Effect of Arctii Fructus on Low Density Lipoprotein Oxidation (우방자 분획물의 저밀도 지질단백질 산화에 미치는 산화에 미치는 영향)

  • Fruits of Arctium lappa L. (Compositae), which has been used as the antiinflammatory, detoxifying and diuretic agents in the folk remedies, was examined on the in vitro oxidation of human low density lipoprotein (LDL). Several lines of evidence indicate that oxidized LDL (Ox-LDL) may promote atherogenesis. Hence. The role of antioxidants in the prevention of LDL oxidation needs to be determined. The activity of fractions of Arctii Fructus treated with oxidized LDL which was incubated with $16\;{\mu}M$ of $Cu^{2+}$ for metal catalyzed oxidation was investigated. The BuOH fraction (4\;ppm) inhibited the oxidative modification of LDL as evidenced by a decrease in the lipid peroxide content (thiobarbituric acid reacting substances activity), the negative charge of LDL (electrophoretic mobility) and increase of the vitamine E content.

Radioimmunoassay of High-Density Lipoprotein in Japanese Quail Serum (메추리혈청 리포단백질의 방사면역분석법)

  • The development of radioimmunoassay (RIA) for high-density lipoprotein (HDL) in Japanese quail serum will contribute to the screening of drugs acting on cholesterol transport. We have developed a double antibody RIA method for J. quail HDL. The first antibody was raised in rabbit by immunization of HDL isolated by the dextrane sulfate-$Mn^{#}$ precipitation method. For the preparation of raclioiodinated antigen, HDL was further purified by combination of electrophoretic procedure. Using the second antibody raised in goat by rabbit IgG, we have furnished the RIA method for HDL. It showed high specificity and sensitivity of working assay range, 0.1-33.mu.g HDL/tube. There was no correlation between the radioimmunoassay of HDL and the enzyme assay of HDL-cholesterol.

Effects of Bursectomy and Thyectomy on the Immunoglobulins in Korean Ogol Chickens (오골계의 Fabricius 낭과 흉선 적출이 Immunoglobulin 에 미치는 영향)

  • The concentration of serum proteins, lipoproteins, leukocytes and lymphocytes in bursectomy and thymectomy Ogol chickens were examined from 5 days to 150 days of age. Concentration of serum proteins and lipoproteins were measured by cellulose acetate electrophoresis. The results summarized as follows : 1. The gamma-globulin concentrations in growing Korean native chickens and Ogol chickens were increased 0.41$\pm$0.01~0.85$\pm$0.05 mg/㎗ and 0.50$\pm$0.01~0.98$\pm$0.08 mg/㎗ from 1 day after to 5 days of age and gradually decreased at 10 days of age, and thereafter increased 0.82$\pm$0.06 mg/㎗, 1.09$\pm$0.04 mg/㎗ at 100 days of age. 2. The serum apha-, beta-, gamma-lipoprotein concentrations of growing Ogol chickens were 74.1$\pm$6.8~240.2$\pm$9.7 mg/㎗, 89.7 5.7~225.8$\pm$11.3 mg/㎗ and 87.6$\pm$4.7-220.3$\pm$10 mg/㎗, respectively. The serum -lipoprotein concentrations from 5 days to 150 days of age Ogol chickens were significantly decreased. 3. The leukocyte and lymphocyte counts in bursectomy and thymectomy Ogol chickens slightly increased from 10 to 100 days of age. However the leukocyte and lymphocyte counts in thymectomy chickens were lower than in untreated chickens. 4. The Ig concentrations of 1 day to 50 days(0.30$\pm$0.03~0.58$\pm$0.04 g/㎗) in bursectomy chickens were similar to those found in untreated chickens and thereafer increased from 50 to 150 days of age(0.58$\pm$0.04~1.21$\pm$0.05 g/㎗). The Ig concentrations in thymectomy chickens increased significantly.

Effects of the Feeding Mixed with Various Levels of n-3 and n-6 Polyunsaturated Fatty Acid on the Lipid Components and Fatty Acid Metabolism of Serum Lipoprotein in Hyperlipidemic Rats (n-3 및 n-6계 다불포화 지방산의 함유비율이 다른 유지가 고지혈증 흰쥐의 혈청 지단백 지질성분 및 지방산 대사에 미치는 영향)

  • This study was designed to observe the effects of the feeding mixed with the sardine oil containing n-3 EPA, DHA and the safflower oil in which rich n-6 linoleic acid on the improvement of the lipids and on the fatty acid matabolism of serum lipoprotein in the dietary hyperlipidemic rats. Experimental oils mixed with 16% butter(control group) and 8% butter+8% olive oil, 8% butter and various level of sardine and safflower oils were administered to the male rats of the Sprague Dawley for 4 weeks. Concentrations of total cholesterol in serum lipoproteins were highest in the LDL, followed by VLDL, HDL and chylomicron components. in the triglyceride concentrations, chylomicron were highest, and much VLDL, LDL, HDL components. There were highest in the HDL, followed by LDL, VLDL and chylomicron in phospholipid concentrations of serum lipoproteins. Concentrations of total cholesterol, triglyceride and phospholipid in serum lipoprotein were lower in the other groups than in the control groups, and particularly, lowest in the group 5(4% sardine oil+4% safflower oil). From this results, the feeding equal quantity mixed oil with n-3 PUFA rich sardine oil and n-6 PUFA rich safflower oil were effective on the improvement of the lipid composition in the serum lipoprotein. It might be due to the effects of appropriate rations of P/S, 0.85 and n-6/n-3P, 2.85 in the test lipids. In the fatty acid composition of serum lipoprotein, ratio of P/S and n-3/n-6P were influenced from the fatty acid compositions of the mixed test lipids. And EPA contents were higher in the chylomicron components.

  • This research was intended to know the influence of ${\omega}3\;and\;{\omega}6$ polyunsaturated fatty acids on rat's brain, kidney, testis tissues and composition of serum lipoprotein. Different kinds of diets containing perilla oil (PO) and red pepper oil (RPO) were administered to the rats: group 1, 10% PO; group 2, 7.5% PO and 2.5% RPO ; group 3, 5% PO and 5% RPO ; group 4, 2.5% PO and 7.5% RPO ; group 5, 10% RPO. The following are the results. 1. No significance of cholesterol concentration in brain was noticed among the groups, but group 5 was the highest and other groups tended to decrease. 2. As for concentration of triglyceride and phospholipid, the groups with more percentage of RPO increased gradually while those with more PO decreased by degrees. 3. No significance of cholesterol level in kidney was found among the groups. About triglyceride level, group 5 was the highest and it was relatively low in groups2, 3 and 4. group 5, however, was the lowest in phospholipid level. 4. As to cholesterol concentration in testis, groups 1 to 4 were low. About triglyceride level, group 1 increased while groups 2 to 4 decreased. 5. Concerning lipoprotein pattern in serum, HDL increased in groups 1 to 3, while LDL decreased gradually with the increase of perilla oil.

The Effect of Blood Nutrient Index on Depression (성인 여성의 혈액 영양소 지표가 우울지표에 미치는 영향)

  • The purpose of this study is that the blood nutrient indicator in adult women, is the depressive indicator. This study recruited 77 pre-menopausal women, and excluding missing values, 53 (mean age 44.2±6.5 years) blood indexes were statistically confirmed and then analyzed the effect of indexes on the Beck Depression Inventory. The Beck's Depression Inventory(BDI)'s average of the study subjects was 7.6±7.2, and most of the depression symptoms were within the normal range. Only two women were very serious and other two women were serious state. As a result of this study, 21 questions of BDI was classified into 5 sub-factors. Of these, 2 factors were considered physical depression, 1 factor was mood depression, 1 factor was cognitive depression, and 1 factor undecided. As a result of the correlation analysis, triglyceride, LDL(low density lipoprotein) cholesterol and potassium showed statistically significant positive relationship with depressive factors. On the other hand, HDL(high density lipoprotein) cholesterol, sodium and chloride ions showed negative relationship with depressive factors. When multiple regression analysis was performed. Considering the depression factors was dependent variable, and triglyceride, cholesterol, blood sugar, and ketone bodies was considered as independent variables. HDL(high density lipoprotein) cholesterol had a statistically significant negative effect on the depression factor 3. Considering the depression factors was dependent variable, and minerals were considered as independent variables. Potassium had a positive effect, and chloride ions had a negative effect on the depressive factor 3. This study was limited to 77 pre-menopausal women. In the future, post-menopausal women, men, and clinical neurological disorders group will be possible.

Effect of Schizandra chinensis Extract in Hyperlipidemic Rats (오미자 추출물이 고지혈증 흰쥐에 미치는 영향)

  • The purpose of this study was to investigate the effect of Schizandra chinensis extract on the serum and hepatic lipids inhyperlipidemic rats fed with high-fat diet. The Schizandra chinensis extract decreased significantly in total serum cholesterol, serum triacylglycerol and heptic triacylglycerol but not in the total hepatic cholesterol. The Schizandra chinensis extract also decreased in very low density lipoprotein increased by the highfat diet without affecting high density lipoprotein cholesterol. These results suggest that Schizandra chinensis extract may be benificial for the regulation of hyperlipidemia.

  • Forteen healthy women, with normal serum cholesterol levels, were supplemented with two egg yolks daily for the 1st week and four egg yolks for the 2nd week to their customary diets for 2 weeks period. Serum cholesterol, triglyceride, phospholipid and lipoprotein fractions were determined. Serum cholesterol and phospholipid levels were not affected by the supplementation of egg yolks. However, triglyceride level was significantly increased by the supplementation of four egg yolks at the end of the 1st week, but the level remained normal range. The serum lipoprotein pattern was influenced by the supplementation of egg yolks : HDL fraction significantly decreased, while LDL and VLDL fractions significantly increased.

  • Acanthopanax species (Araliaceae) traditionally has been used as analgesics, stimulant of immune system, and replenishment of body functions. Acanthopanax divaricatus var. albeofructus is indigenous plant to Korea. The antioxidant activities of compounds from A divaricatus var. albeofructus were determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and thiobarbituric acid reactive substance (TBARS) assay on human plasma low-density lipoprotein (LDL). The triterpenoid and lignan constituents from this plant showed antioxidant activities and the lignan, l-sesamin exhibited the most potent antioxidant activity in Cu$^{2+}$ -induced LDL oxidation.n.
