Characterization of Norepinephrine Release in Rat Posterior Hypothalamus Using in vivo Brain Microdialysis

  • In the present study, we used the microdialysis technique combined with high performance liquid chromatography (HPLC) and electrochemical detection to measure the extracellular levels of norepinephrine (NE) in the posterior hypothalamus in vivo, and to examine the effects of various drugs, affecting central noradrenergic transmission, on the extracellular concentration of NE in the posterior hypothalamus. Microdialysis probes were implanted stereotaxically into the posterior hypothalamus (coordinates: posterior 4.3 mm, lateral 0.5 mm, ventral 8 mm, relative to bregma and the brain surface, respectively) of rats, and dialysate collection began 2 hr after the implantation. The baseline level of monoamines in the dialysates were determined to be: NE $0.17{\pm}0.01,$ 3,4-dihydroxyphenylacetic acid (DOPAC) $0.94{\pm}0.07,$ homovanillic acid (HVA) $0.57{\pm}0.05$ pmol/sample (n=8). When the posterior hypothalamus was perfused with 90 mM potassium, maximum 555% increase of NE output was observed. Concomitantly, this treatment significantly decreased the output of DOPAC and HVA by 35% and 28%, respectively. Local application of imipramine $(50\;{\mu}M)$ enhanced the level of NE in the posterior hypothalamus (maximum 200%) compared to preperfusion control values. But, DOPAC and HVA outputs remained unchanged. Pargyline, an irreversible monoamine oxidase inhibitor, i.p. administered at a dose of 75 mg/kg, increased NE output (maximum 165%), while decreased DOPAC and HVA outputs (maximum 13 and 12%, respectively). These results indicate that NE in dialysate from the rat posterior hypothalamus were neuronal origin, and that manipulations which profoundly affected the levels of extracellular neurotransmitter had also effects on metabolite levels.

Effect of Morindae officinalis Radix extract on muscle fatigue and blood component (단기(短期) 파극(巴戟) 투여(投與)가 생쥐의 근피로(筋疲勞) 및 혈액(血液) 성분(成分)에 미치는 영향(影響))

  • Purpose: This study was conducted to investigate the muscle anti-fatigue effects of Morindae officinalis Radix liquid extract. Methods: 4-6 weeks old ICR rats are used in the study, and we administered the extracts powder of Morinda officinalis concentration of 1, 10, 100mg/0.3 ml to each to rats(MO group) once a day for each 30 and 60 days. After 30 and 60 days, we examined the plasma concentraion of lactate dehydrogenase, glucose. And we measured the persistent time of swimming exercise test and conducted grip strength test Results: We found that there are no significantly differences between the control and the treatment group in the plasma concentration of activity level of lactate dehydrogenase. And we found that in 30days group, level of glucose concentration is significant. In swimming exercise test, as administration time and concetration growed up, swimming time is increased. And in grip strength test, as concentraion is growed up, grip strength is increased. Conclusion: This study show that Morinda officinalis is effective against muscle fatigue.

Effects of Alpha-G Rutin Supplementation in Sperm Freezing Extender on Dog Sperm Cryopreservation

  • This study was designed to investigate the effects of alpha-glucosyl rutin (G-rutin) and its comparative effects with other antioxidants (glutathione: GSH, catalase: CATA and beta-mercaptoethanol : ${\beta}ME$) on dog sperm freezing. In the first experiment (E1), the spermatozoa were diluted in freezing extender supplemented with 0 (control), 0.001, 0.01, or 0.1% G-rutin and frozen using liquid nitrogen ($LN_2$). The progressive motility, reactive oxygen species (ROS) level and apoptosis of spermatozoa were assessed after sperm thawing at $37^{\circ}C$ for 25 sec. In the second experiment (E2), 0.1% G-rutin group was compared with 10 mM ${\beta}-ME$, $5{\mu}M$ GSH and $50{\mu}M$ CATA groups by assaying progressive motility, viability and gene expression of Bcl-2 and SMCP after sperm freezing and thawing. In E1, 0.1% G-rutin group showed higher (P < 0.05) post-thaw progressive motility and lower (P < 0.05) ROS levels. In E2, the expressions of SMCP in G-rutin group were higher (P < 0.05) than in CATA group while Bcl-2 expression of G-rutin group was higher (P < 0.05) than ${\beta}-ME$ and CATA groups. However, there were no significant differences in progressive motility and viability. Therefore, we suggest that G-rutin can be used as a potentially antioxidative supplement in dog sperm freezing extender on the basis of gene expression related to motility and apoptosis as well as ROS level.

Evaluation of the accuracy of dental casts manufactured with 3D printing technique in the All-on-4 treatment concept

  • PURPOSE. The aim of this study is to compare the casts obtained by using conventional techniques and liquid crystal display (LCD) three-dimensional (3D) print techniques in the All-on-4 treatment concept of the edentulous mandibular jaw. MATERIALS AND METHODS. In this study, a completely edentulous mandibular acrylic cast (typodont) with bone-level implants placed with the Allon-4 technique served as a reference cast. In this typodont, impressions were taken with the conventional technique and dental stone casts were obtained. In addition, after scanning the acrylic cast in a dental laboratory scanner and obtaining the Standard Tessellation Language (STL) data, 3D printed casts were manufactured with a 3D printing device based on the design. The stone and 3D printed casts were scanned in the laboratory scanner and STL data were obtained, and then the interimplant distances were measured using Geomagic Control X v2020 (3D Systems, Rock Hill, SC, USA) analysis software (n = 60). The obtained data were statistically evaluated with one-way analysis of variance (ANOVA) and Tukey's pairwise comparison tests. RESULTS. As a result of the one-way ANOVA test, it was determined that the stone casts, 3D printed casts, and reference cast values in all distance intervals conformed to the normal distribution and these values had a significant difference among them in all distance intervals. In Tukey pairwise comparison test, significant differences were found between casts at all distance intervals. In all analyses, the level of significance was determined as .05. CONCLUSION. 3D printed casts obtained with a 3D LCD printing device can be an alternative to stone casts when implants are placed in edentulous jaws. [J Adv Prosthodont 2022;14:379-87]

Eeffect of Wood Xylem Flour in Liquid Culture on Mycelial Biomass of Lentinus lepideus and Lentinus edodes (목분 첨가 액체배양에 의한 잣버섯 및 표고 균사배양의 촉진 효과)

  • This study was carried out to investigate the promoting effect of wood flour on the mycelial growth of Lentinus lepideus and Lentinus edodes. To determine the optimal culture condition, we first examined the tissue origin of pine flour (Pinus densiflora) including needle, bark, root and xylem. Only the xylem-derived flour increased mycelial growth compared to no treatment control. The addition of the xylem flour (5 g/l) showed the highest increase and the glucose level in the basal medium was best at 10 g/l. The smaller particle size of the xylem flour showed the positive effect on mycelial growth; two-fold increase when supplemented with flour of which particle size is less than $106\;{\mu}m$ in diameter compared to $425\;{\mu}m$. The addition of the xylem flour continuously increased the mycelial production for 25 days while mycelia stopped growing within 15 days without the xylem flour. In addition, when woody flour obtained from the different tree species was applied to L. edodes mycelial culture, all treatments accelerated mycelial production compared to the control. Based on all results described above, we conclude that the supplementation of woody flour to culture medium may be an another promising way to increase mycelial production of economically important fungi.

  • In the situation which Russia's ratification of the Kyoto protocol at February,2004, ANNEX I nations must reduce GHG(Green House Gas) discharge rate from 2008 by 2012 to the reduction level at 1990. We introduce the CO₂ ocean sequestration that is one of promising method for getting the stable CO₂ concentration in the atmosphere. There are four categories : ocean transportation technique, ocean initial dissolution technique, ocean deep current evaluation technique, and ocean biological evaluation technique. In this paper, we carried out the fundamental numerical study on the ocean initial dissolution technique, when the Liquidized CO₂ is emitted at the deep ocean, It is very important to the dissolution rate of movable CO₂ interface because it Is directly impact to the ocean organism. In order to investigate the relation of the interface movement and rate of the dissolution, we develope CR(Computational Fluid Dynamics) code that was constructed by the finite volume method based on the unstructured mesh, and a droplet's boundary surface can move and one direction dissolution from disperse phase into continuous phase adopted as its physics be. This study clarifies hydrodynamic relation between solubility and movement of the droplet through the verification of the Cm code.

  • Up to date, demands for satellite including communication are increasing. Advanced countries on space technology such as America, Russia, Europe, Japan, China and so on already had secured launch vehicle technology which can insert a large class satellite to proper orbit. The introduction of technologies on the large class launch vehicle including propulsion system is difficult due to the utilization possibility for defensive reason. The acquisition of indigenous technical expertise on the design and manufacture of valves is believed to contribute to the successful local development of valves for propulsion systems and to significant improvement of local technical level of valve design and development. This paper introduces current status of valves developed by other countries as well as valves developed in domestic. The Developed technology of valves may underlie the construction of engine control systems required for the reliable operation of the KSLV-II engine system and propulsion system.

Plasma Concentrations of Vitamins E and A, and Effects of Vitamin E Supplementation on Oxidative Stress and Immune Status in Korean Non-Insulin Dependent Diabetic Patients

  • Plasma concentrations of Vitamins E and A were measured in 15 non-insulin dependent Korean female subjects and 15 age-matched normal subjects using reversed-phase high-performance liquid chromatography. No differences were found in plasma Vitamin E concentrations between the 2 groups. Plasma Vitamin A concentrations were higher in subjects with non-insulin dependent diabetes melitus (NIDDM). The effects were evaluated of 4 weeks of daily supplementation of 400 mg Vitamin E on plasma levels of these two vitamins. In addition, the effects were observed for Vitamin E supplementation on oxidative stress and immune-related compound productions in non-insulin dependent diabetic patients and control subjects. After treatment with Vitamin E, plasma Vitamin E concentrations were significantly elevated in both groups. Basal plasma thiobarbituric acid reactive substances (TBABS) were identical, and a decreased level of TBARS caused by Vitamin E was observed only in the diabetic group (0.02739$\pm$0.0024 versus 0.01814$\pm$0.0008 nmols malondialdehyde equivalents/dl plasma ; p<0.05). The basal and after-treatment levels of immunoglobulins A, G, M were identical in control and diabetic groups, indicating that Vitamin E did not appear to alter gross humoral responses in this study. However, elevation of Complement 3 ($C_3$) was noticed due to Vitamin E supplementation, revealing a possible effect of vitamin E on one aspect of humoral immunity, Furthermore, an increase in prostaglandin E_2 ($PGE_2$) levels in diabetic patients was normalized by Vitamin E supplementation. This suggests indirectly that the depressed cell-mediated response due to elevated $PGE_2$ could be normalized. For the definitive antioxidant intake recommendations for prevention and treatment of adverse effects of non-insulin dependent diabetes, evidence from intervention trials like this study should be collected. The present data suggests that Vitamin E may oxen some protective effects against oxidative damage and might have beneficial effects of partial immune-stimulation.

Optimization of Bioreactor Operation by Mass Transfer Coefficient (물질전달계수를 이용한 생물 반응기 운전 최적화)

  • The effects of various operating parameters(agitation speed, impeller type, antiform agents, impeller spacing etc.) on air-liquid mass transfer was characterized by volumetric mass transfer coefficient($k_La$). Also, the dual-impeller agitated systems are compared with single-impeller agitated systems with a special focus on its applications for bioreactors, $k_La$ was take over a range of 200~450 rpm of agitation speed, and 0.5~2.5 vvm of air flow rates, for four single impeller and impeller combinations consisting of four impeller types, namely rushton, pitched blade, scaba, intermig were tested. The rushton impeller showed the best $k_La$ as compared with other single impellers. The dual impeller system are found to be superior as compared to single impeller in all aspects, The best combination of the dual impeller was a intermig of axial flow type as an upper impeller and a rushton of radial flow type as a lower part. Also, the control of the DO level with the variation of agitation speed was more efficient than that with an increase in air flow rate. The addition of antiform dropped the $k_La$ very large up to 1g/L regardless the type. PPG was less effect on $k_La$ than other antiforms. The impeller spacing and presence of solute are found very effective on $k_La$. When the $NaNO_3$is presented as solute, the $k_La$ increased approximately 50% then control.

The Changes in Biogenic Amines and Cortisol in Patients with Posttraumatic Stress Disorder After Long-Term Pharmacological Treatment (외상후 스트레스장애 환자에서 장기 치료에 따른 카테콜아민과 코르티솔 변화)

  • Objective : This study was conducted to evaluate the changes in the levels of neurotransmitters and cortisol in patients with chronic posttraumatic stress disorder (PTSD) and to evaluate their correlation with symptoms after long-term pharmacological treatment. Methods : Twenty-eight Vietnam veterans with chronic PTSD and 34 non-PTSD patients were consecutively recruited. The Combat Exposure Scale (CES), Mississippi Scale for Combat-Related Posttraumatic Stress Disorder (M-PTSD), Clinician Administered PTSD Scale (CAPS), Hamilton Rating Scale for Depression (HRSD) and Hamilton Anxiety Scale (HAS) were used to evaluate symptom severity. High performance liquid chromatography (HPLC) was used to measure the plasma levels of epinephrine, norepinephrine, and dopamine, and a radioimmunoassay (RIA) was performed to evaluate the plasma level of cortisol. Results : Plasma cortisol was significantly lower in PTSD patients than in control subjects, while there was no significant difference in plasma epinephrine, norepinephrine and dopamine between the two groups. The scores of M-PTSD, CAPS, HAMD and HAMA were signigicantly higher in PTSD patients than control group. Conclusion : After long-term treatment, the levels of neurotransmitters in PTSD patients returned to within the normal range, and the patients' symptoms showed some improvement. However, the core symptoms of PTSD continued to appear intermittently, and they are thought to be associated with hormonal systems, such as the HPA axis. It is also suggested that PTSD should be considered to be a complex disorder associated with multiple systems and that combinations of the effective medications for each system should be used to treat patients with PTSD.
