Evaluation of Contact Lens Care Regimens in Removing Tear Lipids on Silicone Hydrogel Lens (콘택트렌즈 관리방법에 의한 실리콘하이드로겔렌즈의 지방침전물 제거효과)

  • Purpose: An in vitro study was conducted to compare the lipid cleaning efficacy of care solutions on balafilcon A silicone hydrogel (SiHy) lens. Methods: Lipid spoilation was performed by incubating balafilcon A SiHy lenses in phosphate buffered saline (PBS) containing oleic acid, oleic acid methyl ester and cholesterol. Spoiled contact lenses rinsed with PBS were cleaned with surfactant cleaner, alcohol containing cleaner and multipurpose solution (MPS) respectively and repetitive spoilation and cleaning was conducted up to 14 times. To observe the cleaning effect of ultrasonic wave on the lipid deposit, each spoiled lens was ultrasonicated and then compared with non-sonicated lens. Lipids deposit on the contact lenses was extracted by methanol:chloroform (1:1, v/v) solution. High performance liquid chromatography was used to analyze and quantify lipid deposit extracts. Results: The effectiveness of alcohol containing surfactant cleaner on the lipid deposits was better than that of surfactant cleaner and MPS, and the cleaning efficacy was significantly higher in the ultrasonic wave treated group. Lipid deposits were not removed completely by contact lens care solutions so that lipid deposits increased continuously and cumulatively. Conclusions: The cleaning efficacy of contact lens care solutions was not satisfactory to remove lipid deposits on the SiHy lens that new cleaning products specially designed for SiHy lenses are needed to develop.

The Change in the Amounts of Proteins and Lipids Deposited on Soft Contact Lens Caused by Drinking (음주로 인한 소프트콘택트렌즈의 단백질 및 지질 침착양 변화)

  • Purpose: In the study, the change of protein and lipid deposits on soft contact lens by drinking was investigated. Methods: Fifty male subjects wearing soft contact lens were surveyed whether they felt any discomfort induced by drinking or not. Further, 32 male subjects who has no ocular disease drank 190 mL alcohol. The protein and lipid deposits on soft contact lens (etafilcon A material) of subjects were measured after 4 hours later and compared with those of non-drinking subject. Results: When subjects drink alcohol with soft contact lens on, 58% of subjects answered they experienced the change of lens awareness such as stiffness, blurry sight, dryness and so on. The protein deposit on soft contact lens increased an average of $59.3{\mu}g/lens$ by drinking and the case of more than double in protein deposit was reached in 9 eyes. However, the protein deposited on soft contact lens was lysozyme which was unchanged by drinking. The amounts of cholesterol and methyl oleate after drinking were 85.5% (p=0.25) and 52.6% (p=0.002) of non-drinking's indicating some change of lipid deposit on soft contact lens by drinking. Conclusions: The results showed the composition of protein and lipid deposited on soft contact lens was changed due to drinking. Thus, it is suggested that wearing soft contact lens when drinking might be one of the reasons to feel discomfort.

The Effect of Dietary Fat Levels and Sources of Dietary Fiber on Serum and Liver Lipids of Rats (지방의 섭취량과 첨가된 섬유소의 종류가 흰쥐의 체내 지질 수준에 미치는 영향)

  • The effect of fiber sources on lipid metabolism was investigated in relation to the level of dietary fat. After rats were fed each diet for 4 weeks, all animals were sacrificed to collect the liver, and blood samples by heart puncture. Total lipid, total cholesterol, phospholipid and HDL- cholesterol were determined in serum. Total lipid and total cholesterol were also determined in liver. These results were compared with the histochemical appearances by the method of Oil red-O staining. Higher level of fat in the diet appeared to increase the total lipid levels in serum and more fat in the liver cells were shown from the high fat diet judging from the E.M. pictures. Phospholipid concentration in serum was greater in the rats of high fat groups than those of the control or non-fat groups. High fat level caused to decrease the HDL- cholesterol concentration in serum. Pectin decreased serum and liver lipids and the deposit of fat in the liver cells. HDL-cholesterol levels in serum was increased by pectin. Whereas the cellulose supplementation had no effect to reduce serum and liver total lipid levels, and cellulose did not decreased the deposit of fat in the liver cells. From these results, it is concluded that dietary fat levels led to changes in the lipid metabolism of rats and pectin may exhibit a lowering total lipid contents in serum and liver. Cellulose may slightly elevate serum and liver levels, or have no lowering effect.

Tanshinone I, an Active Ingredient of Salvia miltiorrhiza, Inhibits Differentiation of 3T3-L1 Preadipocytes and Lipid Accumulation in Zebrafish

  • Objectives: Tanshinone I is a bioactive constituent in Salvia miltiorrhiza. At present, the anti-obesity effect and mechanism of tanshinone I are not fully understood. Here we investigated the effect of tanshinone I on lipid accumulation in 3T3-L1 preadipocytes and zebrafish. Methods: Lipid accumulation and triglyceride (TG) content in 3T3-L1 cells were determined by Oil Red O staining and AdipoRed assay, respectively. The expression and phosphorylation levels of adipogenic/lipogenic proteins in 3T3-L1 cells were evaluated by Western blotting. The messenger RNA (mRNA) expression levels of adipogenic/lipogenic markers and leptin in 3T3-L1 cells were measured by reverse transcription polymerase chain reaction (RT-PCR). Lipid accumulation in zebrafish was assessed by LipidGreen2 staining. Results: Tanshinone I at 5 μM largely blocked lipid accumulation and reduced TG content in differentiating 3T3-L1 cells. Furthermore, tanshinone I decreased the expression of CCAAT/enhancer-binding protein-α (C/EBP-α), peroxisome proliferator-activated receptor-γ (PPAR-γ), fatty acid synthase (FAS), acetyl CoA carboxylase (ACC), and perilipin A but also the phosphorylation of signal transducer and activator of transcription-3 (STAT-3) in differentiating 3T3-L1 cells. In addition, tanshinone I increased the phosphorylation of adenosine 3',5'-cyclic monophosphate (cAMP)-activated protein kinase (AMPK) while decreased the intracellular adenosine triphosphate (ATP) content with no change in the phosphorylation and expression of liver kinase-B1 in differentiating 3T3-L1 cells. Importantly, tanshinone I also reduced the extent of lipid deposit formation in developing zebrafish. Conclusions: These findings demonstrate that tanshinone I has strong anti-adipogenic effects on 3T3-L1 cells and reduces adiposity in zebrafish, and these anti-adipogenic effect in 3T3-L1 cells are mediated through control of C/EBP-α, PPAR-γ, STAT-3, FAS, ACC, perilipin A, and AMPK.

  • A surface acoustic wave(SAW) sensor for the detection of odorants has been constructed by depositing various phospholipids and fatty acids onto the surface of the SAW device. Applying the Langmuir-Blodgett technique. it was possible to deposit the optimal number of layer which was found to be between 10 and 20. The characteristics of a SAW device operating at 310 MHz deposited with phosphatidyl choline were analysed. Menthone, amylacetate, acetion, and other organic gases sho7\\\\`ed different affinities to the coated lipids. An explanation is given for differant odorant affinities based on the monolayer properties of phospholipids. The identification of odorants depending on the type of lipid used for coating is discussed in terms of a comparison of their normalized resonant frequency chi It pat terns. Using a number of different lipid-coated SAW devices. odorants can be identified by a computerized pattern recognition algorithm.

Anti-Obesity Effects of a Mixture of Fermented Ginseng, Bifidobacterium longum BORI, and Lactobacillus paracasei CH88 in High-Fat Diet-Fed Mice

  • Ginseng and probiotics have anti-obesity effects in mice fed a high-fat diet (HFD). Absorption of ginsenoside and colonization of probiotics occur in the intestine. In this study, a mixture of fermented ginseng and two probiotics, Bifidobacterium longum BORI and Lactobacillus paracasei CH88, was administered to HFD-fed mice for 9 weeks. The mixture significantly suppressed weight gain (p < 0.05, n = 8) and lipid deposition in the liver and adipose tissues as well as increased the mice's food intake. The adipocyte size of the adipose tissue was significantly decreased in the mixture-fed group, especially when 0.5% fermented ginseng and $5{\times}10^8/ml$ of the two probiotics were used (p < 0.05, n = 10). The expression of TNF-${\alpha}$ in adipose tissue was efficiently downregulated in the mixture-fed group (p < 0.05, n = 4). The supplement also improved the mice's fasting blood glucose levels (p < 0.05, n = 8) and total cholesterol feces excretion (p < 0.05, n = 8). The mixture of fermented ginseng and B. longum BORI and L. paracasei CH88 could have an anti-obesity effect and suppress lipid deposit in the liver and adipose tissues.

The Effect of Sesame Oil, Perilla Oil and Beef Tallow on Body Lipid Metabolism and Immune Response (한국에서 상용되는 식용유지로 사육된 흰쥐의 체내 지방대사 및 면역능력에 대한 연구)

  • The research was designed to study the effect of different fat sources and levels on Body lipid metabolizm and immune responses in Sprague-Dawely strain male rats. These effect of different fat sources compared with sesame Oil, Perilla oil and Beet tallow. Fat sources were divided into 3 groups respectively 7%, 15%, 30% fat level on diet weight basis. The experimental period was 54days. 1) The body weight gain was significantly low in NF group. In Sesame oil group and perilla oil group, low fat level groups were higher than medium, high fat level groups. But in Beef tallow group, high fat level groups were higher than low and Medium groups. 2) The weight of liver, kidney and epididymal fat ped tend to increse with increasing body weight. 3) The contents of triglyceride and total lipid in serum were significantly different with dietary fat sources and Perilla oil group was the lowest. 4) The contents of triglyceride and total lipid in liver were significantly different with dietary fat levels and high fat level group was higher than low fat level group. 5) Perilla oil group, compared with Beef tallow group, showed the higher excretion of cholesterol through feces and the higher deposit of cholesterol in liver. Therefore serum cholesterol level of Perilla oil group was lower than that of Beef tallow group. But eventhough Sesame oil is vegetable oil, Sesame oil did not showed an effect like Perllia oil on serum cholesterol level. 6) Weight of thymus decreased with fat levels particularly in vegetale oil. And it had on effect on mitogen response, mitogen response decreased with fat level in vegetable oil. But in Beef tallow, there was no difference in fat level.

Studies on the Physiological Activities of Caragana chamlagu Roots -Effects on the Hyperlipemia, Hyperglycemia and Liver Damage- (골담초근의 생리활성 -고지질, 고혈당 및 간손상에 미치는 영향-)

  • The studies were attempted to evaluated the therapeutic effects of various fractions(ether, methanol, butanol) of Caragana chamlagu roots on the hyperlipemia induced by feeding the diet containing 1%, cholesterol and 0.5%, cholic acid in rats, and on the hyperglycemia induced by streptozotocin in rats. Also the preventive effects of these fractions were studies on the liver damage in $CCl_4-intoxicated$ rats. The followings were obtained as the results: 1.The butanol fraction was significantly shown to down the serum lipid level in 1%, cholesterol and 0.5%, cholic acid diet-feeding rats and streptozotocin-induced hyperglycemic rats. Cholesterol level in $CCl_4-intoxicated$ rats was reduced in the case of all pre-treated groups. 2.The serum glucose level of streptozotocin-induced hyperglycemic rats was significantly decreased by the administration of various fractions of C. chamlagu roots, and the lipid-peroxidation of pancreas was significantly decreased in the case of administration of these fractions. 3.The activates of s-GOT and s-GPT were decreased by the administration of various fractions, especially in butanol fraction, of C. chamlagu roots in the $CCl_4-intoxicated$ rats. The liver lipid-peroxidation was decreased by administration of 200mg/kg of these fractions in the $CCl_4-intoxicated$ rats. In histological observation, hepatic cellular necrosis and fatty acid deposit were increased remarkably by $CCl_4-intoxication$, but the pretreatment of various fractions of C. chamlagu roots improved the pathological change of parenchymatous cell necrosis and fatty change around centrilobular area of the control.

Effect of Black Ginseng on Body Weight and Lipid Profiles in Male Rats Fed Normal Diets (정상 식이 수컷 흰쥐에서 흑삼이 체중 및 지질에 미치는 영향)

  • Previously we have reported that black ginseng was produced by nine-time repeated steaming 5-year fresh ginsengs at 95${\circ}$C for 3hr in pottery apparatus and drying at 60${\circ}$C for 12${\sim}$36 hr and had an outstanding anticancer activity. This study was designed to examine the ability of black ginseng to suppress body weight gain appetite, and to investigate the effect of black ginseng on lipid profile in male rats fed normal diets. The extract of black ginseng was administered daily for 3 weeks, and the changes of body weight and food intake were measured. After administering the extracts for 3 weeks, the fat weight and serum lipid levels were also measured. Black ginseng persistently suppressed weigh gain and food intake was also reduced, but not significantly; The weight of both epididymal and abdominal fats was decreased In black ginseng-treated rats. Serum triglyceride level in rats administered with black ginseng was also significantly lowered. Therefore, these results reveal that the repeated administration of black ginseng causes a suppression of body weigh gain by decreasing triglyceride level in blood and body fat deposit.

  • Obesity is a metabolic disease associated with multiple hormonal abnormalities. Therefore, obesity management aims at balancing these endocrine malfunctions nowadays. Although many studies proved interactions of hormones related with obesity, there are still lots of controversies. Most of these malfunctions are more pronounced in central, visceral obesity than in peripheral obesity. Recently, it is revealed that a central lesion of endocrine malfunction in human visceral obesity is probably related with a hypersensitivity of hypothalamopituitary-adrenal(HPA) axis. Probably associated with this axis, Insulin and cortisol promote lipid accumulation by expressing lipoprotein lipase activity, while sex hormones and growth hormone exert the opposite effects. Also reviewed was thyroid hormone which is closely related with thermogenesis. Serotonin is prescribed as antidepressant and it is applied to some eating disorders. Recently, leptin made in fat deposit also took attentions in terms of regulator of appetite and messenger of sex signal.
