Diethyl maleate가 N-methyl-N'-nitro-N-nitrosoguanidine에 의해서 유발되는 랫드 위암 발생에 미치는 영향에 관한 병리학적 연구 (The Effects of diethyl maleate on the N-methyl-N'-nitro-N-nitrosoguanidine induced gastric carcinogenesis in rats)

  • This study was carried out to investigate the effects of diethyl maleate(DEM) on the carcinogenesis of forestomach and pyloric glandular stomach in rats caused by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG). A total of 60 male 6-week-old Wistar rats were given twice intragastric injection of MMNG(200mg/kg BW), then were given diets containing 5% NaCl for 3 weeks until 4th week of the experiment. And then the animals of groups of 1 and 2 were placed on diets containing 0.2% DEM for 16 weeks until the end of 20 weeks of the experiment. On the other hand, the animals of groups of 3 and 4 were placed on basal diets for the same periods. The tissues of forestomach and liver of each group were frozen in liquid nitrogen and the activities of quinone reductase(QR) were determined by measurement of the dicoumarol-sensitive reduction of dichloro-indophenol by NADPH at 600nm. All rats were sacrificed at the end of 20 weeks of the experiment. Every animal was fasted for 24 hrs prior to sacrifice. The forestomach was fixed in 10% neutral phosphate buffered formalin for histology and the pyloric gland was fixed in sublimated formalin for immunohistochemistry of pepsinogen 1 altered pyloric gland(PAPG). The final body weight of the group given MNNG and treated with 5% NaCI and DEM was significantly decreased compared with that of the group 4(p<0.05). Food and water consumption rates were not significantly changed. The preneoplastic and neoplastic lesions of the forestomach given MNNG and treated with 5% NaCI and DEM were significantly increased compared to those of the group 4(p<0.0l). The incidence of PAPG in the groups treated with 0.2% DEM was significantly increased compared with that of the group 4(group 1:p<0.01, group 2:p<0.05). The activities of QR of forestomach in the groups treated with 0.2% DEM were significanitly increased compared with those of the group 4(p<0.001), but those of liver were not significant. These results indicate that DEM exert the enhancing effect of forestomach and glandular stomach carcinogenesis in rats pretreated with MNNG and NaCl.

7일령 조기 이유자돈에 대한 액상사료 자동급이기 이용효과 (Studies on the Efficacy of the Newly Developed Automatic Liquid Feeder for 7-day Old Early Weaned Piglets)

  • 축산기술연구소에서 개발한 액상사료 자동급이기의 성능을 검사하고 이를 이용하여 조기 이유자돈의 사육 가능성을 알아보기 위하여 본 연구를 실시하였다. 포유중인 자돈 60두를 공시하여 대조구 자돈들은 21일령 까지 모돈에게 포유시켰다. 나머지는 생후 7일령에 모돈으로 부터 이유시킨 다음 액상사료 자동급이기를 이용하여 21일령까지 액상사료를 자동으로 인공급여 하였다. 그 이후 모든 자돈을 70일령 까지 단계적으로 같은 가루사료를 급여하였다. 그리고, 미국으로부터 수입되어 사용되고 있는 액상사료 자동급이기를 새로이 개발한 액상사료 자동급이기와 비교하여 사용하였다. 자돈들의 성장과 사료이용, 그리고 설사발생과 소장의 융모 변화 등을 조사하였다. 그 결과는 다음과 같다. 생후 7일에 모돈으로 부터 이유한 다음 액상사료 자동급이기를 이용하여 액상사료를 급여한 자돈들의 21일령 체중은 모두 포유를 지속한 자돈들에 비해 모두 유의하게(P〈0.05) 낮았다(5.55 vs 4.97 & 4.98 kg/head). 그 이후 액상사료 급여 자돈들의 성장이 빨라 70일령에 이르러서는 체중이 모돈으로 부터 포유하였던 자돈들에 비해 오히려 유의적으로(P〈0.05) 증가하는 결과를 보여 주었다(24.82 vs. 30.17 & 29.42 kg/head). 시험 전기간 동안의 일당증체량은(346.7 vs 425.8 & 416.1g/head) 모돈으로 부터 포유한 자돈보다 인공 포유 자돈들이 유의하게 증가하였다(P〈0.05). 사료요구율은 시험 전기간 동안 차이가 없었다(1.67 vs 1.78 and 1.84). 자돈들의 설사발생 빈도에 있어 액상사료 자동급이기 간에 차이가 없었다. 소장의 융모 변화를 비교한 결과 액상사료 인공포유 3일째(10일령) 십이지장 융모 높이가 크게 낮아진 것으로 나타났으나 7일째(14일령) 부터 융모가 회복되는 것으로 나타났다. 공장의 융모높이는 7일째(14일령) 크게 낮아진 것으로 나타났으나 14일째(21일령) 부터 융모가 회복되는 것으로 나타났다. 회장의 융모 높이는 처리간에 어떠한 경향을 보이지 않았다. 축산기술연구소에서 개발한 액상사료 자동급이기(NLRI)와 수입한 액상사료 자동급이기(IALF)에서 자돈 성장의 차이는 보이지 않았다. 이상의 결과 새로이 개발한 NLRI로 생후 7일만에 조기이유 시킨 자돈들도 성공적으로 육성할 수 있음을 보여주었다.

Dietary Fatty Acid Increases Body Weight Gain without a Change in Rumen Fermentation in Fattening Cattle

  • Dietary fatty acid including mainly palmitic acid and stearic acid was fed to fattening cattle and its effect on body weight gain, plasma lipid contents and rumen liquid fermentation in vitro was examined. In expt. 1, the effect of dietary fatty acid on body weight gain and plasma lipid concentrations was examined. In the control diet group, cattle were fed 1 kg/day of rice straw and concentrate which satisfied the requirement. In the fatty acid group, cattle were given 250 g/d of fatty acid with the same diet of the control diet group. In the excess concentrate group, cattle were given the same diet of the control diet group plus 735 g/d of concentrate corresponding to the same TDN of 250 g/d of fatty acid. Diets were given for 7 days. Body weight gain of cattle given dietary fatty acid was significantly greater than that of cattle fed only rice straw and concentrate. When dietary fatty acid was added to cattle feed, plasma NEFA and HDL-cholesterol concentrations increased. In expt. 2, the influence of dietary fatty acid on gas production and VFA profile in the rumen liquid was investigated in vitro. In the control group, 10 mg of rice straw and 90 mg of concentrate were incubated in the rumen fluid. In the excess concentrate group, 10 mg of rice straw and 97.5 mg of concentrate were incubated. In the fatty acid group, 10 mg of rice straw, 90 mg of concentrate and 2.5 mg of fatty acids were incubated. The rumen liquid mixed with feed materials was incubated for 24 h and the cumulative gas volume was measured. The VFA profile was also measured. Cumulative gas volume in the rumen liquid with fatty acid was equal to the control. Excess concentrate increased cumulative gas volume compared to the fatty acid group. There was no significant difference in total VFA concentration between experimental diet groups. It is suggested that dietary fatty acid has the potency to improve growth performance in fattening cattle without failure in rumen fermentation.

Flow of Soluble Non-ammonia Nitrogen in the Liquid Phase of Digesta Entering the Omasum of Dairy Cows Given Grass Silage Based Diets

  • An experiment was conducted to quantify the flow of soluble non-ammonia nitrogen (SNAN) in the liquid phase of ruminal (RD) and omasal digesta (OD), and to investigate diurnal pattern in SNAN flow in OD. Five ruminally cannulated Finnish-Ayrshire dairy cows in a $5{\times}5$ Latin square design consumed a basal diet of grass silage and barley grain, and that supplemented with four protein feeds (kg/d DM basis) as follows: skimmed milk powder (2.1), wet distiller' solubles (3.0), untreated rapeseed meal (2.1) and treated rapeseed meal (2.1). Ruminal digesta was sampled using a vacuum pump, whereas OD was collected using an omasal sampling system at 1.0 h interval during a 12 h feeding cycle. Both RD and OD were acidified, centrifuged to remove microbes and precipitated with trichloroacetic acid followed by centrifugation. The SNAN fractions (free amino acid (AA), peptide and soluble protein) in RD and OD were assessed using ninhydrin assay. Free AA, peptide and soluble protein averaged 60.0, 89.4 and 2.1 g/d, respectively, for RD, and 81.8, 121.5 and 2.5 g/d, respectively, for OD. Although free AA flow was relatively high, mean peptide flow was quantitatively the most important fraction of SNAN, indicating that degradation of peptide to AA rather than hydrolysis of soluble protein to peptide or deamination may be the most limiting step in rumen proteolysis. Diurnal pattern in flow of peptide including free AA in OD during a 12 h feeding cycle peaked 1 h post-feeding, decreased by 3 h post-feeding and was relatively constant thereafter. Protein supplementation showed higher flow of peptide including free AA immediately after feeding compared with no supplemented diet. There were no differences among protein supplements in diurnal pattern in flow of peptide including free AA in OD.

Quantification of Karanjin Using High Performance Liquid Chromatography in Raw and Detoxified Karanj (Pongamia glabra vent) Seed Cake

  • Various products of karanj (Pongamia glabra) are utilized for industrial, health and animal agriculture applications in the Indian subcontinent. Despite a rich source of protein (CP, 28-34%), karanj cake was found to be slightly bitter in taste and toxic owing to the presence of flavonoid (Karanjin), restricting its safe inclusion in the livestock diets. Feeding trials with raw cake revealed its poor palatability and adverse performance among different categories of livestock including poultry. The present study was, therefore, aimed to detoxify karanj cake by various physico-chemical methods like solvent extraction, water washing, pressure cooking and alkali and acid treatments. The level of residual karanjin in raw and variously processed cake was quantified using high performance liquid chromatography (HPLC). The raw expeller karanj cake was found to contain about 0.19% of karanjin. Though a non-polar solvent, soxhlet extraction of expeller pressed cake with petroleum ether drastically reduced karanjin content (0.01%). Soaking of cake for 24 h in 1% NaOH (w/w) solution was found to reduce karanjin to a major extent with little further benefit by increasing alkali level. Milder alkalies like lime and fertilizer grade urea reduced the karanjin levels marginally. Similar was the case with mineral acids such as HCl and glacial acetic acid. It was, therefore, concluded that solvent extraction of karanj seeds would be the best method of detoxification as well as for more recovery of oil and karanjin.

죽초액의 첨가가 재래닭의 생산성 및 육질에 미치는 영향

  • This experiment was conducted to investigate the effects of Bamboo Vinegar Liquid(BVL) on performance and meat quality of Korean native chicken(KNC) for 8 weeks. One hundreds KNC were divided into 4 groups with 5 replicates per treatment and placed in pens. Dietary levels of food waste 0%, 1%, 2% and 4% were included in experimental diets. The growth performance and carcass weight of KNC fed BVL supplement diet were significantly different(P<0.05) from the control group. The lipid content, cholesterol content and TBA in breast meat were significantly(P<0.05) decreased by addition of BVL. However, the addition of BVL did not affect the meat color. The saturated fatty acid composition of breast meat was decreased(P<0.05), whereas unsaturated fatty acid was increased(P<0.05) by addition of BVL 1% and 2%. Taste of sensory evaluation was improved by addition BVL. The results of this study indicate that dietary supplemental BVL 1% and 2% may improve the performance and meat quality of KNC. The prevention effect of KNC which challenged with Salmonella gallinarum (1${\times}$10$\^$9/ CFU/ml) was evaluated. A reisolation of S.gallinarum in liver, pathological index of villus and colon were evaluated. There was no reisolation of S.gallinarum in liver and a few pathological index of colon in 4% BVL supplement fed KNC than 1 and 2%. These results indicate that dietary supplemental BVL 4% may improve the prevention effect to salmonellosis in chickens.

Effects of excessive dietary methionine on oxidative stress and dyslipidemia in chronic ethanol-treated rats

  • BACKGROUND/OBJECTIVE: The aim of this study was to examine the effect of high dietary methionine (Met) consumption on plasma and hepatic oxidative stress and dyslipidemia in chronic ethanol fed rats. MATERIALS/METHODS: Male Wistar rats were fed control or ethanol-containing liquid diets supplemented without (E group) or with DL-Met at 0.6% (EM1 group) or 0.8% (EM2 group) for five weeks. Plasma aminothiols, lipids, malondialdehyde (MDA), alanine aminotransferase (ALT), and aspartate aminotransferase were measured. Hepatic folate, S-adenosylmethionine (SAM), and S-adenosylhomocysteine (SAH) were measured. RESULTS: DL-Met supplementation was found to increase plasma levels of homocysteine (Hcy), triglyceride (TG), total cholesterol (TC), and MDA compared to rats fed ethanol alone and decrease plasma ALT. However, DL-Met supplementation did not significantly change plasma levels of HDL-cholesterol, cysteine, cysteinylglycine, and glutathione. In addition, DL-Met supplementation increased hepatic levels of folate, SAM, SAH, and SAM:SAH ratio. Our data showed that DL-Met supplementation can increase plasma oxidative stress and atherogenic effects by elevating plasma Hcy, TG, and TC in ethanol-fed rats. CONCLUSION: The present results demonstrate that Met supplementation increases plasma oxidative stress and atherogenic effects by inducing dyslipidemia and hyperhomocysteinemia in ethanol-fed rats.

The Nutritive Value of Thin Stillage and Wet Distillers' Grains for Ruminants - Review -

  • Thin stillage and distillers' grains are byproducts remaining after alcohol distillation from a fermented cereal grain mash. Both byproducts are used as energy and protein sources for ruminants. Due to its liquid nature, more than 50% of thin stillage bypasses the rumen. Thin stillage can be fed alone or in combination with distillers' grains. However, a better utilization by beef cattle is anticipated when thin stillage replaces water as a fluid source. Ruminal undegraded protein content of distillers' grains is greatly affected by type of cereal grain and by drying. Corn distillers' grains have a higher ruminal undegraded protein content than wheat distillers' grains while dried distillers' grains have a higher ruminal undegraded protein content than the wet distillers' grains. Wet and dried distillers' grains can replace up to 50% of corn grain in beef cattle diets without affecting animal performance. The estimated NEg of corn distillers' grains for beef cattle ranges from 100 to 169% of that of corn. In general, wet corn distillers' grains have a higher NEg value than dried corn distillers' grains and the addition of thin stillage improves the NEg of distillers' grains. Improved performance of ruminats fed distillers' byproducts can be attributed to high digestible fiber content, improved rumen environment and a shift in organic matter digestion from the rumen to the small intestine.

  • The concentrations of $\alpha$-tocopherol in the brain, liver, and serum were studied with and without saponification process between control and vitamin E supplemented rats. Young rats, 80-120g body weight, were fed control and vitamin E supplemented diets, ad libitum, for four weeks. $\alpha$-Tocopherol concentrations were determined by high pressure liquid chromatography. The $\alpha$-tocopherol concentration per wet weight base in the brain tissue was significantly lower than that in the liver. Vitamin E supplementation had no effect on brain $\alpha$-tocopherol levels in contrast to the significant increase in lover $\alpha$-tocopherol concentration with and without saponification is significantly greater in the brain than in the liver or serum. Further study is needed to clarify the nature of interaction or /and binding between $\alpha$-tocopherol and the complex membrane system in brain tissue. It can be speculated from this and other studies that the metabolism and the nature of interaction of $\alpha$-tocopherol with the complex membrane system in brain tissue rich in polyunsaturated fatty acids seems different from that in liver tissue or serum.

Supplemental Fermented Milk Increases Growth Performance of Early-Weaned Pigs

  • Early weaning is a means of breaking the disease cycle from sow to piglet as well as capitalising on the enormous growth potential of the pig. However, the transition from milk to dry diets results in a growth check. Feeding of supplemental milk, fermented to reduce pH and enterotoxigenic bactetial proliferation, may be a means of gradually weaning pigs on to solid feed. This study involved 216 pigs weaned from the sow at 12 days of age, allocated to groups of 6 males and 6 females per weaner pen and allowed ad libitum access to a pelleted diet. In addition, half the pigs were given supplemental fermented skim milk for the first 8 days after weaning. Feeding supplemental fermented milk increased feed intake (104 vs. 157 g DM/d, p=0.011), average daily gain (-3 vs. 112 g/d, p<0.001) and feed conversion efficiency (0.01 vs. 0.81, p=0.003) over the first 8 days after weaning. The improvements observed in the supplemented pigs continued to be augmented such that, by 42 days of age, the pigs that had received supplemental fermented milk were heavier (9.6 vs. 11.5 kg, p=0.003) than their unsupplemented counterparts. Feeding fermented supplemental milk to early-weaned pigs can improve growth performance in the immediate and subsequent post-weaning period.
