The Effect of Variety and Growing Conditions on the Chemical Composition and Nutritive Value of Wheat for Broilers

  • The aim of this study was to examine the effect of variety and growing conditions of wheat on broiler performance and nutrient digestibility. One hundred and sixty-four wheat samples, collected from a wide range of different sources, locations, varieties and years, were analyzed for a range of chemical and physical parameters. Chemical and physical parameters measured included specific weight, thousand grain weight (TG), in vitro viscosity, gross energy, N, NDF, starch, total and soluble non-starch polysaccharides (NSP), lysine, threonine, amylose, hardness, rate of starch digestion and protein profiles. Ninety-four of the wheat samples were selected for inclusion in four bird trials. Birds were housed in individual wire metabolizm cages from 7 to 28 d and offered water and feed ad libitum. Dry matter intake (DMI), live weight gain (LWG) and gain:feed were determined weekly. A balance collection was carried out from 14 to 21 d for determination of apparent metabolizable energy (AME), ME:gain, DM retention, oil and NDF digestibility. At 28 d the birds were sacrificed, the contents of the jejunum removed for determination of in vivo viscosity and the contents of the ileum removed for determination of ileal DM, starch and protein digestibility. The wheat samples used in the study had wide-ranging chemical and physical parameters, leading to bird DMI, LWG, gain:feed, ME:GE, AME content and ileal starch and protein digestibility being significantly (p<0.05) affected by wheat sample. A high level of N fertilizer application to the English and NI wheat samples tended to benefit bird performance, with increases of up to 3.4, 7.2 and 3.8% in DMI, LWG and gain:feed, respectively. Fungicide application also appeared to have a positive effect on bird performance, with fungicide treated (+F) wheat increasing bird DMI, LWG and gain:feed by 6.6, 9.3 and 2.7%, over the non-fungicide treated (-F) wheats. An increase (p<0.1) of 9.3% in gain:feed was also observed at the low seed rate of 40 compared to 640 seeds/$m^2$. It was concluded that the type of wheat sample and environmental growing conditions significantly affects bird performance when fed wheat-based diets.

Management and Development of Seed Rearing in Red Spotted Grouper Epinephelus akaara (양식 붉바리(Epinephelus akaara) 종자의 개발과 사양관리)

  • We investigated seed management and feeding regime of larval red spotted grouper Epinephelus akaara using live food organisms. Fertilized eggs were produced in March 2020 by manipulating photoperiod and water temperature. Broodstock (F0) produced approximately 0.30×106 eggs, of which 0.25×106 were fertilized; first-generation broodstock (F1) produced 2.00×106 eggs, of which 1.90×106 were fertilized. Larvae were reared at temperatures of 21.0-24.5℃. The feeding regime consisted of rotifers Artemia, and commercial pellet feed. From 5-8 days after hatching (dah), rotifers collected from high-density incubation tanks were fed to grouper larvae at a density of 10-20 individuals/mL. From 9-30 dah, rotifers collected from low-density incubation tanks were fed to larvae at a density of 10-20 individuals/mL. Newly hatched Artemia were provided from 20-24 dah and enriched Artemia from 25-50 dah. Commercial pellet feed was given starting at 13 dah. The average mouth size in hatchlings was 263.4±3.7 ㎛ at 5 dah, 406.7±21.0 ㎛ at 11 dah, and 856.9±136.6 ㎛ at 21 dah. At 1 dah, larvae had a mean total length of 1.95±0.06 mm. At 66 dah, the mean TL was 4.0 cm. The final number of surviving red spotted grouper larvae was 218,380 fishes.

Isolation and Characterization of Paralytic Shellfish Poison Detoxification Bacteria (마비성패류독의 생물학적 제독 -1. 마비성패류독 분해세균의 분리 및 세균학적 특성-)

  • For the establishment of biodetoxification method which can be acceptable for live bivalves, paralytic shellfish poison (PSP) detoxification bacteria were isolated from sea water and bivalves, and PSP detoxification activity and optimal growth condition of the isolated strains were investigated. from the bivalve and sea water samples, 8 strains of PSP detoxification bacteria were isolated. Of the isolated strains, CW-6 isolated from sea water shown strong PSP detoxification activity and decomposed completely 18 nmole/g of GTX2 after 3 days incubation in artificial medium. The selected stain CW-6 shown typical characteristics of the Enterobacter sp. and identified as Enterobacter sp, CW-6. Optimal growth condition of the Enterobacter sp. CW-6 were $35^{\circ}C$, pH 7 and $NaCl 1{\%}$, respectively.

Shigellosis

  • Shigellosis is a global human health problem. Four species of Shigella i.e. S. dysenteriae, S. flexneri, S. boydii and S. sonnei are able to cause the disease. These species are subdivided into serotypes on the basis of O-specific polysaccharide of the LPS. Shigella dysenteriae type 1 produces severe disease and may be associated with life-threatening complications. The symptoms of shigellosis include diarrhoea and/or dysentery with frequent mucoid bloody stools, abdominal cramps and tenesmus. Shigella spp. cause dysentery by invading the colonic mucosa. Shigella bacteria multiply within colonic epithelial cells, cause cell death and spread laterally to infect and kill adjacent epithelial cells, causing mucosal ulceration, inflammation and bleeding. Transmission usually occurs via contaminated food and water or through person-to-person contact. Laboratory diagnosis is made by culturing the stool samples using selective/differential agar media. Shigella spp. are highly fragile organism and considerable care must be exercised in collecting faecal specimens, transporting them to the laboratories and in using appropriate media for isolation. Antimicrobial agents are the mainstay of therapy of all cases of shigellosis. Due to the global emergence of drug resistance, the choice of antimicrobial agents for treating shigellosis is limited. Although single dose of norfloxacin and ciprofloxacin has been shown to be effective, they are currently less effective against S. dysenteriae type 1 infection. Newer quinolones, cephalosporin derivatives, and azithromycin are the drug of choice. However, fluoroquinolone-resistant S. dysenteriae type 1 infection have been reported. Currently, no vaccines against Shigella infection exist. Both live and subunit parenteral vaccine candidates are under development. Because immunity to Shigella is serotype-specific, the priority is to develop vaccine against S. dysenteriae type 1 and S. flexneri type 2a. Shigella species are important pathogens responsible for diarrhoeal diseases and dysentery occurring all over the world. The morbidity and mortality due to shigellosis are especially high among children in developing countries. A recent review of literature (KotIoff et al.,1999) concluded that, of the estimated 165 million cases of Shigella diarrhoea that occur annually, $99\%$ occur in developing countries, and in developing countries $69\%$ of episodes occur in children under five years of age. Moreover, of the ca.1.1 million deaths attributed to Shigella infections in developing countries, $60\%$ of deaths occur in the under-five age group. Travellers from developed to developing regions and soldiers serving under field conditions are also at an increased risk to develop shigellosis.

Study on the Reproductive and Developmental Toxicity of 3-MCPD (3-MCPD의 생식ㆍ발생독성에 관한 연구)

  • 3-Monochloro-1,2-propanediol(3-MCPD) is a toxic compound, often present in different foods containing acid hydrolyzed(AH) protein, like seasonings and savory food products. The purpose of the present study was to investigate the effects of 3-MCPD on male fertility, sperm and testosterone secretion. In vivo male fertility test was performed for observing the adverse effects of 3-MCPD on the function of male reproductive system and pregnancy outcome. 0.01, 0.05, 0.25, 1 and 5 mg/kg b.w. of 3-MCPD was given daily by gavage to groups of 15 adult male SD rats for 4 weeks. At the end of pre-treatment period, males were mated overnight with normal females. Following morning, males demonstrating successful induction of pregnancy were sacrificed on that day to assess sperm parameters and histopathology of reproductive organs. The resulting pregnant females were sacrificed on day 20 of gestation to evaluate pregnancy outcome. As a result, four-week paternal administration with 3-MCPD resulted in adverse effects on male fertility and pregnancy outcome without remarkable histopathological changes in testes and epididymides; sperm motility, copulation index and fertility index were markedly decreased in the treated group and numbers of live fetuses showed steep dose-response curves. Also, spermatogenesis was investigated in this experiment. However, no effect was observed on production of sperm in testes treated with 3-MCPD for 4 weeks. Hormone assay was performed for observing the effects of 3-MCPD on testosterone and luteinizing hormone (LH) in blood and testes of male SD rats and cultured primary Leydig cell. In result, significant changes of related hormones did not observed by treatment of 3-MCPD. These results indicated that paternal treatment with 3-MCPD induced spermatotoxic effect, which caused an antifertility on male.

  • The diseased larvae of the flounder, Paralichthys olivaceus, were sampled several times from a fish hatchery located in Cheju Island between December, 1991 and April, 1992. They turned out to be infected with Vibrio sp. and diagnosed as intestinal necrosis of flounder larvae(INFL) based on morphological, biological, and biochemical examinations. INFL was known to be caused by the live food organism infected with Vibrio sp. The optimal growth conditions of the isolates for temperature, pH, and NaCl concentration were $20\sim30^{\circ}C$, 6~8, and 2~4%, respectively. In the drug sensitivity test, the isolates were sensitive to oxytetracycline, nalidixic acid, kanamycin, and novobiocin, but resistant to ampicillin, erythromycin, spiramycin and sulfa-drug.

Nutrient Intake of the Rural Elderly Living in Kyungnam -Focusing on Health and Aging Status, and Life-Satisfaction- (경남 일부지역 농촌노인의 영양섭취조사 -건강과 노화상태 및 생활만족도를 중심으로 -)

  • The purpose of this study was to investigate the difference in nutrient intake according to the level of self-perception of health status, aging status and life satisfaction of the rural elderly. The factors for the study were surveyed by interview method. The subjects were 270 people(71 male, 129 female) aged over 65 years(73.5 $\pm$ 5.6ys) in the Ham-an area. The obtained results as follows : By evaluation of self-perception of health status, 57.5% of subjects answered they are in a bad health condition. The 91.5% of subjects had diseases(rheumatitis & arthritis 31.4%, cardiovascular disease 20.2%, gastric disease 10.2%). The women had more diseases than the men(p < 0.01). The subjects took medical treatment in private hospital(40.5%) and public health centers(35.0%). The men showed better level of aging status(p < 0.001) and life satisfaction index(p < 0.01) than the worsen. Living with spouse influenced the aging status(p < 0.05) and the more pocket money influenced life satisfaction(p < 0.05) and aging status(p < 0.05). The elderly who eat regularly 3 times a day(p < 0.05) and have a good appetite(p < 0.001) appeared to have positive effect on the self perception of health status and aging status. An increasing level of the self-perception of health status and regular exercise worked to improve aging status(p < 0.001). The habits of smoking and alcohol drinking, however had no effect on any index. The self-perception of health status affected the nutrient intake, but only in female elderly. The aging status and the life satisfaction index related overall positively to the intake of nutrients. In conclusion, the study shows that gender did influence nutrient intake in the elderly. The women who live alone rated lowest in social resources and health condition therefore their nutrient intake was also extremely in deficit. For successful aging, a program for rural elderly is needed, i.e. actions to provide minimum economic life, food delivery and psychological/physical health care through regional public health centers.

  • We studied new observation method about take process of rotifer by marine fish larvae. Till now, we can not accurate observation and count of first rotifer feeding day and/or feeding numbers of rotifer by marine fish larvae. Because take rotifer is ingested and disappeared in the digestive system of fish larvae. However we suggest possible observation method for these problems. The trophi is mastication organ of rotifer, and has only one in each rotifer individual. The trophi is left in the mastication organ because sole indigestible organ of rotifer. Therefore we can accurate observation and count of first rotifer feeding day and/or feeding rotifer numbers of marine fish larvae by trophi observation method (RTCM; Rotifer Trophi Counting Method).

Effect of Different Diets on Growth and Survival Rates of Snakehead (Channa striata Bloch, 1797) Larvae

  • A study was conducted to determine growth and survival rates of snakehead (Channa striata) larvae. Live foods such as Artemia salina nauplii, Moina micrura and bloodworm and artificial diet were given to larvae from 1-15 days after hatching as the 1st phase and from 15-30 days as the 2nd phase. In the 1st phase, the growth and survival rates of larvae fed with Artemia nauplii were significantly different from larvae fed with Moina (p < 0.05) with values of 28.5 mg, 49% and 26.7 mg, 31%, respectively. Meanwhile, all larvae fed with bloodworm (Chironomus sp.) or artificial diet (49% protein) died within 10 days of experiment. For the 2nd phase, growth of the fry fed with Artemia or Moina differed significantly from that fed with bloodworm or pellet (200.1, 187.7, 109.6 and 8.2 mg, respectively). Specific growth rate (SGR) of larvae fed with Moina was higher than that of larvae fed with Artemia (17.9 and 17.4% day$^{-1}$, respectively) in the 1st phase. In the 2nd phase, SGRs of larvae fed with Artemia (12.7% day$^{-1}$) or Moina (11.5% day$^{-1}$) were significantly higher than those of larvae fed with bloodworm (8.3% day$^{-1}$) or dry diet (6.1% day$^{-1}$). Generally, Artemia and Moina were suitable food for snakehead larvae during the first month of their life, and artificial diet was much less acceptable and resulted in poor growth and survival.

Fertility and Reproductive & Developmental Toxicity Study on Recombinant Human Epidermal Growth Factor (rhEGF, DWP401) in Rats (재조합 인간상피세포 성장인자(rhEGF, DWP401)가 랫드의 수태능, 태자와 신생자 발달 및 모체기능에 미치는 영향)

  • This study was conducted to investigate for its effects on reproductive and developmental toxicity of recombinant human epidermal growth factor (rhEGF) in Sprague-Dawley rats. Male rats were administered rhEGF at doses of 1, 10, 100, and 1000$\mu$g/kg/day, respective1y, by subcutaneous injection from 63 days before and throughout to mating period until the day before sacrifice. Female rats were administered rhEGF at the same doses from 14 days before mating to day 20 of gestation or to day 21 of lactation. We examined the male and female fertility indices and maternal toxicity of F0 parental animals. Also, we examined the external, visceral, or skeletal malformation of fetuses, growth and development, behavior, and/or reproductive performance of F1 animals. At the highest dose (1,000 $\mu$g/kg), the mean body weights of F0 animals were significantly increased in males and females at 3 or 2 weeks after treatment, respective1y. No clinical signs and food intakes were observed at any time during the experimental period by rhEGF treatment. In autopsy examination, the relative and absolute liver weights significantly increased in both sexes of 1,000 $\mu$g/kg. At the highest dose (1,000 $\mu$g/kg), there was a statistically significant increase of pregnancy period and the number of dead fetuses. Moreover, significant increase of mean fetal body weight and decrease of number of live fetuses, which related to the difficult dilivery were observed in highest dose group. In Fl examination, no adverse effects on external, visceral, and skeletal malformation, physical and functional development, behavior or reproductive ability of Fl animals were observed in any group. Also, there was no significant difference between control and treated groups in copulation or fertility indices of Fl animals. These results indicate that rhEGF had no adverse effect on fertility and reproductive ability of Sprague-Dawley rats.
