Identification of plasma miRNA biomarkers for pregnancy detection in dairy cattle

  • A pregnancy diagnosis is an important standard for control of livestock's reproduction in paricular dairy cattle. High reproductive performance in dairy animals is a essential condition to realize of high life-time production. Pregnancy diagnosis is crucial to shortening the calving interval by enabling the farmer to identify open animals so as to treat or re-breed them at the earliest opportunity. MicroRNAs are short RNA molecules which are critically involved in regulating gene expression during both health and disease. This study is sought to establish the feasible of circulating miRNAs as biomarkers of early pregnancy in cattle. We applied Illumina small-RNA sequencing to profile miRNAs in plasma samples collected from 12 non-pregnant cows ("open" cows: samples were collected before insemination (non-pregnant state) and after pregnancy check at the indicated time points) on weeks 0, 4, 8, 12 and 16. Using small RNA sequencing we identified a total of 115 miRNAs that were differentially expressed weeks 16 relative to non-pregnancy ("open" cows). Weeks 8, 12 and 16 of pregnancy commonly showed a distinct increase in circulating levels of miR-221 and miR-320a. Through genome-wide analyses we have successfully profiled plasma miRNA populations associated with pregnancy in cattle. Their application in the field of reproductive biology has opened up opportunities for research communities to look for pregnancy biomarker molecules in dairy cattle.

A Study on the Hazard and Risk Analysis of Hospital in Korea - Focused on Local Medical Centers (의료기관의 위험도 분석 조사 - 지역공공의료원을 중심으로)

  • The purpose of this study is to analyse the hazard risk by examining the magnitude and severity of each type of hazard in order to mitigate and prepare for disasters in medical facilities. Methods: The hazard risk analysis for hazard types was surveyed for team leaders of medical facilities. The questionnaire analyzed data from 27 facilities, which were returned from 41 Local Medical Centers. Results: When looking at the 'Risk' by category type of hazard, the influence of health safety and fire/energy safety comes first, followed by natural disaster, facility safety, and crime safety. On the other hand, as for 'Magnitude', facility safety and crime safety come first, followed by health safety, fire/energy safety, and natural disasters. Most of the top types of disaster judged to have high hazard in medical facilities are health types. The top five priorities of hazard in medical facilities, they are affected by the geographical and industrial conditions of the treatment area. In the case of cities, the hazard was found to be high in the order of infectious disease, patient surge, and wind and flood damage. On the other hand, in rural areas, livestock diseases and infectious diseases showed the highest hazard. In the case of forest areas, the hazard was high in the order of wildfire, fire accident, lightning, tide, earthquake, and landslide, whereas in coastal areas of industrial complexes, the hazard was high due to fire, landslide, water pollution, marine pollution, and chemical spill accident. Implications: Through the research, standards will be established for the design of hospitals with disaster preparedness, and will contribute to the preparation of preemptive measures in terms of maintenance.

Association Between the Prevalence of Schistosomiasis in Elementary School Students and Their Parental Occupation in Sudan

  • Global efforts to identify groups at high risk for schistosomiasis have mainly concentrated on identifying their geographical distribution. Investigations on the socioeconomic characteristics of high-risk groups are relatively scarce. This study aimed to explore the associations between schistosomiasis among students and their parents' occupations. A nationwide cross-sectional survey was conducted targeting 105,167 students in 1,772 primary schools across Sudan in 2017. From these students, 100,726 urine and 96,634 stool samples were collected to test for Schistosoma haematobium and S. mansoni infection. A multi-level mixed effect analysis was used with age and sex as fixed factors, and school as a random factor. The odd ratios (ORs) of practicing open defecation among farmers' children were almost 5 times higher than their counterparts whose parents were government officials (OR=4.97, 95% confidence intervals (CIs): 4.57-5.42, P<0.001). The ORs of contacting water bodies for watering livestock among farmers' children were more than 4 times higher than those of children whose parents were government officials (OR=4.59, 95% CIs: 4.02-5.24, P<0.001). This study shows that schistosomiasis represents a disease of poverty and that farmers' children constituted a high-risk group.

Prevalence of bovine viral diarrhea virus from Korean native cattle farms in Jeju (제주지역 한우의 소 바이러스성 설사병 바이러스 감염실태)

  • Bovine viral diarrhea virus (BVDV) is an RNA virus belonging to Pestivirus in the family Flaviviridae. BVDV has economic significance for the livestock industry because of its association with acute disease, fetal loss, and birth of persistently infected (PI) animals. This study aimed to investigate the BVDV infection rates in Korean native cattle farms in Jeju for further planning of a BVDV control program in the Jeju Province. BVDV antibodies and antigens were tested in 15,842 sera collected from 302 Korean native cattle herds between January 2014 and June 2017 using enzyme-linked immunosorbent assay (ELISA). Viral antigen was detected by reverse transcription-polymerase chain reaction from 60 sera that were antigen ELISA-positive. BVDV antibodies were found in 90.7% (274/302) herds and 61.1% (9,678/15,842) cows. BVDV antigens were found in 13.2% (40/302) herds and 0.4% (61/15,842) cows. The oldest animal group (> 8 years) exhibited the highest sero-positive rates (91%), while the youngest animal group (< 1 years) had the highest antigen positivity rates (0.52%). Of the 60 antigen-positive sera, BVDV types 1 and 2 were found in 36 and 12 sera, respectively. Additionally, six animals were considered to be PI as BVDV was continually detected in annual examination.

Management Guidelines and the Structure of Vegetation in Natural Monuments Koelreuteria Paniculata Community (천연기념물 모감주나무군락의 식생구조와 관리제언)

  • This study analyzed vegetation structure of natural monuments Koelreuteria paniculata community in search of a conservation and management plan. Plant sociological analysis of Koelreuteria paniculata community indicates that it can be classified into Achyranthes japonica subcommunity and Rhodotypos scandens subcommunity and Trachelospermum asiaticum var. intermedium subcommunity. While Koelreuteria paniculata community of Ahnmyeondo is composed of sub tree layer and herb layer, those of Pohang and Wando are composed of tree layer, Sub tree layer, shrub layer, herb layer. The results of tree vitality analysis showed that those in Ahnmyeondo appeared to be relatively low when compared to those in Pohang and Wando-gun. This can be understood in two different aspects: disease and insects vulnerability due to a relatively simple structure and lack of competitive species, and decreased vitality / natural branch losses due to crown competition arising from high density. The result of soil characteristics analysis showed that soil texture, soil pH, organic matter, $p_2O_5$, exchange positive ion were sufficient for tree growth while total nitrogen was not, so that discretion would be needed for fertilizer application. As there were damages of disease and inscet, but only for 10~15% of the entire area; it still requires consistent preconsideration. The study suggests the management methods for preservation of Koelreuteria paniculata community. First, securing designated areas is necessary in order to minimize environment deterioration due to surrounding development. Especially, for sections with decreased areas, expansion of designated areas through land purchase should also be considered. Second, artificial interference may affect the livestock. Therefore, monitoring of artificial interference is necessary, based on which protection projects must be conducted. Third, from analysis of young plants which influence the maintenance mechanisms of Koelreuteria paniculata community, a decrease compared to the prior year was observed; investigation is needed. Therefore, an active management policy through status examination of livestock such as germination and young plants is necessary.

Enterotoxigenic Bacteroides fragilis-Associated Diseases and Detection (Enterotoxigenic Bacteroides fragilis에 의한 질환과 검출)

  • These commensal intestinal bacteria can enhance the immune system and aid in nutrient absorption but can also act as opportunistic pathogens. Among these intestinal bacteria, the anaerobic Bacteroides fragilis are divided into enterotoxigenic B. fragilis (ETBF) which secrete the B. fragilis toxin (BFT) and non-enterotoxigenic B. fragilis (NTBF) which do not secrete BFT. ETBF can cause diarrhea and colitis in both humans and livestock but can also be found in asymptomatic individuals. ETBF is predominantly found in patients with inflammatory diarrheal diseases and traveller's diarrhea. Several clinical studies have also reported an increased prevalence of ETBF in human patients with inflammatory bowel disease (IBD), colitis and colorectal cancer. In small animal models (C57BL/6 wild-type mice, germ-free mice, multiple intestinal neoplasia (Min) mice, rabbits and Mongolian gerbils), ETBF have been found to initiate and/or aggravate IBD, colitis and colorectal cancer. BFT induces E-cadherin cleavage in intestinal epithelial cells resulting in loss of epithelial cell integrity. Subsequent activation of the ${\beta}$-catenin pathway leads to increased cellular proliferation. In addition, ETBF causes acute and chronic colitis in wild-type mice as well as enhances tumorigenesis in Min mice via activation of the Stat3/Th17 pathway. Currently, ETBF can be detected using a BFT toxin bioassay and by PCR. Advances in molecular biological techniques such as real-time PCR have allowed both researchers as well as clinicians to rapidly detect ETBF in clinical samples. The emergence of more sensitive techniques will likely advance molecular insight into the role of ETBF in colitis and cancer.

Evaluation of Forage Production of Sorghum for High-Yielding Hybrid (우량 품종의 선발을 위한 사초용 수수의 생산성 평가)

  • Livestock farmers face several limitations when using the government recommended sorghum cultivars for forage because of the limited seed supply. Therefore, the objective of the this study is to evaluate, select, and recommend the best high-yielding hybrid as the government recommended cultivars. The agronomic characteristics and forage yield of three cultivars (cv. P947, cv. KF429 and cv. SS405) of forage sorghum were evaluated at two locations (Suwon and Sunghwan) for 2 years (1999${\sim}$2000). 'KF429' was susceptible to foliar disease, while 'SS405' had less lodging resistance among the sorghum cultivars tested. When compared with 'P947' (recommended cultivar), dry matter yield of 'KF429' and 'SS405' cultivars increased by 16% and 75%, respectively. The crude protein of 'P947'(9.4%) was higher than other cultivars. The percentage of ADF and NDF were the lowest in 'KF429' cultivar. The results of forage performance experiment indicate that 'KF429' and 'SS405' are recommended as the government recommended sorghum cultivars due to a high lodging tolerance, disease resistance, and high dry matter yield.

Comparison of hematological values of conventional pigs and transgenic pigs supressed in immune rejection response (일반돼지와 면역 거부반응이 억제된 형질전환돼지의 혈액 성상 비교)

  • Blood test is a useful tool in establishing medical treatment for livestock. It provides information such as disease diagnosis, treatment effects, prognostic judgment, and health status. This study compared the value of erythrocytes and leukocytes among conventional, transgenic miniature, and transgenic conventional pigs aged six months to 24 months. Further, it analyzed the aspects of hematological value changes according to the pigs' ages. As a result, the number of red blood cells (RBC), which include hemoglobin, and hematocrit, and the number of white blood cells (WBC), which include neutrophils, and lymphocyte, were high among transgenic miniature pigs, compared with the conventional and transgenic conventional pigs. Conventional pigs showed similar values of RBC and WBC regardless of transgenesis. In comparing their age, the RBC decreased as the age increased compared with the pigs among all the three groups aged of 6~12 months. On the other hand, WBC and neutrophils showed no significant difference regardless of different ages among all the three groups. However, various counts in RBC and WBC were mostly found to be higher in each age in transgenic miniature pigs than in conventional and transgenic conventional pigs. The results of this study show that the values of RBC and WBC were generally higher in transgenic miniature pigs than in conventional and transgenic conventional pigs. Based on this research, hematological values can be widely used in diagnosing diseases or checking the health status of transgenic pigs that are used as disease models, organ transplant source and alike.

A Survey of Epidemic Diseases in Horses Imported into South Korea between 2003 and 2008 (2003년부터 2008년까지 한국에 수입된 말에서 전염성 질병의 실태조사)

  • South Korea is susceptible to foreign diseases due to its high rate of livestock importation. The purpose of this study was to investigate the infectious conditions of contagious disease of horses imported into South Korea from other countries. The horses were tested for contagious equine metritis (CEM), equine infectious anemia (EIA), equine piroplasmosis (EP), equine viral arteritis (EVA), vesicular stomatitis (VS), dourine, and glanders. The prevalence of these infectious diseases in 6,650 horses imported from 24 countries between 2003 and 2008 was reviewed by the National Veterinary Research and Quarantine Service. Seropositive results were found for EIA, EP, EVA, dourine and glanders: 3/6,189 serum samples tested were EIA-positive, 37/6,005 samples tested by complement fixation (CF) were EP-positive, 28/6,043 samples tested by virus neutralization (VN) were EVA-positive, 4/2,071 serum samples tested by CF were positive for dourine, and 4/1,950 samples tested by CF were positive for glanders. No contagious equine metritis or vesicular stomatitis was detected. In total, 76/6,650 imported horses tested positive for an infectious disease. Notably, 4/6 sera (66.6%), all taken from horses imported from Tanzania, were positive for glanders. This is the first report of glanders infection in horses from Tanzania since 1996.

Growth Characteristics and Productivity of New Orchardgrass(Dactylis glomerata L.) Variety "Jangbeol 102" (오차드그라스 신품종 "장벌 102호"의 생육특성과 수량성)

  • "Jangbeol 102" is a new orchardgrass(Dacrylis glomerata L.) variety developed by the National Livestock Research Institute(NLRI) in 2002. To develope the new variety of orchardgrass, 5 superior clones were selected and polycrossed for seed production. Agronomic growth characteristics and forage production of "Jangbeol 102" were examined at Suwon from 1995 to 1998, and regional trials were conducted in Suwon, Namwon and Pyungchang from 1999 to 2002 and Jeju from 2000 to 2002, respectively. "Jangbeol 102" showed semi-erect growth habit in fall and spring and medium to long type in length of flag leaf and upper internode. Plant height of "Jangbeol 102" was similar to that of standard variety, "Ambassador" and heading date was I day faster as 11th May compared to Ambassador. Characters such as winter hardiness, regrowth, moisture tolerance, disease resistance of "Jangbeol 102" were stronger or better than those of Ambassador, specially in regrowth and disease resistance. "Jangbeol 102" showed 18% higher dry matter yield(13,430kg/ha) compared to Amabassador. Nutritive value was appeared to be similar in both varieties.assador.
