finally, D3 watered with 225mL for 2 weeks and 275mL from 4 to 5 weeks after planting could be selected on the purpose of both plant growth and essential oil production. Essential oil content of sweet basil was increased in response to water stress. For increase of essential oil yield, oil synthesis could be raised by giving water stress just before harvesting.

Clearance and ingestion rate according to different culture condition in the sulf clam, Tresus keenae (사육조건에 따른 왕우럭, Tresus keenae의 여수율과 먹이섭취률)

  • In juvenile size, the maximum clearance rate and ingestion rate were 15mm, which is the 70.8 mL/h, 4245.0×104cells/h, respectively. But the lowest clearance rate and ingestion rate were found at 5mm, which is the 4.1 mL/h, 246.0×104cells/h, respectively. The highest clearance rate and ingestion rate of AFDW seen at 7.5mm, with 3.3 mL/mg AFDW/h, 196.0×104cells/mg AFDW/h, respectively. According to the results of water temperature, clearance rate and ingestion rate were lowest at 10℃, showing the 0.3 mL/h, 20.3×104cells/h, respectively. The highest clearance rate and ingestion rate were 25℃, which is the 16.6 mL/h, 993.4×104cells/h, respectively. But in 30℃, clearance rate and ingestion rate decreased to the 12.9 mL/h, 772.6×104cells/h, respectively. With respect to the food density, the experiment groups with 30×104 cells/mL and 240×104 cells/mL showed the clearance rate of 21.1 mL/h and 2.3 mL/h, respectively. But the experimental group with 120×104 cells/mL or more showed decreased clearance rate. While, the maximum ingestion rate was at 60×104 cells/m, showing the 876.2×104cells/h.

Isolation, Identification, and Probiotic Properties of Lactobacillus reuteri HY701 from Human Feces

  • Strain HY701 was isolated from human feces for probiotic use by selecting highly resistant isolates to artificial gastric acid and bile acid. Strain HY701 was identified as Lactobacillus reuteri using 16S rDNA sequencing, and tentatively named L. reuteri HY701. The resistance of L. reuteri HY701 to artificial gastric acid (PH 2.5) was high with a survival rate of over 90%. L. reuteri HY701 also showed high tolerance to artificial bile acid after incubation in artificial gastric acid. Using the API ZYM test kit, the carcinogenic enzymes (${\beta}$-glucuronidase and (${\beta}$-glucosidase were not detected with L. reuteri HY70l, while the beneficial enzyme (${\beta}$-galactosidase was weakly detected. L. reuteri HY701 was sensitive to $100\;{\mu}g/mL$ nisin, $20\;{\mu}g/mL$ roxithromycin, $15\;{\mu}g/mL$ erythromycin, but resistant to $20\;{\mu}g/mL$ streptomycin, $10\;{\mu}g/mL$ tetracycline, $20\;{\mu}g/mL$ ciprofloxacin, $20\;{\mu}g/mL$ nystatin, $20\;{\mu}g/mL$ gentamycin, $10\;{\mu}g/mL$ doxycycline, $10\;{\mu}g/mL$ chloramphenicol, and $20\;{\mu}g/mL$ ampicillin. L. reuteri HY701 was shown to possess bactericidal activity as it inhibited the growth of Listeria monocytogenes ATCC 19111 and Escherichia coli JM109 completely within 24 hr of incubation. These results indicate that L. reuteri HY701 could be used as a probiotic strain.
