Effect of the Acute Phase Response on the Performance and Superoxide Dismutase Activity in Broiler Chicks Fed on Dietary Krill Meal (사료 중 크릴 밀을 급여한 육계의 생산성과 SOD 활성에 미치는 급성기 반응의 영향)

  • Two experiments were conducted to examine the effects of the acute phase response on the performance and superoxide dismutase(SOD) activities in liver and erythrocyte of broiler chicks fed dietary krill meals A and B in experiment 1 and krill meal A in experiment 2. The experimental diets are basal diet based on yellow corn and soybean meal and diets substituted 2.0% of krill meal A or B with soybean meal of the basal diet, respectively. Day-old birds fed on the experimental diets and the acute phase response(immunological stress) was activated in the birds on 8-day of age by alternate day injection i.p. with 3 doses the Salmonella typhymurium lipopolysaccharide(LPS) in saline. The values during the acute phase response were compared with those controls injected with saline. The performance; daily gain, feed intake, and feed efficiency were different between dietary krill meal A and B in birds during the acute phase response and in the control. The acute phase response increased relative liver and spleen weights. Recovery of birds from the immunological stress was different between krill meals. Dietary krill meals increased activities of MnSOD and Cu/ZnSOD in erythrocyte cytosols during the actute phase response. Dietary krill meals did not affect the PHA-p response. The results indicated that the dietary krill meals may accentuate oxidative stress during the acute phase response.

Biological Activity of Organic Solvent Fractions from Lythrum salicaria L. (Root) (털부처꽃 뿌리로부터 조제된 분획물의 생리활성)

  • Root extract of Lythrum salicaria reported a hepato-protective effect on $CCl_4$-induced liver toxicity of rat was prepared into fractions such as n-hexane up layer (HA), n-hexane down layer (HB), diethyl ether (E), ethylacetate (EA), n-butanol (B) and water (W). Fractions prepared were tested their activities in vitro and in vivo condition. All of the fractions showed effective antioxidant asctivities on DPPH radical and $CuSO_4$-induced oxidation of human low density lipoprotein and E fraction showed the highest inhibitory effect (98.1% at $50\;{\mu}g/m{\ell}$) on linoleic acid autoxidation at $40^{\circ}C$, which was more effective than $\alpha$-tocopherol (82.4%). Five fractions (H = HA plus HB, E, EA, B, and W, 150 mg/kg/day) were fed into Sprague Dawley, male rats for 4 days, which were intoxicated with intra-peritoneal injection of carbon tetrachloride ($1\;m{\ell}/kg$ in corn oil) at the 4th day and were sacrificed in 24 hrs. Serum tumor necrosis factor-alpha (TNF-$\alpha$), a proinflammatory cytokine, elevated with $CCl_4$-intoxication in negative control group ($83\;pg/m{\ell}$) was significantly decreased in E fraction-supplemented group ($18\;pg/m{\ell}$). Cu, Zn-superoxide dismutase (SOD) activity increased in negative control group (0.12 U/mg protein) was decreased in E fraction (0.07 U/mg protein). From the results, it is suggested that ether fraction from root extract of L. salicaria would be a potent antioxidant candidate for ameliorating liver injury induced by chemical intoxicant.

Effect of Maengjong-Juk Extract Coated Rice Supplementation on Antioxidative System in Rabbit Fed High Cholesterol Diet (맹종죽 추출물 코팅쌀 식이가 고콜레스테롤 섭취 토끼의 항산화 시스템에 미치는 영향)

  • To evaluate the antioxidative effect of rice coated with maengjong-juk extract in vivo system, rice coated with maengjong-juk extract diets were fed to NZW rabbit for 16 weeks and lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content in tissues were measured. TBARS contents in liver and spleen were significantly decreased in maengjong-juk extract diet group compared to control group, while those in kidney and heart tissue were not significantly different. Maengjong-juk extract diet suppressed the protein oxidation significantly in liver, spleen, kidney and heart tissues. Hepatic total SOD, Cu$.$Zn-SOD and Mn-SOD activities of maengjong-juk extract diets were significantly higher than those of control diet. GSH-Px and catalase activities of maengjong-juk extract diet were higher than those of control, while GR activities show no significant difference between the two groups. Total hepatic glutathione content was significantly increased by maengjong-juk extract diet. According to this study, many antioxidative materials and phytochemicals in maengjong-juk extracts seems to protect tissues from oxidative stress by stimulating antioxidative systems in atherosclerotic rabbit fed high cholesterol diet.

The Degree of Lipid Oxidation of Rat Liver Fed Peroxidized Lipid and Its Effects on Anti-Oxidative System (과산화지질의 투여가 흰쥐 간의 산화와 항산화계에 미치는 영향)

  • Accumulation of peroxidized lipid, fed or injected in the body of rats was investigated and the effect of peroxidized lipid on the antioxidative system was studied also. Three groups each having six of Sprague-dawley rats were raised for 8 weeks. the peroxide value(POV) of diet fed to the control and the peroxidized group was 5.47 and 22.14meq/kg , respectively. Injected group was given the control diet and peroxidized linoleic acid(POV 31.81meq/kg) was injected into the peritoneal area three times a week. The POV, MDA, and protein carbonyl values of the peroxidized and the injected group (experimental groups) were significantly higher (p<0.05) than those of the control group. Cu, Zn-SOD and M-SOD activity of the experimentla groups increased 1.6 times that of control group at 4 th week. and decreased by 60% of their activityafter 8 weeks of feeding (p<0.05) . Catalase activity, glutathione and Vt, E contents of the experimental groups were significantly lower (p<0.05) than those of the control group during 8 weeks. The accumulation of peroxidcized lipid in liver were ovserved both in the fed or the injected group. The increased of enzyme activity of the experimental group during 4 weeks suggests ianadaptation of antioxidative system to get rid of the peroxidized lipid. Decrease of enzyme activity and glutathione observed as the peroxidized lipid lipid accumulation proceeded further, however, seems to indicate the oxdiative damage of enzyme and protein . Determination of the protein carbonyl content may be used as a method for measuring the oxidative damaging effect of peroxidized lipid.

Cloning of Human Liver Cytosolic Sialidase from Genomic DNA Using Splicing by Overlap Extension and Its Characterization

  • Cytosolic sialidase (Neu2), a member of the sialidase family that is responsible for hydrolysis of sialic acid from the terminal position of sialoglycoconjugates, is poorly expressed in skeletal muscle and not detected in any other adult tissues. Thus, we isolated Neu2 cDNA using splicing by overlap extension (SOEing). In order to further characterize this enzyme, a His-tagged derivative was expressed in the bacterial expression system and purified by $Ni^{2+}$-affinity chromatography. A recombinant product of approximately 42 kDa had sialidase activity toward 4-methyl-umbelliferyl-$\alpha$-D-N-acetylneuraminic acid (4MU-NeuAc). The optimal pH and temperature of the recombinant Neu2 for 4MU-NeuAc was 6.0 and $37.5^{\circ}C$, respectively. The metal ions, such as $Cu^{2+}\;and\;Cd^{2+}$, showed strong inhibitory effect on the activity of the enzyme. The enzyme efficiently hydrolyzed the gangliosides GM3 and GD3 and had relatively low activities on ganglioside GD1a and GD1b, $\alpha$2-3 sialyllactose, and sialylated glycoproteins such as fetuin, transferrin, and orsomucoid, but had hardly any activities on $\alpha$2-6 sialyllactose and ganglioside GM1 and GM2. We concluded that the recombinant Neu2 has a sialidase activity toward glycoproteins as well as gangliosides.

EFFECTS OF RARE EARTH NITRATE ON INTERNAL ORGANS AND MINERAL ELEMENTS IN THE SERUM OF BROILER CHICKENS

  • This paper reports the effects of rare earth nitrate (REN) on the growth of internal organs and mineral elements in serums of broiler chickens. The REN used is mixture of light RE containing mainly La, Ce, Pr and Nd 4 elements. 40 eight-week old AA broiler chickens were divided randomly into four groups that their treatments were respectively 0, 20, 200 and 2000 mg REN/kg feed and the duration of the experiment was 60 days. Responses were measured in terms of internal organic weight and tissue, contents of mineral elements in serums. There were no significant differences between the organ ratios and tissue changes of the 20, 200 mg REN/kg groups and the control group. The organs included the heart, liver, kidneys, lungs, testicles, thyroids, adrenal glands, pancreas, tonsies of caecum, stomachus glandularis, duodenum, ileum and bursa of Fabricius. The liver and thyroid indices of 2000 mg REN/kg group were significantly higher than those of the control group (p < 0.01, p < 0.05) and the adrenal gland index lower (p < 0.05). The pathlolgic changes in the heart, kidney and thyroid were more serious than that of the control group. No significant differences occurred between the contents of K, Na, Mg elements in the serum of all groups, with the exception of the content of Ca, which was higher in the 2000 mg/kg group (p < 0.05). The contents of all trace elements including Mn, Zn and Cu, but except Fe, in the serums of all the treated groups were significantly higher than those of the control group (p < 0.01).

  • Four isoforms of calcium binding proteins containing 2 EF hand motifs and a dynein light chain-like domain in the human liver fluke Opisthorchis viverrini, namely OvCaBP1, 2, 3, and 4, were characterized. They had molecular weights of 22.7, 21.6, 23.7, and 22.5 kDa, respectively and showed 37.2-42.1% sequence identity to CaBP22.8 of O. viverrini. All were detected in 2- and 4-week-old immature and mature parasites. Additionally, OvCaBP4 was found in newly excysted juveniles. Polyclonal antibodies against each isoform were generated to detect the native proteins in parasite extracts by Western blot analysis. All OvCaBPs were detected in soluble and insoluble crude worm extracts and in the excretory-secretory product, at approximate sizes of 21-23 kDa. The ion-binding properties of the proteins were analyzed by mobility shift assays with the divalent cations $Ca^{2+}$, $Mg^{2+}$, $Zn^{2+}$, and $Cu^{2+}$. All OvCaBPs showed mobility shifts with $Ca^{2+}$ and $Zn^{2+}$. OvCaBP1 showed also positive results with $Mg^{2+}$ and $Cu^{2+}$. As tegumental proteins, OvCaBP1, 2, and 3 are interesting drug targets for the treatment of opisthorchiasis.

Characterization of Copper/Zinc-Superoxide Dismutase (Cu/Zn-SOD) Gene from an Endangered Freshwater Fish Species Hemibarbus mylodon (Teleostei; Cypriniformes)

  • Gene structure of copper/zinc-superoxide dismutase (Cu/Zn-SOD; sod1) was characterized in Hemibarbus mylodon (Teleostei; Cypriniformes), an endangered freshwater fish species in Korean peninsula. Full-length cDNA of H. mylodon SOD1 consisted of a 796-bp open reading frame sequence encoding 154 amino acids, and the deduced polypeptide sequence shared high sequence homology with other orthologs, particularly with regard to metal-coordinating ligands. Genomic structure of the H. mylodon sod1 gene (hmsod1; 1,911 bp from the ATG start codon to the stop codon) was typical quinquepartite (i.e., five exons interrupted by four introns); the lengths of the exons were similar among species belonging to various taxonomic positions. The molecular phylogeny inferred from sod1 genes in the teleost lineage was in accordance with the conventional taxonomic assumptions. 5'-flanking upstream region of hmsod1, obtained using the genome walking method, contained typical TATA and CAAT boxes. It also showed various transcription factor binding motifs that may be potentially involved in stress/immune response (e.g., sites for activating proteins or nuclear factor kappa B) or metabolism of xenobiotic compounds (e.g., xenobiotic response element; XRE). The hmsod1 transcripts were ubiquitously detected among tissues, with the liver and spleen showing the highest and lowest expression, respectively. An experimental challenge with Edwardsiella tarda revealed significant upregulation of the hmsod1 in kidney (4.3-fold) and spleen (3.1-fold), based on a real-time RT-PCR assay. Information on the molecular characteristics of this key antioxidant enzyme gene could be a useful basis for a biomarker-based assay to understand cellular stresses in this endangered fish species.

High Vitamin E Supplement is Needed to Have an Anticarcinogenic Effect of Fish Oil (어유와 비타민 E 보강 수준이 쥐간의 전암성 병변에 미치는 영향)

  • The influences of fish oil and different levels of vitamin I supplement on hepatocellular chemical carcinogenesis have been studied. Male Sprague-Dawley rats received diethylnitrosamine (DEN)(200mg/kg body weight) and were subjected to two-thirds partial hepatectomy to induce murine chemical hepatocarcinogenic procedure. Placental glutathione S-transferase(GST-P) positive foci area, antioxidant enzymes(Cu/Zn-superoxide dismutase(SOD), catalase, glutathione reductase (GR), total- glutathione peroxidase (TGPx), glutathione S -transferase (GST)), glucose 6-phosphatase (G6Pase) activities, and lipid peroxidation of microsomes(thiobarbituric acid reactive substances (TBARS)) were measured. Experimental animals were fed 15% corn or fish oil with 0, 40, 1,000, 10,000IU vitamin E /kg diet for 8 weeks. Vitamin E supplements decreased the area of GST-P positive foci in both groups. The higher the vitamin E levels, the smaller the area of GST-P positive foci were noticed. Compared to 0 IU vitamin E, 40 IU in corn oil and 1,000 IU in fish oil groups were effective in decreasing G57-P positive foci area. Fish oil groups tended to have smaller area of GST-P positive foci. fish oil groups showed lower body weight, lower activities of Cu/Zn-SOD and TGPx, higher TBARS contents, higher activities of GST, catalase, G6Pase, GR and higher liver/body ratio than corn oil groups. As the level of vitamin I increased, GST-P positive foci count, catalase activities, and TBARS tended to decrease. G6Pase activities tended to increase in both groups. At higher vitamin E levels, GST activities tended to decrease in fish oil groups. These results suggest that vitamin I has suppressive offects on hepatocellular chemical carcinogenesis probably through antioxidant eH:cts decreasing TBARS contents, $H_2O$$_2$, and organic peroxides. fish oil tended to have greated suppressive offects than corn oil on hepatocellular carcinogenesis. A proton induced X-ray emission (PIXE) analysis of concentration of major/trace and toxic elements in broiler gizzard and flesh of Tehsil Gujar Khan area in Pakistan

  • Ten gizzard and three flesh samples of the broiler were collected from different locations in Tehsil Gujar Khan District Rawalpindi, Pakistan. The samples were dried, crushed and ground. Pellets were prepared by pressing the powder of the samples and that of the Bovine liver 1577c reference material obtained from NIST, USA. Proton induced X-ray emission (PIXE) installed at National Center for Physics, Islamabad, Pakistan has been used as a reliable and improved technique to determine concentration of various major/trace and toxic elements e.g. S, Cl, K, Ca, Cl, Fe, Cu, Mn, Co, Zn, Ti, Cd, Ga, Cr, V and Ni, in the Gizzard and Flesh samples of the broiler. The concentrations of all the detected elements in the samples are statistically significant. The certified and measured values of the elements in the reference material were in agreement with each other within a deviation of 7%. S, Cl, K and Ca are within tolerable limits and are good for human consumption. Ti, V, Cr, Mn, Fe, Co, Ni, Cu and Zn were more than the acceptable limits of World Health Organization, WHO whereas Ga, As, Sn, Sb and Pb are not detected in most of the samples.
