Numerical analysis was performed on the enzyme transport and the flow fields in order to predict the effectiveness of urokinase injection regimens in clot dissolution. The species and momentum transport equations were numerically solved for the case of uniform perfusion of enzyme into a fibrin clot for an arterial thrombus and a deep vein thrombus models. In order to predict the thrombus lysis efficiency of continuous and forced intermittent injections, enzyme perfusion and clot lysis were simulated for the different injection velocities. Intermittent injection showed faster clot lysis compared to continuous perfusion, and lysis efficiency was increased as injection velocity increased.
감마선을 조사(照射)하거나 조사하지 않은 두 경우의 Bacillus Subtilis를 대상으로 하여 박테리아 세포 붕괴(lysis)를 유도한 후 방출된 유기 분자가 pH 7 조건에서 석영 용해 속도에 미치는 영향을 조사하였다. 시간이 경과하며 석영과 박테리아 혼합 슬러리에서 용존 유기탄소(dissolved organic carbon; DOC) 함량이 증가하였으며 이는 박테리아 투입량과 대체적으로 비례하는 것으로 보아 박테리아 세포 붕괴의 결과인 것으로 판단되었다. 방사선을 조사하지 않은 박테리아를 투입하였을 경우, 시간이 경과하며 박테리아를 투입하지 않은 화학적 비교 슬러리에 비해 높은 함량의 규소가 용출되어 나왔다. DOC 함량과 용해되어 나온 규소 함량간에 나타난 좋은 상관관계는 규소 용출의 원인이 박테리아 세포 붕괴에 의해 방출된 DOC에서 비롯되었음을 나타낸다. 한편 방사선을 조사한 박테리아의 세포 붕괴 산물은 방사선을 조사하지 않은 경우에 비하여 단위 DOC 함량당 매우 높은 농도의 규소를 용출시켰다. 이 때 관찰되는 규소 용출은 방사선이 조사되었을 때 교란된 박테리아 내부의 유기 분자가 방사선을 조사하지 않은 박테리아에 비하여 석영을 보다 효과적으로 용해할 수 있는 유기 분자로 변화하였기 때문으로 판단된다. 이 결과는 고준위 방사성 폐기물 처 분장에서 누출된 핵종이 지표 생태계에 도달하는데 소요되는 시간을 예측할 때 처분장 주변 대수층 암석의 풍화 속도 촉진에 미치는 박테리아 세포 붕괴의 영향을 고려해야 함을 나타낸다.
Characteristics of sophorolipid and rhamnolipid were evaluated as antifungal agents against plant pathogenic fungi. Eight percent of mycelial growth of plant pathogen (Phytophthora sp. and Pythium sp.) was inhibited by 200 mg/l of rhamnolipid or 500 mg/l of sophorolipid, and zoospore motility of Phytophthora sp. decreased by $90\%$ at 50 mg/l of rhamnolipid and $80\%$ at 100 mg/l of sophorolipid. The effective concentrations for zoospore lysis were two times higher than those of zoospore motility inhibition. The highest zoospore lysis was observed with Phytophthora capsici; $80\%$ lysis at 100 mg/I of di-rhamnolipid or lactonic sophorolipid, showing the dependency of structure on the lysis. In the pot test, the damping-off disease incidence ratio decreased to $42\%\;and\;33\%$ of control value at 2,000 mg/l sophorolipid and rhamnolipid, respectively. These results showed the potential of microbial glycolipid biosurfactants as an effective antifungal agent against damping-off plant pathogens.
We present a novel cell deformability monitoring chip based on the digitally measured cell lysis rate which is dependent on the areal strain of the cell membrane. This method offers simple cell deformability monitoring by automated high-throughput testing system. We suggest the filter design considering the areal strain imposed on the cell membrane passing through the filter array having gradually increased orifice length. In the experiment using erythrocytes, we characterized the cell deformability in terms of average fracture areal strain which was $0.24{\pm}0.014\;and\;0.21{\pm}0.002$ for normal and chemically treated erythrocytes, respectively. We also verified that the areal strain of 0.15 effectively discriminates the deformability difference of normal and chemically treated erythrocytes, which can be applied to the clinical situation. We compared the lysis rates and their difference for the samples from different donors and found that the present chips can be commonly used without any calibration process. The experimental results demonstrate the simple structure and high performance of the present cell deformability monitoring chips, applicable to simple and cost-effective cell aging process monitoring.
어류 에드워드감염증에 대한 예방 재조합 ghost 백신을 개발하기 위한 연구의 일환으로 어류 병원성 세균인 Edwardsiella trada를 대상으로 플라스미드 형질전환을 실시하고 형질전환 안정성을 평가하였으며, 형질 도입된 재조합 ghost 세균의 E-lysis 유전자 발현을 분석하였다. E. tarda를 대상으로 한 ghost 유도는 대장균에 비해 상대적으로 장시간의 반응 시간이 요구되며 lysis의 개시가 지연된 점을 고려 시 발현된 E protein의 용해 능력 또는 E-gene의 전사발현 양이 E. tarda에서 다소 약화되는 것으로 나타났다. 그러나 대장균에 비해 ghost 유도 속도가 다소 낮음에도 불구하고 반응이 완성되었을 시점에서의 E. tarda의 ghost 효율은 대장균과 전혀 차이가 없이 99.99% 이상의 유도효율을 나타내었다.
고형암에서의 종양용해증후군은 드물지만 발생하면 높은 사망률을 보이는 치명적인 합병증으로 외국에서는 지금까지 약 50여명의 증례가 보고되어 왔다. 우리 나라에서도 종종 발생한다고 알려져 있지만 실제로 뒷받침 할만한 증례를 찾아 보기가 힘든 실정이었다. 이에 저자들은 소세포폐암에서 항암화학요법 도중 발생한 치명적 종양 용해증후군 1예를 경험하였기에 문헌 고찰과 함께 이를 보고하는 바이다.
Digitonin is a strong hemolysin and glycyrrhizin has protective activity against the deterring effect of other hemolytic saponins. The interaction of these saponins with liposomes was studied as a function of cholesterol in membrane. In the case of multilamellar vesicles, which act as ideal osmometers, digitonin distrupted the barrier function of liposomes composed of phosphatidyl choline, dicetyl phosphate and cholesterol, however, did not influence on cholesterol-lacking liposomes. Glycyrrhizin had similar effect on liposomes irrespective of cholesterol in membrane. In the test with large unilamellar vesicles, digitonin increased the lysis with increasing cholesterol content in membrane, but glycyrrhizin showed no detectable change in cholesterol-containing liposomes. These results suggest that incorporation of cholesterol into liposomes increases the susceptibility to digitonin, resulting in lysis of liposomes, and that the inhibitory effect of glycyrrhizin against other hemolytic saponins in cholesterol-independent.
A more rapid and efficient method to extract RNA from Ecklonia cava Kjellman (Laminariales, Phaeophyceae) was introduced in this study. Each step of the procedure was evaluated and the optimal concentration of each chemical in the lysis solution was determined. Tissue pulverization with PVPP and β-mercaptoethanol in the lysis solution were not essential for RNA extraction of this species. The highest yield and purity of E. cava RNA were obtained by the lysis solution containing 1% CTAB, 1 M NaCl, 0.7% PVP, 10mM EDTA and 100mM Tris-Cl (pH 9.0). Approximately 8μg of RNA was obtained from 200 mg of ground tissue. The ratios of the absorbance at 260 nm and 280 nm were from 1.6 to 1.8 and those of at 230 nm and 260 nm were from 1.8 to 2.0. The extracted RNAs obtained in this study turned out to have a sufficient quality for cDNA synthesis.

As our population ages and the rate of spine surgery continues to rise, the use epidural lysis of adhesions (LOA) has emerged as a popular treatment to treat spinal stenosis and failed back surgery syndrome. There is moderate evidence that percutaneous LOA is more effective than conventional ESI for both failed back surgery syndrome, spinal stenosis, and lumbar radiculopathy. For cervical HNP, cervical stenosis and mechanical pain not associated with nerve root involvement, the evidence is anecdotal. The benefits of LOA stem from a combination of factors to include the high volumes administered and the use of hypertonic saline. Hyaluronidase has been shown in most, but not all studies to improve treatment outcomes. Although infrequent, complications are more likely to occur after epidural LOA than after conventional epidural steroid injections.
The tumour lysis syndrome (TLS) is a group of metabolic abnormalities caused by rapid and unexpected release of cellular components into the circulation as a result of massive destruction of rapidly proliferating malignant cells. It usually develops in patients with hematologic malignancies like acute lymphoid leukemia, non-Hodgkin and Burkitt's lymphoma after initiation of chemotherapy or may, rarely, occur spontaneously. Though TLS is seldom observed in relation to solid tumours, there have been reports of connections with examples such as lung, liver, breast, gastric carcinomas. The clinical manifestations of TLS include hyperuricemia, hyperkalemia, hyperphosphatemia and hypocalcemia. These indications if untreated lead to life-threatening complications such as acute renal failure, cardiac arrhythmias, seizures, and eventually death due to multiorgan failure. Therefore early detection of TLS is of vital importance. This can be accomplished by identification of high risk patients, implementation of suitable prophylactic measures andmonitoring of the electrolyte levels in patients undergoing chemotherapy.
