Effects of Potassium Deficiency on C and N Metabolism during Regrowth of Italian Ryegrass (Lolium multiflorum L.) (칼륨 결핍이 이탈리안 라이그라스 재생기간동안의 탄소와 절소의 대사에 미치는 영향)

  • To investigate C and N metabolisms in response to potassium-deficient stress during regrowth of Italian ryegrass(Lolium multiflorum L.), C and N metabolites were analyzed at day 0 (cutting date), 6, 12 and 24 days after defoliation. K-sufficient (control, +K) and K-absent (-K) nutrition solutions were applied from 7 days before defoliation, and continued for one cycle of 24 days-regrowth period. During 24 days of regrowth dry matter of regrowing shoots and remaining tissues were not significantly different between +K and -K treatment. In remaining stubble, all C compounds in both +K and -K treatment largely decreased (69% to 84% of the initial level) during the first 6 days of regrowth, and then rapidly recovered. The decline of soluble sugars and fructan in roots for the first 6 days much less in the -K medium. Amino acids, soluble and insoluble proteins in stubble also feel down during the first 6 days, thereafter actively replenished in both +K and -K treatment. The decline of nitrate in stubble prolonged to 12 days of regrowth. Initial amounts of all N compounds in roots were significantly lower in the -K medium. Higher accumulation of amino acids and soluble protein in roots in the -K medium was observed after 12 days of regrowth. In regrowing shoots, 3 all carbohydrates increased with a very similar pattern for both treatments. Nitrate was not significantly different between two treatments. Depress of soluble protein accumulation in -K medium was noteworthy after 12 days of regrowth. These results indicated that an active utilization of organic reserves occurred to support regrowth even under K deficient condition with a similar extent with K sufficient condition.

Efficient Fertile Plant Regeneration from Protoplasts of Javanica Rice and Their Ploidy Determination by Flow Cytometry (Javanica 벼 원형질체로 부터 효율적인 식물체 재분화와 flow cytometry에 의한 ploidy 검정)

  • The Southeast Asian javanica rice variety Tinawen was investigated for efficient protoplast culture and plant regeneration from cell suspension-derived protoplasts using a feeder cell culture method. Feeder cells of both Lolium multiflorum and Oryza ridleyi, either alone, or in combination, were employed and plants were regenerated from protoplast-derived colonies on several plant regeneration media. Dehydration of protoplast-derived colonies was also investigated as a means of enhancing plant regeneration. In the presence of L. multiflorum or O. ridleyi feeder cells, the protoplast plating efficiency ranged from 0.09% to 1.48%, depending on the feeder cell type and the age of the cell suspension. L. multiflorum feeder cells induced approximately 6-fold higher plating efficiency compared with those of O. ridleyi. The plant regeneration frequencies were 19.3-31.7% with L. multiflorum, 13.0-18.0% with O. ridleyi and 18.0-22.0% with a mixture of both in various plant regeneration media when protoplast-derived colonies were dehydrated, while for the non-dehydrated colonies, the values were 2.0-7.0%, 3.0-5.0% and 0-4.0%, respectively. Flow cytometric analysis of 34 protoplast-derived plants showed that the majority of plants were diploids and only 2 plants were tetraploids. The plants which were transferred to glasshouse were fertile.

A Comparative Study on the Dry Matter Yield and Nutritive value of Festulolium braunii (Festulolium braunii의 건물수량 및 사료가치 비교 연구)

  • This experiment was conducted to evaluate the forage yield and quality potential of Festulolium braunii (Festuca pratensis Huds. $\times$ Lolium multiflotum Lam) as forage source in Korea. The experimental design was a split plot design with four replications from Feb. 1994 to Jan. 1996 at Hohenheim uniwrsity. Main plots were three forage species {Fesruca pratensis(Cosmos), Lolium mult~~orum(Lemtal) and Festulolium braunii(Pau1ita)) and subplots were three N levels (150,300 and 450kgha). No differences in dry matter(DM) yields were noted between Festulolium braunii and Lolium multj7orum, but DM yields for Festulolium braunii was 138 more than for Fesruca prafensis(P<0.01 ). Total DM yields over all species increased linearly with increasing N levels. The peak of DM yields over 2 years mean at 450kglha N levels were higher for Lolium mulriJotum(l5,724kg) than for Festulolium braunii(14,730kg) and for Festuca pratensis(l3,68 l kg) (P<0.01). OMD, ME, NEL and other minerals of Fesrulolium braunii were slightly higher and CP, Ca and Mg slightly lower than thsoe of Fesruca pratensis, but CP, OMD, NEL and minerals were absolutly higher for Fesrulolium braunii than for Lolium mulriflorum(P<0.01). It appears that Festulolium braunii has higher potential for improvement of the forage quality than Fesruca prarensis or Lolium rnulrijlorum. The content of CP, CF and minerals were increased, wherease OMD, ME and NEL were decreased in all species with increasing N levels. Yields of CP, DOM and NEL over 2 yean mean were increased in all species with increasing N levels.   • As an initial step for future genetic manipulations to improve forage characteristics of Italian ryegrass (Lolium multiflorum Lam.), an efficient tissue culture system was established and the factors affecting plant regeneration were evaluated. MS medium containing 5mg/L 2,4-D was optimal for embryogenic callus induction from mature seed and had a strong effect on successive plant regeneration. The plant regeneration frequency was observed at above 70% when embryogenic calli induced were transferred to N6 medium supplemented with 1mg/L 2,4-D and 5mg/L BA. Among several basic media tested, MS and N6 medium were optimal for callus induction and plant regeneration, respectively. Genotype was an important factor in plant regenerability. 'Jeanne' showed the highest regeneration frequency of 73%. A short tissue culture period and high-frequency regeneration system established in this study will be useful for molecular breeding of Italian ryegrass through genetic transformation.

Plant Regeneration from Protoplasts of Indica Rice (Indica 벼의 원형질체들로부터 식물체 재분화)

  • An efficient protocol for plant regeneration from protoplasts of the indica rice variety IR43 has been developed. The procedure involved plating of embryogenic suspension-derived protoplasts on the surface of a filter membrane overlaying agarose-embedded feeder cells. Lolium multiflorum cell suspensions were preferable to these of Oryza ridleyi as feeder cells and Lolium suspensions supported colony formation from up to 0.68% of the protoplasts, depending on the age of cell suspensions. Plant regeneration frequency was significantly improved by using maltose alone or in a 1:1(w/w) combination with sucrose as carbohydrate source and a simple dehydration treatment using a high concentration of agarose in the regeneration medium. Medium containing maltose or maltose mixed with sucrose increased the plant regeneration frequency compared with medium containing sucrose alone. The plant regeneration frequency was increased to 30.7 to 70.7% following dehydration treatment, while the non-treated controls showed a regeneration frequency of 3.1 to 30.6%. Protoplast-derived plants were transferred to the glasshouse, flowered with morphologically normal.
