Effect of Propolis on the Activity of Antioxidant Enzymes in Rat Liver Irradiated by X-ray

  • We investigated the effect of propolis on the activity of antioxidant enzymes in rat liver exposed by X-ray irradiation. The dosage of propolis showed the effect of lowering the concentration of superoxide anion in irradiated rat liver, suggesting that propolis has a significant role to remove superoxide anion as an antioxidant and/or by activating the antioxidant enzyme. The activities of superoxide dismutase (SOD) and glutathione reductase (GR), disturbed by X-ray irradiation, were restored in 30 days to normal status in the group which dosed propolis before X-ray irradiation. Interestingly, catalase (CAT) and glutathione peroxidase (GPOX) activities were highly increased with feeding propolis to rat compared to untreated group, whereas glutathione s-transferase (GST) activity was little affected. Taken together, it suggests that the propolis has a protective role in the rat liver cells against X-ray irradiation by increasing and recovering the activities of antioxidant enzymes.

Characterization of 1,4-Benzoquinone Reductase from Bovine Liver

  • 1,4-Benzoquinone reductase was purified to electrophoretic homogeneity from bovine liver, and the purified enzyme found to have a molecular mass of 29 kDa, as determined by sodium dodecyl sulfate- polyacrylamide gel electrophoresis The enzyme exhibited pH optimum between 8.0 and 8.5. The apparent fm for 1,4-benzoqulnone was 1.643 mM, and the apparent Km for NADH was 1.837 mM. Various divalent cations, such as Hg$\^$2+/, Cu$\^$2+/, and Zn$\^$2+/, exhibited strong inhibitory effects. The enzyme activity was also strongly inhibited by quercetin, dicumarol, and benzoic acid. Incubation of the enzyme with N-bromosuccinimide and pyridoxal 5’-phosphate led to inhibitions of 100% and 99%, respectively. Accordingly, these results suggest that trypto-phan and Iysine residues are Involved at or near the active sites of the enzyme.

청간산(淸肝散)이 흰쥐의 알코올 대사(代謝) 효소(酵素) 활성도(活性度)에 미치는 영향(影響) (Effects of Chunggansan on Detoxication of Alcohol by Activity of Enzyme in Rats)

  • Chunggansan was tested for the effects on detoxication mechanism of alcohol. Chunggansan was treated firstly into samples, and then ethanol intoxicated animal models were set with them. The administration of Chunggansan to the rats increased proportionally in alcohol dehydrogenase activities in liver in relation to the level of concentration and days of treatment. Especially, the alcohol dehydrogenase was the most active when the concentration of extract was 200mg/kg and it was 7th day. The enzyme activities of alcohol dehydrogenase and aldehyde dehydrogenase in liver highly increased in Chunggansan pre-medicating group compared to that of ethanol treated group. Also, the blood ethanol concentration in rats was considerably decreased. In conclusion, Chunggansan recovers the damage of liver due to acute alcohol intoxication by the increased enzyme activities of alcohol dehydrogenase and aldehyde dehydrogenase.

흰쥐의 간(肝) 조직(組織)에서 녹용(鹿茸) 약침(藥針) 제제(製劑)의 항산화작용(抗酸化作用)에 관(關)한 연구(硏究) (Effects of Cervus elaphus for herb-acupuncture solution on Antioxidation in Rat‘s liver)

  • Cervus elaphus for herb-acupuncture solution(CEHAS) was tested for the effects of free radical generating enzyme and lipid peroxidation in rat's liver. In vitro, levels of lipid peroxide in tissues of liver were proportionally decreased to concentration of CEHAS. They were much more decreased. when lipid peroxidation was induced with ferrous iron $(Fe^{-2})$. Also, enzyme activities of xanthine oxidase were decreased. The ratio of type conversion of xanthine oxidase was lowered, too. But, here was not special changes on enzyme activities of aldehyde oxidase. These results suggest that CEHAS decrease the activities of free radical generating enzymes such as xanthine oxidase which form lipid peroxide.

  • In order to utilize marine processing waste which would normally be discarded, cod liver protein was hydrolysed by ${\alpha}$-chymotrysin, and the hydrolysate was investigated for the new angiotensin I converting enzyme (ACE) inhibitor. Thy hydrolysate was separated into three major types, with molecular weight cut-off (MWCO) values less than 10 kDa, 5 kDa and 1 kDa of ultrafiltration membranes, respectively. ACE inhibitory peptides were isolated from the fractions passed through MWCO 1 kDa membrane, and purified by using ion-exchange chromatography on a SP-Sephadex C-25 column, gel filtration on a Sephadex G-15 column, and HPLC on an ODS column. The purity was identified with capillary electrophoresis. The amino acid sequences of two peptides were Met-Ile-Pro-Pro-Tyr-Tyr (IC50=10.9 ${\mu}$M) and Gly-Leu-Arg-Asn-Gly-Ile (IC50=35.0 ${\mu}$M)

솔잎추출물이 고지방식이를 급여한 흰쥐의 혈청, 간장의 효소 및 간조직구조에 미치는 영향 (Effects of Pine Needle Extracts on Enzyme Activities of Serum and Liver, and Liver Morphology in Rats Fed High Fat Diet)

  • The effects of pine needle extracts on lipid contents and antioxidative enzyme activities in liver of rat were evaluated. Thirty male Sprague-Dawley rats were divided into five groups and fed high fat diets for four weeks. Each group was administered with pine needle extract in the following doses: control, water ; WE-3, hot water extract(3% PN) ; WE-6, hot water extract(6% PN) ; AE-3, acetone extract(3% PN) ; AE-6, acetone extract(6% PN). The results obtained from the experiment were as follows: GOT activities were not significantly different among experimental groups but GPT activities were significantly low in the experimental groups compared to the control group. Liver superoxide dismutase(SOD) activity of pine needle extract administered groups was higher than that of control group. Catalase activities of liver had a similar tendency to SOD activities, but were not significantly different among the groups. Liver TBARS of WE-3 WE and AE-6 groups were slightly lower than those of other groups. Microscopic observation of liver tissue revealed that pine needle extracts increased cellular swelling, which was markedly increased in WE-6 group compared with control group.

Purification of Aldose Reductase and Decolorization of Dye by the Enzyme

  • Aldose reductase was purified to electrophoretic homogeneity from porcine liver. The purified enzyme was a monomer of 36 kDa. The enzyme was strongly inhibited by $Cu^{2+}\;and\;Mg^{2+}$ ions. Incubation of the enzyme with pyridoxal 5'-phosphate led to complete inhibition of enzymatic activity, suggesting that lysine residue is involved at or near the active site of the enzyme. The enzyme exhibited a broad substrate specificity. Furthermore, the enzyme was capable of decolorizing Alizarin, an anthraquinone dye.

  • 오징어 가공 부산물인 오징어 간의 효율적 이용을 위하여 오징어 간을 이용하여 액젓을 제조하고 이들 액젓의 ACE 저해효과를 살펴보았다. 액젓의 ACE 저해활성은 12개월까지는 점차적으로 증가하였으나 그 이후에는 저해활성이 둔화되어 거의 일정한 저해활성을 유지하였다. 숙성 액젓 중 15개월째 액젓($IC_{50}=29.66\;{\mu}g$)을 한외여과막으로 통과시켜 회수한 분자량 10,000 Da 이하의 저분자물질을 Bio-gel P-2 gel chromatography를 행하여 ACE 저해효과를 가지는 3개획분을 분취하였다. 이들 획분 중에서 ACE 저해효과가 가장 높은 B 획분을 SuperQ-Toyopearl 650S column을 이용한 음이온 교환크로마토그래피에 의해 B-1의 활성획분을 분리하였다. 획분 B-1의 아미노산 조성은 lysine, glycine 및 proline의 함량이 가장 많아 전체의 약 85%를 차지하였으며 $IC_{50}$$5.46\;{\mu}g$으로 나타났다.

고지방 섭취 흰쥐에서 대두 이소플라본 섭취가 혈액 내 Nitrite 함량과 항산화 효소 활성에 미치는 영향 (Effects of Soy Isoflavone Intake on Nitrite Content and Antioxidant Enzyme Activities in Male Rats Fed High-Fat Diet)

  • This study was performed to investigate the effect of soy isoflavone on plasma nitrite concentration and the antioxidant enzyme activities of erythrocyte and the liver using adult male rats fed high fat diet. Seven-week old male Sprague Dawley rats were divided into three groups and fed high fat diet (15% beef tallow, 1 % cholesterol; control: IF0) or high fat diets containing isoflavone 80 ppm (IF80) or 320 ppm (IF320) for 10 weeks. Plasma nitrite concentration as a vasodilator, and antioxidant enzyme activities in erythrocytes and the liver were measured. Plasma nitrite concentration was increased by 45% and 35%, respectively, in IF80 and IF320 than in IF0 group. Erythrocyte catalase, glutathione peroxidase (GPx) and glutathione reductase (GR) activities increased by 31 %, 30% and 40% in IF320 compared to IF0 group. Especially, erythrocyte GR activity increased by 61 % in IF80 group. However, catalase activity in the liver was decreased in IF80 group. GPx and GR activities in the liver were not differ among groups. The results suggest that soy isoflavone have the protective effect against risk factors related with cardiovascular disease by improving vasodilator factor, nitrite, and antioxidant enzyme activities in blood. Effects of Dietary Fatty Acid and Protein Sources on Serum Protein Profiles and Liver Functional Enzyme Activities in Rats with DMBA-Induced Mammary Tumors

  • This study was conducted to examine the effects of dietary sources of fatty acids and protein on serum protein profiles, hepatic functional enzyme activities, mammary tumor incidence and tumor weight in 7, 12-dimethylbenz($\alpha$)anthracene (DMBA)-treated rats. The sources of dietary fatty acids were 18n6 (rich in linoleic acid), 18n3 (rich in linolenic acid) and 22n3 (rich in DHA) : sources of dietary protein were casein (C) and soy protein isolate (S). mammary tumors (MTs) were chemically induced by DMBA (9 mg/100 g body weight) which was gastrically intubated at 7 weeks of age. Each experimental diet was given for the following 25 weeks. Casein-fed rats (group C) exhibited significantly higher levels of weight gain and FER (food efficiency ratio) than did group S. Group C showed higher levels of serum protein and globulin, and higher albumin/globulin (A/G) ratios than group S. Liver functional enzyme activities (GOT, GPT, ALP, LDH, $\gamma$-GT) and LDH/GOT ratios were not influenced by dietary protein. GPT activity was lower in the group given 18n3, and ALP activity was lower in the group given 18n6. The incidence and total number of MTs appeared to be lower in the group given 22n3 than in the group given 18n3 or 18n6, even though the average weight of MTs was highest in the group given 22n3, The average weight of MTs was higher in the C group than in the S group. MT incidence had a positive correlation with LDH activity and LDH/GOT ratio. The average weight of MTs had a negative correlation with serum albumin levels and A/G ratios, and a positive correlation with ALP activity. This research suggests that the measurement of serum protein profiles and liver functional enzyme activities may be utilized to monitor the development of mammary tumors.
