Effects of Lycii Fructus Water Extracts on the Lead-Induced Nephrotoxicity in Rats (구기자(枸杞子) 추출물이 납 투여(投與)에 의한 흰쥐의 신장(腎臟) 손상(損傷)에 미치는 영향(影響))

  • This study was designed to investigate the effects of Korean Lycii Fructus water extract in Pb-administered rats. The Pb exposed rats were given 100 ppm and 200 ppm in the distilled water. Sixty male Sprague-Dawley rats weighing between 90 and 110g were blocked into 6 groups according to body weight. The control group was fed a normal diet, without lead. The experimental groups, which was fed a normal diet plus 100 ppm and 200 ppm lead, and one group received a normal diet plus Lycii Fructus water extracts. The results: the Food intake, the weight gain, and the kidney weight content in the cadmium added groups were lower than those in the Lycii Fructus water extracts group. The contents of Pb in the kidneys of the rats were determined by using ICP(lnductively Coupled Plasma Spectrophotometer). The accumulation of lead in the kidney was lower in the Lycii Fructus water extracts group. The Plasma levels of renin activity was higher in the lead administration groups, as compared with the Lycii Fructus water extracts. Plasma levels of aldosterone activity was higher in the lead administration group, as compared with Lycii Fructus water extracts. These results suggest that Lycii Fructus water extracts has a lowering effects on the accumulation of pb on kidney and it is believed that the Lycii Fructus water extracts have some protective effects on lead-induced nephrotoxicity in rats, but the mechanism of these effects was obscure.

  • The aim of this experiments was to investigate the effect of Lycii Fructus water extracts on the blood pressure, plasma renin activity and plasma levels of atrial natriuretic peptide in two-kidney one clip Goldblatt hypertensive rats. Systolic blood pressure was decreased significantly after administration of Lycii Fructus water extracts. Plasma levels of atrial natriuretic peptide was increased significantly after adminstration of Lycii Fructus water extracts. Plasma renin activity was not changed after adminstration of Lycii Fructus water extracts. Thease results suggested that the depressor response after adminstration of Lycii Fructus water extracts were related with the changes of the plasma levels of atrial natriuretic peptide.

Immunopotentiating Activity of Water Extracts of Some Crude Drugs (수종 생약 수침 엑기스의 면역증강 작용에 관한 연구)

  • Immunopotentiating activity of water extract of Lycii Fructus, Rubus Fructus, Torilis Fructus, Schizandrae Fructus and Cuscutae Semen were investigated using mouse fed with normal diet and protein deficient diet. Body weight, ratio of spleen or thymus to body weight, number of total peritoneal exudate cell, content of total serum protein and albumin content were decreased in the mouse fed with protein deficient diets as compared to the mouse fed with normal diet. Regardless of the nutritional condition, the supplement of the water extract of Lycii, Rubus Torilis, Schizandrae Fructus or Cuscutae Semen increased the ratio of spleen to body weight. The increment of this ratio was significant by the supplement of water extracts of Lycii and Torilis Fructus in mouse fed with protein deficient diet. The supplement of water extracts of Torilis Fructus to the mouse fed with protein deficient diet increased the content of total serum protein and albumin content significantly. Similar results were obtained with Lycii, Rubus, Schizandrae Fructus and Cuscutae Similar. The electrophorogram shows the difference in the concentration of some serum protein constituents between the mouse fed with normal diet and protein deficient diet.

  • Pharmacological effects of water extracts of Lycii Fructus, Lycii Folium and Lycii Cortex Radicis from Lycium chinensis were investigated. Lycii Folium significantly protected the hepatic function from damages orally caused by $CCl_4 $administration in mice and had a strong hypoglycemic effect in hyperglycemic mice induced by streptozotocin. Lycii Fructus decreased the blood pressure rise associated with the growth of normal rats. Lycii Cortex Radicis had a strong hypoglycemic effect in hyrerglycemic mice induced by streptozotocin. Also, hypolipidemic effects in hyperlipidemic rats induced by 1% cholesterol fed-diet and 75% fructose were significantly observed by oral administration of water extracts of Lycii Fructus, Lycii Folium and Lycii Cortex Radicis.

  • This study was designed to investigate the effects of Korean Lycii fructus water extract in Al (Aluminum) administered rats. Forty-eight male Sprague-Dawley rats were divided into six groups: Control group, water extract group with 3% Lycii fructus, 1000 and 2000 ppm of Al groups, and 1000 and 2000 ppm of Al with 3% Lycii fructus water extract group. The Al content of rat tissue in the Al administered group was lower than that in rat tissue in the Al with 3% Lycii fructus water extract group. Plasma levels of renin and aldosterone activity was higher in the Al administration group, compared with the 3% Lycii fructus water extract group and Al administered group. Aspartate amino transaminase and alanine amino transaminase activities were elevated in the Al administered group and lower in the 3% Lycii fructus water extract group. Lactate dehydrogenase was lower in the 3% Lycii fructus water extract Al group than in the Al group. Choline acetyltransferase was higher in the 3% Lycii fructus water extract Al group than in the Al group.

Effect of Water Extracts of Ganoderma lucidum, Camellia sinensis and Lycii fructus on the Lipid Stability of Walnut (영지, 녹차 및 구기자 물 추출물이 호도 지방질의 안전성에 미치는 영향)

  • The lipid stability of walnut added with water extracts of Ganoderma lucidum, Camellia sinensis ana Lycii fructus was studied. The results are as follows:1. when the extracts were added to walnut, the lipid stability was improved. The antioxidative activities of water extract of Camellia sinensis and Lycii fructus were considerably higher than that of Ganoderma lucidum. The antioxidative activity was decreased in the rank order Lycii fructus>Camellia sinensis>Ganoderma lucidum. 2. This clearly suggests that water extract of Lycii fructus at 4$^{\circ}C$ Bx level can be used as an alternative natural antioxidant for the lipid stability of walnut.

Antioxidative and Antihypertensive Effects of Lycii fructus Extracts (구기자(Lycii fructus) 추출물의 항산화와 항고혈압 효과)

  • The physiological activity of Lycii fructus extracts were examined. Total phenolic contents in the ethanol extracts (9.5 mg/g) of Lycii fructus were higher than that of water extracts (8.7 mg/g). The chlorogenic acid ($1.7{\mu}g$ in water extracts and $1.3{\mu}g$ in $60\%$ ethanol extract) was the most abundant phenolic compound as analyzed by HPLC. The ABTS [2,2'-azinobis (3-othylbenfothiaznoline-6- sulfornic acid)] radical decolor-ization electron donating ability (DPPH) and antioxidant protection factor (PF) were determined for extracts from Lycii fructus. Water extract ($76.7\%$ on ABTS, $92.6\%$ on DPPH and 1.1 on PF) showed higher inhibition rate than $60\%$ ethanol extracts ($52.8\%,\;88.8\%$ and 1.0). Thiobarbituric acid reactive substance (TBARS) was determined as $1.5{\times}10^{-3}\;{\mu}M$ in $60\%$ ethanol extract. Ethanol extracts was more effective in decreasing TBARS than water extracts. The water extracts from Lycii fructus had higher angiotensin converting enzyme (ACE) inhibition activity than ethanol extracts. The result will be useful for functional foods application and under-standing the physiological activities of Lycii fructus.

  • In the previous screening on antidiabetic effect of Lycii fructus by glucose transport method using $N_2$-STZ diabeted rat model, each extracts showed the potent antidiabetic activity. We obtained three compounds isolated from the water fraction, EtOAc fraction and n-BuOH fraction of Lycii fructus in the present work and their structures were identified as 1-carboxy-N,N,N-trimethylmethanaminium hydroxide inner salt, 2,4(1H,3H)-pyrimidinedione and 3,3',4',5,7-pentahydroxyflavone-3-rutinoside . Among the constituents separated from Lycii fructus, 2,4(IH,3H)-pyrimidinedione, 3,3',4',5,7-pentahydroxyflavone-3-rutinoside and ascorbic acid were shown a remarkable antidiabetic effect.

Cardiovascular Biofunctional Activity and Antioxidant Activity of Gugija (Lycium chinensis Mill) Species and Its Hybrids (구기자 품종과 교배종의 부위별 심혈관관련 질환 예방 기능성 및 항산화 활성비교)

  • To develop new high valuable Gugija (Lycium chinensis), biofunctionalities of Gugija standard species and its hybrids were investigated and compared with each water extracts and methanol extracts from Lycii Fructus, Lycii Folium and buds and Lycii Cortex Radicis. Among various biofunctionalities of Gugija standard species, antioxidant activity was showed the highest in methanol extracts from buds of Cheongwoon species (93%) and antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity was 84.1% in the water extracts from Lycii Cortex Radicis of Cheongyang NO.7. Futhermore, methanol extracts from Lycii Cortex Radicis of Myungan A-2 hybrid showed 93.1% of antioxidant activity and 96.9% of ACE inhibitory activity was also showed in the methanol extracts from Lycii Fructus of DO148-72(A11) hybrid. However, fibrinolytic activity and anticholesteromia HMG-CoA reductase inhibitory activity were weak or not detected in almost of Gugija standard species and its hybrids. Therefore, we finally selected Cheongwoon Gugija standard species (buds) and Myungan A-2 hybrid (Lycii Cortex Radicis) as good antioxidant sources and also DO148-72 (A11) hybrid (Lycii Fructus) as excellent antihypertensive ACE inhibitior sources for manufacturing functional food product.

Effects of Coating Syrup with Water-Soluble Extracts of Gugija(Lycii fructus) on the Quality Characteristics of Yukwa (구기자의 수용성 추출물을 첨가한 집청액이 유과의 품질특성에 미치는 영향)

  • In this study, Yukwa(YU-G) was prepared by coating in syrup with water-soluble extracts of Gugija(Lycii fructus). The Yukwa samples were stored with PE film packing with air for 12 weeks at room temperature, after which the quality and shelf-life of the Yukwa were assessed. During storage, the moisture contents of YU-G were higher than those of the Yukwa due to being coated in syrup without water-soluble extracts of Gugija(YU-NG), whereas the instrumental textural hardness values of the YU-G were lower than those of the YU-NG. Color determination indicated that YU-NG and YU-G assumed a darker color after storage, becoming both redder and yellower. The peroxide and acid values increased abruptly in the YU-NG during storage, but increased slowly in the YU-G. During the 12 weeks of storage after preparation, the peroxide and acid values of YU-G remained at less than 40 meq/kg and 4 KOH mg/g, respectively. Thus, the experimental method of syrup coating with water-soluble extracts of Gugija rendered the YU-G fairly stable against fat rancidity. The YU-G evidenced generally higher overall acceptability than YU-NG when stored.
