Separation and Identification of Antimicrobial Substances from Green Tea Extracts (녹차추출물로부터 항균물질의 분리 및 구조동정)

  • Green tea has been shown to have multifunctional health-promoting properties including cholesterol level control an antidiabetic effect and anticancer, antioxidant, and antimicrobial properties, inboth in vivo and in vitro experiments.We earlier reported antifungal and antibacterial effects of green tea extract by investigating cell membrane functions. The purpose of the present study was to purify and identify antimicrobial substances from green tea extract. Such materials were extracted from green tea (Camellia sinensis. var. sinensis) and purified by high-performance liquid chromatography. The antimicrobial substances in the extract were identified as epicatechin gallate and epigallocatechin gallate by nuclear magnetic resonance spectrophotometry.

Characteristics Evaluation of Moving Picture Blur Noise for Liquid Crystal Display (액정 디스플레이의 동화상 퍼짐 노이즈 특성 평가)

  • The moving picture blur noise of LCD is measured and the characteristics of it are evaluated. From the results, blur noise is generated only when the sight line moves while pursuing the moving image and blur noise is not generated when the sight line is fixed. In addition, decrease of gray level by the image blur has a linearity and velocity dependence. The blur noise simulator based on this experimental results is developed. From the results of blur noise simulation, the faster the moving speed of image is, the more blur noise has increased and these agree with the results of measurement. In the result of simulating blur noise characteristics by the duty ratio control of backlight, noise is reduced by lowering of the duty ratio. but the blur noise increases if there is in the both of adjacent two frames. Moreover, the case of doubling the frame rate to 120[Hz], decreasing the moving speed of the image by making an new image between the adjacent two frames brings the reduction of blur noise.

  • Korean Space Launch Vehicle (KSLV-II) Propellant Supply System charges liquid oxygen and kerosene to each propellant tank for the stages. To charge the launch vehicle propellant tank safety, the propellant charge flow rates and scenarios should be defined. First, the Propellant Supply System was modeled with 1D flow analysis program. The control valve capacity and orifice size were calculated by performing the 1D steady state simulation. Second, the 1D transient simulation was performed by using the steady state simulation results. As propellants were being charged at the each tank, the increased tank liquid level decreases the charge flow rate. Consequently, the proposed supply system satisfies the required design charging conditions.

  • This paper presents the development status of a subscale precooled turbojet engine "S-engine" for the hypersonic cruiser and space place. S-engine employs the precooled-cycle using liquid hydrogen as fuel and coolant. It has $23cm{\times}23cm$ of rectangular cross section, 2.6 m of the overall length and about 100 kg of the target weight employing composite materials for a variable-geometry rectangular air-intake and nozzle. The design thrust and specific impulse at sea-level-static(SLS) are 1.2 kN and 2,000 sec respectively. After the system design and component tests, a prototype engine made of metal was manufactured and provided for the system firing test using gaseous hydrogen in March 2007. The core engine performance could be verified in this test. The second firing test using liquid hydrogen was conducted in October 2007. The engine, fuel supplying system and control system for the next flight test were used in this test. We verified the engine start-up sequence, compressor-turbine matching and performance of system and components. A flight test of S-engine is to be conducted by the Balloon-based Operation Vehicle(BOV) at Taiki town in Hokkaido in October 2008. The vehicle is about 5 m in length, 0.55 m in diameter and 500 kg in weight. The vehicle is dropped from an altitude of 40 km by a high-altitude observation balloon. After 40 second free-fall, the vehicle pulls up and S-engine operates for 60 seconds up to Mach 2. High altitude tests of the engine components corresponding to the BOV flight condition are also conducted.

Hippuric Acid Levels in Paint Workers at Steel Furniture Manufacturers in Thailand

  • Background: The aims of this study were to determine hippuric acid levels in urine samples, airborne toluene levels, acute and chronic neurological symptoms, and to describe any correlation between urinary hippuric acid and airborne toluene. Methods: The hippuric acid concentration in the urine of 87 paint workers exposed to toluene at work (exposed group), and 87 nonexposed people (control group) was studied. Study participants were selected from similar factories in the same region. Urine samples were collected at the end of a shift and analyzed for hippuric acid by high performance liquid chromatography. Air samples for the estimation of toluene exposure were collected with diffusive personal samplers and the toluene quantified using gas-liquid chromatography. The two groups were also interviewed and observed about their work practices and health. Results: The median of the 87 airborne toluene levels was 55 ppm (range, 12-198 ppm). The median urinary hippuric acid level was 800 mg/g creatinine (range, 90-2547 mg/g creatinine). A statistically significant positive correlation was found between airborne toluene exposure and urine hippuric acid levels (r = 0.548, p < 0.01). Workers with acute symptoms had significantly higher hippuric acid levels than those who did not (p < 0.05). It was concluded that there was a significant correlation between toluene exposure, hippuric acid levels, and health (p < 0.001). Conclusion: There appears to be a significant correlation between workers exposure to toluene at work, their urine hippuric acid levels, and resulting symptoms of poor health. Improvements in working conditions and occupational health education are required at these workplaces. There was good correlation between urinary hippuric acid and airborne toluene levels.

MicroRNA super-resolution imaging in blood for Alzheimer's disease

  • We propose a novel blood biomarker detection method that uses miRNA super-resolution imaging to enable the early diagnosis of Alzheimer's disease (AD). Here, we report a single-molecule detection method for visualizing disease-specific miRNA in tissue from an AD mice model, and peripheral blood mononuclear cells (PBMCs) from AD patients. Using optimized Magnified Analysis of Proteome (MAPs), we confirmed that five miRNAs contribute to neurodegenerative disease in the brain hippocampi of 5XFAD and wild-type mice. We also assessed PBMCs isolated from the whole blood of AD patients and a healthy control group, and subsequently analyzed those samples using miRNA super-resolution imaging. We detected more miR-200a-3p expression in the cornu ammonis 1 and dentate gyrus regions of 3 month-old 5XFAD mice than in wild-type mice. Additionally, miRNA super-resolution imaging of blood provides AD diagnosis platform for studying miRNA regulation inside cells at the single molecule level. Our results present a potential liquid biopsy method that could improve the diagnosis of early stage AD and other diseases.

Change in three dry rangeland species growth and soil properties by compost application

  • There are different types of compost used as soil conditioners and fertilizers. Plants can have different responses to different forms of compost. This field study was performed to examine the effects of different types of compost on growth factors of three dry rangeland species (Atriplex, $Atriplex$ $lentiformis$; Saltwort, $Seidlitzia$ $rosmarinus$; Haloxylon, $Haloxylon$ $persicum$) and soil properties. The experiment was conducted in the Fars Province of Iran during the year 2010-2011. Compost applications consisted of compost tea, solid compost (SC), solid and liquid mixture (MX) and no compost as the control. The study was a factorial experiment based on a randomized complete block design with 3 replications. The results showed that all the tested compost applications enhanced the growth traits of all three species. It was also demonstrated that the use of compost significantly increased the organic matter (1% probability level [PL]), nitrogen concentration (5% PL), phosphorous (5% PL) and potassium (5% PL) concentrations of the soil. The soil's pH level was unchanged (range, 7.3 to 7.6), and the sodium concentration was also significantly decreased (1% PL) by the use of compost. The higher responses were observed in canopy volume and soil sodium and the lower were observed in stem diameter and soil pH level. Among the three plants in the study, Atriplex showed the best response to the application of compost. Based on the results of this study, it can be recommended that the best compost application to increase growth and improve soil condition is the mixed compost (MX) for Atriplex and the SC for Saltwort and Haloxylon.

Pest Control and Analysis of Residual Pesticides of Mulberry Fruit and Leaf against Popcorn Disease by Cultivated Type and Region (오디 재배양식 및 지역에 따른 균핵병 방제 효과와 농약 잔류 특성)

  • Background: As the Positive List System (PLS) is implemented in broad application to agricultural products, attention to the correct use of pesticides is also needed in the production of mulberry leaves and fruit. In this regard, three types of pesticides against mulberry popcorn disease were applied 2 - 3 times both in the field and greenhouses to prepare safety standards. Residual pesticide analysis was conducted on mulberry fruits and leaves. Methods and results: Three pesticieds, thiophanate-methyl, thiophanate-methyl·triflumizole and fluopyram registered as PLS pesticides for mulberry popcorn disease, were sprayed in the Wanju and in Buan regions, after which residual pesticide analysis was conducted using high-performance liquid chromatography (HPLC). As three pesticieds were either undetected or below the permissible level in mulberry fruit, demonstrating that they were suitable for safe spraying. However, 5.6 mg/kg of thiophanate-methyl was detected in the greenhouse after three application, which was slightly above maximum residue limit (MRL). Furthermore the level of thiophanate-methyl·triflumizole was higher than 0.2 mg/kg ("Gwasang No. 2" variety, spraying twice) or similar to 0.09 mg/kg ("Daesim" variety, spraying thrice) the permissible level (0.1T mg/kg) as the thiophanate-methyl was detected in mulberry leaves in the greenhouse. Conclusions: The spraying frequency for controlling mulberry popcorn disease in greenhouses should be limited to two times or less, especially when mulberry leaves are treated with thiophanate-methyl·triflumizole careful consideration is required if the leaves are to be used as food materials.

Studies on the Anti-bacterial, Anti-inflammatory and Anti-oxidant Effect of BPH (비피(鼻皮) 증류액의 항균, 소염, 항산화 작용에 대한 연구)

  • Objectives : The purpose of the study is to test the antibacterial, anti-inflammatory and antioxidant effects of BPH, which is composed of Pini Densiflorae Nodi Lignum and Querci Acutissimae Fructus, Angelicae Gigantis Radix, Cnidii Rhizoma, Angelicae Dahuricae Radix, Angelicae Tenuissimae Radix. Method : Antibacterial and anti-inflammatory effects of BPH on Propionibacterium acnes, one of anaerobic bacteria species were evaluated by measuring the levels of 2,2-diphenyl-1-picrylhydrazyl (DPPH) elimination and lipid peroxidation. Result : When BPH was applied to CCD-986sk (Human normal fibroblast) to confirm the level of cytokine(tumor necrosis factor-alpha, interleukin-8), its level increased in proportion to that of BPH's concentration, which indicated dose-dependent relationship. Using the Disk diffusion to measure the bacterial growth inhibition zone varying BPH concentration, it was found that the antibacterial effect of BPH was less than that of erythromycin, the control group, but was higher than that of saline, and it increased with higher concentrations. In a liquid culture medium containing BPH, the growth rate of Propionibacterium acnes was decreased by more than 10% at 25% BPH. After adding P. acnes to THP-1 monocyte, and treated it with BPH, and measuring the concentration of TNF-a and IL-8, it was observed that the amount of TNF-alpha and IL-8 significantly decreased depending on the level of BPH concentration. The ability to eliminate DPPH increased with higher BPH concentration. The inhibition of lipid peroxidation was increased by BHT treatment in a dose-dependent manner. Conclusion : Using Propionibacterium acnes, an anaerobic bacteria, we confirmed that BPH has antibacterial, anti-inflammatory and antioxidant effects.

  • The effects of liquid culture of Coriolus versicolor on the body weight gain, lipid metabolism, protein levels and enzyme activities were studied in growing male rats. Sprague-Dawley rats were divided into four groups; control group (C. versicolor free water), 10%, 20% or 30% C. versicolor groups (10%, 20% or 30% C. versicolor in water) according to the levels of C. versicolor supplementation. The rats were fed ad libitum each of the experimental beverage for 5 weeks. The body weight gain, food intake, food efficiency ratio, and organ weight of the all C. versicolor groups were similar to those of the control group. There were no differences in the concentrations of triglyceride, total cholesterol, HDL-cholesterol, total protein and albumin, and the activities of GOT, GPT, and alkaline phosphatase of the serum among the experimental groups. However, the atherogenic index of the 30% C. versicolor groups were significantly lower than those of the control group, and the serum HDL-cholesterol/total cholesterol ratio increased.
