마우스에서 사염화탄소로 유발된 급성 간독성에 대한 EDTA 및 EGTA의 보호효과 (Protective effects of EDTA and EGTA against CCl4-induced acute hepatotoxicity in mice)

  • This study investigated the protective effects of ethylene glycol-bis(${\beta}$-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA), an extracellular calcium chelator, and ethylenediaminetetraacetic acid (EDTA), which chelates calcium and most metal ions, against carbon tetrachloride ($CCl_4$)-induced acute hepatotoxicity in mice. Mice were treated with EGTA or EDTA at a dose of 20 (low) or 100 mg/kg (high) subcutaneously 1h before $CCl_4$ administration. The mice were fasted and sacrificed 18h after $CCl_4$ treatment. Blood samples were collected from the carotid artery by decapitation under light ether anesthesia. Serum alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), triglyceride (TG), and cholesterol levels were measured. Malondialdehyde (MDA) production was determined as an index of lipid peroxidation in the liver. The liver, kidneys, and spleen were weighed. We also evaluated the histopathological changes in the liver in each group. The relative weights of the liver were significantly higher in the $CCl_4$-treatment group than in the normal group, except in the high-EDTA treatment group. EGTA and EDTA treatment caused a significant decrease in serum ALP, ALT, and AST levels. Of all of the doses of EGTA and EDTA tested, the high-EDTA dose resulted in the most remarkable inhibitory action. The protective effect in the high-EDTA-treatment group was confirmed histopathologically. The low-EGTA-treatment group showed a significant decrease in serum TG and cholesterol levels. Liver MDA levels were significantly decreased in the EGTA (20 mg/kg) and EDTA (20, 100 mg/kg) groups. These results suggest that EDTA, which chelates both calcium and metal ions, confers better protection in $CCl_4$-induced acute liver damage than does EGTA, a calcium chelator.

지황(地黃)의 품질평가 (Quality Evaluation of Various Dried Roots of Rehmannia glutinosa)

  • The dried roots of Rehmannia glutinosa (RG) have been used in the traditional medicine for treatment of diabetes, fever and dysuria, etc. In order to investigate the quality evaluation of the dried roots of RG, we conducted the physico-chemical and biological evaluation method. The amount of catalpol from commercial samples is very diverse about $0.00{\sim}3.89%$ by using HPLC method, because it is easily decomposed by processing of RG. So, we should try to identify the correlation with the contents of catalpol and biological activities of RG. We chose 3 samples which were a wide difference of catalpol contents between each sample (Sample-I; 3.4%, Sample-II; 2.8%, Sample-III; 0.05%). Sample-I and Sample-II were found to be more effective than Sample-III on the DPPH free radical scavenging effect and inhibitory effect on $H_2O_2-catalyzed$ lipid peroxidation in rat liver in vitro. And, Sample-I and Sample-II exhibited more significant effects than Sample-III on accelerating actions of the small and large intestinal transport, and diuretic action in mice. So, it is suggested that the quantitative determination of catalpol should be required for the tandardization of the dried roots of RG.

고려인삼(高麗人蔘)의 노화억제작용(老化抑制作用)에 관한 연구(硏究) 제1보(第1報) 홍삼(紅蔘)과 백삼(白蔘)의 노화억제작용(老化抑制作用) 비교연구(比較硏究) (Studies on the Anti-aging Action of Korean Ginseng (I) Comparative Study of Red and White Ginsengs on Anti-aging Action)

  • 홍삼(紅蔘)과 백삼(白蔘) 항산화작용(抗酸化作用)에 의한 노화억제작용(老化抑制作用)을 구명(究明)하기 위하여 80% ethanol extract 및 수포화(水飽和)부탄올로 이행(移行)한 saponin을 시료(試料)로 하여 in vitro 및 in vivo 실험(實驗)을 통하여 홍삼(紅蔘)과 백삼(白蔘)의 노화억제작용(老化抑制作用)을 비교(比較)하였다. DPPH에 대한 전자공여능(電子供與能)(EDA), TBA value 및 peroxide value에 의한 과산화지질생성억제작용(過酸化脂質生成抑制作用), superoxide dismutase 및 peroxidase의 활성측정(活性測定)등으로 홍삼(紅蔘)과 백삼(白蔘)의 노화억제작용(老化抑制作用)을 비교(比較)한 결과(結果), 홍삼(紅蔘)은 장기간(長期間)에 걸친 연속투여(連續投與)에서 백삼(白蔘)보다 과산화지질생성억제작용(過酸化脂質生成抑制作用). superoxide dismutase 및 peroxidase의 활성(活性)을 유의성(有意性)있게 증가(增加)하였다. 생체내실험(生體內實驗)에서 홍삼(紅蔘)이 백삼(白蔘)보다 노화억제작용(老化抑制作用)이 현저한 것은 증삼(蒸參) 및 가공처리중(加工處理中) saponin pattern의 변화(變化)에 기인(起因)하며 백삼(白蔘)의 PT/PD ratio가 0.401인데 반(反)해 홍삼(紅蔘)의 PT/PD ratio가 0.561로서 triol saponin이 증가(增加)되었기 때문이다.
