구기자분말을 첨가한 생면의 품질특성 (Quality Characteristics of Wet Noodle with Lycii fructus Powder)

  • 구기자 분말을 1, 3, 5 및 7% 첨가한 복합분으로 생면을 제조하여 아밀로그램특성, 조리특성, 조직감, 관능검사 등 생면의 물성과 총균수 변화를 통한 저장성을 조사하였다. 아밀로그램특성은 구기자 분말 첨가비율이 높아질수록 호화개시 온도는 $61^{\circ}C$(대조구), $61.5^{\circ}C$(1%), $62^{\circ}C$(3%), $62^{\circ}C$(5%), $62.5^{\circ}C$(7%)로 증가하였고, 최고점도시 온도는 $91.5^{\circ}C$(대조구), $91^{\circ}C$(1%), $90.5^{\circ}C$(3%), $90^{\circ}C$(5%), $89^{\circ}C$(7%)로 감소하였으며, 최고점도는 875 B.U.(대조구) 720 B.U.(1%), 565.5 B.U.(3%), 475 B.U.(5%), 395 B.U.(7%)로 각각 감소하였다. 조리면의 중량과 부피는 구기자 분말 첨가량이 증가함에 따라 감소하였고, 조리 손실량은 증가하였다. 구기자 분말을 첨가한 생면을 조리 후 texturometer에 의한 기계적 texture 특성을 측정한 결과 구기자 분말 첨가량이 증가할수록 견고성, 응집성, 씹힘성, 탄성 모두 감소하였다. 조리면의 관능검사결과 구기자 분말을 3%첨가구가 조직감을 제외한 외관, 맛, 전체적인 기호도 면에서 가장 높은 평가를 받았다. 생면의 저장성은 상온 $(20{\pm}4^{\circ}C)$ 저장시에는 대조구와 구기자 분말 첨가구간에 유의적인 차이를 보이지 않고 빠르게 총균수가 증가 하였으며, 저온$(5{\pm}1^{\circ}C)$ 저장시에는 대조구가 저장 15일에 $6.6{\times}10^4\;cfu/g$에 도달한 것에 비해 구기자 분말 첨가구는 저장 20일 이후에도 $4{\times}10^4\;cfu/g$에 도달하여 구기자 분말 첨가에 따라 저장기간이 연장되는 결과를 보였다.

  • 구기자추출물(枸杞子抽出物)을 시료(試料)로 mouse의 독성실험(毒性實驗), 실험적(實驗的)으로 유발(誘發)시킨 가토(家兎)의 간장기능장애(肝臟機能障碍) 및 alloxan부하당뇨병(負荷糖尿病)에 미치는 영향(影響)을 실험(實驗)하여 다음과 같은 결과(結果)를 얻었다. 1. Mouse의 급성독성실험(急性毒性實驗)에서 복강투여(腹腔投與)의 경우 평균치사량(平均致死量)은 12.17g/kg이었다. 2. $CC1_4$로 간장기능장애(肝臟機能障碍)가 유발(誘發)된 가토(家兎)에 구기자추출물투여(枸杞子抽出物投與)는 대조군(對照群)에 비하여 유의성(有意性)있는 회복도(回復度)를 보였는데 특(特)히 대량투여군(大量投與群)에서 더욱 빨랐다. 1) GPT활성도(活性度) 및 total bilirubin은 시료투여군(試料投與群)에서 아주 우수(優秀)한 저하효과(低下效果)를 나타냈다. 특(特)히 GPT활성도(活性度)에서 200mg/kg은 제(第)8일(日)째부터, 800mg/kg은 제(第)6일(日)째부터 유의성(有意性)이 있었다. 2) Alkaline phosphatase치는(値) 대조군(對照群)에 비하여 커다란 차이를 보이지 않았다. 3) Alloxan으로 유발(誘發)한 가토(家兎)의 고혈당(高血糖)에 대하여 800mg/kg는 아주 탁월한 혈당강하효과(血糖降下效果)를 나타냈다. 즉(卽) 제(第)4일(日)째부터 대조군(對照群)에 비하여 유의성(有意性)이 있었으며 제(第)12일(日)째는 정상치(正常値)에 거의 접근(接近)하였다. 또한 GPT활성도(活性度)는 차츰 저하(低下)되어 제(第)6일(日)째부터 저하효과(低下效果)가 뚜렷하였다.

지골피와 생강 정제 추출물이 고지방식이 흰쥐의 체내 지질과 혈청 Cytokine 수준에 미치는 영향 (Effects of the purified extracts from Lycii Cortex Radicis and ginger on lipid statusand serum cytokine levels in rats fed high fat diet)

  • The present study was to investigate the effects of Lycii Cortex Radicis (LCR), the root bark of lycium (Lycium chenese Miller) and ginger (Gin) on body lipid status and serum levels of cytokines. Sprague-Dawley (SD) male rats weighing $193.6{\pm}16.8g$ were divided into five groups, including one low fat (LF) and four high fat groups, i.e. HF-Control, HF-LCR, HF-Gin and HF-LCR + Gin groups. Diets for HF-LCR, HF-Gin and HF-LCR + Gin groups contained purified extracts having 0.2 g LCR tyramine, ginerol and 0.1 g tyramine plus 0.02 g gingerol per kg, respectively. Compared with those of the HF-Control total serum cholesterol level decreased, and HDL-cholesterol level increased in the HF-LCR group and serum triglyceride levels decreased in the three experimental groups fed the purified extracts. Liver cholesterol level was lower in the HF-LCR group than the HF-Control group, but triglyceride levels, which were increased by high fat diets were not changed by significantly by LCR or ginger extracts. Fecal lipid excretion was higher in the HF-LCR and HF-Gin groups, but cholesterol excretion was lower in the HF-Gin group than in the HF-Control group. The activities of liver cytosolic glucose-6-phosphate dehydrogenase and malic enzyme were lower in the HF-LCR + Gin group than in the HF-Control group. Serum adiponectin levels did not differ among the five groups, while leptin level was lower in the HF-Gin group and C-reactive protein levels were lower in the HF-Gin and the HF-LCR + Gin groups than in the HF-Control group. It is concluded that LCR can be utilized as an ingredient for lipid-lowering functional foods in the form of purified extract and addition of small amount of ginger extract would be useful for reducing one of the inflammatory cytokines to help prevent atherosclerosis.

비알코올성 지방간 세포 모델에 대한 택사, 산사, 구기자, 울금, 단삼, 인진의 효능 비교 (Comparison of the Therapeutic Efficacy of Rhizoma Alismatis, Fructus Crataegi, Fructus Lycii, Radix Curcumae, Radix Salviae Miltiorrhizae, Herba Artemisiae Scopariae on the Experimental Cellular Model of Nonalcoholic Fatty Liver Disease)

  • Objectives : We try to compared the efficacy of six herbal medicines, Rhizoma Alismatis (RA), Fructus Crataegi (FC), Fructus Lycii (FL), Radix Curcumae (RC), Radix Salviae Miltiorrhizae (RSM), and Herba Artemisiae Scopariae (HAS), constituting KHchunggan-tang which was previously proven to be hepatoprotective on non-alcoholic fatty liver disease with combined properties of cellular steatosis, ROS production, and cytoprotection. Methods : HepG2 cells were pretreated with aqueous extracts of the six herb medicines at concentrations of 1, 10, 50 and 100 ${\mu}g/ml$ each, and treated with 0.5 mM palmitate consecutively. After 21 hrs, cell viability was assessed using MTT assay, and the percentage of cells with sub-G1 DNA content was measured using fluorescence-activated cell sorting after propidium iodide staining. Results : The first three extracts, RA, FC, and FL restored cell viability reduced by palmitate in MTT assay, and RA, FC, FL and RC inhibited palmitate-induced apoptosis in sub-G1 analysis. FL showed relatively weak potential only at tested maximal dose, and RA showed the greatest higher efficacy on this experimental cellular model of nonalcoholic fatty liver disease. Conclusions : According to this comparative experiment, Rhizoma Alismatis seems to have the most powerful potential among the six herbs constituting KHchunggan-tang, and consecutive further study seems to be required for more standardized and effective clinical application of KHchunggan-tang for treatment of non-alcoholic fatty liver disease.

당뇨환자를 위한 새로운 식이요법제에 관한 연구 (Studies on a New Alimentotherapy for Diabetic Patients)

  • 상엽과 상심자, 구기자, 인삼 등은 당뇨에 효능이 있는 것으로 널리 알려진 약재들이다. 본 실험에서는 기본 실험을 통하여 상엽, 상심자, 구기자, 인삼, 차가버섯의 최적 배합비와 최적의 추출방법으로 엑기스 형태로 만들어진 소당엑스$^{TM}$와 이를 특허 출원한 방법으로 백미에 코팅한 소당미$^{TM}$를 사용하여 시험한 결과 Alloxan으로 당뇨를 유도한 마우스, 정상인, 그리고 당뇨환자 모두에서 혈당이 저하되었음을 확인하였다. 따라서 소당엑스$^{TM}$와 소당미$^{TM}$는 당뇨환자의 혈당 조직 및 합병증 예방에 효과적이므로 당뇨식이요법제로서 이용될 수 있는 건강 식품으로 사료된다.

사육탕가미방(四六湯加味方)이 streptozotocin으로 유발(誘發)된 백서(白鼠)의 실험적(實驗的) 탕뇨(糖尿)에 미치는 영향 (Effect of Saryuktangkamibang on Streptozotocin-induced Diabetic Rat)

  • Objectives : The objective of this study is to observe the effect of Saryuktangkamibang, Saryuktang with Liriopis Tuber, Lycii Fructus, Schizandrae Fructus, and Bombyx Batryticatus, on the experimental diabetes. Methods : In order to induce diabetes experimentally, injected streptozotocin to the vein in the tail of rats and then treated oral administration of Saryuktangkamibang water extracts. In the measurement of the variation levels of glucose, ALP, creatinine, and BUN concentration for each concentration levels for serum (0.2g/100g, 0.4g/l00g, 0.6g/100g), concentration level of glucose significantly decreased in 0.4g/l00g concentration level of Saryuktangkamibang. With this 0.4g/l00g concentration level of Saryuktangkamibang, the following conclusion was derived from the measurement of the serum levels of glucose. ALP, AST, ALT, creatinine, and BUN concentration for the injection of each 4th, 11th, and 18th day. Results : 1. In the measurement for each concentrations, the glucose concentration level in the serum significantly decreased on the 9th day in the Saryuktangkamibang-dosed group. 2. The creatinine and BUN concentration level in the serum significantly decreased on the 9th day in the Saryuktangkamibang-dosed group to which 0.6g/100g injected. 3. In the measurement for each dosage dates, the glucose concentration level in the serum significantly decreased on the 11th day in the Saryuktangkamibang-dosed group to which 0.6g/100g injected. 4. The AST concentration level in the serum significantly decreased on the 11th day in the Saryuktangkamibang-dosed group. Conclusion : Saryuktangkamibang, Saruktang with Liriopis Tuber, Lycii Fructus, Schizandrae Fructus, and Bombyx Batryticatus, has an effect of hypoglycemia, with no hepato-renal dysfunction. For the experimental results above, in that nephrotoxicity significantly decreased on the 0.6g/100g concentration, another research such as study for effect in other ranges and study for effect of dosage term except the concentration range of this study is necessary. Also dosage of appropriate concentration of Saryuktangkamibang is thought to provide valuable aid to cure on diabetes.

Streptozotocin으로 유도된 당뇨 흰쥐에서 하소에 사용되는 한약의 항당뇨 효과 비교 연구 (Anti-diabetic effect of medicinal plants used for lower wasting-thirst in streptozotocin-induced diabetic rats)

  • Objectives : To prove the channel-tropism theory in herbology, we investigated the anti-diabetic effect of six herbal plants used for lower wasting-thirst in streptozotocin-induced diabetic rats. Methods : Diabetes was induced in male Sprague-Dawley rats by consecutive injection of streptozotocin (30 mg/kg i.p.) for 5 days. The rats were divided into normal control, diabetic control, and diabetic treatment with Lycii Radicis Cortex (LRC, 300 mg/kg); Corni Fructus (CF, 300 mg/kg); Bombyx Batryticatus (BB, 50 mg/kg); Lycii Fructus (LF, 300 mg/kg); Phellodendri Cortex (PC, 300 mg/kg); Epimedii Herba (EH, 300 mg/kg); and glibenclimide (10 mg/kg) as a reference drug. Herbal extracts or reference drug were administered orally for 28 days. The changes of body weight, food intake and water intake, and serological markers such as blood glucose, serum total cholesterol, triglyceride (TG), blood urea nitrogen (BUN), and creatinine (Cr) were measured. Results : The decrease of body weight and the increase of food and water intake in STZ-induced diabetic rats was improved by the administration of CF and LF. Also, the enhancement of blood glucose and serum total cholesterol, TG, BUN and Cr in STZ-induced diabetic rats was significantly inhibited by the administration of CF, BB, LF and glibenclimide. On the other hand, EH strongly inhibited the increase of BUN and Cr in the sera of STZ-induced diabetic rats. Conclusions : These results suggest that among six herbal medicines used lower emaciation of emaciation-thirst disease, CF, BB, LF and EH show a characteristics including the channel-tropism theory.

천연산물로부터 항산화물질의 탐색

  • 30 kinds of natural products were considered for developing natural antioxidants by improved D.O. analyzing method including simple calculation of Area Under Curve. Several natural products such as Cimicifuge Rhizoma, Epimedii Herba, Atractylodis Rhizoma Alba, Acori Graminei Rhizoma, Mori Cortex Radicis, Aurantii Nobilis Pericarpium were bore nearly same antioxidant effects compared to synthetic powerful antioxidant BHT and also expressed powerful antioxidant effect than ${\beta}-carotene$ such as Eucommiae Cortex, Cinnamomi Cortex, Angelicae Gigantis Radix, Lycii Furctus, Acanthopanacis Cortex, Sophorae Radix, Paeoniae Radix, Geranii Herba. Another method of DPPH was performed for searching natural antioxidant from natural product. Sophorae Radix, Puerariae Radix, Aurantii Nobilis Pericarpium, Atractylodis Rhizoma Alba, Acori Graminei Rhizoma, Corni Fructus, Angelicae Gigantis Radix, Paeoniae Radix were carried higher antioxidant capacity than ${\beta}-carotene$ by DPPH method.

랫드와 마우스에서 연생호보액의 급성 경구 독성 시험에 관한 연구 (Acute Toxicity Study of Yan-Sheng Health Liquid in Rats and Mice)

  • The acute toxicity of orally administered Yan-Sheng health liquid (YSHL), water extracts from twelve Chinese drugs (Cervi Cornu, Lonicerae Flos, Foeniculi Fructus, Glycyrrhizae Radix, Liriopis Tuber, Raphani Semen, Bombyx, Ginseng Radix alba, Cinnamomi loureirii Cortex, Epimedii Herba, Zingiberis Rhizoma, Lycii Fructus) was evaluated in male and female Sprague-Dawley rats and ICR mice. Rats and mice aging 5 weeks were gavaged with 0, 2.0, 3.0, 4.4, 6.7, 10.0, 66.7, or 100.0 ml/kg of YSHL. No animal died by oral treatment and no toxic symptom was observed in the treated animals during 5 days. The body weight of the treated animals was not significantly different from the controls. The results of macroscopic examination on the organs of the treated animals revealed no abnormal findings. Therefore, it was concluded that YSHL was practically non-toxic when it was orally administered to rats and mice, and its LD50 was suspected to be greater than 100 ml/kg in rats and mice.
