Effect of Lycii Fructus Powder on Lipid Metabolism in 1% Cholesterol Fed Rats (구기자 분말 첨가 식이가 1% 고콜레스테롤 급여 흰쥐의 지질 대사에 미치는 영향)

  • This study examined the effects of 2% Lycii fructus powder (LFP) supplementation on lipid metabolism in rats. Male Sprague-Dawley rats were administered 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia. Then, 40 rats were divided into four diet groups: a normal diet group (NC), high cholesterol diet group (HC), normal diet plus 2% Lycii fructus powder (NC-LFP) group, and high cholesterol diet plus 2% Lycii fructus powder (HC-LFP) group. The HC group presented higher growth rates and liver weights than NC and NC-LFP however, growth rates and liver weights in the 2% LFP administered groups gradually decreased. HC also showed increased serum triglyceride, total cholesterol, free cholesterol, and LDL-cholesterol levels and decreased atherogenic index values, HDL-cholesterol, and phospholipid levels, whereas LFP group showed decreased serum triglyceride, total cholesterol, free cholesterol, and LDL-cholesterol levels as compared to HC. There were no differences in serum triglyceride, phospholipid, LDL-cholesterol, HDL-cholesterol, and free cholesterol concentrations between the normal diet groups (NC and NC-LFP). The high cholesterol diet groups (HC and HC-LFP) had significant increases in serum alanine aminotransferase (ALT), asparate aminotransferase (AST), alkaline phosphatase (ALP), and lactate dehydrogenase (LDHase) activities. And the 2% LFP administered groups had lower hepatic concentrations of total cholesterol and triglycerides than the HC groups. Overall, the results suggest that Lycii fructus powder has hypochloesterolemic effects by reducing serum and liver cholesterol contents.

Development of Extracts of Lycii folium Having High Antioxidant Activity (항산화 활성이 높은 구기엽 추출물 제조)

  • The aim of this study was to analyze the antioxidant properties of Lycii folium extracts prepared from different solvents. Lycii folium were extracted with water, 80% ethanol (80% EtOH), 80% methanol (80% MeOH) and 100% methanol (100% MeOH) in water bath at $40^{\circ}C$. The antioxidant activity of the extracts was evaluated using DPPH, hydroxyl and hydrogen radical scavenging activities, and SOD-liked activity. Total phenolic acid contents were 1.085 mg/mL in 100% MeOH, 1.382 mg/mL in 80% EtOH, 1.420 mg/mL in 80% MeOH and 1.084 mg/mL in water. DPPH radical scavenging activity of the extracts were 65.60% in 80% EtOH, 56.80% in 80% MeOH, 83.85% in 100% MeOH and 54.65% in water. Hydroxyl radical scavenging activities were 66.65% in 100% MeOH, 73.13% in 80% ethanol, 73.58% in 80% MeOH and 70.73% in water. Hydrogen radical scavenging activity of the extracts prepared from Lycii folium were 11.70% in 100% MeOH, 33.73% in 80% EtOH, 35.40% in 80% M eOH and 23.86% in water. SOD-liked activity of the extracts prepared from Lycii folium was 71.58% in 100% MeOH, 74.29% in 80% EtOH, 88.46% in 80% MeOH and 67.47% in water. Our result showed that Lycii folium extracts prepared from 80% methanol were found to be promising biomaterials with antioxidant effects.

Effects of Lycii Fructus on the Ovariectomized Osteoporosis of Rats (구기자(枸杞子)가 난소적출(卵巢摘出) 흰쥐의 골조직대사(骨組織代謝)에 미치는 영향(影響))

  • Objectives: Osteoporosis is one of the most common metabolic bone disorders in the menopausal stage, a disease signified by decrease of bone mass. The purpose of this study was to learn how Lycii Fructus influences SD rats with osteoporosis caused by the removal of the ovaries. Methods: After the removal of the ovaries from 24 female Sprague-Dawley rats, they were divided into a normal group, a control group, and a Lycii Fructus (25mg/100g) administration group. After 8 weeks, the rats were sacrificed and their weights, albumin, AST, ALT, ALP, total-cholesterol, triglyceride, P, Ca, T4, E2, weight of a femur, ash content of the tibia, and area and thickness of trabecular bone were measured. Results: Regarding the density of ALT, ALP, and estradiol from serum, the Lycii Fructus group was significantly decreased compared to the control group, and the density of phosphorous and calcium also decreased slightly. Moreover, albumin, triglyceride, and T4 showed an increasing propensity, but AST and total-cholesterol showed a decreasing propensity, but not significant. Regarding the variation of bone, femur weight and ash content of tibia, the Lycii Fructus group was increased compared to the control group but significance wasn't shown. The Lycii Fructus group increased significantly over the control group in area and thickness of trabecular bone and osteoclast number. Conclusions: From the results of the above study, Lycii Fructus should be effective for osteoporosis treatment and prevention.

Antioxidative and Antihypertensive Effects of Lycii fructus Extracts (구기자(Lycii fructus) 추출물의 항산화와 항고혈압 효과)

  • The physiological activity of Lycii fructus extracts were examined. Total phenolic contents in the ethanol extracts (9.5 mg/g) of Lycii fructus were higher than that of water extracts (8.7 mg/g). The chlorogenic acid ($1.7{\mu}g$ in water extracts and $1.3{\mu}g$ in $60\%$ ethanol extract) was the most abundant phenolic compound as analyzed by HPLC. The ABTS [2,2'-azinobis (3-othylbenfothiaznoline-6- sulfornic acid)] radical decolor-ization electron donating ability (DPPH) and antioxidant protection factor (PF) were determined for extracts from Lycii fructus. Water extract ($76.7\%$ on ABTS, $92.6\%$ on DPPH and 1.1 on PF) showed higher inhibition rate than $60\%$ ethanol extracts ($52.8\%,\;88.8\%$ and 1.0). Thiobarbituric acid reactive substance (TBARS) was determined as $1.5{\times}10^{-3}\;{\mu}M$ in $60\%$ ethanol extract. Ethanol extracts was more effective in decreasing TBARS than water extracts. The water extracts from Lycii fructus had higher angiotensin converting enzyme (ACE) inhibition activity than ethanol extracts. The result will be useful for functional foods application and under-standing the physiological activities of Lycii fructus.

Quality Characteristics of Fish Pastes Containing Lycii fructus Powder (구기자 분말 함유 어묵의 품질 특성)

  • This study was performed to aid promotion of fish paste containing Lycii fructus powder as a food. The tested concentrations of Lycii fructus powder were 0, 1, 3, 5, and 7%. All the fish paste samples had 37% water content. The L values of the samples decreased as the concentration of Lycii fructus powder increased(p<0.001), and the a- and b-value increased(p<0.001). All samples with 3 mm of thickness had good flexibility and did not break even after four times of folding. In texture meter tests, the cohesiveness increased, but gumminess and brittleness decreased according to increasing concentration of Lycii fructus' powder. In the sensory evaluation, scores for color, flavor, pleasant taste, hardness and adhesiveness increased with increasing powder concentration; however oily taste was decreased. The 3% Lycii fructus powder sample had the highest acceptance scores in terms of appearance, flavor, taste, texture and overall acceptability. The results suggest that Lycii fructus powder can be applied to fish paste for both quality and functionality.

Cardiovascular Biofunctional Activity and Antioxidant Activity of Gugija (Lycium chinensis Mill) Species and Its Hybrids (구기자 품종과 교배종의 부위별 심혈관관련 질환 예방 기능성 및 항산화 활성비교)

  • To develop new high valuable Gugija (Lycium chinensis), biofunctionalities of Gugija standard species and its hybrids were investigated and compared with each water extracts and methanol extracts from Lycii Fructus, Lycii Folium and buds and Lycii Cortex Radicis. Among various biofunctionalities of Gugija standard species, antioxidant activity was showed the highest in methanol extracts from buds of Cheongwoon species (93%) and antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity was 84.1% in the water extracts from Lycii Cortex Radicis of Cheongyang NO.7. Futhermore, methanol extracts from Lycii Cortex Radicis of Myungan A-2 hybrid showed 93.1% of antioxidant activity and 96.9% of ACE inhibitory activity was also showed in the methanol extracts from Lycii Fructus of DO148-72(A11) hybrid. However, fibrinolytic activity and anticholesteromia HMG-CoA reductase inhibitory activity were weak or not detected in almost of Gugija standard species and its hybrids. Therefore, we finally selected Cheongwoon Gugija standard species (buds) and Myungan A-2 hybrid (Lycii Cortex Radicis) as good antioxidant sources and also DO148-72 (A11) hybrid (Lycii Fructus) as excellent antihypertensive ACE inhibitior sources for manufacturing functional food product.

Effects of Lycii Fructus and Lycii Folium Extracts on Osteoporosis in Ovariectomized Rats (구기자와 구기엽 추출물이 난소적출 흰쥐의 골다공증에 미치는 영향)

  • The effects of Lycii fructus and Lycii folium on osteoporosis and serum cholesterol levels were tested in ovariectomized (OVX) rats. Twenty-four female Sprague-Dawley rats were divided into four groups: Sham group (sham-operated), Control group (OVX, ovariectomized model), LCF group (Ovx+Lycii fructus extract), and LCL group (OVX+Lycium folium extract). After 8 weeks, the OVX ($330{\pm}5.39$ g), LCF ($315{\pm}2.99$ g), and LCL ($318{\pm}2.06$ g) groups showed increased body weight compared with sham group ($281{\pm}1.71$ g). The levels of serum osteocalcin (OC) also increased in the LCF ($444.6{\pm}26.9$ ng/mL) and LCL ($407{\pm}18.9$ ng/mL) groups compared with the OVX group ($107{\pm}3.52$ ng/mL). The activities of serum alkaline phosphatase (ALP) increased in the LCF ($108{\pm}2.7$ U/L) and LCL ($407{\pm}18.9$ ng/mL) groups compared with the OVX group ($95{\pm}2.9$ U/L). Stereomicroscopy found that the low bone density that developed in the OVX group was significantly reversed in the LCF and LCL groups after 8 weeks. We also obtained molecular-based in vivo evidence that supports a mechanism of action involving novel estrogen receptor ($ER{\alpha}$) modulator in the uterus. We found that expression of ER${\alpha}$ mRNA in the OVX rat uterus was elevated by Lycium chinense. These results suggest that Lycii fructus and Lycii folium administered to rats during 8 weeks after oophorectomy may partially recover postmenopausal osteoporosis or delay the progression of osteoporotic changes.

Effects of Mixture of Lycii Radicis Cortex and Moutan Cortex on Corticotropin-Releasing Factor, c-Fos, and Tyrosine Hydroxylase in Forced Swimming Test (지골피(地骨皮)와 목단피(牧丹皮) 복합제(複合劑)가 강제수영부하실험에서 CRF, c-Fos와 Tyrosine Hydroxylase에 미치는 영향)

  • Objectives : The goal of this study was to investigate the antidepressant effects of Mixture of Lycii Radicis Cortex and Moutan Cortex on the change of HPA-Axis and Catecholamic system. Methods : We were performed the Forced Swimming Test. The expressions of Corticotropin-Releasing Factor (CRF) and c-Fos at paraventricular nucleus (PVN), and tyrosine hydroxylase (TH) at ventral tegmental area (VTA) and locus coeruleus (LC) were evaluated by immunohistochemical method. Results : The duration of immobility in the Forced Swimming Test was significantly decreased in A100, A400(p<0.05~p<0.01). CRF expressions at PVN was significantly decreased in A400(p<0.05).No other group showed significant difference in expression of c-Fos at PVN comparing with control group. TH expressions at VTA was significantly decreased in A100 and A400, respectively(p<0.001). TH expressions at LC was significantly decreased in A100(p<0.01). Conclusions : According to the above results, Mixture of Lycii Radicis Cortex and Moutan Cortex has antidepressant effects via the reduction of CRF expression and the Catecholamine System activity.

  • Some physical properties and chemical compositions of Lycii Cortex depending on the various extracting conditions were investigated for their changes during extraction with ethanol and water. Solid matter and contents of total sugar in water extracts were higher than those of ethanol extract. but contents of tannic acid in ethanol extracts was higher than that in water extracts. The methanol extract of the Lycii Cortex were fractionated with ethyl ether. ethyl acetate. n-butanol and water. The yield was higher in the order of water > n - butanol > ethyl ether and ethyl acetate fraction. Turbidity of water extraction was higher than that of ethanol extraction.

Skin Whitening Effects of Extracts from Angelicae Gigantis Radix and Lycii fructus Ethanol Extracts (당귀와 구기자 에탄올 추출물의 피부 미백작용)

  • Objective : The purpose of this study was to investigate the synergistic effect of Angelicae gigantis Radix (AG) and Lycii fructus (LF) ethanol extracts on skin-whitening effects. Method : LFAG extracts were prepared by extracting with 80% ethanol. The efficacy of LFAG was judged by measurement of cell viability, tyrosinase activity, melanin production, tyrosinase and microphthalmia-associated transcription factor (MITF) expression in B16F10 murine melanoma cells by lipopolysaccharides (LPS) treatment. Results : Each extract (LF or AG) inhibited the tyrosinase activity in a dose-dependent manner. The co-treatment of LFAG extracts ($25{\mu}g/mL$ LF plus $25{\mu}g/mL$ AG) markedly suppressed the LPS-induced cellular tyrosine activity, melanin production, tyrosinase and MMP-1 expression in B16F10 murine melanoma cells. These suppressive effects were synergistically increased by their combination. Conclusions : With these observations, we suggest that the extracts from Lycii fructus and Angelica gigantis Radix could be potent natural materials for whitening skin.
