Effects of Atelectasis on Surface Activity of lung in Rabbits (무기폐(無氣肺)가 폐표면활성도(肺表面活性度)에 미치는 영향에 관(關)한 실험적(實驗的) 연구(硏究))

  • The effects of atelectasis on surface activity of lung extracts were examined in rabbits. Experimental atelectasis was produced in rabbits by artificial pneumothorax and surface tension properties were measured on saline extracts of lung 24 hrs, 48 hrs, one week and four weeks after the induction of pneumothorax. The results were as follows; 1) The minimum surface tension of excised lung extracts 24 hrs after pneumothorax was significantly increased to 28.3 0.41 dynes/cm, and the stability index was significantly decreased to 0.30 from normal value of 0. 87. 2) In the group which was re-expanded 24 hours, later from pneumothorax the surface activity was returned to almost normal range 24 hrs after reexpansion of collapsed lung, 3) When the atelectasis was continued by mechanical means, the extracts of atelectatic lung showed progressive decrease in surface activity, but it was found that surface activity returned to normal level after four weeks even the presence of atelectasis. 4) These observations suggest to us that atelectasis per se does not cause an increase in surface tension of lung extracts, and even in prolonged atelectasis the re-expansion of collapsed lung may be possible when the mechanical cause of atelectasis was excluded.

  • On the study of surface activity in excized lung extracts of various pulmonary diseases, following facts were concluded. 1]The minimum surface tension measured in lung extracts of tuberculous tissue surrounding cavitary lesion was 26.3dyne/cm and its stability index was 0.53. 2]Macroscopically almost normal lung tissue at a distance of tuberculous lesion in same lobe revealed 21.3 dyne/cm of minimum surface tension in extracts and its stability index showed 0.66. This low surface activity may be due to the chronic pneumonitis microscopically. 3] In the atelectatic lung which had been collapsed by chronic empyema the extracts revealed much higher minimum surface tension in 27.3 dyne/cm and its stabillry index revealed the least value of 0.47 without correlation of duration of disease. This suggests that the longstanding collapsed lung may be soon collapsed even after mechanical full expansion because of lack of surfactant.

  • In an attempt to clarity the effect of X-irradiation on the activity of surfactant in rabbits, and also to observe the possible difference when the irradiation was made in single or fractionated dose, X-ray in dose of 900 r was irradiated to rabbits either in single or fractionated dose of 300 r each day for three day, Tension-area diagram of lung extract was recorded automatically by a modified Langmuir-Wilhelmy balance with a synchronized recording system designed in this department. The surface tension of lung extract was measured at 1,3,5,24, and 48 hours post-irradiation, and the results were compared with the non-irradiated normal group. The result obtained are summerized as follows: 1) The maximal surface tension, minimal surface tension, width of the tension-area diagram at the surface area of 40% in lung extract, and stability index of the normal rabbit lung extracts were 40.73 dynes/cm, 8.96 dynes/cm. 20.71 dynes/cm, and 1.28 respectively. 2) Activity of surfactant was significantly depressed by X-irradiation, and the pattern of depression was more prominent in the single irradiation group than in the fractionated group. 3) It was found that the changes of the width of the tension-area diagram at the surface area of 40% in lung extract and stability indices corresponded well with that of the maximal or minimal surface tension.

  • The skin is related the lung as follows ; 1. The lung control skin and give a nutrition to skin. The skin and hair 'hab' (合) the lung. 2. As a damage of lung is a damage of skin, skin and hair 'chi-lag' (聚落) 3. A defensive energy of lung give a nutrition to skin, and strengthen the body surface. 4. Dryness damage the lung, and so damage the skin. According to the above findings, we know the body surface are closely related to the lung functionally.

  • The effect of reticuloendothelial hyperfunction on hypothermic preservation of lung was studied in dogs. In order to evaluate the viability after hemodynamic_ load in preserved isolated lung, observations were made on the rate of increase in weight, degree of edema,compliance and surface activity of lung. The results obtained as follows: l. In the group of activating of the reticuloendothelial system by injection of sodium thiosulfate intravenously before pneumonectomy and infusion of naphthionine through the pulmonary artery before hypothermic preservation of isolated lung the limit of preservation was eight hours whereas four hours in non-treated control group. 2.Therefore the method of activating of the reticuloendothelial system before and after pulmonary resection seems effective in preserving for prolonging the period of preservation of lung by means of inhibition of pulmonary edema. 3. Pulmonary surface activity is expected to be valuable as a method in evaluation of the viability of preserved lung along with compliance and rate of increase in weight of lung.

  • It is well known that mammalian alveolar membrane is covered with a very thin layer of surfactant film which characteristically reduces surface tension of alveolar membrane, and maintains alveolar stability. Since Clements in 1957 demonstrated that the surfactant is extractable by mincing the lung tissue in saline, various studies on the subject have been succeeded by many workers. However, the effect of radiation on the surfactant is not well clarified. Present study was attempted to observe the effect of x-irradiation on the activity of surfactant in rabbits. X-ray in dose of 300 r, 600 r or 900 r was irradiated to the chest of rabbits. The lung was removed from normal or irradiated rabbits sacrificed by arterial blood shedding, and lung-saline extract, adding 3 grams of lung tissue to 50 mili-liters of saline, was prepared by means of Vertis homogenizer. Tension-area diagram of lung extract was recorded automatically by a modified Langmuir-Wilhelmy balance with a synchronized recording system designed in this department. The surface tension of lung extract was measured at 1st, 2 nd, 3 rd, 7 th and 15 th post-radiation day in 300 r irradiated group, at 3 rd, 7 th and 15 th post-radiation day in 600 r irradiated group, 3 rd and 7 th post-radiation day in 900 r irradiated group respectively. For the histo-pathological study, lung tissue preparations were made in all irradiatiated groups on the day of experiment and in normal group. The results obtained are summarized as follows: 1. The minimal surface tension, maximal surface tension and stability index of normal rabbits lung extracts were 7.68 dynes/cm, 38.84 dynes/cm, and 1.39 respectively. 2. The activity of surfactant was depressed prominently by x-irradiation. However, increase in the dose of x-irradiation did not show any significant change in the degree of surfactant activity suppression. The most marked depression in surfactant was observed at the third post-radiation day in all irradiated groups. 3. Activity of surfactant depressed by x-irradiation showed a tendency of recovering to normal on 15 th post-radiation day. 4. The tendency of change in activity of surfactant following x-irradiation was somewhat correlative with histo-pathological changes. But the degree of depression of surfactant by x-irradiation did not correspond to the degree of histo-pathological changes, and recovery of lung tissue from radiation damage, tissue edema and congestion, seemed to be followed by restoration of surfactant activity. 5. The width of the tension-area diagram was measured at the surface area of 40% in lung extract of normal and x-irradiated rabbits. And it was found that the changes of the width corresponded well with that of minimum surface tension and of stability index in all normal and x-irradiated groups.

The result$ thus obtained are summarized as follows: I The maximal surface tension, minimal surface tension, width of the tension·area diagram at the surface area of 40% in the lung extract and stability index of the normal rabbit long extract were 40.73 dynes/cm, 8.96 dynes/cm, 20.71 dynes/cm and 1.28, respectively. II. When 900r of X-ray was irradiated to the lung in vitro, 1) The maximal and minimal surface tensions did not differ noticeably from the normal at 1,3, and 5 post-irradiation hours, but the minimal surface tension increased significantly at 24 and 48 hours Post-irradiation. 2) The width of the tension area at the surface area of 40% showed a tendency of decrease throughout the experiment. 3) The stability index showed no significant change at 1,3 and 5 post-irradiation hours,but at 24 and 48 hours post-irradiation a significant decrease was observed comparing with the control. III. Activity of surfactant was significantly depressed by X·irradiation in vitro especially at 24 and 48 hours post-irradiation.

  • The skin(皮膚) separates epidermis(皮) and dermis(膚) in Oriental medicine. epidermis and hair come from ectoderm as dermis comes from mesoderm in embryology. Epidermis and hair belong to the Metal(金) and dermis belongs to the Earth(土) in Oriental medicine. The lung is related to the skin and hair. The skin surface is nourished by the essence from the lung, and the. lung has the function of sending wei-qi(衛氣.: defensive Energy) to the skin surface to guarantee body resistance. The skin takes nourishment from blood(血) and express by qi(氣). The supply of nutrients to the lung and skin relies on the function of the spleen, pancreas and subcutis. Flaming of the heart fire may scorch the essence of the lung-metal, inducing dermatosis.

  • In this research, we developed a biosensor to detect lung cancer-specific biomarker using Anodic Aluminum Oxide (AAO) chip based on interference and nano surface plasmon resonance (nanoSPR). The nano-porous AAO chip was fabricated $2{\mu}m$ of pore-depth by two-step anodizing method for surface uniformity. NanoSPR has sensitivity to the refractive index (RI) of the surrounding medium and also provides simple and label-free detection when specific antibodies are immobilized to the Au-deposited surface of nano-porous AAO chip. To detect the lung cancer-specific biomarker, antibodies were immobilized on the surface of the chip by Self Assembled Monolayer (SAM) method. Since then lung cancer-specific biomarker was applied atop the antibodies immobilized layer. The specific reaction of the antigen-antibody contributed to the change in the refractive index that cause shift of resonance spectrum in the interference pattern. The Limit of Detection (LOD) was 1 fg/ml by using our nano-porous AAO biosensor chip.

Effects of Hantaan Virus and $IFN-{\gamma}$ on Induction of Surface ICAM-1 in Primary Cultured Human Nasal Epithelial Cells and Human Lung Fibroblasts

  • The primary culture of human nasal epithelial cells was performed using the inferior nasal turbinate tissues, and infected with Hantaan virus to examine the hypothesis of airborne transmission of Hantaan virus in humans. The primary culture cells were identified as epithelial cells by morphologic and immunologic analyses. The viral antigens were detected in the primary human nasal epithelial cells infected with Hantaan virus by immunofluorescence staining. The ICAM-1 induction by Hantaan virus or $IFN-{\gamma}$ was examined in the primary human nasal epithelial cells and human lung fibroblasts (WI-38). Hantaan virus induced the surface ICAM-1 in WI-38 cells in a time-dependent manner, and $IFN-{\gamma}$ induced the surface ICAM-1 in a dose-dependent manner in HNEC and WI-38 cells. These results revealed that the human nasal epithelial cells are susceptible to Hantaan viral infection supporting the hypothesis of airborne transmission of Hantaan virus in humans. The human lung fibroblasts also might have an important role in the pathogenesis of Hantaan virus through the induction of ICAM-1.
