Anti-inflammatory activity of a sulfated polysaccharide isolated from an enzymatic digest of brown seaweed Sargassum horneri in RAW 264.7 cells

  • BACKGROUND/OBJECTIVES: Sargassum horneri is an edible brown alga that grows in the subtidal zone as an annual species along the coasts of South Korea, China, and Japan. Recently, an extreme amount of S. horneri moved into the coasts of Jeju Island from the east coast of China, which made huge economic and environmental loss to the Jeju Island. Thus, utilization of this biomass becomes a big issue with the local authorities. Therefore, the present study was performed to evaluate the anti-inflammatory potential of crude polysaccharides (CPs) extracted from S. horneri China strain in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. MATERIALS/METHODS: CPs were precipitated from S. horneri digests prepared by enzyme assistant extraction using four food-grade enzymes (AMG, Celluclast, Viscozyme, and Alcalase). The production levels of nitric oxide (NO) and pro-inflammatory cytokines, including tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-$1{\beta}$ were measured by Griess assay and enzyme-linked immunosorbent assay, respectively. The levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), nuclear factor (NF)-${\kappa}B$, and mitogen-activated protein kinases (MAPKs) were measured by using western blot. The IR spectrums of the CPs were recorded using a fourier transform infrared spectroscopy (FT-IR) spectrometer. RESULTS: The polysaccharides from the Celluclast enzyme digest (CCP) showed the highest inhibition of NO production in LPS-stimulated RAW 264.7 cells ($IC_{50}$ value: $95.7{\mu}g/mL$). Also, CCP dose-dependently down-regulated the protein expression levels of iNOS and COX-2 as well as the production of inflammatory cytokines, including TNF-${\alpha}$ and IL-$1{\beta}$, compared to the only LPS-treated cells. In addition, CCP inhibited the activation of NF-${\kappa}B$ p50 and p65 and the phosphorylation of MAPKs, including p38 and extracellular signal-regulated kinase, in LPS-stimulated RAW 264.7 cells. Furthermore, FT-IR analysis showed that the FT-IR spectrum of CCP is similar to that of commercial fucoidan. CONCLUSIONS: Our results suggest that CCP has anti-inflammatory activities and is a potential candidate for the formulation of a functional food ingredient or/and drug to treat inflammatory diseases.

Medial Retracted Large Rotator Cuff Tears (내측으로 퇴축된 대범위 회전근 개 파열)

  • Medially retracted large-sized rotator cuff tears includes large-sized tears, massive tears and irreparable tears. Generally arthroscopic repair or open repair of rotator cuff tears is used in reparable tears. However, arthroscopic repair requires long period practice and endurance. In irreparable tears, arthroscopic debridement, partial repair, latissimus dorsi transfer and retrograde arthroplasty can be the option. Arthoscopic debridement gives temporal relief who experienced improvement in pain and increase in range of motion after subacromial local anesthetic injection. Also arthroscopic partial repair gives good results in irreparable cases, especially in suprascapular nerve traction neurapraxia. Tendon transfer can be used in mild to moderate muscle weakness in shoulder abduction for long term treatment. Pectoralis major transfer can be used in anterosupeior tears and latissimus dorsi transfer can be used in posterosuperior tears. Reverse shoulder prosthesis is used in extreamly weakened shoulder pseudoparalysis. The authors discussed the method of arthroscopic repair in irreparable tears. The debridement, partial repair, and tendon transfer could be used in medially retracted large-sized rotator cuff tears.

Analysis and Improvement for Manual to Protect Mountain Disaster in Urban Area (도심지 토사재해 예방을 위한 기존 매뉴얼 분석과 개선 방안)

  • More than 70 percent of the Korean territory consists of mountain area so development of mountain district is essential to urbanize continuously. Thus, technological developments for risk factors and standards and manuals must be needed to prevent mountain disaster. Risk Management Manual should be made and operated in government legislation related to national disaster, but there is still no Emergency Management Standard Manual and Emergency Response-Practical Manual to prevent mountain disaster. This study suggests the improvement plans that are legislated but not established cleary in the field of disaster in urban area. The main items are like as 1) adaptable standard and practical manual to prevent mountain disaster in urban area, 2) reinforcement between managing department and interagency vertically and horizontally in central and local government organization, 3) Personal SOP (Standard Operating Procedure) not EOP (Emergency Operation Plan), 4) considering 13 items selected by Ministry of Public Safety and Security, 5) schematization with personal action plan, 6) check list to do in the event of mountain disaster, and 7) regular practice per quarter.

A Study on the Experience of Hands-on Workers' Case Management in the Organization for Preventing Prostitution (성매매방지기관 실무자의 사례관리 경험에 대한 연구)

  • The purpose of this study is to investigate the problems brought about by applying the case management of prostitution in practice, considering its special characteristics. The study is based on the actual experience and understanding of the hands-on workers toward its case management, who are working at the organization for preventing prostitution. To do this, data collected for the research is analyzed by an inductive approache. Participants are selected in the Counseling Centers for Prostitution, General Supporting Facilities, Supporting Centers for Self-help, and Youth Facilities. Three organizations in each type of the centers or facilities are respectively chosen. Twelve participants are composed of six hands-on workers in charge of the case management and six managers supervising it(called the director of the Center in some Centers). According to the analysis, the main issues of their case management experiences are divided into 5 topics and then sub-divided into 12 ones in 3 areas. Each theme of the topics is 'Steady & ordinary, not special tasks', 'Co-existence of a burden & thrilling', 'Sad, but pretending to be serene', 'Know-hows through practical experiences', 'Nevertheless, fill my future with hopes'. On the basis of the result, some policies and instructions necessary to actively revitalize the case management are suggested: expanding professional training centers based on the field, preparing government level customized-services in medical treatments and laws, creating jobs & intensifying job training, setting up various cooperative systems with local organizations via active networking, establishing super-vision systems, and so on.

Successful Aging and Work Activities during Elderly Period: Focused on the Case of Ogawanosho Oyakimura in Japan (노년기의 성공적 노화와 취로 활동 : 일본의 오가와노쇼 오야키무라 사례를 중심으로)

  • The purpose of this study is to raise the necessity of an alternative awareness toward economic activities as the work form of social participation by the elderly and their corresponding educational tasks. For this aim, the concept of Japan's 'rewarding work participation' and its practical cases have been examined. As a study method, the characteristics of Japan's state and local government elderly policy projects have been examined through literature research. Next, the cases of the elderly having established an autonomous company and practiced Ogawanosho Oyakimura were researched based on on-site visits and interviews. As this study has suggested that the economic activities of the elderly have to be carried out from the perspective of successful aging and these activities have been conducted through 'rewarding work participation', the following features have been clarified. First, a converted image of the elderly is necessary. The elderly do have the experiences worthy of social utilization. Second, the educational and managerial policies which turn the elderly's experiences into social values are necessary. Third, a flexible awareness toward the corresponding works of the elderly is needed. In other words, the works which have reflected mentally solitary tendency and social discontinuity features of the elderly and taken into account the demands of the elderly generation who desire to become an influential and meaningful existence are necessary. Thus, future elderly education need to come up with the policies and practices toward work education from the viewpoint of rewarding work participation.

Research on flood risk forecast method using weather ensemble prediction system in urban region (앙상블 기상예측 자료를 활용한 도시지역의 홍수위험도 예측 방안에 관한 연구)

  • Localized heavy storm is one of the major causes of flood damage in urban regions. According to the recent disaster statistics in South Korea, the frequency of urban flood is increasing more frequently, and the scale is also increasing. However, localized heavy storm is difficult to predict, making it difficult for local government officials to deal with floods. This study aims to construct a Flood risk matrix (FRM) using ensemble weather prediction data and to assess its applicability as a means of reducing damage by securing time for such urban flood response. The FRM is a two-dimensional matrix of potential impacts (X-axis) representing flood risk and likelihood (Y-axis) representing the occurrence probability of dangerous weather events. To this end, a regional FRM was constructed using historical flood damage records and probability precipitation data for basic municipality in Busan and Daegu. Applicability of the regional FRMs was assessed by applying the LENS data of the Korea Meteorological Administration on past heavy rain events. As a result, it was analyzed that the flood risk could be predicted up to 3 days ago, and it would be helpful to reduce the damage by securing the flood response time in practice.

Experimental Investigation on Post-Fire Performances of Fly Ash Concrete Filled Hollow Steel Column

  • In structural engineering practice, understanding the performance of composite columns under extreme loading conditions such as high-rise bulding, long span and heavy loads is essential to accuratly predicting of material responses under severe loads such as fires or earthquakes. Hitherto, the combined effect of partial axial loads and subsequent elevated temperatures on the performance of hollow steel column filled fly ash concrete have not been widely investigated. Comprehensive test was carried out to investigate the effect of elevated temperatures on partial axially loaded square hollow steel column filled fly ash concrete as reported in this paper. Four batches of hollow steel column filled fly ash concrete ( 30 percent replacement of fly ash), (HySC) and normal concrete (CFHS) were subjected to four different load levels, nf of 20%, 30%, 40% and 50% based on ultimate column strength. Subsequently, all batches of the partially damage composite columns were exposed to transient elevated temperature up to 250℃, 450℃ and 650℃ for one hour. The overall stress - strain relationship for both types of composited columns with different concrete fillers were presented for each different partial load levels and elevated temperature exposure. Results show that CFHS column has better performance than HySC at ambient temperature with 1.03 relative difference. However, the residual ultimate compressive strength of HySC subjected to partial axial load and elevated temperature exposure present an improvement compared to CFHS column with percentage difference in range 1.9% to 18.3%. Most of HySC and CFHS column specimens failed due to local buckling at the top and middle section of the column caused by concrete crushing. The columns failed due to global buckling after prolong compression load. After the compression load was lengthened, the columns were found to fail due to global buckling except for HySC02.

Relevance between oral health life practices and oral discomforts in elderly school participants in W urban-rural complex city (도농복합도시 W시 노인대학생의 구강건강생활 실천도와 구강불편감과의 관련성)

  • This study aims to analyze the relevance between oral health practices and oral discomforts of the elderly in an urban-rural area by surveying the elderly in welfare centers for the Elderly-continued oral health care program. The result shows that the elderly brush their teeth under regular oral care, and practice oral health life by scaling for the prevention of periodontal disease. Most of the elderly who drink alcohol have experienced implant care and tend to quit drinking after the treatment, and seemed to get their oral discomfort relieved. However, no relevance is found between implant experiences and social discomfort. Furthermore, the elderly who had bad breath expressed pronunciation, taste, pain and chewing discomfort, and social discomfort (P>.05). The educational level of the elderly did not have an impact on oral discomfort, but smoking, chewing and bad breath discomfort seemed to be related to social discomfort(P>.05). Therefore, since oral discomfort of the elderly causes social discomfort which decreases their quality of living, we recommend oral health departments of local governments to help the elderly maintain happy lives by continuing to study the practical use of reducing oral discomfort.

A Network Analysis on the Trend of Pressing Dementia Management Policy: Focusing on the Prevention of Dementia (치매관리정책의 언론보도 경향에 대한 네트워크 분석: 치매예방을 중심으로)

  • The purpose of this study is to identify the tendency of media reports on the dementia management policy in Korea and to suggest policy implications such as prevention of dementia, improvement of awareness, and management of dementia through network analysis. We analyzed the linkage structure between the main texts centered on the number of citations of the main language related to the dementia management policy and the centrality and mediation as the research procedures and methods. As a result of the analysis, first, a 'micro' perspective is needed to explain practically. Second, it is desirable to understand the dementia management policy in the context of community. Third, the network structure of key words such as 'dementia management policy' suggests the possibility of research study in academic research in future research. Therefore, the phenomenon of dementia management policy will contribute to the direction of future dementia management policy, not local or temporary.

Unsupervised Non-rigid Registration Network for 3D Brain MR images (3차원 뇌 자기공명 영상의 비지도 학습 기반 비강체 정합 네트워크)

  • Although a non-rigid registration has high demands in clinical practice, it has a high computational complexity and it is very difficult for ensuring the accuracy and robustness of registration. This study proposes a method of applying a non-rigid registration to 3D magnetic resonance images of brain in an unsupervised learning environment by using a deep-learning network. A feature vector between two images is produced through the network by receiving both images from two different patients as inputs and it transforms the target image to match the source image by creating a displacement vector field. The network is designed based on a U-Net shape so that feature vectors that consider all global and local differences between two images can be constructed when performing the registration. As a regularization term is added to a loss function, a transformation result similar to that of a real brain movement can be obtained after the application of trilinear interpolation. This method enables a non-rigid registration with a single-pass deformation by only receiving two arbitrary images as inputs through an unsupervised learning. Therefore, it can perform faster than other non-learning-based registration methods that require iterative optimization processes. Our experiment was performed with 3D magnetic resonance images of 50 human brains, and the measurement result of the dice similarity coefficient confirmed an approximately 16% similarity improvement by using our method after the registration. It also showed a similar performance compared with the non-learning-based method, with about 10,000 times speed increase. The proposed method can be used for non-rigid registration of various kinds of medical image data.
