Application of Molecular Biology to Rumen Microbes -Review-

  • Molecular biological techniques that recently developed, have made it possible to realize some of new attempts in the research field of rumen microbiology. Those are 1) cloning of genes from rumen microorganisms mainly in E. coli, 2) transformation of rumen bacteria and 3) ecological analysis with nonculturing methods. Most of the cloned genes are for polysaccharidase enzymes such as endoglucanase, xylanase, amylase, chitinase and others, and the cloning rendered gene structural analyses by sequencing and also characterization of the translated products through easier purification. Electrotransformation of Butyrivibrio fibrisolvens and Prevotella ruminicola have been made toward the direction for obtaining more fibrolytic, acid-tolerant, depoisoning or essential amino acids-producing rumen bacterium. These primarily required stable and efficient gene transfer systems. Some vectors, constructed from native plasmids of rumen bacteria, are now available for successful gene introduction and expression in those rumen bacterial species. Probing and PCR-based methodologies have also been developed for detecting specific bacterial species and even strains. These are much due to accumulation of rRNA gene sequences of rumen microbes in databases. Although optimized analytical conditions are essential to reliable and reproducible estimation of the targeted microbes, the methods permit long term storage of frozen samples, providing us ease in analytical work as compared with a traditional method based on culturing. Moreover, the methods seem to be promissing for obtaining taxonomic and evolutionary information on all the rumen microbes, whether they are culturable or not.

경주 서부동 출토 두레박의 수종 분석과 보존 처리 (Characterization and Conservation of a Well-bucket Excavated from Seobu-dong, Kyungju)

  • 경주시 서부동에서 출토된 두레박에 대한 수종 식별과 보존 처리를 실시하였다. 오랜 매장 기간 동안 유물은 분해되어 최대 함수율이 614-1050%에 달하는 분포를 보이는 매우 심한 분해 상태를 나타내었으며 유물의 제작에 사용된 목재는 소나무였다. 유물의 치수 안정화 효과를 얻기 위한 보존 처리제로 슈크로오스를 사용하였다. 초기 농도를 10%로 하여 30, 50, 70%로 20%씩 농도를 상승하였으며 총 8 주간 처리하였다. 유물은 농도를 상승시킴에 따라서 순조롭게 중량이 증가하였으며 약제의 농도가 30%, 50%에 달하였을 때 특히 높은 중량 증가율을 나타내었다. 보존처리 후 두레박은 에폭시계 수지로 접합하여 형태를 복원하였으며 처리 후 유물은 건조나 수축 변형이 관찰되지 않았다.

  • Epidermoid cysts are benign tumors that account for approximately 1% of intracranial tumors. In very rare cases, temporally located extradural intradiploic epidermoid cysts can cause neurological symptoms and skull perforation. Herein, we report the case of a 34-year-old woman who underwent successful treatment of an epidermoid cyst in the temporal region accompanied by neurological symptoms. Accurate radiological evaluation and complete removal of the tumor and capsule play a vital role in ensuring favorable long-term outcomes. Computed tomography and magnetic resonance imaging scans can provide an accurate assessment of the extent of intracranial expansion and invasion of the cerebral parenchyma, as well as enabling the precise localization and characterization of the bone defect and mass. In addition, collaborative surgery with a neurosurgeon is required for cases involving intracranial expansion and dural invasion.

질소산화물의 토양배출량 추정과 지구 환경에 미치는 대기화학적 특성 연구 (Characterization of NOx Emission from Soils in Southwest Korea and Their Atmospheric Chemistry)

  • The soil NO flux measurements in Korea were made from 17 May 1997 to 16 June 1997 on grass land at Kunsan National University in southwestern Korea by using flow-through chamber technique. The experiment was conducted in an effort to determine the role of natural emissions of NO on rural atmospheric photochemistry, and to understand the soil NO emission mechanism with respect to soil parameters. Soil NO fluxes were measured every minutes and averaged in every 15 minutes as well as soil temperature. Soil samples were analyzed for $NO_3^-, NH_4^+$, and moisture in soil. Soil nitrate was not detected in most times, and total N-containing was limited in site soils. There was a optimum range of soil moisture and temperature for soil NO flux. The overall average of soil NO emission rates were found to be 1.30 $\pm 0.92 ngNm^{-2}s^{-1}$ (n=1219), and ranged from 0.01 ngNm^{-2}s^{-1}$ to 5.62 ngNm^{-2}s^{-1}$. Diurnal variation of soil NO emission was typical, which was in higher level during daytime, and was in lower level over the night. NO flux showed a strong soil temperature dependence $(r^2=0.78)$, but not with soil moisture and soil N-containing during this experimental period; NO fluxes increased exponentially as soil temperature increased. In order to assure the relevant relationship between soil NO flux and the soil parameters, long-term soil flux measurement on different types of land use should be planned and conducted continuously.

  • Root-form endosseous implants which are in use today have a variety of materials, designs and surface characteristics. Among them, pure titanium surface implants and titanium matrix coated with HA are popular as well as are available in many studies. Rate of clinical success is obviously lower in jaw with cancellous bone than dense bone. In order to increase the rate of success in poor bone quality. More advanced techniques of implant surgery and surface treatment of implant fixture body have been developed. As a successful result, the installation of HA coated implant in bone quality type III or IV became highly successful. Since most clinical studies were performed without knowing the characteristics of HA coated implants, it has been impossible to come up with proper clinical data. Therefore the characterization of HA coated implants is essential to understand long term clinical performance and the predictability of HA coated implant system Our results showed that HA coated implants had the success rate at 93.7% in bone quality type III, IV for 3.8 years, and the fixture of Steri-Oss showed more stability with time.

Assessment of Corrosion Lifetime of a Copper Disposal Canister Based on the Finnish Posiva Methodology

  • In this paper, an approach developed by the Finnish nuclear waste management organization, Posiva, for the construction license of a geological repository was reviewed. Furthermore, a computer program based on the approach was developed. By using the computer program, the lifetime of a copper disposal canister, which was a key engineered barrier of the geological repository, was predicted under the KAERI Underground Research Tunnel (KURT) geologic conditions. The computer program was developed considering the mass transport of corroding agents, such as oxygen and sulfide, through the buffer and backfill. Shortly after the closure of the repository, the corrosion depths of a copper canister due to oxygen in the pores of the buffer and backfill were calculated. Additionally, the long-term corrosion of a copper canister due to sulfide was analyzed in two cases: intact buffer and eroded buffer. Under various conditions of the engineered barrier, the corrosion lifetimes of the copper canister due to sulfide significantly exceeded one million years. Finally, this study shows that it is necessary to carefully characterize the transmissivity of rock and sulfide concentration during site characterization to accurately predict the canister lifetime.

Characterization of Physiological Changes in $S3H5/\gamma{2bA2}$ Hybridoma Cells During Adaptation to Low Serum Media

  • Physiological changes of the murine hybridoma cell line $S3H5/\gamma{2bA2}$ during adaptation to RPMI 1640 medium with 1%(v/v) fetal bovine serum were characterized in terms of cell growth, antibody production, morphology, and metabolic quotients. Cells adapted to 1% serum medium in T-flasks became sensitive to shear induced by mechanical agitation and required at least 5% serum in the medium or spent medium for cell growth in spinner flasks, while cells adapted to 10% serum medium in T-flasks could grow in 1% serum medium in spinner flasks. Consequently, long-term adaptation to low serum media may not give the expected growth enhancement. After adaptation to 1% serum medium, changes in cell morphology were observed. The cells in 10% serum medium were uniform and circular, while cells in 1% medium were irregularly shaped. The DNA contents, which were measured by flow cytometry, were almost constant among the cells in the range of 1% to 10%. Further, no significant changes in energy metabolism and specific monoclonal antibody production rate were observed among these cells.

  • Ti metal has superior mechanical properties along with biocompatibility, but it still has the problem of bio-inertness thus forming weaker bond in bone/implant interface and long term clinical performance as orthopaedic and dental devices are restricted for stress shielding effect. On the other hand, despite the excellent biodegradable behavior as being an integral constituent of the natural bone, the mechanical properties of ${\beta}$-tricalcium phosphate $(Ca_3(PO_4)_2;\;{\beta}-TCP)$ ceramics are not reliable enough for post operative load bearing application in human hard tissue defect site. One reasonable approach would be to mediate the features of the two by making a composite. In this study, ${\beta}$-TCP/Ti ceramic-metal composites were fabricated by spark plasma sintering in inert atmosphere to inhibit the formation of $TiO_2$. Composites of 30 vol%, 50 vol% and 70 vol% ${\beta}$-TCP with Ti were fabricated. Detailed microstructural and phase characteristics were investigated by FE-SEM, EDS and XRD. Material properties like relative density, hardness, compressive strength, elastic modulus etc. were characterized. Cell viability and biocompatibility were investigated using the MTT assay and by examining cell proliferation behavior.

Avalanche Hot Source Method for Separated Extraction of Parasitic Source and Drain Resistances in Single Metal-Oxide-Semiconductor Field Effect Transistors

  • Separate extraction of source ($R_S$) and drain ($R_D$) resistances caused by process, layout variations and long term degradation is very important in modeling and characterization of MOSFETs. In this work, we propose "Avalanche Hot-Source Method (AHSM)" for simple separated extraction of $R_S$ and $R_D$ in a single device. In AHSM, the high field region near the drain works as a new source for abundant carriers governing the current-voltage relationship in the MOSFET at high drain bias. We applied AHSM to n-channel MOSFETs as single-finger type with different channel width/length (W/L) combinations and verified its usefulness in the extraction of $R_S$ and $R_D$. We also confirmed that there is a negligible drift in the threshold voltage ($V_T$) and the subthreshold slope (SSW) even after application of the method to devices under practical conditions.

DEEP-South: Lightcurves of Near Earth Asteroids from Year One Operations

  • Deep Ecliptic Patrol of the Southern Sky (DEEP-South) observations have been conducted officially during the off-season for exoplanet search since October 2015. Most of the allocated time for DEEP-South is devoted to targeted photometry, Opposition Census (OC), of Near Earth Asteroids (NEAs) to increase the number of such objects with known physical properties. It is efficiently achieved by multiband, time series photometry. This Opposition Census (OC) mode target objects near their opposition, with km-sized PHAs in the early stage and goes down to sub-km objects. Continuous monitoring of the sky with KMTNet is optimized for spin characterization of various kinds of asteroids, including binaries, satellites, slow/fast- and non-principal axis-rotators, and hence is expected to facilitate the debiasing of previously reported lightcurve observations. We present the preliminary lightcurves of NEAs from year one of the DEEP-South with our long term plan.
