Effect on Insecticide Susceptibility of Lissorhoptrus oryzophilus Fed on Carotenoid-Biofortified Rice Variety (비타민 A 강화벼 급이가 벼물바구미(Lissorhoptrus oryzophilus)의 살충제 감수성에 미치는 영향)

  • BACKGROUND: The carotenoid-biofortified (PAC) rice was generated by inserting phytoene synthase (Psy) and carotene desaturase (Crtl) genes isolated from Capsicum annuum cv. Nockwang and Pantoea ananatis into the genome of a conventional variety of rice (Nakdongbyeo). In our present study, we studied the effects on insecticide susceptibility of Rice Water Weevil (Lissorhoptrus oryzophilus). METHODS AND RESULTS: The L. oryzophilus were fed on carotenoid-biofortified (PAC) rice and its near non-genetically modified (GM) counterparts (Nakdongbyeo) under $25{\pm}1^{\circ}C$, humidity of $60{\pm}5%$, and photoperiod 16L:8D for more than 60 days. Ten adults were soaked in the Clothianidin SC solution for 5 second in different concentrations, and were detected the mortalities after 24, 48 and 72 hours respectively. Every experiment was conducted with three replications. The cumulative mortalities of L. oryzophilus exposed to Clothianidin SC were similar between two types of feed administration. CONCLUSION: The results of this study suggested that carotenoid-biofortified rice might not affect the insecticide susceptibilities of Lissorhoptrus oryzophilus.

Seasonal Change in Rice water Weevil, Lissorhoptrus oryzophilus Kuschel (Coleoptera : Curculionidae), in Rice Field (중부지방에 있어서 벼물바구미의 년중 발생 경과)

  • Seasonal occurrence in each stages of rice water weevil, Lissorhoptrus oryzophilus Kuschel, was studied in paddy field transplanted on May 25, Siehung, 1989. The peak of overwintered adults was late May an that of newly emerged adults was early August. Oviposition occurred in late May and late June and its peak was observed on early June. Larvae were observed from early June to early August and pupation occurred from early July to late August. The first adults of newly emerged rice water weevil was observed on early July and its emergence peak was early August.

  • The rice water weevil, Lissorhoptrus oryzophillus are major pests of aquatic rice plant throughout the country. In field, we examined efficacy of Steinernema carpocapsae and Xenorhabdus nematophilus against rice water weevil, Lissorhoptrus oryzophilus. Steinernema carpocapsae showed 71-74.2% mortality after 15days in the field. Also symbiotic bacteria Xenorhabdus nematophilus isolated from Steinernema carpocapsae showed 33.3-58.3% mortality after 15days at vat against rice water weevil, Lissorhoptrus oryzophilus.

Effect of Temperature on Reproduction and Development of Rice Water Weevil, Lissorhoptrus oryzophilus (Coleoptera: Curculionidae) (벼물바구미의 생식과 발육에 미치는 온도의 영향)

  • This study was carried out to test the effects of temperatures between 2$0^{\circ}C$ and 3$0^{\circ}C$ on the reproduction and development of the rice water weevil, Lissorhoptrus oryzophilus. Preoviposition periods were much longer (17.2 to 51.0 days) in the overwintering adult females collected in March than those collected in May, regardless of temperature. Oviposition periods, however, were longer (16.9 to 22.0 days) in the adult females collected in May than those collected in March at the same temperatures. The longer oviposition period observed in the females collected in May were directly associated with higher fecundity. Egg periods were shortened from as temperature increased, but the hatching rate was highest (100%) at 27$^{\circ}C$. The developmental periods from egg to adult were shortened as temperature increased : from 77.9 days at 2$0^{\circ}C$ to 38.3 days at 3$0^{\circ}C$. The developmental zero point temperature (T) and the total effect temperature (K) for egg were 16.3$^{\circ}C$ and 62.1 dgree days, respectively. The T and K from egg to adult emergence were 13.9$^{\circ}C$ and 577.6 dgree days, respectively. The adult females of the first generation did not oviposit at 2$0^{\circ}C$, but did at $25^{\circ}C$ and 3$0^{\circ}C$. The intrinsic rate of natural increase (r$_{m}$) increased as temperature augmented. Net reproductive rate (Ro) per generation was highest (75.3) at $25^{\circ}C$.>.

Analysis of junction site between T-DNA and plant genome in Lissorhoptrus oryzophilus resistance GM rice (벼물바구미 (Lissorhoptrus oryzophilus) 내충성 GM 벼에서 T-DNA와 게놈의 인접부위 분석)

  • Four transgenic rice lines harboring insect-resistant gene cry3A showed ideal field performances characterized by high considerable resistance to rice water weevil (Lissorhoptrus oryzophilus Kuschel). In this study, we estimated the insertion number of foreign genes, and analyzed the flanking sequences of T-DNAs in rice genome. As a result, T-DNA of BT12R1 line was inserted in exon region of rice chromosome 10. Two copies of T-DNAs were inserted in line BT12R2. BT12R3 line was analyzed at only left border flanking sequence. BT12R4 line was confirmed one copy of foreign gene insertion at the position 24,516,607 ~ 24,516,636 of rice chromosome 5, accompanied by a deletion of 30 bp known genomic sequences. This intergenic position was confirmed none of expressed gene and any deletion/addition of T-DNA sequence. In conclusion, these molecular data of rice water weevil resistant Bt rice would be used to conduct the biosafety and environment risk assessment for GM crop commercialization.

Impact of Pesticide Treatment on an Arthropod Community in the Korean Rice Ecosystem

  • An arthropod community in a rice ecosystem was surveyed to determine the impact of two insecticides frequently used in Korean rice ecosystems: carbofuran 3GR, which targets the rice water weevil, Lissorhoptrus oryzophilus Kuschel (Coleoptera: Curculionidae) in the early season and fenobucarb EC, which targets the brown planthopper, Nilaparvata lugens ($St{\aa}l$) (Hemiptera: Delphacidae) in the mid- and late seasons, respectively. Overall, the application of the insecticides reduced density of total arthropods by 48.4% compared to the untreated field, but their impact on each functional group were different. Carbofuran GR treatment on 1 June reduced the L. oryzophilus population significantly until mid-season. The population of filterfeeding chironomids was also reduced by 50%, whereas the spider population was less disturbed. Fenobucarb EC treatment on 16 August significantly reduced N. lugens and detrivorous entomobryid populations until the late season. Both web-building and wandering spiders were also significantly disturbed by fenobucarb EC although the impact differed according to their behavioral differences. While the population of web-building spiders significantly decreased over time, that of wandering spiders recovered from the disturbance a few weeks later.

Feeding Plants of Rice Water Weevil (벼물바구미의 가해식물)

  • Feeding plants of rice water weevil(RWW) adult and larvae were observed. Thirteen families including 104 species were found as host plants of RWW adult, and 74 species of them were newly identified as its host. Thmedia triandra var. japonica, Luzula capitata, Agropyron tsukushiense var. ransiens, and Zoysiama crostachya were most favorable hosts of adult RWW. Host plants for larvae were 9 species of weeds including Cperus nipponicus, Eleccharis kuroguwa, Sagittaria trifolia, and Claviceps purpurea in the rice paddy fields.

Influence of Rice Water Weevil (Lissorhoptrus oryzophilus) Damage on Growth and Grain Quality in Rice (벼물바구미 피해정도가 벼 생육 및 미질에 미치는 영향)

  • A field experiment was conducted to investigate the influence of water weevil damage on growth and grain guality in rice. Plant height was shortened, number of tillers per hill decreased and maximum tillering stage delayed in damaged field by rice water weevil. Heading date was delayed by 1-2days, culm lenght shortened, panicle lengthened, number of panicles decreaced, number of panicles per spikelet increased, ripened grain ratio decreased in damaged field by rice water weevil. Yield per 10a was decreased 4-22% in damaged field compared with that in undamaged field by rice water weevil, yield loss in late transplanting (June 23) was larger than that in early transplanting (May 20). Imperfect kerml rice, especially green kernel rice, was increased in damaged field by rice water weevil.

Comparison on the Time of Occurrence of Major Rice Insect Pests Based on Growing Degree Day in Northern Part of Korean Peninsula (북방농업지대에서 유효적산온도를 이용한 벼 해충의 발생시기 비교)

  • This study was carried out to compare on the time of occurrence of 6 major rice insect pests [Lissorhoptrus oryzophilus Kusche, Oulema oryzae Kuwayama, Sogatella furcifera (Horvath), Nilaparvata lugens Stal., Cnaphalocrocis medinalis (Guenee), Mythimna separata Walker] in northern part of the Korean peninsula. Using growing degree days of the insects, we evaluated the number of occurrence generations and the time of occurrence. Over-wintering insects such as L. oryzophilus and O. oryzae showed different occurrence periods in northern regions. The occurrence period of the first generation adults was later in northeast regions than in Korean middle regions and more later both in northern alpines and in northern parts of east sea. In addition, the first adults of S. furcifera, N. lugens, C. medinalis, and M. separata occurred between June and early August. However, from late August to September, these insects showed the different occurrence periods in northern regions. Especially, the second adults of N. lugens were not occurred and the second to third generation adults of S. furcifera, C. medinalis, and M. separata showed similar occurrence properties. Based on these properties, the occurrence of major rice insect pests will be less in northeast regions, northern regions of east sea, northern inlands, and northern alpines of the Korean peninsula. However, comparing with their occurrences in northern regions of Gyenonggi and Gangwon provinces, the rice insect pests may show similar occurrence pattern in mid-korean mountains except for pyunggang and yangduk regions as well as in the southern and northern regions of Suyang-san.
