Effects of Dance Sports on Body Composition, Serum Lipids and Leptin in the Middle-Aged Women (댄스 스포츠가 중년여성의 혈중지질 및 렙틴에 미치는 영향)

  • The purpose of this study was to examine the effects of Dance Sports on body composition, blood level of lipids and leptin in the middle-aged women. The 24 subjects for this study were divided into three groups by the % fat classification non-obese control group (n=8, CG), obese group with Dance Sports (n=8, OG), and normal group with Dance Sports (n=8, NG). They performed Dance Sports with 60∼80% of HRmax, for 60min a day and three times per week during 12 weeks. The body composition, blood levels of total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride and leptin were examined before and after Dance Sports. The percent body fat was decreased in OG and NG significantly. The blood levels of HDL-cholesterol were increased in OG and NC after the Dance Sports. The blood levels of triglyceride was significantly decreased in the same groups. Other serum lipids were not changed significantly. The blood levels of leptin were decreased in OG and NC after the Dance Sports. But the lowering effect of Dance Sports in the obese group (OG) was not improved compared with that in the normal group (NG), suggesting that Dance Sports on the obese group (OG) has no further improving effect in the change of blood lipid and leptin. These results suggest that Dance Sports has a beneficial effect on the change of blood levels of HDL-cholesterol and leptin.

The Efficacy of Lowering Blood Glucose Levels Using the Extracts of Fermented Bitter Melon in the Diabetic Mice (당뇨 마우스에서 여주발효추출물의 혈당 강하 효능)

  • Momordica charantia, commonly known as bitter melon, has interesting pharmacological activities such as anticancer, antiviral, antibacterial, anti-inflammatory, analgesic, and antioxidant. As supported by recent scientific reports on the beneficial effects of M. charantia, it is one of the most promising functional plants for diabetes today. In this study, we fermented the bitter melon with lactic acid bacteria and investigated the capability of controlling diabetic conditions by decreasing the blood glucose levels. After extracting the fermented bitter melon with hot water or ethanol, we tested several biological activities using mouse models. When we tested the efficacy of the glycemic control, the extracts of fermented bitter melon significantly lowered the blood glucose levels of the alloxan-induced diabetic mice. We also found that the lactic acid bacteria-fermented bitter melon protected liver damages from the treatment of alloxan monohydrates and maintained low levels of triglycerides and high levels of HDL cholesterol in these mouse models. These results suggest that our approach on fermenting bitter melon and the extracts of fermented bitter melon could lead to the possibility of the development of functional foods that contain the effectiveness of controlling blood glucose and lipid levels as well as preventing liver damages.

Effects of Lactobacillus plantarum CIB 001 on Lipid Metabolism of Hypercholesterolemic Rats (김치에서 분리한 Lactobacillus plantarum CIB 001 급여가 고콜레스테롤혈증 흰쥐의 지질대사에 미치는 영향)

  • The aim of the present study was to assess the anti-hypercholesterolemic effect of bile salt hydrolase-producing Lactobacillus plantarum CIB 001 (KCTC 11717 bp) in rats fed a high-cholesterol diet. Four treatment groups of rats (n=5) were fed experimental diets: a normal diet (ND), a ND plus L. plantarum CIB 001(NDL) at $5.0-7.5{\times}10^9$ colony forming unit (CFU)/day, a high-cholesterol diet (HCD), as well as a HCD plus L. plantarum CIB 001 (HCDL) at $5.0-7.5{\times}10^9$ CFU/day for 6 weeks. Compared with the HCD group, the HCDL group demonstrated a decrease in serum triglyceride (p<0.05), total cholesterol (p<0.05), and the corresponding HDL-cholesterol concentration increased at a rate of 40% (p<0.05). The HCDL group also induced a decrease in liver inflammation and steatosis. The present results suggest that supplementation of L. plantarum CIB 001 can have short-term (6 weeks) effects on blood lipids and liver injury, as well as on the atherogenic index and cardiac risk factors.

Bioequivalence Evaluation of Lovastatin Tablets (로바스타틴 정제의 생물학적 동등성 평가)

  • Lovastatin is a lipid lowering agent for the treatment of hypercholesterolemia and belongs to a new class of pharmacologic compounds called the 3-hydroxy-3-methylglutaryl coenzyme A (HMG CoA) reductase inhibitors. By competitively inhibiting HMG CoA reductase, lovastatin disrupts the biosynthesis of cholesterol in hepatic and peripheral cells and increases the synthesis of high-density-lipoprotein HDL) receptors. Following oral administration, the lactone ring of lovastatin is hydrolysed to the active inhibitor of HMG CoA reductase, lovastatin acid. Lovastatin is known to have poor oral absorption and wide individual variation. In this study, bioequivalence test of two lovastatin formulations, the test drug ($Lovaload^{TM}$, Chong Kun Dang Pharmaceutical Co.) and the reference drug ($Mevacor^{TM}$, Chung Wae Pharmaceutical Co.) were conducted according to the guidelines of Korea Food and Drug Administration (KFDA). A total of 18 healthy male volunteers, $31.90\pm3.60$ years old and $72.17\;7.88$ kg of body weight in average, were evaluated in a randomized crossover manner with a 2-week washout period. Concentrations of lovastatin acid in plasma were measured upto 12 hours following a single oral administration of eight tablets (20 mg of lovastatin per tablet) by high-performance liquid chromatography with UV detection at 238 nm. The area under the concentration-vs-time curve from 0 to 12 hours $(AUC_{0-12h})$ was calculated by the trapezoidal summation method. The statistical analysis showed that there are no significant differences in $AUC_{0-12h),\;C_{max}\;and\;T_{max}$ between the two formulations ($6.72\%,\;1.52\%,\;and\;0.88\$, respectively). The least significant differences between the formulations at $\alpha$=0.05 were less than $20\%\;(11.65\%,\;19.73\%,\;and\;14.81\%\;for\;AUC_{0-12h},\;C_{max}\;and\;T_{max}$, respectively). The $90\%$ confidence intervals for these parameters were also within $\pm20\%\;(-1.50{\leq}{\delta}{\leq}15.00$, $-12.50{\leq}{\delta}{\leq}15.50,\;and\;-9.64{\leq}{\delta]{\leq}11.40{\leq}\;for\;\;AUC_{0-12h}$ ,$C_{max}\;and\;T_{max}$, respectively). In conclusion, the new generic product $Lovaload^{TM}$ was proven to be bioequivalent with the reference drug.

Effect of In Vitro Antioxidant Properties and Extract of Corn Husk on Serum Lipids in Mice (옥수수 껍질 분말의 In Vitro 에서의 항산화 특성과 고지방식이로 유도된 Mice의 혈청지질 수준에 미치는 영향)

  • This study was performed to verify the effects of corn husk powder on serum lipid levels of high-fat diet-induced mice from a nutritional viewpoint. Powder of corn husks has been used as a folk remedy from ancient times in Korea. In the case of corn husk measurement of TPC (p<0.05), ABTS+ radical scavenging activity of trolox (p<0.05), DPPH radical scavenging activity (p<0.05), and measured value of FRAP (p<0.0001) were higher in ethanol extract than water extract. Weight of livers was significantly higher in the control group and high-fat diet group compared to groups fed 10% and 20% corn husk powders with high-fat diet (p<0.0001). For blood HDL-cholesterol concentration, mice fed 10% corn husk powder with high-fat diet ($160.00{\pm}16.17mg/dL$) showed significantly higher levels compared with the other three groups (p<0.05). Mice fed 10% and 20% corn husk powders with high-fat diet showed high numerical values of $13.50{\pm}2.51$ and $11.25{\pm}2.87mg/dL$, respectively. For triglyceride concentration in blood, mice fed 10% ($83.50{\pm}19.30mg/dL$) and 20% ($88.50{\pm}22.25$) corn husk powder with high-fat diet showed very low levels compared to the control group and high-fat diet group. In the case of leptin, the high-fat diet group showed a significantly high level of $5.88{\pm}3.53ng/dL$. Groups fed 10% and 20% corn husks with high-fat diet showed low numerical values of $2.72{\pm}2.34ng/dL$ and $1.10{\pm}0.62ng/dL$, respectively (p<0.05). Corn husk powder increased high HDL-cholesterol concentration in blood while lowering triglyceride concentration and leptin secretion. Therefore, powder of corn husks has utility in the food industry and food processing.

Improvement of High-fat Diet-induced Obesity by Xanthigen in C57BL/6N Mice (잔티젠(Xanthigen)에 의한 고지방식이로 유도된 비만 개선 효과)

  • Obesity is a risk factor for numerous metabolic diseases. Recently, naturally occurring compounds that may improve obesity have received increasing attention. Xanthigen is a mixture of fucoxanthin and punicic acid derived from brown seaweed and pomegranate seed, respectively, which have been traditionally used for lipid-lowering effects in humans. In this study, we investigated whether Xanthigen attenuates high-fat diet-induced obesity in C57BL/6N mice. The mice were fed on a normal diet (ND), high-fat diet (HFD), HFD plus 1% Xanthigen or HFD plus 1% green tea extract (GTE) for 11 weeks. Food efficiency ratio (FER) and body weight were significantly reduced in mice fed HFD plus Xanthigen compared to HFD-fed mice. Consistent with the results in body weight change, Xanthigen also significantly decreased the weights of epididymal adipose tissue, retroperitoneal adipose tissue, and liver in HFD plus 1% Xanthigen-fed mice. The serum level of low-density lipoprotein (LDL)-cholesterol was significantly decreased in HFD plus Xanthigen-fed mice compared to HFD-fed mice. These results suggest that Xanthigen may be useful in the development of a functional health food for anti-obesity.

Effects of ${\beta}-glucan-enriched$ Barley Fraction on the Lipid and Cholesterol Contents of Plasma and Feces in Rat (보리 ${\beta}-glucan$ 농축획분 섭취가 흰쥐 혈장과 변의 지질 및 콜레스테롤 함량에 미치는 영향)

  • Cholesterol-lowering effects of ${\beta}-glucan-enriched$ barley fraction were investigated in rats fed 0.5% cholesterol and barley as a fiber source. Male rats of Sprague-Dawley strain were divided into six groups and fed different diets for 5 weeks: normal (cholesterol-free), control (cellulose 5%), fiber-free, and three groups containing ${\beta}-glucan-enriched$ barley fractions. Although plasma cholesterol and triglyceride levels were not significantly different among different diet groups, ${\beta}-glucan-enriched$ barley fraction-fed groups had higher HDL-cholesterol concentrations than the cellulose control group after 5 weeks of experiment. The fecal excretion of cholesterol and triglyceride was significantly increased by barley diets. Fecal concentrations of cholesterol and triglyceride in cellulose control group were 38.2 and 2.6 g/day, respectively, whereas those of barley-fed groups were $52.4{\sim}59.2$ and $6.8{\sim}9.5$ g/day, respectively, during the experimental period.

A Case Report on Korean Medical Treatment of a Patient with Abnormal Liver Function Induced by Rovazet (로바젯정으로 유발된 간수치 상승 환자의 한방 치료 증례 보고)

  • Objective: This study evaluated a clinical case of a patient with Rovazet-induced abnormal liver function treated with intradermal acupuncture. Method: A 59-year-old woman with abnormal liver function induced by Rovazet ceased taking the drug and instead received intradermal acupuncture treatment. After 15 days, clinical symptoms and laboratory test results were evaluated. Results: Aspartate aminotransferase and alanine aminotransferase levels decreased, and clinical symptoms such as nausea and fatigue improved after treatment with intradermal acupuncture and cessation of the Rovazet. Conclusion: This study meaningfully examined a clinical case of a patient with abnormal liver function induced by a lipid-lowering agent, which is uncommon in Korea. Unfortunately, the lack of clear evidence and the use of only one case are limitations of the study.

Establishment of Winterizing Conditions and Analysis of Component Composition of Winterizing By-product in Corn Oil (옥수수기름의 탈납조건 확립 및 탈납부산물의 성분조성 분석)

  • Optimal winterizing condition of com oil was cooling temperature of bleached oil from $105^{\circ}C$ to $40^{\circ}C$. Then, filtering it after keeping $1{\sim}2^{\circ}C$ by lowering the temperature gradually with treating perlite of 0.3%(w/w) amount about bleached oil and stirring. We could measure that triglyceride(TG) that extracted from lipid components from spent perlite(SP) obtained through filtration after winterizing by SACC method is major causing materials of clouding in com oil. The result of separating TG fraction by agentation TLC was that it classified into 4 kinds -U3, SU2, S2U, S3 type and the most were U3 type. From this, it's easy to identify cause of clouding in com oil is TG fraction and most of them form wax materials that can observed. The results were they kept clear appearance at $0^{\circ}C$ generally during 39.6 to 96.5 hours, especially the result of A sample that had the lowest temperature condition while they have some difference by condition of treating temperature.

The Effects of Seaweed Gongjindan on Estrogen like Activities, Platelet Aggregation and Serum Lipid Levels in Ovariectomized Rats (해조공진단의 에스트로겐 유사활성 및 난소를 절제한 흰쥐의 혈소판 응집과 혈청 내 지질 조성에 미치는 영향)

  • Estrogen like activities were evaluated using ethanol and hot water extracts of gongjindan (GJD) and 3 types of brown algae by an in vitro detection system. Laminaria japonica and Undaria pinnatifida represented statistically significant (p<0.001) estrogen like activities, while GJD and Ecklonia stolonifera did not. Furthermore, activities of hot water extracts ($500{\mu}g/ml$) of Laminaria japonica and three mixture (Ecklonia stolonifera : Undaria pinnatifida : Laminaria japonica, 1: 1: 1) were stronger than that of $10^{-7}M$ $17{\beta}$-estradiol. These results suggest that Laminaria japonica contain estrogen like compounds. Based on these results, the prepared seaweed GJD were used for animal experiments about the effects on inhibition of platelet aggregation and serum lipid levels in ovariectomized rats. 9-week Old female SD-rats were randomly assigned to 3 groups as sham-operated rats, ovariectomized rats and ovariectomized rats that were treated with GJD. The rats were placed on prescribed diets for 5 weeks following ovariectomy. Total cholesterol and triglyceride contents on serum decreased in the SHAM group compared to the OVX-CON group. 5 weeks feeding of GJD resulted in significant lowering of triglyceride and a decreasing tendency of total cholesterol level. The level of HDL-cholesterol on serum increased significantly by feeding diets containing the GJD. The ability of platelet aggregation of group treated with GJD was less than that of the OVX-CON group. These results suggest that the beneficial effects of seaweed gongjindan may be used to improve menopausal syndrome.
