장려품종 콩의 영양저해 인자 및 리폭시게나아제 특성 (The Contents of Antinutritional Factors and Lipoxygenase Activity of the Recommended Soybean Varieties in Korea)

  • 장려품종 콩 19종에 함유된 몇 가지 영양저해 인자의 함량과 리폭시게나아제의 역가를 조사하였다. 복부팽만인자인 라피노오스와 스타키오스의 함량은 각각 $0.74{\sim}1.58%\;3.34{\sim}5.30%$ 범위이었으며 힐, 백운, 장백 등의 품종에서 이 두 종류 당의 합량이 높았다. 콩에 함유된 트립신 저해제는 $21.2{\sim}37.0mg$ TI/g, 피트산 형태로 존재하는 인은 건물 100g 당 $337{\sim}605mg$ 정도로 품종에 따라 큰 차이를 보였다. 한편, 콩에 함유된 리폭시게나아제의 역가는 $163.6{\sim}403.5unit$로 팔달, 장백, S-133등의 품종이 높은 값을 나타내었다.

Lipoxygenase 결핍 콩의 가공 및 관능 특성 (Processing and Sensory Characteristics of Lipoxygenase-Deficient Soybeans)

  • 본 실험은 콩비린내의 주요 생성효소인 LOX가 결핍된 콩의 가공시 특성을 살펴봄으로 식품학적 이용 가능성을 검토해 보고자 실시하였다. 몇가지 가공상태의 콩의 LOX 활성을 측정한 결과 전지콩분말, 탈지콩분말, 콩단백의 순으로 LOX의 활성이 감소되는 것으로 나타났는데 특히 탈지와 콩단백 제조시 황금콩의 활성 저하가 가장 현저하였다. 콩나물 재배 시에는 황금콩과 진품콩의 자엽 부분에서 LOX 활성이 상당히 나타났다. 두유 제조시에는 진품콩과 진품콩 2호로 만든 경우 황금콩 두유 보다 콩비린내가 적게 난다고 평가되었으며, 콩나물에서는 진품콩 2호가 배축 신장율과 수율은 떨어졌으나 콩나물 비린내는 적은 것으로 평가되었다. 아이스크림 제조 시에는 황금콩 탈지분을 이용한 경우 탈지분으로 만든 대조군 아이스크림에 비하여 콩비린내가 많이 나는 것으로 감지되었으나 진품콩 2호 탈지분을 이용한 경우에는 콩비린내를 거의 감지하지 못하였다. 이상에서 LOX 결핍 콩은 가공시비린내 개선의 효과를 나타내어 가공식품으로서의 이용 가능성을 나타내 주었다.

Characteristic Changes in Brown Rice (Oryza sativa L.) Cultivars of 3 Ecotypes During Different Storage Conditions

  • The aim of this study investigated the fluctuations of 3 characters from 3 ecotypes [early ripening (ER), middle ripening (MR), and late ripening (LR)] of 20 Korean brown rice cultivars in different storage systems [time: 12 and 24 weeks, temperature: low ($10^{\circ}C$) and room ($25^{\circ}C$)]. With increase of storage time and temperature, lipoxygenase activity, and fat acidity increased, whereas germination rate was reduced. ER cultivars exhibited the highest lipoxygenase activity of $35.49{\pm}2.46$ unit/mg protein during 24 weeks storage at $25^{\circ}C$, followed by LR ($32.73{\pm}1.33$) and MR ($32.66{\pm}1.62$) cultivars. The amounts of fat acidity also were observed by the same order (ER: $20.40{\pm}2.12$>LR: $19.68{\pm}1.86$>MR: $19.64{\pm}1.35$ mg KOH/100 g). Germination rate slightly decreased with increase of time and temperature (MR>LR>ER), but MR and LR cultivars showed the most significant changes (ER: $60.90{\pm}23.47%$, MR: $32.66{\pm}13.95%$, and LR: $32.53{\pm}5.87%$). On the basis of above results, MR cultivars were evaluated the highest quality, because high lipoxygenase activity, high fat acidity, and low germination rate have deteriorated in quality and generated off-odor. Thus, MR cultivars might be very important sources in food processing and stored dietary supplement aspects.

고형상의 모델시스템에 있어서 리놀레산의 산화에 미치는 리폭시게나아제, 카로틴, 토코페롤 및 수분활성의 영향 (Effect of Lipoxygenase, ${\beta}-Carotene$, ${\alpha}-Tocopherol$ and Water Activity on the Oxidation of Linoleic Acid in Starch-Solid Model System)

  • 고형상의 모델시스템에서 리놀레산의 산화에 영향을 미치는 여러가지 인자들 즉 리폭시게나아제, 수분활성도, ${\beta}$-카로틴, ${\alpha}$-토코페롤등에 대해 관찰하여 다음과 같은 결과를 얻었다. 리폭시게나아제에 의한 리놀레산의 산화반응은 aw 0.72에서 aw 0.25에 보다 잘 일어났다. 그러나 이 시스템에 ${\beta}$-카로틴이나 ${\alpha}$-토코페롤을 반응시키게 되면 리놀레산의 산화를 크게 저해하였으며 ${\beta}$-카로틴보다는 ${\alpha}$-토코페롤이 지질과 산화 과정에서 항산화작용이 더 컸다. 즉, 유사한 조건에서 ${\beta}$-카로틴은 24시간의 유도기간을 보여 준 반면 ${\alpha}$-토코페롤은 72시간까지도 유도기간이 지속되었다. ${\beta}$-카로틴의 항산화작용은 수분활성도의 영향을 크게 받았으며 수분활성도가 높을수록 항산화 작용이 뛰어났고 또한 ${\beta}$-카로틴의 농도가 높을수록 CDA 생성이 적었다.

Cumin(Cuminum cymium L.) seed로부터 정제한 $2-hydroxyethyl-{\beta}-undecenate$의 항치주염 효과의 효소학적 평가 (Enzymatic Assessment of $2-Hydroxyethyl-{\beta}-undecenate$ Purified from Cumin (Cuminum cymium L.) Seed for Anti-periodontitis)

  • 본 연구자들은 Cumin(Cuminum cymium L.) seed로부터 정제한 $2-hydroxyethyl-{\beta}-undecenate$의 항치주염 활성을 사람의 백혈구를 이용하여 염증 매개물질 $LTB_4$ 및 PGs의 생성 효소인 5-lipoxygenase와 cyclooxygenase, 및 조직파괴 효소인 collagenase와 elastase의 저해 효과를 규명하여 다음과 같은 결론을 얻었다. HPS의 항치주염 활성을 사람의 백혈구를 이용하여 염증 매개물질 $LTB_4$ 및 PGs의 생성 효소인 5-lipoxygenase와 cyclooxygenase 및 조직파괴효소인 collagenase와 elastase의 저해효과를 규명한 결과, HPS은 $5\;{\times}\;10^{-2}\;M$ 첨가 시 97%의 PMNL로부터 $LTB_4$ 생합성이 저해되었으며 이때 $IC_{50}$ 값은 $2\;{\times}\;10^{-4}\;M$ 이었다. 균질화된 사람의 PMNL 5-lipoxygenase 및 시판 정제 5-lipoxygenase 활성에 대한 HPS의 저해 효과는 $5\;{\times}\;10^{-2}\;M$첨가 시 92%의 효소 활성이 저해되었다. 또한 효소 활성에 대한 $IC_{50}$ 값은 $2.5\;{\times}\;10^{-4}\;M$이었으며, 정제 5-lipoxygenase에 대한 $IC_{50}$ 값은 $2.3\;{\times}\;10^{-4}\;M$이었다. 또한 백혈구 COX 활성에 대한 $IC_{50}$ 값은 $5.1\;{\times}\;10^{-4}\;M$이었고, 정제 COX에 대한 $IC_{50}$ 값은 $2.3\;{\times}\;10^{-4}\;M$이었으며, 백혈구 collagenase 활성에 대한 $IC_{50}$ 값은 $2\;{\times}\;10^{-3}\;M$이었고 정제 collagenase에 대한 $IC_{50}$ 값은 $5\;{\times}\;10^{-2}\;M$이었다. 백혈구 elastase의 경우 $5\;{\times}\;10^{-2}\;M$ 첨가 시 66%의 활성이 저해된 반면 정제 elastase의 경우 $5\;{\times}\;10^{-2}\;M$ 첨가 시 25%의 효소 활성이 저해되었다. 또한 백혈구 elastase 활성에 대한 $IC_{50}$ 값은 $7.5\;{\times}\;10^{-3}\;M$이었다. 인체 치은세포에 대한 독성 시험 결과 $5\;{\times}\;10^{-2}\;M$의 HPS 첨가시 세포의 활성은 배양 2일째 47.83%로 나타났으며, $1\;{\times}\;10^{-2}\;M$의 HPS 첨가시에도 68.53%로 나타나 비교적 세포독성이 강한 것으로 나타났다.

Biological Significance of Essential Fatty Acids/Prostanoids/Lipoxygenase-Derived Monohydroxy Fatty Acids in the Skin

  • The skin displays a highly active metabolism of polyunsaturated fatty acids (PUFA). Dietary deficiency of linoleic acid (LA), an 18-carbon (n-6) PUFA, results in characteristic scaly skin disorder and excessive epidermal water loss. Although arachidonic acid (AA), a 20-carbon (n6) PUFA, is metabolized via cyclooxygenase pathway into predominantly prostaglandin $E_2(PGE_2)$ and $PGF_{2{\alpha}}$, the metabolism of AA via the 15-lipoxygenase (15-LOX) pathway, which is very active in skin epidermis and catalyzes the transformation of M into predominantly 15S-hydroxyeicosatetraenoic acid (15S-HETE). Additionally, the 15-LOX also metabolizes the 18-carbon LA into 13S-hydroxyoctadecadienoic acid (13S-HODE), respectively. Interestingly, 15-LOX catalyzes the transformation of $dihomo-{\gamma}-linolenic$ acid (DGLA), derived from dietary gamma-linolenic acid, to 15S-hydroxyeicosatrienoic acid (15S-HETrE). These monohydroxy fatty acids are incorporated into the membrane inositol phospholipids which undergo hydrolytic cleavage to yield substituted-diacylglycerols such as 13S-HODE-DAG from 13S-HODE and 15S-HETrE-DAG from 15S-HETrE. These substituted-monohydroxy fatty acids seemingly exert anti-inflammatory/antiproliferative effects via the modulation of selective protein kinase C as well as on the upstream/down-stream nuclear MAP-kinase/AP-1/apoptotic signaling events.

Inhibition of Tyrosinase and Lipoxygenase Activities by Resveratrol and Its Derivatives from Seeds of Paeonia lactiflora

  • Previously, a methanol extract from seeds of Paeonia lactiflora was shown to have a potent inhibitory activities against tyrosinase and soybean lipoxygenase (SLO). Seven stilbenes, trans-resveratrol-4-Ο-$\beta$-D-glucoside, trans resveratrol, trans-$\varepsilon$-viniferin, cis-$\varepsilon$-viniferin, gnetin H, suffruticosol A and B were isolated from the seeds as active principles for inhibition of the enzymatic activity. Among them, the resveratrol trimer, gnetin H exhibited the most potent inhibitory activities against tyrosinase and SLO, respectively. Additionally, the resveratrol dimers, trans-$\varepsilon$-viniferin and cis-$\varepsilon$-viniferin exhibited significant inhibitory activity against the two oxidative enzymes. Meanwhile, three other stilbene derivatives, such as trans-resveratrol, suffruticosol A and suffruticosol B had also weak inhibition activity. The least inhibitory activity was observed in transresveratrol-4-Ο-$\beta$-D-glucoside. These results suggest that resveratrol dimers and trimer in the seeds of Paeonia lactiflora are potentially useful therapeutic agents against pathological disorders such as hyperpigmentation and inflammation.

마늘의 가공 조리 방법에 따른 lipoxygenase 활성도 저해 효과 (Inhibition of Lipoxygenase Activity by the Extract of Various Processed Garlic)

  • Bioactivity of the extract from various processed garlic was evaluated based on the inhibition of lipoxygenase(LPO), and the effect of some stabilizers on the bioactivity was investigated. Water, ethanol or chloroform extract of 30 min boiled garlic showed 75%, 76% or 70% inhibition, respectively, compared to extracts of fresh garlic. In pickled garlic, LPO inhibition decreased gradually during storage. Chloroform extract of 40 day-stored pickled garlic inhibited LPO by 77%, and even on 60th day storage it still retained inhibitory effect of 73%, compared to that of fresh garlic. Meanwhile, the powdered (freeze-dried) garlic showed more bioactivity(80%) than the other processed garlics, and moreover, the jrreversible/unstable components seem to be stabilized by freeze-drying. The optimum pH for stabilization of bioactive components in garlic macerate was pH 3 for 48 hr incubation and pH 11 for 6 hr incubation. In addition, the effect of NaCl was not so great, although but maximal stabilization was observed at 150 mM. Stabilizing effect of $\alpha$-tocopherol was markedly great, and at 6mM it showed 308% stabilizing effect after 48hr incubation. More stabilizing effect was observed at lower concentrations of ascorbic acid($\leq$0.6mM) than higher concentrations. The stabilizing effect of soybean oil was found to be remarkable only during initial period(6 hr) of incubation.

제1형 과민 반응에 미치는 고려인삼의 영향 (Effects of Panax ginseng on Type I Hypersensitivity)

  • Effects of Panax ginseng on allergic reactions were studied uslng various in vivo and in vitro experimental models such as 48-hr passive cutaneous anaphylaxis, mediators-induced skin reactions, histamine release from rat peritoneal mast cells, hexosaminidase release from RBL-2H3 cells, and lipoxygenase assay . In all of anti-allergic experiments we conducted, ginseng components (50% ethanol extract or ginseng total saponin or ginsenosides) extracted from Korean red ginseng, did not show significant anti-allergic actions. In 48-hr passive cutaneous anaphylaxis and mediators-induced skin reactions, 50% ethanol extract did not suppress hypersensitivity reactions. Total saponin, 50% ethanol extract, and 8 major ginsenosides did not show inhibitory effects on lipoxygeanse activity. Ginseng total saponin did not inhibit histamine release from rat peritoneal mast cells. All of the ginseng components mentioned above were also tested on RBL-2H3 cells, but none of them inhibited hexosaminidase release from this cell line. These results suggest that Panax ginseng does not have effects on allergic reactions at the level of 50% ethanol extract or total saponin used. All of 8 major saponin components tested ($Rb_1$, $Rb_2$, Rc, Rd, Re, Rf, $Rg_1$, $Rg_2$), did not inhibit lipoxygenase activity and degranulation events.
