It is well known that poly(glutamate)s with long n-alkyl side chains form thermotropic liquid crystalline state by melting of the side-chain crystallites and also poly(glutamate)s such as poly(${\gamma}-benzyl\;L-glutamate$ )(PBLG), poly(${\gamma}-n-alkyl\;L-glutamate$), etc. in solvent form the isotropic, biphasic and liquid crystalline phases which contains cholesteric and columnar liquid crystalline forms depending on the polypeptide concentration. Although there is no diffusion study for PBLG in liquid crystalline state, because it is difficult to observed $^{1}H$ spectrum of PBLG in liquid crystalline state for its very short $^{1}H\;T_{2}$. In this study, the diffusional behavior of rodlike PBLG in concentrated solution is successfully elucidated as studied by the field-gradient $^{1}H$ NMR methods.

As an effective separation method for biomolecules, aqueous two-phase systems based on ionic liquids were suggested. Hydrophobic ionic liquids are more expensive and viscous in spite of their usage in the ionic liquid/water biphasic extraction compared with hydrophilic ionic liquids. In case of aqueous two-phase systems using hydrophilic ionic liquids, they can be diluted in aqueous phase. Experimental results show that aqueous two phase systems can be formed by adding appropriate amount of ionic liquids to aqueous salts solutions. The viscosity of ionic liquid aqueous phase is proportional to the cation chain length in ionic liquids. It is founded that the ionic liquid based aqueous two phase systems are effective for the separation of biomolecules such as acrylic acid.

The effects of aliphatic hydrocarbons (n-hexadecane and n-dodecane) on the volumetric oxygen mass transfer coefficient $(k_L\;a)$ were studied in flat alveolar airlift reactor and continuous stirred tank reactors (CSTRs). In the flat alveolar airlift reactor, high aeration rates (>2vvm) were required in order to obtain efficient organic-aqueous phase dispersion and reliable $k_La$ measurements. Addition of 1% (v/v) n-hexadecane or n-dodecane increased the $k_La$ 1.55- and 1.33-fold, respectively, compared to the control (superficial velocity: $25.8{\times}10^{-3}m/s$, sparger orifice diameter: 0.5 mm). Analysis of the gas-liquid interfacial area a and the liquid film mass transfer coefficient $k_L$ suggests that the observed $k_La$ increase was a function of the media's liquid film mass transfer. Addition of 1% (v/v) n-hexadecane or n-dodecane to analogous setups using CSTRs led to a $k_La$ increase by a factor of 1.68 and 1.36, respectively (superficial velocity: $2.1{\times}10^{-3}m/s$, stirring rate: 250 rpm). These results propose that low-concentration addition of oxygen-vectors to aerobic microbial cultures has additional benefit relative to incubation in purely aqueous media.

The folding of recombinant hepatitis B virus X-protein (rHBx) solubilized from Escherichia coli inclusion bodies was investigated. By sequential dialysis of urea, rHBx was folded into its native structure, which was demonstrated by the efficacy of its transcriptional activation of the adenovirus major late promoter (MLP), fluorescence spectroscopy, and circular dichroism (CD) analysis. The decrease in CD values at 220 nm and a corresponding blue shift of the intrinsic fluorescence emission confirmed the ability of rHBx to refold in lower concentrations of urea, yielding the active protein. Equilibrium and kinetic studies of the refolding of rHBx were carried out by tryptophan fluorescence measurements. From the biphasic nature of the fluorescence curves, the existence of stable intermediate states in the renaturation process was inferred. Reverse phase-high performance liquid chromatography (RP-HPLC) analysis further demonstrated the existence of these intermediates and their apparent compactness.

아민 수용액을 이용한 CO₂ 포집 공정의 문제점은 재생에 따른 높은 에너지 요구량이다. 이러한 단점을 극복하기 위해서 본 연구에서는 디에틸렌트리아민(diethylenetriamine, DETA) 수용액에 이소프로판올(isopropanol, IPA)을 도입한 상분리 흡수제 적용에 따른 CO₂ 포집 특성을 고찰하였다. 20 wt% DETA 수용액에서 IPA 조성이 20 wt%를 이상이 되면 CO₂를 흡수함에 따라 흡수제의 상이 CO₂-농축상과 CO₂-희박상으로 액-액 분리된다. 그 이유는 CO₂와 DETA가 반응하여 형성된 염의 IPA에 대한 낮은 용해도 때문이다. DETA 수용액에서 IPA 조성이 증가하면 CO₂-희박상의 부피에 대한 CO₂-농축 상의 부피비가 증가하게 되고 이에 따라 CO₂-희박상에서의 CO₂ 농축 정도가 높아지게 된다. 20 wt% DETA + IPA + 물 흡수제를 이용하여 흡수탑에서 CO₂를 흡수한 결과 20 wt% DETA 수용액 흡수제에 비해 CO₂ 흡수능 및 흡수속도가 모두 높은 것으로 확인되었다. DETA + IPA + 물로 구성된 상분리 흡수제를 CO₂ 포집에 적용할 경우 상분리 따른 CO₂ 농축과 CO₂-농축상의 부피 감소에 따른 재생에너지 저감을 기대할 수 있다.

The hexane and ether extracts from the roots of Angelica dahurica caused a significant inhibition of hepatic drug-metabolizing enzyme (DME) activity. Through systematic fractionation by $SiO_2\;column$ and vacuum liquid chromatography monitoring by bioassays, three furanocoumarins, phellopterin, byakangelicin and tert-O-methylbyakangelicin were isolated as active principles. These components have biphasic responses, both inhibitory and inducing effects on DME system. Tert-O-methyl byakangelicin was found to have the strongest enzyme inhibitory potency.

본 연구에서는 이온성 액체인 염화콜린에 5가지의 금속염화물을 첨가하여 루이스산 이온성 액체 촉매를 제조하고, 이 촉매를 사용하여 대두유로부터 바이오디젤을 합성하였다. 먼저 단독의 금속염화물인 염화주석과 염화아연, 염화알루미늄, 염화철(III), 염화구리(I) 촉매에 대하여 363~423 K 온도 범위에서 에스터 교환 반응의 반응성을 조사하였다. 5가지의 금속염화물 중 염화주석이 우수한 촉매 활성을 나타내었고, 이러한 경향과 같이 5가지의 루이스산 이온성 액체 촉매 중 $[Me_3NC_2H_4OH]Cl-2SnCl_2$의 촉매가 403 K에서 14시간 동안 유지:메탄올:촉매의 몰 비율 1:12:0.9인 조건으로 최대 91.1%의 높은 반응수율을 나타내었다. 단독의 염화주석 촉매와는 달리, $[Me_3NC_2H_4OH]Cl-2SnCl_2$의 촉매는 반응 후 액체-액체 이상계를 형성하여 반응물과 생성물로부터 쉽게 분리할 수 있으며, 5회 이상 재사용 후에도 활성이 거의 감소하지 않았다. 이러한 결과는 촉매의 수분에 대해 안정성과 강한 루이스 산성도의 특성에 기인한 것으로 생각된다. 또한 촉매에 대한 반응시간과 촉매 및 메탄올 몰 비율 등의 반응변수들에 대한 영향이 조사되었다.

In this study, we developed and validated a sensitive analytical method to quantify baphicacanthin A in mouse plasma using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The standard calibration curves for baphicacanthin A ranged from 0.5 to 200 ng/mL and were linear, with an r² of 0.985. The inter- and intra-day accuracy and precision and the stability fell within the acceptance criteria. Besides, we investigated the pharmacokinetics of baphicacanthin A following its intravenous (5 mg/kg) and oral administration (30 mg/kg). Intravenously injected baphicacanthin A showed biphasic elimination kinetics with high clearance and volume of distribution values. Furthermore, baphicacanthin A showed a rapid absorption but low aqueous solubility (182.51±0.20 mg/mL), resulting in low plasma concentrations and low oral bioavailability (2.49 %). Thus, we successfully documented the pharmacokinetic properties of baphicacanthin A using this newly developed sensitive LC-MS/MS quantification method, which could be used in future lead optimization and biopharmaceutic studies.

항생제의 서방형 전달을 위해 황산겐타마이신 gentamicin sulfate (GS)을 함유한 poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) 제형을 제조하였다. 본 연구에서는 제형의 두께, hydroxyvalerate (HV) 농도, 초기 약물함유량 및 첨가제의 함유에 따른 약물 방출거동의 변화를 조사하였다. 전자주사현미경을 이용하여 제형의 표면형태와 매트릭스 내부 약물의 조성을 관찰한 결과 약물방출 전과 후 모두 거칠고 다공성인 형태를 가짐을 알 수 있었다. 또한, HV와 첨가제의 함량이 증가할수록 약물이 고분자 매트릭스에 더 조밀하게 배열함을 관찰하였고, 이러한 구조가 약물의 방출에 영향을 미침을 알았다. HPLC를 이용하여 약물의 방출량을 측정한 결과, 모든 제형이 복합적인 방출 방향을 나타내었고, 일부 매트릭스는 30일 동안 거의 영차에 가까운 방출거동을 보였다. 이상의 결과에서 우리는 제형의 두께, 고분자 매트릭스의 조성, 첨가제의 함유량 등을 조절함으로 약물 방출을 제어할 수 있음을 확인하였다.
