우유와 쏘세지 등 몇가지 동물성 식품의 보존에 미치는 오골계 난백 lysozyme의 효과를 검토하기 위하여 이들을 0.05% lysozyme 으로 처리한 후 $20^{\circ}C$, $30^{\circ}C$ 및 $37^{\circ}C$에 저장하면서 경시적으로 이들의 pH와 휘발성 염기질소 함량 및 생균수의 변화를 조사 하였다. 생우유를 lysozyme 으로 처리하여 $20^{\circ}C$에서 48시간 저장 하였을 때 휘발성 염기질소 함량이 63mg%로 제일 낮아 보존효과가 있었고 $30^{\circ}C$에서는 lysozyme 과 glycine의 혼용처리시 그 보존효과가 상승하였다. 또한 시판우유와 어묵에서도 lysozyme처리에 의한 보존효과가 있었으며 특히 솔빈산을 첨가하지 않은 쏘세지에 lysozyme 을 처리하여 $5^{\circ}C$에 저장 하였을 때 휘발성 염기질소 함량이 약 150 mg%로 비교적 낮았다.
Bacillus sp. TA-11에 대한 오골계 난백 lysozyme과 일반 난백 lysozyme의 용균성을 비교분석 하였다. 오골계 난백 lysozyme의 용균활성은 Bacillus sp. TA-11를 5$0^{\circ}C$에서 18시간 정치배양한 대수기 후기의 세포에 대하여 가장 높았고, lysozyme의 농도는 0.25%가 최적이었다. 또한 lysozyme의 최적반응 pH와 온도는 각각 4.5와 35$^{\circ}C$였다. 일반 난백 lysozyme의 용균활성은 시험균주를 24시간 배양한 정지기의 세포에 대하여 가장 높았고 lysozyme의 최적 농도는 0.5%였으며 반응 최적 pH와 온도는 각각 5.5와 4$0^{\circ}C$이었다.
We have isolated and characterized a new insect chicken type (c-type) lysozyme gene from the common cutworm, Spodoptera litura. The full-length cDNA of Spodoptera lysozyme is cloned by rapid amplification of cDNA ends PCR (RACE-PCR). The isolated cDNA consists of 1039 bp including the coding region for a 142-amino acid residue polypeptide, which included a signal peptide of 21-amino acid residue and a mature protein of 121-amino acid residue. The predicted molecular weight of mature lysozyme and its theoretical isoelectric point from amino acid composition is 13964.8 Da and 9.05, respectively. The deduced amino acid sequence of Spodoptera lysozyme gene shows the highest similarity (96.7%) to Spodoptera exigua lysozyme among other lepidopteran species. Amino acid sequence comparison with other the c-type lysozymes, Spodoptera lysozyme has the completely conserved $Glu^{32}$ and $Asp^{50}$ of the active site and eight Cys residues are completely conserved in the same position as that of other lepidopteran lysozymes.
난백으로부터 lysozyme을 효과적으로 추출하기 위하여 난백의 전처리와 이온교환수지의 선택 및 전처리 등 추출조건에 관한 연구를 실시한 결과는 다음과 같다. 난백의 점도저하를 위하여 2,000 rpm의 속도로 30분간 균질하고 45분간 초음파처리를 실시한 결과 난백의 겉보기 점도가 3 cp까지 떨어졌으나 pH의 변화에 따라서는 점도의 차이가 크지 않았다. lysozyme의 추출을 위한 이온교환수지의 선택을 위하여 Amberlite IRC-84 등 6종류의 수지에 대한 Lysozyme 흡착력을 검토한 결과 CM-Sepdex C-25>Duolte C-464>Amberlite C-50>Dowex MSC-1>Amberlite IRC-50>Amberlite IRC-84의 순으로 lysozyme 흡착력이 높았으나 난백에의 적용시 CM-Sephadex C-25의 경우에는 난백의 유출이 용이하지 않은 것으로 나타나 Duolite C-464가 가장 적합한 것으로 나타났다. 양이온교환수지인 Duolite C-464 의 양이온형태 결정을 위하여 $Na^{+}$, $NH_4$$^{+}$. $K^{+}$ form으로 만들어 lysozyme 흡착력을 검토한 결과 $Na^{+}$ > $NH_4$$^{+}$ > $K^{+}$의 순으로 나타나 $Na^{+}$ form으로 하는 것이 가장 효과적인 것으로 나타났다. Duolite C-464의 $Na^{+}$form을 이용하여 lysozyme을 추출할 경우 rinse buffer는 pH6.5의 0.1M sodium phosphate buffer, eluting solution은 10% ammoniun sulfate가 효율적이었다. Lysozyme을 추출한 전, 후 난백의 가공적성을 측정한 결과 기포력은 추출전의 난백액에 대하여 85.3%로서 다소 떨어졌으나 가열한 난백겔의 경우에는 추출전, 후 색택의 변화가 크지 않았으며 조직감의 경우에도 hardness가 다소 떨어졌으나 elasticity는 오히려 13%가량 증가되었다.
Lysozyme is a protein found in egg white, tears, saliva, and other secretions. As a marketable natural alternative to preservatives, lysozyme can act as a natural antibiotic. In this study, we have isolated Bacillus licheniformis TIB320 from soil, which contains a lysozyme gene with various features. We have cloned and expressed the lysozyme in E. coli. The antimicrobial activity of the lysozyme showed that it had a broad antimicrobial spectrum against several standard strains. The lysozyme could maintain efficient activities in a pH range between 3 and 9 and from $20^{\circ}C$ to $60^{\circ}C$, respectively. The lysozyme was resistant to pepsin and trypsin to some extent at $40^{\circ}C$. Production of the lysozyme was optimized by using various expression strategies in B. subtilis WB800. The lysozyme from B. licheniformis TIB320 will be promising as a food or feed additive.
양이온 교환수지를 이용하며 추출한 난백 lysozyme의 분리방법에 따른 역가측정과 역가 측정방법에 따른 차이를 비교 검토하고 pH와 열에 대한 안정성을 구명하였다. Lysozyme의 역가를 측정하는데 있어서 시료 무게당 lysozyme량으로 정의할 경우는 순도 높은 표준품의 구입이 필요하나 반복에 따른 측정 오차의 범위가 적었다. 반면 시료 무게당 units으로 할 때 오차의 범위는 조금 넓었지만 간편 신속하게 측정할 수 있었다. 난백 lysogyme을 pH6.3인 0.066M 인산염 완충액에 넣어 $37^{\circ}C$에서 2시간동안 항온시켰을 때 Micrococus lysodeiktius의 용균작용이 가장 활발히 일어났다. CM Sephadex C-25로 O.D. 1.0이상의 용액에서 회수한 lysozyme의 역가는 36,000units/mg으로 처리구 중 가장 높았다. 추출 lysozyme은 가열함에 따라 역가가 감소하였으며 인산염 완충액에서 $100^{\circ}C$, 15분간 가열하였을 때 35%이상이 감소하였다. 추출한 lysozyme용액을 pH별로 $100^{\circ}C$에서 15분간 가열했을 때 산성영역에서는 매우 안정되었으나 pH6.0 이상의 알칼리 영역에서 급격히 불활성화 되었다.
Immobilization of lysozyme on chitosan non-woven using glutaraldehyde(GA) was investigated. For this, 100 % chitosan non-woven was prepared as novel support for the enzyme immobilization. In addition, free lysozyme activity was examined depending on various pH and temperature by measuring time. Moreover, the optimum immobilization conditions depending on various pH, temperature, immobilization time and lysozyme concentration was evaluated. In addition, thermal stability and storage stability of immobilized lysozyme were measured. The characteristics of immobilized lysozyme was examined by FT-IR, surface morphology, and MTT assay. The results are follows: the optimal immobilization of lysozyme were pH 7.0, $25^{\circ}C$, lysozyme concentration 1.5 mg/ml, immobilization time 240 min. The immobilized lysozyme showed higher thermal stability than the free trypsin. The immobilized lysozyme activity was retained 80 % of its initial activity at $4^{\circ}C$ over 30 days of storage. The lysozyme was immobilized effectively on chitosan non-woven by observation of surface morphology.

We have isolated and characterized Agrius convolvuli cDNA encoding a c-type lysozyme. The cDNA sequence encodes a processed protein of 139 amino acid residues with 19 amino acid residues amino-terminal signal sequence and 120 amino acid residues mature sequence. The amino acid residues responsible for the catalytic activity and the binding of the substrate are conserved. Agrius lysozyme has a high identity to Manduca sexta. Recombinant A. convolvuli lysozyme was expressed in Escherichia coli BL21(DE3) pLysS cells for pGEX 4T-1 expression vector. Their optimal conditions for the fusion protein expression and purification were screened. Lysozyme gene amplified with primers ACLyz BamHI and ACLyz XhoI was ligated into the pGEX 4T-1 vector, which contained the glutathione S-transferase(GST) gene for fusion partner. The fusion protein was induced by IPTG and identified by SDS-PAGE analysis. Molecular weight of the fusion protein was estimated to be about 45 kDa. Recombinant lysozyme, fused to GST, was purified by glutathion-Sepharose 4B affinity chromatography. Western blot analysis of this protein revealed an immunoreactivity with the anti-Agrius lysozyme.
To understand molecular mechanisms of mouse mammary gland involution, clones were isolated by differential screening of a cDNA library. Partial sequences of a clone showed 100% identity to cDNA sequences of mouse lysozyme P gene. Northern analysis was performed to examine expression levels of lysozyme mRNA in mammary gland at several physiological states. Expression of lysozyme gene was induced at involution day 5 compared with lactating stage. High levels of lysozyme mRNA were also detected at virgin tissues. Two types of separate genes, P and M lysozyme, have been known in mouse, and we found that both lysozyme P and M genes were expressed in mammary tissues by reverse transcriptase-polymerase chain reaction. The lysozyme enzyme activity determined by lysoplate assay was also higher in involuted mammary tissues compared with lactating tissues, showing a similar trend to its mRNA levels. Lysozyme is an antimicrobial protein and involved in host defense mechanism. The increase in lysozyme gene expression may help to prevent microbial infection during mammary gland involution at which stage the residual milk in the mammary gland provides good nutritional sources for microbial growth.
