자초(Lithospermum erythrorhizon)추출물의 투여가 Galactosamine으로 유도된 간손상에 미치는 보호 효과 (Protective Effect of Lithospermum erythrorhizon on Galactosamine Induced Liver Injury)

  • 본 연구는 galactosamine 투여로 인해 유발된 흰쥐의 간독성에 미치는 자초추출물의 효과를 알아보기 위해 자초 추출물을 각각 200 mg/kg과 500 mg/kg을 매일 1회씩 7일간 복강내 주사한 후, 마지막 자초 추출물 투여일에 galactosamine을 주사하여 다음과 같은 결론을 얻었다. AST, ALT, ALP, GGT와 LDH의 활성도에서는 LE200군이 GalN군에 비해 유의적으로 감소하였다. 그리고 조직학적 변화에서, GalN군은 염증세포의 침윤과 국소적인 괴사성 변화가 나타났지만, LE군들에서는 GalN 투여에 의한 변화가 상당히 경감되었다. 또한 미세구조적 변화에서는 GalN군은 미토콘드리아의 변형, 지방적의 증가, 조면소포체의 분절 등 세포소기관의 변형을 유발하였다. 그러나 LE군들에서는 대조군과 유사한 소견을 보여 주었다. 이상의 결과들로 보아, 자초추출물은 galactosamine 투여로 간독성을 유발시킨 흰쥐의 간 조직에 장애를 주지 않으면서 간독성을 완화시키는 효과가 있다고 사료된다.

자초(Lithospermum erythrorhizon) 추출물이 UVB로 조사된 생쥐 피부에 미치는 영향 (The Effect of Lithospermum erythrorhizon Extracts in UVB-Irradiated Mouse Skin)

  • 본 연구는 UVB 조사로 인해 손상된 피부에 있어서 자초추출물의 유효성을 검증하기 위해 시도되었다. 생후 6주령된 제모한 C57BL/6 마우스를 대상으로 대조군, UVB 조사군(UVB군), UVB조사 후 자초추출물처치군(UVB+Le군)으로 구분하여 24시간, 48시간, 72시간, 120시간, 168시간의 시간대별로 관찰하였다. 경표피수분손실량을 측정한 결과, UVB+Le군이 UVB군 보다 시간이 경과함에 따라 TEWL이 감소하였다. 특히 168시간군에서 유의하게 낮게 나타났다(p<0.05). 멜라닌 양 측정 결과, UVB+Le군이 UVB군 보다 낮게 나타났으나 통계학적으로 유의성은 없게 나타났다(p>0.05). 홍반 지수 측정 결과, UVB+Le group 24시간, 48시간, 72시간군에서 UVB group 보다 유의하게 낮게 나타났다(p<0.05). 주사전자현미경적 관찰 결과, UVB group 24시간군에서는 UVB group보다 팽윤현상이 완화되었다. 48시간군에서는 가피형성, 72시간군에서는 규칙적인 판상구조, 120시간군에서 새로운 각질세포 생성, 168시간군에서는 얇은 섬유망으로 덮혀 있는 것이 관찰되었다. 투과전자현미경적 관찰 결과, UVB+Le group은 층판소체의 증가와 층판소체의 재형성이 UVB group보다 모든 군에서 촉진되었다. 특히 165시간군에서는 지질이중막의 구조가 거의 회복되어졌다. 이상의 모든 실험결과를 통해 자초추출물이 UVB 조사로 손상된 생쥐 피부를 회복시키는 효과가 있는 것으로 사료된다.

Gromwell (Lithospermum erythrorhizon) Attenuates High-Fat-Induced Skeletal Muscle Wasting by Increasing Protein Synthesis and Mitochondrial Biogenesis

  • Gromwell (Lithospermum erythrorhizon, LE) can mitigate obesity-induced skeletal muscle atrophy in C2C12 myotubes and high-fat diet (HFD)-induced obese mice. The purpose of this study was to investigate the anti-skeletal muscle atrophy effects of LE and the underlying molecular mechanism. C2C12 myotubes were pretreated with LE or shikonin, and active component of LE, for 24 h and then treated with 500 μM palmitic acid (PA) for an additional 24 h. Additionally, mice were fed a HFD for 8 weeks to induced obesity, and then fed either the same diet or a version containing 0.25% LE for 10 weeks. LE attenuated PA-induced myotubes atrophy in differentiated C2C12 myotubes. The supplementation of LE to obese mice significantly increased skeletal muscle weight, lean body mass, muscle strength, and exercise performance compared with those in the HFD group. LE supplementation not only suppressed obesity-induced skeletal muscle lipid accumulation, but also downregulated TNF-α and atrophic genes. LE increased protein synthesis in the skeletal muscle via the mTOR pathway. We observed LE induced increase of mitochondrial biogenesis and upregulation of oxidative phosphorylation related genes in the skeletal muscles. Furthermore, LE increased the expression of peroxisome proliferator-activated receptor-gamma coactivator-1 alpha and the phosphorylation of adenosine monophosphate-activated protein kinase. Collectively, LE may be useful in ameliorating the detrimental effects of obesity-induced skeletal muscle atrophy through the increase of protein synthesis and mitochondrial biogenesis of skeletal muscle.

Suppressive effects of Lithospermum erythrorhizon extracts on lipopolysaccharide-induced activation of AP-1 and NF-κB via mitogen-activated protein kinase pathways in mouse macrophage cells

  • A variety of anti-inflammatory agents have been shown to exert chemopreventive activity via targeting of transcription factors such as NF-${\kappa}B$ and AP-1. Lithospermum erythrorhizon (LE) has long been used in traditional oriental medicine. In this study, we demonstrated the inhibitory effects of LE extracts on lipopolysaccharide (LPS)-stimulated production of inflammatory cytokines. As an underlying mechanism of inhibition, LE extracts reduced LPS-induced transactivation of AP-1 as well as NF-${\kappa}B$ in mouse macrophage cells. Electrophoretic mobility shift assays indicated that LE extracts inhibited the DNA binding activities of AP-1 and NF-${\kappa}B$. In addition, phosphorylation of $I{\kappa}B-{\alpha}$ protein was suppressed by LE extracts. Moreover, LE extracts inhibited c-Jun N-terminal kinase and extracellular signal-regulated signaling pathways. Our results suggest that the anti-inflammatory activity of LE extracts may be mediated by the inhibition of signal transduction pathways that normally lead to the activation of AP-1and NF-${\kappa}B$. These inhibitory effects may be useful for chemoprevention of cancer or other chronic inflammatory diseases.

식물체(솔잎, 자초)의 에탄올 추출물이 유탁액의 지방산화에 미치는 영향 (Effect of Ethanol Extracts in Pinus densiflora, Lithospermum erythrorhizon on the Lipid Oxidation of Oil Emulsion)

  • This study was carried out to investigate the effects of ethanol extracts on lipid oxidation of oil emulsion. The results are as follows; The scavenging ability of plant extracts for hydroxyl radical was found, and plant extracts played an important role as a strong chelating agents to bind iron if Fe2+ ion exists in oil emulsion. Pinus densiflora(PD), Lithospermum erythrorhizon(LE) and PD+LE acted as strong chelating agents to bind iron to reduce lipid oxidation in oil emulsion. The content of Fe2+ ion in ethanol extracts from LE and PD+LE were significantly higher(p<0.05) than that of ethanol extracts from PD. The content of total iron has same tendency. The ascorbic acid content of PD(16.36ppm) was slightly higher than those of LE(13.08ppm). Electron donating ability of PD was significantly higher(p<0.05) than those of LE. However, the superoxide(SOD) like ability of LE showed a little higher than those of LE and PD+LE, which means the strong antioxidant activity of LE. The nitrite scavenging effects were dependent on pH value, however, they decreased as pH value increased. Especially, they almost didn't show the nitrite scavenging effect in pH 6.0. In conclusion, the PD and LE extracts may be used as natural antioxidant sources to reduce lipid oxidation in oil emulsion.

LPS로 유도한 RAW 264.7 세포의 염증반응에서 자초(紫草)의 항염증 효과 (The anti-inflammatory effect of Lithospermum Erythrorhizon on lipopolysaccharide - induced inflammatory response in RAW 264.7 cells)

  • Objective : Lithospermum Erythrorhizon (LE) has been used as an anti-bacterial and anti-inflammatory agent. However, it is unclear that LE aqueous extract could show the anti-inflammatory effects in RAW 264.7cells. The purpose of this study was to investigate the anti-inflammatory effect of aqueous extract from LE on lipopolysaccharide (LPS) - induced inflammatory response. Methods : To measure out the cytotoxicity of LE, we performed the MTT assay. To evaluate the anti-inflammatory effects of LE, we examined the inflammatory mediators such as nitric oxide (NO), prostaglandin E2 ($PGE_2$) and pro-inflammatory cytokines (tumor necrosis factor (TNF)-${\alpha}$, interleukin, (IL)-$1{\beta}$ and (IL)-6) on RAW 264.7 cells. We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B (NF-${\kappa}B$) activation by western blot. Results : Aqueous Extract from LE itself did not have any cytotoxic effect in RAW 264.7 cells. Aqueous extract from LE inhibited LPS-induced productions of inflammatory mediators such as NO, $PGE_2$, and pro-inflammatory cytokines including TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 in RAW 264.7cells. In addition, LE inhibited the phosphorylation of p38 kinases (p38), c-Jun $NH_2$-terminal kinase (JNK), and NF-${\kappa}B$ activation in RAW 264.7 cells. Conclusion : LE down-regulated LPS-induced production of inflammatory mediators through the inhibition of p38, JNK and NF-${\kappa}B$ activation. Taken together, these results could provide the evidence for the anti-inflammatory effects of LE. Therefore, LE may be a novel target in the management of inflammation and help to support a potential strategy for prevention and therapy of inflammatory diseases.

오디 및 자초추출물의 피부 광노화 보호효과 (Protective Effect of Mulberry and Lithospermum erythrorhizon Extracts on Anti-aging against Photodamage)

  • 본 연구에서 적색의 색소 성분을 함유한 오디 및 자초 추출물의 피부 광노화 보호효과를 조사하였다. 오디 추출물의 총 안토시아닌 함량을 분석한 결과 4.92 mg/g이었고 HPLC 분석을 통한 자초 추출물의 shikonin 함량은 9.58 mg/g으로 나타났다. 오디 및 자초추출물의 전자공여능은 각각 84.32%와 57.46%로 분석되어 오디가 우수하였으며, superoxide radical 소거능은 오디 추출물이 76.34%로 자초추출물 61.55%보다 항산화 활성이 높았다. 오디 및 자초추출물의 ORAC 값은 각각 $545.37{\mu}moles$ TE/g과 $427.18{\mu}moles$ TE/g으로 분석되어 오디 추출물의 항산화 활성이 높음을 알 수 있었다. HS68 세포에서의 UVB 조사군과 비교 시 MMP-1 생산 억제율은 오디 및 자초 추출물 $200{\mu}g/mL$ 첨가 시 각각 68.6%, 32.7%로 분석되었다. 무모쥐를 이용한 UVB 광노화 피부 동물 모델에서 오디 및 자초 추출물의 피부조직의 변화를 조사한 결과, 시료 처리군이 무처리군에 비해 홍반, 주름 두께 및 깊이, 표피 비후, 콜라겐 각질층, 표피층이 감소되었으며 수분함량은 증가하였다. 총 주름 깊이의 경우, 오디 추출물 7% 처리군과 자초 추출물 5% 처리군에서 대조군과 비교 시 30% 가량 억제되어 in vitro에서 뿐만 아니라 in vivo에서도 오디 및 자초 추출물이 UVB에 의한 피부 광노화 보호효과가 있음을 확인하였다.

자초(紫草)가 HaCaT 세포의 사이토카인 유전자 발현에 미치는 영향 (Effects of Lithospermum erythrorhizon on the cytokine gene expression in human keratinocytes)

  • Objective : Propionibacterium acnes (P. acnes) is a major pathogenic bacteria for acne vulgaris. This study was performed to evaluate the effects of Lithospermum erythrorhizon extracts on the inflammatory cytokines gene expression by P. acnes in human keratinocytes, HaCaT cell line. Methods : Anti-bacterial activity and cytotoxicity of LE extracts was analyzed by agar plate culture and XTT assay. The cytokines gene expressions were assessed by real time RT-PCR for IL-8, MCP-1 and TNF-${\alpha}$. During the cell culture and treatments, amounts of secreted TNF-${\alpha}$ were measured by ELISA. Translocation of transcription factor NF-${\kappa}B$ from cytoplasm into nucleus was observed by immunocytochemistry and confocal microscopy. Results : There were no anti-bacterial effects and cytotoxicity as high as $1,000{\mu}g/ml$ of LE extracts in XTT assay. Transcription levels of inflammatory cytokines, IL-8, MCP-1 and TNF-${\alpha}$ were increased by P. acnes in HaCaT. LE extracts decreased the upregulated gene transcription levels. However, amounts of secreted TNF-${\alpha}$ were similar in HaCaT cells with P. acnes and LE extracts. Translocation of NF-${\kappa}B$ into nucleus by P. acnes was significantly inhibited by LE extracts. Conclusions : From the results of this study, LE extracts have anti-inflammatory effects on HaCaT cells by P. acnes that decreased the mRNA expressions of IL-8, MCP-1 and TNF-${\alpha}$. This anti-inflammatory effects of LE extracts could provide the potential of therapeutic substance for acne vulgaris.

  • There was an increasing interest that herbal medicine and natural material extracts were proved processes of antioxidant, cosmeceutical activity and the other effects. The aim of this study was to assess the antioxidant of extraction of four kinds from Prunus armeniaca L., Reynoutria elliptica, Curcuma aromatica, Lithospermum erythrorhizon. RE (Reynoutria elliptica) and CA (Curcuma aromatica) have good electron donating ability. The water and ethanol extract of RE at a 100 ppm concentration showed over 70%, the water extract at 500 ppm concentration showed 83% and the ethanol extract at 100 ppm concentration showed 86% of CA. Xanthine oxidase inhibition activity of the water extract of LE (Lithospermum erythrorhizon) at a 1,000 ppm concentration showed over 44%, on the other hand, RE showed in all lowest effect and there was no inhibition activity of a couple more extracts. In the measurement of nitrite scavenging activity, all extracts showed highly scavenging activity. Especially the water and ethanol extract of RE showed over 99% at 500 ppm, also LE showed over 40% at 10 ppm concentration.

한약재 추출물이 김치 관련 유산균의 성장과 김치의 숙성에 미치는 효과 (Effect of Medicinal Herbs' Extracts on the Growth of Lactic acid bacteria isolated from Kimchi and Fermentation of Kimchi)

  • 김치의 숙성을 지연시킬 목적의 일환으로 항균효력이 있는 것으로 판단되는 한약재 35종을 ethanol로 추출하여 김치 숙성관련 유산균에 대한 항균효력을 검토한 후 항균효력이 있는 한약재를 김치에 첨가하여 김치의 선도에 미치는 영향을 관찰하였다. 한약재 35종 중 오미자, 단삼, 감초, 자초, 고삼 등 5종에서는 비교적 강하게 김치 숙성관련 유산균의 증식 억제력을 관찰할 수 있었으며 사상자, 솔잎, 백작약, 오갈피, 오배자 등 5종의 한약재 추출물은 일부 유산균의 생육만을 억제하는 경향을 나타내었다. 특히 고삼, 자초추출물이 강한 억제도를 나타내었다. 4종의 한약재 추출물을 김치에 1% 수준으로 첨가하여 $10^{\circ}C$에서 숙성시키면서 변화를 관찰한 결과 pH, 산도, 미생물의 변화는 숙성 15일까지는 다소 억제하는 경향을 보였으나 15일째 이후에는 별다른 차이를 관찰할 수 없었다. 한약재를 첨가한 김치에 대한 관능검사는 자초, 감초, 단삼 첨가구의 경우에는 대조구와 비슷한 경향을 보였으나 고삼 첨가구의 경우에는 대조구에 비해 기호도가 감소하였다.
