Bombesin-based Radiopharmaceuticals for Imaging and Therapy of Cancers Expressing Gastrin-releasing Peptide Receptor

  • Bombesin has a high binding affinity to gastrin-releasing peptide receptor (GRPR) and can be used as a targeting ligand in GRPR-related cancers. Because GRPR is overexpressed in prostate cancer, bombesin analogues have been investigated extensively for diagnosis and treatment of prostate cancer. In nuclear medicine, bombesin derivatives labeled with radiometals such as 55/57Co, 64Cu, 68Ga, 99mTc, and 177Lu or radiohalogen such as 131I and 211At were developed as markers for early detection of tumors and theragnostic tool for cancer treatment. This review focuses on the introduction of bombesin-based radiopharmaceuticals that are studied in pre-clinical or clinical research.

감마선 및 엑스선의 피이크 분석 (Peak Analysis of Gamma-ray and X-ray)

  • 핵분열(核分裂) 및 방사화생성물(放射化生成)物)에서 방출(放出)하는 여러가지 감마선들은 핵연료(核燃料)를 파괴(破壞)하지 않고도 추출(抽出)할 수 있는 많은 정보(情報)를 포함(包含)한다. 그러나 반도체(半導體) 검출기(檢出器)에서 얻은 복잡한 스펙트럼에서 이러한 정보를 추출(抽出)하기가 용역(容易)하지 않기 때문에 전산(電算)코드의 사용(使用)이 필요(必要)하게 된다. 본(本) 연구(硏究)에서는 그동안 국제적(國際的)으로 널리 보급(普及)되어 사용(使用)되는 감마선 분석(分析) 프로그램들의 장점(長點)을 취하여, 감마선은 물론 X-선(線)의 스펙트럼도 피팅 (fitting)하여 피이크의 중심과 면적을 정확(正確)히 계산(計算)할 수 있는 전산코드 CAERI를 개발(開發)하였다. CAERI는 FORTRAN으로 쓰여있고, 특히 고유복사선폭(固有輻射線幅)(natural line width) 을 무시할 수 없는 X-선(線)의 피이크 표현함수(表現函數)인 Voigt 함수(函數)에 대해서는, 다른 X-선(線) 분석(分析) 프로그램들이 사용(使用)한 간단한 근사식(近似式) 대신에, 더욱 정확(正確)한 무한급수근사식(無限級數近似式)을 사용하였다. 특히 CAERI 는 U이나 Pu과 같은 중원소(重元素)의 핵종분석시(核種分析時)에 직면(直面)하는 감마선과 U이나 Pu의 X-선(線)이 임의(任意)로 간섭(干涉)하며 공존(共存)하는 복잡한 스펙트럼까지도 취급(取扱)할 수 있다. $^{177m}Lu$감마선과 $^{235}U\;U_{\alpha}$X-선(線)의 시험(試驗)스펙트럼을 피팅하여, 다른 프로그램들의 피팅결과와 비교했을 때 좋은 일치(一致)를 보았다.

치료용 방사성동위원소 사용 시 단일 세포의 핵 크기 변화에 따른 흡수선량 분석 (Analysis of Absorbed Dose on the Nucleus Size Change of Single Cells using Therapeutic Radioisotopes)

  • 표적 방사성핵종 치료(targeted radionuclide therapy, TRT)는 방사성의약품을 사용하여 종양세포를 치료하는 방법이다. 인체의 조직을 구성하는 세포와 핵은 구형과 타원형으로 구성되어 있지만, 암세포의 경우 다양한 형태의 세포 형태로 이루어져 있다. 이에 본 연구는 몬테카를로 방법을 통해 표적 방사성핵종 치료 시 사용되는 베타선 방출 핵종을 대상으로 단일 세포 내 세포핵의 크기 변화에 따른 세포 구성 영역별 흡수선량을 분석하였다. 세포는 5 ㎛, 10 ㎛ 두 가지의 크기의 구 형태로 선정하였으며, 세포의 내부 구성은 세포핵, 세포질, 세포 표면으로 구분하였으며, 세포핵의 크기 증가에 따른 흡수선량을 평가하였다. 그 결과, 표적 방사성핵종 중 177Lu이 세포 모든 구획에서 가장 높은 선량을 나타냈으며, 세포 내 핵의 비중이 증가함에 따라 세포 표면의 흡수선량은 증가되었으나, 세포질과 세포핵의 흡수선량은 감소하는 경향을 보였다. 따라서 표적 방사성핵종 치료 시 암세포의 크기를 고려한 방사성핵종의 선택과 적절한 방사능량 결정이 중요할 것으로 판단된다.

Simple measurement the chelator number of antibody conjugates by MALDI-TOF MS

  • Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS) is one of the powerful methods that enable analysis of small molecules as well as large molecules up to about 500,000 Da without severe fragmentation. MALDI-TOF MS, thus, has been a very useful an analytical tool for the confirmation of synthetic molecules, probing PTMs, and identifying structures of a given protein. In recent nuclear medicine, MALDI-TOF MS liner ion mode helps researcher calculate the average number of chelator(or linkage) per an antibody conjugate, such as DOTA-(or DFO-) trastuzumab for labeling a medical radioisotope. This simple technique can be utilized to improve the labeling method and control the quality at the development of antibody-based radiopharmaceuticals, which is very effected to diagnosis and therapy for in vivo tumor cells, with radioisotopes like $^{89}Zr$, $^{64}Cu$, and 177Lu. To minimize the error, MALDI-TOF MS measurement is repeatedly performed for each sample in this study, and external calibration is carried out after data collection.

외식서비스에 대한 소비가치가 소비자만족에 미치는 영향: 중국 프랜차이즈 레스토랑 소비자경험을 중심으로 (Effect of Consumption Value of Restaurant Service on Consumer Satisfaction: Focusing on Consumer Experiences at a Chinese Franchise Restaurant)

  • This study examined consumer experiences on consumption value for the restaurant service and the effect of consumption value on consumer satisfaction. Consumption value was composed of four values: functional, social, emotional, and epistemic. Participants consisted of 407 consumers who live in Shanghai and have experience eating at Haidilao restaurant. The results of this study suggested that the perceived consumption value of the restaurant service varies by the socio-demographic backgrounds of consumers. Consumer satisfaction is affected by the epistemic, emotional, and functional values of the restaurant service. The effect of epistemic value on consumer satisfaction with restaurant service was the highest among all consumption values followed by emotional and functional values. There was no significant effect of social value on consumer satisfaction. This study showed that consumers pursue various values in the restaurant services which had a greater influence on consumer satisfaction than the functional value related to the primary purpose of restaurant services. As the consumer experience diversifies along with the globalized food service industry, it is necessary to improve the consumer satisfaction of restaurant services by investigating what values and experiences consumers pursue. The results of this study contribute to a consumer-oriented service design of restaurants by illuminating consumers' experiences with consumption values that influence consumers' satisfaction with the overall restaurant service.

몬테카를로 방법을 이용한 치료용 방사성동위원소 사용 시 단일 세포에 대한 선량 분석 (Analysis of Radiation Dose on Single Cells Using Therapeutic Radioisotopes Using the Monte Carlo Method)

  • Targeted radionuclides treatment (TRT) requires the establishment of treatment plans that consider various factors, such as the type of radionuclides, target organs, and administration methods. For this reason, in this study, the absorption dose of a single cell was analyzed according to the type of radioisotope used to treat target radionuclides. In this study, a simulation was performed on beta rays used in the treatment of target radionuclides at the cell level using MCNPX (ver. 2.5.0). First, according to the calculation formula, the beam path according to the type of radioisotope for treatment was calculated. Second, the amount of self-radiation by beta rays emitted from cell diameters of 5 ㎛ and 10 ㎛ cell nuclei was evaluated. As a result, it showed a high range proportional to the maximum energy of the beta-ray, and the highest self-dose distribution from 177 Lu radiation sources among therapeutic radioisotopes. This was analyzed as a result that is inversely proportional to the maximum energy of the beta-ray, and it suggests that the selection of a nuclide considering the range of the beta-ray is necessary in the treatment of target radionuclides in the future.

Comparative in vivo biodistributions of nanoparticles and polymers of 177lutetium-labeled hyaluronic acids in mice during 28 days

  • Hyaluronic acid (HA) has been investigated for biomedical and pharmaceutical applications. This study was conducted to determine the distributions of HA nanoparticles (NPs; size 350-400 nm) and larger HA polymers in mice at intervals after application. $^{177}Lutetium$ (Lu)-labeled HA-NPs or HA polymers were intravenously injected (5 mg/kg) into male ICR mice, and radioactivity levels in blood and target organs were measured from 0.25 h to 28 days post-injection. In blood, the radioactivities of HA-NPs and HA polymer peaked at 0.5 h after injection but were remarkably decreased at 2 h; subsequently, they maintained a constant level until 6 days post-injection. HA-NPs and HA polymers were observed in the liver, spleen, lung, kidney, and heart (in ascending order) but were seldom observed in other organs. After 3 days, both the HA-NP and HA polymer levels showed similar steady decreases in lung, kidney, and heart. However, in liver and spleen, the HA-NP levels tended to decrease gradually after 1 day and both were very low after 14 days, whereas the HA polymer level accumulated for 28 days. The results indicate that HA-NPs, with their faster clearance pattern, may act as a better drug delivery system than HA polymers, especially in the liver and spleen.

Chemical and Textural Properties in Commercial Fermented Soybean Curds of Sufu

  • A survey aiming to find out the chemical and textural properties of commercial fermented soy bean curd called sufu was conducted. Sixteen brands of plain sufu produced in the Northern or the Southern part of China were collected and examined for their crude protein, crude fat, texture profiles, free amino acids, and free fatty acid contents. Twenty-one free amino acids were extracted and derivatized using a commercial kit followed by separation and analyzed by the gas chromatography-mass spectrometry (GC-MS). Similarly, ten free fatty acids were extracted using alumina, eluted, separated and analyzed. The content ranges of crude fat and protein were 22~36% and 31~38%, respectively. In texture profile analysis, ranges of the texture parameters were 131~493 g (hardness), 0.4~0.5 (cohesiveness), -137 to -50 gs (adhesiveness), 0.6~1 (springiness), 47~220 g (gumminess) and 32~177 g (chewiness). Twenty-one different free amino acids, especially alanine, glycine, $\alpha$-aminobutyric acid, valine, leucine, allo-isoleucine, aspartic acid, glutamic acid and lysine in large amount, as well as ten fatty acids in total, notably linoleic acid (9-octadecanoic acid), oleic acid (9,12-octadecadienoic acid), linolenic acid (9,12,15-octadecadienoic acid), hexadecanoic acid and octadecanoic acid were found. This information provides important quality reference ranges for product developers and manufacturers to optimize and produce the plain sufu.

치매(痴?)의 침구치료(鍼灸治療)에 대(對)한 문헌적(文獻的) 고찰(考察) (The Philological Study on Pathology, Acupuncture and Moxibustion of Dementia)

  • Objectives : This study is designed to investigate the pathology, acupuncture and moxibustion therapy applicable of the dementia through literature of oriental medicine. Results : The findings of this study are as follows ; 1. The cause of dementia is internal damage of seven emotions(七情內傷), deficiency of the kidney essence(腎精不足), retention of dampness in the mental(痰迷心竅). 2. The acupuncture meridians used often for dementia are $Tok-maek(GV),\;Chok-t'aeyang\;Pang-gwang-Ky\check{o}ng(BL),\;Chok-yangm\check{o}yng\;Wi-Ky\check{o}ng(ST),\;Su-gwor\check{u}m\;Shimp'o-Ky\check{o}ng(PC),\;Su-so\check{u}m\;Shim-Ky\check{o}ng(HT)$. 3. The spots on the body suiTable for acupuncture used for therapy of the dementia are 98 points and extra points are 6 points, total therapeutic acupunture points are 104 points. And acupunture points are frequently used Shinmun(神門-HT7, 24), Paek'oe(百會-GV20, 19), Chok-samni(足三里-ST36, 18), Sugu(水溝-GV26, 17), P'ungnyung(豊隆-ST40, 16), Taech'u(大椎-GV14, 14), $Sam\check{u}mgyo(三陰交-SP6,\;14)$, Shimsu(心兪-BL15, 14), $Taen\check{u}ng(大陵-PC7,\;11)$, Nogung(勞宮-PC8, 11), Sosang(少商-LU11, 10), Kansa(間使-PC5, 10), Naegwan(內關-PC6, 10). 4. The acupoints used often on moxibustion are Paek'oe(百會-GV20), Chang-gang(長强-GV1), Taech'u(大椎-GV14), Kansa(間使-PC5). Conclusion : It is recommended that further study of many sided investigations in the future.
