Antiobesity Effect of Major Korean Spices (Red Pepper Powder, Garlic and Ginger) in Rats Fed High Fat Diet

  • Commonly used spices in Korean cooking (red pepper powder, garlic, ginger) were evaluated for anti-obesity properties and effects on triglyceride (TG) and cholesterol in blood and adipose tissues in rats fed a high fat (20%) diet. SD rats were raised for four weeks on either a normal diet (ND, based on the AIN-93M diet), high fat diet (HFD, supplemented with 16% lard oil in the ND), or diets containing 5% red pepper powder (RPP), garlic or ginger. These spices significantly decreased weight gain compared to HFD, but garlic and ginger showed a greater effect on reducing weight gain than RPP. The weights of liver and epididymal and perirenal fat pads in garlic and ginger diet groups were lower than those of the HFD groups (p < 0.05). The garlic and ginger also decreased triglyceride and cholesterol contents in liver and epididymal and perirenal fat pad, reversing the higher levels seen in HFD. RPP, garlic and ginger supplemented diets were effective in lowering serum triglyceride and cholesterol levels (p< 0.05). These results indicated that garlic and ginger more effectively suppressed the effects of HFD on body fat gain and lipid values of adipose tissues and serum than RPP.

Changes of Plasma and Hepatic Lipids, Hydroxy-Methyl-Glutaryl CoA Reductase Activity and Acyl-CoA : Cholesterol Acyltransferase Activity by Supplementation of Hot Water Extracts from Rosa rugosa, Crataegus pinnatifida and Polygonum cuspidatum in High-Cholesterol Fed Rats

  • Lipid lowering properties from three plant water extracts, Rosa rugosa, Crataegus pinnatifida and Polygonum cuspidatum, were tested by supplementing a 1% high-cholesterol diet with them in rats. Plasma triglyceride levels in Rosa fugosa, Crataegus pinnatifida and Polygonum cuspidatum groups were significantly lower compared to that of the control. by 29% , 24% and 47% respectively. hepatic trigylceride levels in Rosa rugosa and Crataegus pinnatifida groups were significantly lower compared to the control by 11% and 15% respectively. Hepatic HMG-CoA reductase activity in Rosa rugosa group was significantly greater compared to the control by 406%. Hepatic ACAT activity was significantly lower in Polygonum cuspidatum group compared to the control by 28%. by multiple regression results, only plasma cholesterol was associated significantly (p<0.05) with liver HMG-CoA reductase activity. Plasma cholesterol explained 12% of thevariance of the liver HMG-CoA redctase activity. In conclusion, we have showen that hot water extracts from Rosa rugosa, Crataegus pinnatifida and Polygonum cuspidatum lowered plasma triglycerides in rats fed on a high-cholesterol diet. Data suggests that these extracts could potentially prevent or treat hypertriglyceridemia induced by a high fat diet and fatty liver.

Lipid lowering activity of Ponciri Fructus and Aurantii Fructus Immaturus on hyperlipemia rats induced by Triton WR-1339. (국산과 중국산 지실(枳實)의 Triton WR-1339 유발 고지혈증(高脂血症) 흰쥐에 대한 효능 연구)

  • Objective : In order to elucidate hyperlipidemic effects of the fruits of Poncirus trifoliata and Citrus aurantium, our experimental study was performed on hyperlipidemia rats induced by Triton WR-1339. Method : The hyperlipidemia rats induced by intraperitonial injection of triton WR-1339 were treated with the fruits of P. trifoliata (rare and preparata) and C. aurantium 50 and 200mg/kg. The amount of triglyceride. total cholesterol. HDL-cholesterol, LDL-cholesterol, AST. and ALT levels were measured. Results: The levels of triglyceride. total cholesterol and LDL-cholesterol were significantly reduced in fruits of P. trifoliata (rare and preparata) and C. aurantiu. Simvastatine treated group compared with those of control but the level of HDL-cholesterol was incresed. The level of AST and ALT were not significantly reduced by the treatment to the fruits of P. trifoliata (rare and preparata) and C. aurantium. Conclusion : In the hyperlipidemia rats induced by triton WR-1339. the fruits of P. trifoliata (rare and preparata) and C. aurantium showed a hyperlipidemic effect.

The Factors Affecting Recurrence of Symptoms after Infrainguinal Arterial Endovascular Angioplasty

  • Background: This study reports the result of endovascular treatment for arterial occlusive disease limited to femoropopliteal lesions, focusing on the recurrence of symptoms instead of patency. Methods: This was a retrospective, single-center study. From April 2007 to November 2011, 48 limbs in 38 patients underwent endovascular stenting or balloon angioplasty to treat femoropopliteal arterial occlusive disease. The factors affecting the recurrence of symptoms were analyzed. Results: The mean age of the patients was $69.60{\pm}7.62$ years. Among the baseline characteristics of the patients, initial hyperlipidemia was the most important factor affecting the recurrence of symptoms (relative risk=5.810, p=0.031). The presence of a dorsal arch was also a significant factor (relative risk=0.675, p=0.047). Conclusion: The major factors that affect the recurrence of symptoms after endovascular treatment for femoropopliteal arterial occlusive lesions are hyperlipidemia and the presence of a dorsal arch. Therefore, the usage of lipid-lowering agents after endovascular treatment and taking the presence of a dorsal arch into consideration are important elements of managing the recurrence of symptoms.

Shigella flexneri Inhibits Intestinal Inflammation by Modulation of Host Sphingosine-1-Phosphate in Mice

  • Infection with invasive Shigella species results in intestinal inflammation in humans but no symptoms in adult mice. To investigate why adult mice are resistant to invasive shigellae, 6~8-week-old mice were infected orally with S. flexneri 5a. Shigellae successfully colonized the small and large intestines. Mild cell death was seen but no inflammation. The infected bacteria were cleared 24 hours later. Microarray analysis of infected intestinal tissue showed that several genes that are involved with the sphingosine-1-phosphate (S1P) signaling pathway, a lipid mediator which mediates immune responses, were altered significantly. Shigella infection of a human intestinal cell line modulated host S1P-related genes to reduce S1P levels. In addition, co-administration of S1P with shigellae could induce inflammatory responses in the gut. Here we propose that Shigella species have evasion mechanisms that dampen host inflammatory responses by lowering host S1P levels in the gut of adult mice.

The Effects of Wolbiga-banha-tang on Obese Rats Fed a High-fat Diet (월비가반하탕이 고지방식이로 유도된 비만 쥐에 미치는 영향)

  • This study investigated the effects of Wolbiga-banha-tang (WBBT) on high fat diet-induced rats. We divided the rats into 4 groups: normal (fed a general-fat diet), control (fed a high-fat diet and no treatment), $WBBT{\times}1$ (fed a high-fat diet and 250 mg/kg of Wolbiga-banha-tang extracts), and $WBBT{\times}2$ (fed a high-fat diet and 500 mg/kg of Wolbiga-banha-tang extracts). We measured the body weight, total cholesterol, HDL cholesterol, LDL cholesterol, triglycerides, free fatty acids, total lipids, AST, ALT, and CBC in the serum of the obese rats for 7 weeks. There was a significant decrease in the control group, $WBBT{\times}1$, and $WBBT{\times}2$ in triglycerides and free fatty acids. The results suggest that WBBT is effective for lowering triglycerides and free fatty acids. Therefore, WBBT may be applied to the treatment of lipid metabolism in obesity and further clinical studies are needed.

  • NADPH is an essential co-factor for fat and cholesterol biosynthesis. However, the role of cytosolic NADP$\^$+/-dependent isocitrate dehydrogenase (IDPc), a putative NADPH producer, in the control of the fat and cholesterol metabolism has not been assessed. Here we report that increased or decreased IDPc expression in 3T3-Ll fat cells promoted or retarded adipogenesis, respectively. Furthermore, overexpression of IDPc in transgenic mice exhibited fatty liver, hypertriglyceridemia, hypercholesterolemia and obesity by increasing NADPH production leading to subsequent stimulation of acetyl-coenzyme A and malonyl-coenzyme A consumption. In contrast, administrations of a synthetic IDPc inhibitor, DAl1004, to ob/ob mice effectively reduced body weight with lowering cholesterol and triglyceride levels. In addition, a positive relationship (${\gamma}$ = 0.69, $\rho$<0.0l) between plasma IDPc activity and body mass indexes was observed in 98 randomly-selected human volunteers. Our findings strongly indicate that NADPH produced by IDPc plays an important role in controlling body fat and lipid biosynthesis.

Antihyperlipidemic and Antioxidant Effects of Poncirus trifoliata (탱자 열매 추출액의 항산화 및 지질저하효과)

  • Lipid-lowering and antioxidant effects of Poncirus trifoliata extract were investigated in hyperlipidemic rat. Concentrations of FFA and triglyceride in plasma were decreased in Poncirus trifoliata extract treatment groups. Plasma total cholesterol and LDL-cholesterol were lower in Poncirus trifoliata extract treatment groups. However HDL-cholesterol showed no significant difference in all treatment groups. Concentrations of liver total cholesterol and triglyceride were decreased in Poncirus trifoliata extract treatment groups. Concentrations of plasma and liver TBARS showed a low values in Poncirus trifoliata extract treatment groups. The values of GSH-Px and CAT activities were increased in the Poncirus trifoliata extract treatment groups. However the values of SOD activity showed no significant difference in all treatment groups.

  • Oryza sativa cv. Heugjinjubyeo, an anthocyanin-pigmented rice variety, is well known to contain high levels of bioactive phytochemicals, anthocyanin, quinolone alkaloids and phenolic acids. Here, we studied the inhibitory effect of Oryza sativa cv. Heugjinjubyeo bran on the absorption of dietary fat in streptozotocin-induced diabetic Sprague-Dawley male rats. For these experiments, experimental animals were divided into four groups: normal, diabetic-control and two experimental groups that were fed 1.0 g or 2.0 g/kg body weight/day of Oryza sativa cv. Heugjinjubyeo bran supplement for 14 days. As a result, liver glycogen levels increased significantly by 65% and 32% in groups receiving 1.0 g and 2.0 g/kg body weight/day, respectively, compared to diabetic-control. Liver cholesterol levels were significantly lower by 8.3% and 14.5% in the groups fed 1.0 g and 2.0 g of anthocyanin- pigmented rice extracts, respectively.

Effects of Leonurus Sibiricus L. Extract on Diet-induced Hypercholesterolemia in Rats (익모초 추출물이 흰쥐의 식이성 고콜레스테롤혈증에 미치는 영향)

  • This study was performed to investigate the cholesterol-lowering effect of liquid extract from Leonurus sibiricus Linne (LS). SD rats were fed atherogenic diet for 6 weeks. During the experimental period, LS liquid extract (3 g/kg and 6 g/kg) was orally given to the rats. The liver weight and levels of total cholesterol, LDL-cholesterol, and triglycerides in serum were significantly increased in rats fed with atherogenic diets. Oral treatment of LS liquid extract to the rats fed atherogenic diet significantly reduced the liver weights and significantly decreased the levels of total cholesterol, LDL-cholesterol, and triglycerides. The level of serum HDL-cholesterol was significantly increased at a high dose group compared with rats fed normal diet chow. These results indicate that LS liquid extract has the potential to restore altered lipid profiles induced by cholesterol rich diets and prevent the development of atheroclerosis.
