Studies on Ganglioside GM3 and Sialidase Activity in Human Fetal Liver

  • Ganglioside GM3 and sialidase activities in human fetal liver have been investigated. Gangliosides were extracted from fetal livers by the Folch-Suzuki method and analyzed by high-performance thin layer chromatography (HPTLC). GM3 increased, but lactosylceramide (LacCer) decreased predominantly over the developmental stages. Sialidase in human fetal liver was mainly localized in the lysosomal fraction and its activity was high in the earlier stages of development. The optimum pH for this enzyme was 4.3~4.4. Sialidase was more active with the ganglioside mixture than with GM3, sialyllactose or fetuin. Fetal liver sialidase was still active (20% activity) in the presence of 25% methanol. These results suggested that the changes of the ganglioside GM3 and sialidase activity may be involved in the regulation of cell growth in human fetal liver during development.

미역 (Undaria pinnatifida) 국수의 투유가 랫트 간장중의 활성산소 및 제거효소에 미치는 영향 (Effect of Brown Algae (Undaria pinnatifida)-Noodle on Oxygen Radicals and Their Scavenger Enzymes in Liver of Sprague-Dawley Rats)

  • 간장중의 활성산소 및 제거효소의 활성에 미치는 영향을 평가하기 위하여 미역의 건조분말을 $10{\%},\;20{\%},\;40{\%}$-첨가 조제한 미역국수 ($10{\%},\;20{\%},\;40{\%}\;FBA-noodle$)를 4주동안 SD계 흰쥐에 투여하여 미역국수의 생리작용을 평가하였다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 투여그룹의 mitochondria의 ${\cdot}OH$ 생성은 대조그룹 대비 각각 $20{\%},\;25{\%},\;35{\%}$의 현저한 OH 생성 억제효과가 인정되었다. 또한 $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 투여그룹의 microsome의 ${\cdot}OH$ 생성도 대조그룹 대비 $12{\~}20{\%}$의 유의적인 ${\cdot}OH$ 생성 억제효과가 인정되었다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 microsome의 $H_2O_2$의 생성은 유의적인 억제효과를 인정할 수 없었지만, cytosol중의 $O_2^({\cdot}-)$ 생성은 $20{\%},\;40{\%}$ FBA-noodle에서 $10{\%}$$O_2^({\cdot}-)$의 생성 억제효과가 인정되었다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle 투여그룹의 간장 mitochondria의 Mn-SOD 활성은 대조그룹 대비 $10{\~}15{\%}$의 Mn-SOD 활성의 증가효과가 인정되었다. 간장 microsome의 Mn-SOD 활성은 $10{\%} 및 20{\%}$ FBA-noodle 투여그룹은 Mn-SOD 활성의 유의적인 증가 효과를 인정할 수 없었지만, $40{\%}$ FBA-noodle 투여그룹은 $12{\%}$의 유의적인 Mn-SOD 활성 증가효과가 인정되었다. 또한 $10{\%},\;20{\%},\;40{\%}$ FBA-noodle의 투여그룹의 간장 cytosol의 Cu, Zn-SOD 활성은 대조그룹 대비 $10{\~}20{\%}$의 Cu, Zn-SOD 활성의 유의적인 증가효과가 인정되었다. 한편 간장 cytosol의 글루타치온 퍼옥시다아제 (GSHPx) 활성을 평가하여 보면 $10{\%}, 20{\%}, 40{\%}$ FBA-noodle 투여그룹의 cytosol의 GSHPx 활성은 대조그룹 대비 $20{\~}40{\%}$의 매우 효과적인 GSHPx활성의 증가효과가 인정되었다. $10{\%}, 20{\%}, 40{\%}$ FBA-noodle 투여그룹의 mitochondria의 LPO의 함량을 비교하여 보면 $10{\%}$ FBA-noodle은 유의적인 LPO의 생성 억제효과를 인정할 수 없었지만, $20{\%},\;40{\%}$ FBA-noodle은 $10{\%}$의 유의적인 LPO 생성 억제효과가 인정되었다. $10{\%},\;20{\%},\;40{\%}$ FBA-noodle 투여그룹의 microsome의 LPO의 함량은 $10{\%}$ FBA-noodle은 유의적인 LPO의 생성 억제효과를 인정할 수 없었지만, $20{\%} 및 40{\%}$ FBA-noodle은 약 $10{\~}12{\%}$의 유의적인 LPO 생성 억제효과가 인정되었다.

Recent insights into the role of ChREBP in intestinal fructose absorption and metabolism

  • Fructose in the form of sucrose and high fructose corn syrup is absorbed by the intestinal transporter and mainly metabolized in the small intestine. However, excess intake of fructose overwhelms the absorptive capacity of the small intestine, leading to fructose malabsorption. Carbohydrate response element-binding protein (ChREBP) is a basic helix-loop-helix leucine zipper transcription factor that plays a key role in glycolytic and lipogenic gene expression in response to carbohydrate consumption. While ChREBP was initially identified as a glucose-responsive factor in the liver, recent evidence suggests that ChREBP is essential for fructose-induced lipogenesis and gluconeogenesis in the small intestine as well as in the liver. We recently identified that the loss of ChREBP leads to fructose intolerance via insufficient induction of genes involved in fructose transport and metabolism in the intestine. As fructose consumption is increasing and closely associated with metabolic and gastrointestinal diseases, a comprehensive understanding of cellular fructose sensing and metabolism via ChREBP may uncover new therapeutic opportunities. In this mini review, we briefly summarize recent progress in intestinal fructose metabolism, regulation and function of ChREBP by fructose, and delineate the potential mechanisms by which excessive fructose consumption may lead to irritable bowel syndrome.

Cleistocalyx nervosum Extract Ameliorates Chemical-Induced Oxidative Stress in Early Stages of Rat Hepatocarcinogenesis

  • Purpose: To study the effect of Cleistocalyx nervosum extract (CE) on diethylnitrosamine (DEN) and phenobarbital (PB) induced oxidative stress in early stages of rat hepatocarcinogenesis. Materials and Methods: Male Wistar rats were divided into 4 groups, with Group 1 as a negative control and Group 2 was a positive control receiving DEN injections once a week and PB in drinking water for 6 weeks. Two weeks before DEN initiation and PB treatment, Groups 3 and 4, were fed with 500 and 1000 mg/kg of CEs, respectively, for 8 weeks. Results: A number of GST-P-positive foci, preneoplastic lesions, in the liver were markedly increased in carcinogen administered rats, but was comparatively decreased in rats treated with 1000 mg/kg of CE. The CE reduced malondialdehyde in serum and in the livers of rats treated with DEN and PB. Moreover, CE significantly increased the activities of glutathione peroxidase and catalase in rat liver. Conclusions: CE appeared to exert its chemopreventive effects by modulating antioxidant status during DEN and PB induced early stages of hepatocarcinogenesis in rats.

Augmentation of antioxidant system: Contribution to antimalarial activity of Clerodendrum violaceum leaf extract

  • Reactive oxygen species are known to mediate various pathological conditions associated with malaria. In this study, the antioxidant potential of Clerodendrum violaceum leaf extracts, an indigenous antimalarial remedy, was evaluated. Total phenol, flavonoid, selenium, vitamins C and E contents of Clerodendrum violaceum leaf extracts were determined. The free radical scavenging activities of the extracts against DPPH, superoxide anion and hydrogen peroxide coupled with their reducing power were also evaluated in vitro. Moreover, responses of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) in a rodent malaria model to a 4-day administration of Clerodendrum violaceum leaf extracts were also evaluated. The methanolic extract was found to contain the highest amounts of antioxidant compounds/element and also demonstrated the highest free radical scavenging activity in vitro. The results showed a significant decrease (p < 0.05) in SOD and CAT activities with a concurrent significant (p < 0.05) increase in GPx and GR activities in both erythrocytes and liver of untreated Plasmodium berghei NK65-infected animals compared to the uninfected animals. The extracts were able to significantly increase (p < 0.05) SOD and CAT activities and significantly reduce (p < 0.05) GPx and GR activities in both the liver and erythrocytes compared to those observed in the untreated infected animals. The results suggest the augmentation of the antioxidant system as one of the possible mechanisms by which Clerodendrum violaceum extract ameliorates secondary effects of malaria infection, alongside its antiplasmodial effect in subjects.

인삼 지용성분획의 고혈당 강하작용에 관한 연구 (Study on the Hypoglycemic Action of the Fat Soluble Fraction of Panax ginseng C.A. Meyer in Streptozotocin Induced Diabetic Rats)

  • We attempted in this study to understand the hypoglycemic action of the fat soluble fraction of red ginseng roots in streptozotocin injected diabetic rats, through its actions on several enzymes relating to carbohydrate metabolism of the 1eve1 to compare with those of ginsenosides in streptozotocin injected diabetic rats. It was realized that the increased level of glucose, ketone bodies, lactate, nonesterified fatty acids and triacylglycerol in blood was significantly decreased and the decreased liver glycogen content of streptozotocin injected rats were appreciably moderated by intraperitoneal injection of the fat soluble fraction of red ginseng roots as shown in the saponin injected diabetic rats. The deceased activities of liver enzymes relating to carbohydrate metabolism such as phosphofructokinase, glucokinase, glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase and acetyl CoA carboxylase of streptozotocin induced diabetic rats were also sufficiently modified by the intraperitoneal injection of the above fat soluble fraction as shown in the ginsenoside injected streptozotocin induced rats.
