Systemic Eosinophil Infiltration by Ovine Lungworm Infection (양 폐충 감염증에 의한 전신 호산구 침윤증)

  • Case 1 was showed severe caughing and nasal discharge, who was one of 5-month-old 18 male sheep taking inspection for MCF experiment. Pathological examination of case 1 was conducted. Macroscopic observations were the foamy sticky fluids in nasal and tracheal cavity, the grey spots with 2-4 mm diameter on the surface of lung. Histopathological observations were severe eosinophil and other round cell infiltration in general organs including lung, trachea, small intestine, large intestine, liver. Also, in the lung, there were parasite-cutting lesions in some alveolar spaces and bronchioles. Following these observations, case 1 was diagnosed as ovine lungworm infection.

Aspergillus fumigatus infection in Jackass penguin (자카스펭귄에서 발생한 Aspergillus fumigatus 감염증)

  • A three years old male Jackass penguin(Spheniscus demersus) showed anorexia, depression, respiratory problem for a few days, followed by clinical treatment for 20 days resulted in death. Grossly, multiple white nodules measuring 2~3 mm in diameter were observed in the surface of and inside lung, liver, kidney, thoracic wall. Microscopically, granuloma formations were observed in the lung and liver. The margin of granuloma surrounded by connective tissue barrier and infiltrated lymphocytes, and also observed giant cell near the granuloma. By Periodic acid Schiff reaction, hyphae were detected in granuloma of lung and liver. This case was a chronic and systemic aspergillosis caused by Aspergillus fumigatus infection in a Jackass penguin at a zoo.

Airway Mucus: Its Components and Function

  • The airway surface liquid (ASL), often referred to as mucus, is a thin layer of fluid covering the luminal surface of the airway. The major function of mucus is to protect the lung through mucociliary clearance against foreign particles and chemicals entering the lung. The mucus is comprised of water, ions, and various kinds of macromolecules some of which possess the protective functions such as anti-microbial, anti-protease, and anti-oxidant activity. Mucus glycoproteins or mucins are mainly responsible for the viscoelastic property of mucus, which is crucial for the effective mucociliary clearance. There are at least eight mucin genes identified in the human airways, which will potentially generate various kinds of mucin molecules. At present, neither the exact structures of mucin proteins nor their regulation are understood although it seems likely that different types of mucins are involved in different functions and might also be associated with certain airway diseases. The fact that mucins are tightly associated with various macromolecules present in ASL seems to suggest that the defensive role of ASL is determined not only by these individual components but rather by a combination of these components. Collectively, mucins in ASL may be compared to aircraft carriers carrying various types of weapons in defense of airborne enemies.

Development of Voltammetric Nanobio-incorporated Analytical Method for Protein Biomarker Specific to Early Diagnosis of Lung Cancer (폐암 조기 진단을 위한 단백질 바이오마커 측정용 전압-전류법 기반의 나노바이오 분석법 개발)

  • In this article, a portable and cost-effective voltammetric biosensor with nanoparticles was developed for the measurements of heterogeneous nuclear ribonucleoprotein A1 protein (hnRNP A1) biomarker which can potentially be used for lung cancer diagnosis. Gold nanoparticles were first electrodeposited onto screen printed carbon electrode (SPCE) followed by immobilizing a single stranded DNA aptamer specific to hnRNP A1 onto the electrode surface. Ethanolamine was also used when immobilizing DNA aptamer on the surface to prevent signals from non-specific adsorption events. Sequential injection of hnRNP A1 biomarker and anti-hnRNP A1 conjugated with alkaline phosphatase (ALP) onto the aptamer chip surface allows to form the sandwich complex of DNA aptamer/hnRNP A1/ALP-anti-hnRNP A1 on the electrode surface which further reacted with 4-aminophenyl phosphate (APP). The electrocatalytic reaction of the enzyme, ALP, and the substrate, APP, resulting in the oxidative current response changes at -0.05 and -0.17 V (vs. Ag/AgCl) against the hnRNP A1 concentration was measured using cyclic and differential pulse voltammetry, respectively. The Au nanoparticles-integrated voltammetric biosensor was applied to analyze human normal serum solutions possibly suggesting potential applicability for lung cancer diagnosis.

Effects of Lonicerae Flos Extracts on LPS-induced Acute Lung Injury (금은화가 LPS로 유발된 급성 폐 손상에 미치는 영향)

  • Objective : The object of this study was to observe the effects of Lonicerae Flos (LF) aqueous extracts on lipopolysaccharide (LPS)-induced rat acute lung injury. Method : Five different dosages of LF extracts were orally administered once a day for 28 days before LPS treatments, and then all rats were sacrificed after 5 hour-treatment of LPS. Eight groups of 16 rats each were used in the present study. The following parameters caused by LPS treatment were observed ; body weights, lung weights, pulmonary transcapillary albumin transit, arterial gas parameters (pH, $PaO_2$ and $PaCO_2$) bronchoalveolar lavage fluid (BALF) protein lactate dehydrogenase (LDH), and proinflammatory cytokines tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$) contents, total cell numbers, neutrophil and alveolar macrophage ratios, lung malondialdehyde (MDA), myeloperoxidase (MPO), proinflammatory cytokines TNF-${\alpha}$ and IL-$1{\beta}$ contents. In addition, the histopathologic changes were observed in the lung in terms of luminal surface of alveolus, thickness of alveolar septum, number of polymorphonuclear neutrophils. Result : As results of LPS-injection, dramatical increases in lung weights, pulmonary transcapillary albumin transit increases, increases in $PaCO_2$, decreases in pH of arterial blood and $PaO_2$, increases of BALF protein, LDH, TNF-${\alpha}$ and IL-$1{\beta}$ contents, total cells, neutrophil and alveolar macrophage ratios, TNF-${\alpha}$ and IL-$1{\beta}$ contents increases were detected with decreases in LSA and increases of alveolar septum and PMNs numbers, respectively as compared with intact control. These are means that acute lung injuries (resembling acute respiratory distress syndrome) are induced by treatment of LPS mediated by inflammatory responses, oxidative stress and related lipid peroxidation in the present study. However, these LPS-induced acute lung injuries were inhibited by 28 days continuous pretreatment of 250 and 500mg/kg of LF extracts. Because of lower three dosages of LF treated groups, 31.25 and 62.5 and 125mg/kg did not showed any favorable effects as compared with LPS control, the effective dosages of LF in LPS-induced acute lung injuries in the present study, is considered as about 125mg/kg. The effects of 250mg/kg of LF extracts showed almost similar effects with ${\alpha}$-lipoic acid 60mg/kg in preventing LPS-induced acute lung injuries. Conclusion : It seems that LF play a role in protecting the acute respiratory distress syndrome caused by LPS.

Primary Signet Ring Cell Carcinoma of the Lung - Report of Two Cases - (폐에 발생한 원발성 인환세포암종 - 2예 보고 -)

  • Signet ring cell carcinoma is a variant of adenocarcinoma and has been rarely reported in the lung as a primary site. Recently, we experienced two cases of primary signet ring cell carcinoma in the lung without any other extrapulmonary lesion. Sputum cytology was performed and the tumor cells which have eccentrically located nuclei and abundnat mucinous cytoplasm were dispersed in diffuse sheets. On resected specimen, the signet ring cells occupied about $50{\sim}80%$ of all tumor cell nests. Histochemical staining revealed that the mucin produced by tumor cells was mostly carboxylated acid mucins. Ultrastructurally, the tumor cells contained variable sized membrane-bound mucin granules with weak central osmilophilic density and showed numerous surface microvilli, which represented that tumor cells arose from bronchial epithelial cells. In general, this tumor has diffusely infiltrative nature and the prognosis is fatal due to widespread metastasis before clinical discovery.

  • The cellular change of the pulmonary tuberculous lesions may be divided into two groups,exudative and proliferative form by their course and fate. In the most cases, the patients usually have very much complex type of cellular changes. Therefore, the shadows of the chest films in pulmonary tuberculosis are also much variable in nature. And Daniel said that knowledge of the pathology of tuberculosis and an appreciation of the method of progression and healing are essential to proper interpretation of the films. Author, having reviewed 33 cases of resected tuberculous lung obtained in N.M.T.H. for one year from Oct. '75 to Sep. '76 by surgical managements, classified the Pathological findings such as: 1) caseation only, 2) tuberculoma, 3) atelectatic lung 4) cavitary lesion and 5) atelectasis with cavity, and examined the relationship between the roentgenological characteristics of the chest films and the pathological process of tuberculous lesions of the resected lungs, The result were obtained as follows. (1) Tuberculoma was commonly appeared in $S_2$ segment in right and $S_6$ segment in left. (2) Atelectasis and destroyed lung were more commonly appeared in left lung than right, and their containing rate of cavity was 82%. (3) Cavities were mostly appeared in $S_1$ and $S_2$ segments of both lung and the appearance-rate of cavity on $S_6$ segment was higher in left than right. And among the cavitary lesions of the resected lung, cavity was not seen in the preoperative chest films in 22%. (4) The configuration, thickness and sharpness of the walls of cavities, which revealed the cavitary shadows in the preoperative chest films, were mostly depended on the degree of increased collagenous fiber of the wall, existence of perifocalitis, and more or less of the caseous masses on the inner surface of the cavity wall.

Automated Detection and Volume Calculation of Nodular Lung Cancer on CT Scans (CT 영상에서 결절성 폐암의 자동추출 및 체적계산)

  • This paper describes automated methods for the detection of lung nodules and their volume calculation on CT scans. Gray-level threshold methods were used to segment the thorax from the background and then the lung parenchymes from the thoracic wall and mediastinum. A scanning-ball algorithm was applied to more accurately delineate the lung boundaries, thereby incorporating peripheral nodules contiguous to pleural surface within the segmented lung parenchymes. The lesions which have the high gray value were extracted from the segmented lung parenchymes. The selected lesions include nodules, blood vessels and partial volume effects. The discriminating features such as size, solid-shape, average, standard deviation and correlation coefficient of selected lesions were used to distinguish true nodules from pseudo-lesions. Volume and circularity calculation were performed for each identified nodules. The identified nodules were sorted in descending order of the volume. These method were applied to 621 image slices of 19 cases. The sensitivity was 95% and there was no false-positive result.

Effects of Red-Koji Fermented Scutellariae Radix Extracts on Lipopolysaccharide-induced Rat Acute Lung Injury (홍국발효 황금이 Lipopolysaccharide 유발 급성 폐손상에 미치는 영향)

  • In the present study, the possibility of whether the pharmacological effects of Scutellariae Radix Aqueous Extracts(SR) were favorably changed by report that lipopolysaccharide(LPS)-induced rat acute lung injury was treated with Red-Koji(Monascus purpureus 12002) fermentation. Three different dosages of Red-Koji fermented SR extract(fSR), 125, 250 and 500 mg/kg were orally administered once a day for 28 days before LPS(Escherichia coli 0111:B4) treatments, and then 5 hours after LPS treatment(500 ${\mu}g$/head, intra trachea instillation), all rats were sacrificed. Changes in the body weights, lung weights, pulmonary transcapillary albumin transit, arterial gas parameters(pH, $PaO_2$ and $PaCO_2$) bronchoalveolar lavage fluid(BALF) protein, lactate dehydrogenase(LDH) and proinflammatory cytokine tumor necrosis factor-${\alpha}$(TNF-${\alpha}$), interleukin-$1{\beta}$(IL-$1{\beta}$) contents, total cell numbers, neutrophil and alveolar macrophage ratios, lung malondialdehyde(MDA), myeloperoxidase(MPO), proinflammatory cytokine TNF-${\alpha}$ and IL-$1{\beta}$ contents were observed with histopathology of the lung, changes on luminal surface of alveolus(LSA), thickness of alveolar septum, number of polymorphonuclear neutrophils(PMNs). As results of LPS-injection, dramatical increases in lung weights, pulmonary transcapillary albumin transit increases in $PaCO_2$, decreases in pH of arterial blood and $PaO_2$, increases of BALF protein, LDH, TNF-${\alpha}$ and IL-$1{\beta}$ contents, total cells, neutrophil and alveolar macrophage ratios, lung MDA, MPO, TNF-${\alpha}$ and IL-$1{\beta}$ contents increases were detected with decreases in LSA and increases of alveolar septum and PMNs numbers, respectively as compared with intact control. Especially fSR 125 mg/kg showed quite similar favorable effects on the LPS-induced acute lung injuries as compared with 60 mg/kg of ${\alpha}$-lipoic acid and 250 mg/kg of SR. The results suggest that over 125 mg/kg of fSR extracts showed favorable effects on the LPS-induced acute lung injury mediated by their antioxidant and anti-inflammatory effects. Moreover, increases of the pharmacological effects of SR on LPS-induced acute lung injury were observed by Red-Koji fermentation in this study, at least 2-fold higher.

Histopathological study of uremia in a juvenile shepherd dog (어린 shepherd 개에서 요독증의 조직 병리학적 연구)

  • A 7 month old male shepherd was presented with anorexia, diarrhea, and salivation, and euthanized due to no clinical improvement. Grossly, ulcers were seen on ventral surface of tongue and stomach. The mineralization was obvious in the intercostal space, lung and inner surface of aorta. Kidney was pale, firm and irregular. Histopathology confirmed uremic pneumonitis, end stage kidney and hyperplasia of parathyroid. This case was the typical uremic condition of juvenile dog, which was characterized the diffuse soft tissue mineralization due to hyperparathyroidism secondary to chronic renal failure.
