Low-dose Radiation Induces Antitumor Effects and Erythrocyte System Hormesis

  • Objective: Low dose radiation may stimulate the growth and development of animals, increase life span, enhance fertility, and downgrade the incidence of tumor occurrence.The aim of this study was to investigate the antitumor effect and hormesis in an erythrocyte system induced by low-dose radiation. Methods: Kunming strain male mice were subcutaneously implanted with S180 sarcoma cells in the right inguen as an experimental in situ animal model. Six hours before implantation, the mice were given 75mGy whole body X-ray radiation. Tumor growth was observed 5 days later, and the tumor volume was calculated every other day. Fifteen days later, all mice were killed to measure the tumor weight, and to observe necrotic areas and tumor-infiltration-lymphoreticular cells (TILs). At the same time, erythrocyte immune function and the level of 2,3-diphosphoglyceric acid (2,3-DPG) were determined. Immunohistochemical staining was used to detect the expression of EPO and VEGFR of tumor tissues. Results: The mice pre-exposed to low dose radiation had a lower tumor formation rate than those without low dose radiation (P < 0.05). The tumor growth slowed down significantly in mice pre-exposed to low dose radiation; the average tumor weight in mice pre-exposed to low dose radiation was lighter too (P < 0.05). The tumor necrosis areas were larger and TILs were more in the radiation group than those of the group without radiation. The erythrocyte immune function, the level of 2,3-DPG in the low dose radiation group were higher than those of the group without radiation (P < 0.05). After irradiation the expression of EPO of tumor tissues in LDR group decreased with time. LDR-24h, LDR-48h and LDR-72h groups were all statistically significantly different from sham-irradiation group. The expression of VEGFR also decreased, and LDR-24h group was the lowest (P < 0.05). Conclusion: Low dose radiation could markedly increase the anti-tumor ability of the organism and improve the erythrocyte immune function and the ability of carrying $O_2$. Low-dose total body irradiation, within a certain period of time, can decrease the expression of hypoxia factor EPO and VEGFR, which may improve the situation of tumor hypoxia and radiosensitivity of tumor itself.

  • 저선량의 감마선 조사(0-9.1 kGy)에 의하여 옥수수녹말의 고유 점도는 감소, 겉보기 아밀로오스 함량, 물 결합능력, 팽윤력과 용해도는 증가하였다. RVA 또는 아밀로그래프 점도는 조사선량이 증가함에 따라 감소하였으나 DSC에 의한 흡열 전이에는 큰 영향이 없었다. 감마선 조사한 옥수수 전분을 이용한 저항전분 생성은 효과가 없었다.

  • 저선량 ${\gamma}$선 조사가 기내 유기한 대지와 수미 감자 소괴경의 맹아율과 식물체 생장 및 생리활성에 미치는 효과 등을 알아보고자 감자 소괴경의 저장기간별로 ${\gamma}$선을 0.5${\sim}$30Gy 까지 조사하여 pot실험을 수행하였다. 감자 소괴경의 휴면과 생육에 대한 저선량 ${\gamma}$선 효과는 품종과 저장기간에 따라 다소 차이가 있었다. 대지 품종의 경우 대체로 2,4,8Gy에서 대조구에 비해 높은 맹아 출현율을 보였고, 온실 재배에서는 4,8,16Gy에서 비교적 양호한 생육과 수량 증가 효과를 보였다. 수미 품종에서의 맹아 출현은 2Gy와 4Gy에서 촉진되었고 0.5Gy와 4Gy에서 양호한 생육 및 수량을 보였다. 엽록소 함량은 저선량 ${\gamma}$선 조사구에서 대조구에 비해 뚜렷한 증가효과를 보이지 않았다. 이에 저선량 ${\gamma}$선이 감자의 휴면타파와 생육촉진에 효과가 있음을 확인할 수가 있었다.

Understanding Phytosanitary Irradiation Treatment of Pineapple Using Monte Carlo Simulation

  • Purpose: Pineapple is now the third most important tropical fruit in world production after banana and citrus. Phytosanitary irradiation is recognized as a promising alternative treatment to chemical fumigation. However, most of the phytosanitary irradiation studies have dealt with physiochemical properties and its efficacy. Accurate dose calculation is crucial for ensuring proper process control in phytosanitary irradiation. The objective of this study was to optimize phytosanitary irradiation treatment of pineapple in various radiation sources using Monte Carlo simulation. Methods: 3-D geometry and component densities of the pineapple, extracted from CT scan data, were entered into a radiation transport Monte Carlo code (MCNP5) to obtain simulated dose distribution. Radiation energy used for simulation were 2 MeV (low-energy) and 10 MeV (high-energy) for electron beams, 1.25 MeV for gamma-rays, and 5 MeV for X-rays. Results: For low-energy electron beam simulation, electrons penetrated up to 0.75 cm from the pineapple skin, which is good for controlling insect eggs laid just below the fruit surface. For high-energy electron beam simulation, electrons penetrated up to 4.5 cm and the irradiation area occupied 60.2% of the whole area at single-side irradiation and 90.6% at double-side irradiation. For a single-side only gamma- and X-ray source simulation, the entire pineapple was irradiated and dose uniformity ratios (Dmax/Dmin) were 2.23 and 2.19, respectively. Even though both sources had all greater penetrating capability, the X-ray treatment is safer and the gamma-ray treatment is more widely used due to their availability. Conclusions: These results are invaluable for optimizing phytosanitary irradiation treatment planning of pineapple.

방사선 조사 마우스에서 학습기억 장애에 대한 홍삼의 효과 (Effect of Red Ginseng on Radiation-induced Learning and Memory Impairment in Mouse)

  • Previous studies suggest that even low-dose irradiation can lead to progressive cognitive decline and memory deficits, which implicates, in part, hippocampal dysfunction in both humans and experimental animals. In this study, whether red ginseng (RG) could attenuate memory impairment was investigated through a passive-avoidance and object recognition memory test, as well as the suppression of hippocampal neurogenesis, using the TUNEL assay and immunohistochemical detection with markers of neurogenesis (Ki-67 and doublecortin (DCX)) in adult mice treated with a relatively low-dose exposure to gamma radiation (0.5 or 2.0 Gy). RG was administered intraperitonially at a dosage of 50 mg/kg of body weight, at 36 and 12 h pre-irradiation and at 30 minutes post-irradiation, or orally at a dosage of 250 mg! kg of body weight/day for seven days before autopsy. In the passive-avoidance and object recognition memory test, the mice that were trained for one day after acute irradiation (2 Gy) showed significant memory deficits compared with the sham controls. The number of TUNEL-positive apoptotic nuclei in the dentate gyrus (DG) was increased 12 h after irradiation. In addition, the number of Ki-67- and DCX-positive cells was significantly decreased. RG treatment prior to irradiation attenuated the memory defect and blocked apoptotic death as well as a decrease in the Ki-67- and DCX-positive cells. RG may attenuate memory defect in a relatively low-dose exposure to radiation in adult mice, possibly by inhibiting the detrimental effect of irradiation on hippocampal neurogenesis.

  • In some situations, for example at very low doses, in microbeam irradiation experiments, or around high energy heavy ion tracks, use of the absorbed dose to describe the energy transferred to the irradiated target can be misleading. Since absorbed dose is the expected value of energy per mass it takes into account all of the targets which do not have any energy deposition. In many situations that results in numerical values, in Joules per kg, which are much less than the energy deposited in targets that have been crossed by a charged particle track. This can lead to confusion about the biochemical processes that lead to the consequences of irradiation. There are a few alternative approaches to describing radiation that avoid this potential confusion. Examples of specific situations that can lead to confusion are given. It is concluded that using the particle radiance spectrum and the exposure time, instead of absorbed dose, to describe these irradiations minimizes the potential for confusion about the actual nature of the energy deposition.

  • The electrical characteristics of solid state devices such as the bipolar junction transistor (BJT), metal-oxide semiconductor field-effect transistor (MOSFET), and other active devices are altered by impinging photon radiation and temperature in the space environment. In this paper, the threshold voltage, the breakdown voltage, and the on-resistance for two kinds of MOSFETs (200 V and 100 V of $V_{DSS}$) are tested for ${\gamma}-irradiation$ and compared with the electrical specifications under the pre- and post-irradiation low dose rates of 4.97 and 9.55 rad/s as well as at a maximum total dose of 30 krad. In our experiment, the ${\gamma}-radiation$ facility using a low dose, available at Korea Atomic Energy Research Institute (KAERI), has been applied on two commercially available International Rectifier (IR) products, IRFP250 and IRF540.

Antioxidant Effect of Annexin A-1 Induced by Low-dose Ionizing Radiation in Adipose-derived Stem Cells

  • Radiation therapy is one of the primary options for the treatment of malignant tumors. Even though it is an effective anti-cancer treatment, it can cause serious complications owing to radiation-induced damage to the normal tissue around the tumor. It was recently reported that normal stem cell response to the genotoxic stress of ionizing radiation can boost the therapeutic effectiveness of radiation by repairing damaged cells. Therefore, we focused on annexin A-1 (ANXA1), one of the genes induced by low-dose irradiation, and assessed whether it can protect adipose-derived stem cells (ADSCs) against oxidative stress-induced damage caused by low-dose irradiation and improve effectively cell survival. After confirming ANXA1 expression in ADSCs transfected with an ANXA1 expression vector, exposure to hydrogen peroxide (H2O2) was used to mimic cellular damage induced by a chronic oxidative environment to assess cell survival under oxidative conditions. ANXA1-transfected ADSCs demonstrated that increased viability compared with un-transfected cells and exhibited enhanced anti-oxidative properties. Taken together, these results suggest that ANXA1 could be used as a potential therapeutic target to improve the survival of stem cells after low-dose radiation treatment.

Field-in-Field Technique을 이용한 두경부암의 접합부위 선량개선에 관한 고찰 (Field-in-Field Technique to Improve Dose Distribution in the Junction of the Field with Head & Neck Cancer)

  • 목 적: 두경부암의 치료에 있어 상부 두경부의 양측면조사면과 하경부의 전방조사면의 접합부위에 균등한 선량을 조사하는 것은 매우 중요하다. 접합부위의 선량분포개선을 위하여 하경부 전방조사면의 치료시 Field-in-Field technique을 이용하여 부족선량(under dose)과 초과선량(over dose)으로 인한 선량불균등을 개선하고 일반치료와의 비교를 통하여 두경부암치료에 적용하고자 한다. 대상 및 방법: 상부 두경부의 양측면 조사시 빔의 확산으로 일어나는 입사점과 출사점의 선량차이를 알아보기 위하여 인체모형팬톰을 이용하였다. 인체모형팬톰을 전산화단층촬영하고 전산화치료계획에서 관심점의 선량비교를 시행하였고, 하경부 접합부위의 선량비율을 계산하여 이를 보정하였다. 조사면 접합부위의 선량분포를 알아보기 위하여 하경부의 접합부위에 저감도 필름을 놓고 일반적인 치료인 상부 두경부의 양측면조사와 하경부의 전방조사시 선량분포를 측정하였다. 또한, 상부 두경부 양측면 조사에 따른 빔의 확산을 고려한 Field-in-Field technique을 이용하여 하경부 전방조사를 할 때의 접합부위의 선량분포 차이를 측정하여 비교하였다. 접합부위의 관심점 선량을 알아보기 위하여 열형광선량계를 이용하여 인체모형팬톰내의 관심점에서의 선량변화를 비교, 분석하였다. 결 과: 전산화치료계획에서 하경부의 접합부위에 Field-in-Field technique을 적용하여 치료계획시 상부 두경부 양측면 조사와 선량합성을 한 경우 부족선량 영역의 선량이 4.7~8.65% 이상 증가하였다. 초과선량 영역의 선량도 2.75~10.45% 감소하였다. 또한, 저감도 필름을 이용한 측정에서는 부족선량영역에서 11.3% 증가, 초과선량영역에서 5.3% 감소한 것으로 나타났다. 열형광선량계를 이용한 관심점선량측정에서도 Field-in-Field technique 적용시 부족선량을 최소 7.5%에서 최대 17.6%까지 보정해주는 것으로 나타나 불균등한 선량분포를 개선할 수 있었다. 결 론: 전산화치료계획시 빔의 확산을 고려한 Field-in-Field technique을 적용하면 접합부위의 선량보정을 통해 냉점(cold spot)과 온점(hot spot)을 줄일 수 있었으며 특히, 빔의 확산에 따른 입사점의 부족선량을 보정할 수 있었다. 본 실험을 통해 Field-in-Field technique의 임상적용시 경부임파절의 저선량으로 인한 임파절전이에 대한 위험도를 감소시킬 수 있을 것으로 사료된다.

백서의 좌골신경 손상에 미치는 저출력 레이저의 효과 (IR-Laser) (Effect of Infrared Low Dose Laser on Injured Sciatic Nerve of Rats)

  • 신경손상에 마치는 저출력 적외선 레이저의 효과를 백서의 좌골신경을 이용하여 조직학적으로 살펴 본 결과를 아래와 같이 요약한다. 1) 동일개체에서 레이저 치료군과 비치료군 사이에 뚜렷한 조직학적 차이는 발견할 수 없었다. 2) 레이저 치료군에서는 조직의 구조(tissue organization)가 비교적 잘 보존되어 있었으며, 혈관의 증식은 비치료군에 비해 풍부하였다. 3) 레이저 치료군과 비치료군의 좌골신경 지배 근육조직 소견에서 뚜렷한 차이는 발견할 수 없었으며, 양군 모두에서 부분적인 근육위축 현상을 관찰할 수 있었다. 4) 레이저 치료군에서는 비치료군에 비해 신경외막(epineurium)의 비후가 뚜렷하였다.
