Cryopreservation of winter vegetation buds of Betula platyphylla var. japonica in liquid nitrogen (자작나무 동아의 액체질소 내 초저온 보존)

  • In woody plant germplasms, using prefrozen dormant buds for materials is one way to achieve successful cryopreservation. The protocol of cryopreservation for White birch (Betula platyphylla var. japonica) winter vegetative buds is the following. First, the branches of White birch were collected in January 20, when the vegetative buds were still in a state of quiescence. The winter buds with about 5㎜ of xylem tissue were removed from the branches. They were dehydrated to moisture contents about 44% by air dry treatment. The buds were prefrozen, with the temperature being decreased by 5∼-20$\^{C}$ and then transfered to the LN(liquid nitrogen) maintained below -l96$\^{C}$. After cryopreservation, the vegetative buds were rapidly thawed in a water bath at 40$\pm$5$\^{C}$. In this case, the cell survival rate of samples was about 86%. After sterilization, buds were then cultured on MS medium. These results demonstrate the feasibility for cryopreservation of winter vegetation buds of Betula platyphylla var. japonica.

  • As hypersonic flight speeds and engines efficiencies increase, heat loads on an aircraft and it's engine increase. Because the temperature of the air flow is too high to cool the aircraft structure at hypersonic flight speeds, it is essential to use the aircraft fuel as the primary coolant. Endothermic fuels are liquid hydrocarbon aircraft fuels which are able to absorb the heat loads by undergoing endothermic reactions, such as thermal and catalytic cracking. The endothermic reactions are improved by catalysts which change the extent of reaction and product distribution. At high temperature, liquid hydrocarbons would lead to coke formation that can reduce the effectiveness of heat exchanger and cause rapid degradation of the catalyst, thus endothermic capacity of endothermic fuels is limited to the temperature at which coke doesn't form. In this study, the essential cooling technologies by applying endothermic fuels and the properties of the endothermic fuels are described.

  • Polymer electrolyte membrane fuel cells (PEMFCs) are regarded as a promising alternative to replace the existing automotive power sources. To get high performance and long-term durability for PEMFC systems, novel water management is essential. To this end, a comprehensive understanding of dynamics of the liquid water droplets within an operating PEMFC plays an important role. In this work, direct visualization of dynamic behaviors of the water droplet in the ex situ unit flow channel of a PEMFC including gas diffusion layer (GDL) is carried out as one of the fundamental studies for novel water management. Water droplet dynamics such as the movement and growth of liquid water droplets are mainly presented. Effects of GDL characteristics and inlet air flow rate on the water droplet transport and its removal from the flow channel are also discussed. The data obtained in this study can contribute to build up the fundamental operating strategy including balanced water removal capacity for automotive PEMFC systems.

  • With the tightening of environmental regulations (i.e., IMO Tier III), natural gas (NG) has been spotlighted as an eco-friendly fuel with few air pollutants other than nitrogen oxides (NOx) and sulfur oxides (SOx). For reasons of economic efficiency, it is mainly stored and transported in a liquid state at $-163^{\circ}C$, which is a cryogenic temperature, using a liquefied gas storage tank. Accordingly, it is necessary to reduce the boil-off gas (BOG) occurrence due to the heat flow according to the temperature difference between the inside and outside of the storage tank. Therefore, in this study, a BOG measurement test on an independent-type storage tank made up of SUS304L was carried out. The test results showed the tendency for BOG occurrence according to the temperature under different filling ratios.

  • In this paper, insulations characteristics by temperature changes($+30{\sim}-40[^{\circ}C]$) of $SF_6$ gas in the experimental chamber were studied. From this result, The breakdown characteristics classify the vapor stage of $SF_6$ according the Paschen's law, the gas & liquid coexisted stage of voltage value increases & much deviation and the VB low stage as the interior of chamber gets filled with mixture of $SF_6$ that are not liquefacted and remaining air which couldn't be ventilated. In addition the ability of insulation of liquid $SF_6$ was higher than that of the highly pressurized $SF_6$ gas. In this research, we want to provide the base data on designing insulation of high-temperature superconductor and the cryogenic equipments by investigating the insulation characteristics of $SF_6$.

Damage Effects Modeling by Chlorine Leaks of Chemical Plants (화학공장의 염소 누출에 의한 피해 영향 모델링)

  • This study describes the damage effects modeling for a quantitative prediction about the hazardous distances from pressurized chlorine saturated liquid tank, which has two-phase leakage. The heavy gas, chlorine is an accidental substance that is used as a raw material and intermediate in chemical plants. Based on the evaluation method for damage prediction and accident effects assessment models, the operating conditions were set as the standard conditions to reveal the optimal variables on an accident due to the leakage of a liquid chlorine storage vessel. A model of the atmospheric diffusion model, ALOHA (V5.4.4) developed by USEPA and NOAA, which is used for a risk assessment of Off-site Risk Assessment (ORA), was used. The Yeosu National Industrial Complex is designated as a model site, which manufactures and handles large quantities of chemical substances. Weather-related variables and process variables for each scenario need to be modelled to derive the characteristics of leakage accidents. The estimated levels of concern (LOC) were calculated based on the Gaussian diffusion model. As a result of ALOHA modeling, the hazardous distance due to chlorine diffusion increased with increasing air temperature and the wind speed decreased and the atmospheric stability was stabilized.

Theoretical approach on the heating and cooling system design for an effective operation of Li-ion batteries for electric vehicles (전기구동 자동차용 리튬이온 배터리의 고효율 운전을 위한 냉방 및 난방 시스템 설계에 대한 이론적 접근법)

  • This study is aiming to suggest the effective thermal management system design technologies for the high voltage and capacity battery system of the electricity driven vehicles and introduce the theoretical designing methods. In order to investigate the effective operation of the battery system for the electricity driven vehicles, the heat generation model for Li-ion battery system using the chemical reaction while charging and discharging was suggested and the thermal loads of the heat sources (air or liquid) for cooling and heating were calculated using energy balance. Especially, the design methods for the cooling and heating of the battery system for maintaining the optimum operation temperature were investigated under heating, cooling and generated heat (during charging and discharging) conditions. The battery thermal management system for the effective battery operation of the electricity driven vehicles was suggested reasonably depending on the variation of the season and operation conditions. In addition, at the same conditions under summer season, the cooling method using the liquid and active cooling technique showed a relatively high capacity, while cooling method using the passive cooling technique showed a relatively low capacity.

  • This paper presents the development status of a subscale precooled turbojet engine "S-engine" for the hypersonic cruiser and space place. S-engine employs the precooled-cycle using liquid hydrogen as fuel and coolant. It has $23cm{\times}23cm$ of rectangular cross section, 2.6 m of the overall length and about 100 kg of the target weight employing composite materials for a variable-geometry rectangular air-intake and nozzle. The design thrust and specific impulse at sea-level-static(SLS) are 1.2 kN and 2,000 sec respectively. After the system design and component tests, a prototype engine made of metal was manufactured and provided for the system firing test using gaseous hydrogen in March 2007. The core engine performance could be verified in this test. The second firing test using liquid hydrogen was conducted in October 2007. The engine, fuel supplying system and control system for the next flight test were used in this test. We verified the engine start-up sequence, compressor-turbine matching and performance of system and components. A flight test of S-engine is to be conducted by the Balloon-based Operation Vehicle(BOV) at Taiki town in Hokkaido in October 2008. The vehicle is about 5 m in length, 0.55 m in diameter and 500 kg in weight. The vehicle is dropped from an altitude of 40 km by a high-altitude observation balloon. After 40 second free-fall, the vehicle pulls up and S-engine operates for 60 seconds up to Mach 2. High altitude tests of the engine components corresponding to the BOV flight condition are also conducted.

  • In this study, preliminary design of a high-altitude test facility (HATF) was performed to simulate the high-altitude environment using a rocket engine that liquid oxygen and kerosene were used as the propellant. Experimental facility consists of vacuum chamber, supersonic exhaust diffuser, heat exchanger, ejector and gas generator. The vacuum chamber was simulated and maintained high-altitude environmental pressure by supersonic exhaust diffuser. Combustion gas of the rocket engine was cooled by water at heat exchanger after that the mixed gas was emitted to the air by ejector. The ejector which was operated by the steam generator using 75% ethanol and liquid oxygen as propellants and water for steam maintains a vacuum condition.

Hippuric Acid Levels in Paint Workers at Steel Furniture Manufacturers in Thailand

  • Background: The aims of this study were to determine hippuric acid levels in urine samples, airborne toluene levels, acute and chronic neurological symptoms, and to describe any correlation between urinary hippuric acid and airborne toluene. Methods: The hippuric acid concentration in the urine of 87 paint workers exposed to toluene at work (exposed group), and 87 nonexposed people (control group) was studied. Study participants were selected from similar factories in the same region. Urine samples were collected at the end of a shift and analyzed for hippuric acid by high performance liquid chromatography. Air samples for the estimation of toluene exposure were collected with diffusive personal samplers and the toluene quantified using gas-liquid chromatography. The two groups were also interviewed and observed about their work practices and health. Results: The median of the 87 airborne toluene levels was 55 ppm (range, 12-198 ppm). The median urinary hippuric acid level was 800 mg/g creatinine (range, 90-2547 mg/g creatinine). A statistically significant positive correlation was found between airborne toluene exposure and urine hippuric acid levels (r = 0.548, p < 0.01). Workers with acute symptoms had significantly higher hippuric acid levels than those who did not (p < 0.05). It was concluded that there was a significant correlation between toluene exposure, hippuric acid levels, and health (p < 0.001). Conclusion: There appears to be a significant correlation between workers exposure to toluene at work, their urine hippuric acid levels, and resulting symptoms of poor health. Improvements in working conditions and occupational health education are required at these workplaces. There was good correlation between urinary hippuric acid and airborne toluene levels.
