Influence of Deletions in the Apoemulsan Gene Cluster on Acinetobacter venetian us RAG-l Polysaccharide Biosynthesis

  • Apoemulsan is a biopolymer with potent emulsification activity, produced by Acinetobacter venetian us RAG-1 (RAG-1). The wee gene cluster is responsible for apoemulsan biosynthesis. The analysis of (i) a putative polysaccharide copolymerase mutant (${\Delta}wzc$), (ii) a putative polymerase mutant (${\Delta}wzy$), and (iii) an apoemulsan-deficient variant (${\Delta}2$) indicated that the wee gene cluster controls the synthesis of two polysaccharides: high molecular weight (HMW) and low molecular weight (LMW). LMW polysaccharide of wee origin was present in LPS isolated from RAG-1 cells, suggesting a link to the Lipid A-core of LPS molecules. SDS-PAGE analysis indicated that apoemulsan is copurified with LPS polysaccharide, with implications in the emulsification activity of RAG-1 polymer.

  • Hypertension is considered to be caused by a complicated combination of genetic and environmental factors. Atrial natriuretic peptide (ANP) has been to suppress renin activity and inhibit the synthesis and release of aldosterone. Therefore, Abnormalities of this peptide caused by genetic variation may be influence the blood pressure. The aim of present study was to examine the relationship between hypertension and Sca I RFLP of ANP gene in Korean population. The genotype distribution of this RFLP was significantly different between normotensives and hypertensives (P<0.05). However, this genetic marker was not significantly associated with any anthropometric parameters or plasma lipid concentrations in our study group. Therefore, our result suggest that Sca I RFLP of ANP gene may be useful as genetic marker in the ethiology of hypertension in Korean population, independent of any cardiovascular risk. factors studied.

지용성 Benz[cd]indole 엽산길항제의 합성 및 In Vitro 항암활성 (Synthesis of Lipophilic Benz[cd]indole Antifolates and Their Antitumor Activity In Vitro)

  • Nonclassical aminobenz[cd]indole antifolates 4, 5 and 6, in which the glutamic acid moiety of the classical antifolates is substituted by 2-phenylglycinamide or 3-aminobenzamide, were synthesized and their in vitro antitumor activity was evaluated. The purpose of this substitution is that the lipophilicity is enhanced due to the aromatic ring of the target compounds for the passive transport through lipid membrane of cells while the hydrogen bonding of the amide is retained in the active site of the enzyme, thymidylate synthase, where the glutamate is originally present. The target compounds were highly cytotoxic against tumor cell lines of murine and human origin with micromolar to nanomolar $IC_{50}$ values. Most effective was compound 4 ($N^6-methyl-N^6$-[4-[(${\alpha}$(S)-aminocarbonylbenzyl) aminocarbonyl]benzyl]-2,6-diaminobenz[cd]indole)with $IC_{50}$ of 2 nM against SW480, human colon adenocarcinoma cell line, which is 650-fold more potent than the reference compound 3.

Synthesis of NBD-Labeled DOTAP Analog to Track Intracellular Delivery of Liposome

  • A DOTAP analog labeled by NBD on the head group (DTNBD) was designed and synthesized to label DOTAP liposome. The structure was confirmed by $^1H$ NMR and FAB-MS, and the fluorescence of the newly synthesized DT-NBD was observed by fluorescent microscopy. The transfection efficiency of DOTAP liposome containing DT-NBD was comparable to commonly used NBD PE in COS7 and MCF7 cells. Furthermore, the level of cellular uptake and fluorescent intensity of fluorescent liposome containing DT-NBD was higher than NBD PE. Therefore, the novel NBD-labeled DOTAP analog seems to be effectively used for investigation of the cellular interaction and transfection mechanism of DOTAP liposome.

EFFECT OF DITHIOL MALONATE DERIVATIVES (DMDs) ON CARBON TETRACHLORIDE-INDUCED HEPATOTOXICITY IN PRIMARY CULTURES OF ADULT RAT HEPATOCYTES

  • Protective effects of dithiol malonate derivatives (DMDs), YH-100, YH-150 and YH-439 on carbon tetrachloride-induced hepatotoxicity were investigated in primary rat hepatocytes culture. Treatment of DMDs to hepatocytes culture did not affect total cytochrome P-450 content and ECOD and AHH activities. Protein and RNA synthesis was also similar to control. Meanwhile, DMDs significantly decreased LDH release and in vitro lipid peroxidation induced by $CCI_4$. Accumulation of cellular triglyceride and decreased secretion of VLDL from liver cells by $CCI_4$ treatment were also significantly protected.

엽록체로 향하는 단백질 (Proteins Heading for the Chloroplast)

  • The chloroplast has been the prime light-energy harvesting organelle on earth. It also carries out several key metabolic processes, such as lipid synthesis and nitrogen metabolism. Even though the chloroplast has its own genome, its coding capacity can afford only dozens of proteins, and most of the proteins functioning in the chloroplast are imported from the cytosol where nuclear encoded chloroplast genes are synthesized on free cytosokic ribosomes. Precursor proteins synthesized on cytosolic ribosomes have transit peptides at the amino termini of the proteins, and the transit peptide is sufficient to transfer chloroplast proteins from the cytosol into the chloroplast. When comparing amino acid sequences duduced from the nucleotide sequences of the clones of the chloroplast proteins, high homologies can be found among the transit peptides of proteins with the same function. Overall amino acid compositions of the transit peptides show amphiphilic characters of the transit peptides, and the amphiphilicity indicates that three dimensional structure of the transit peptide is responsible for the translocation of the chloroplast proteins.

Anti-Obesity Effects of Red Beet Extract

  • Obesity is caused by the accumulation of triglycerides in adipocytes by the differentiation and lipid synthesis process of pre-adipocytes, and excessive accumulation of adipocytes by the activated Adipogenesis process within the differentiated cells. Therefore, inhibiting the differentiation of adipocyte cells or controlling the adipogenesis process is known as an effective treatment method for obesity. This study evaluates the inhibition of Red beet root extract on pancreatic lipase and pre-adipocyte cell differentiation. Also it evaluates the Red beet root extract activities on C/EBP-𝛼,𝛽, and PPAR-𝛄. The experiments proved that the Red beet root extract inhibits pancreatic lipase by concentration dependency. Further, in 3T3-L1 inhabitation experiment, it was found Red beet root extract inhibited adipocyte formation. Red beet root extract also inhibits the expression of C/EBP-𝛼, C/EBP-𝛽, and PPAR-𝛾 which effect the process of adipocytic differentiation. We therefore concluded that RBE has a high potential to further studies on anti-obesity effect.

  • Diacylglycerol acyltransferase (DGAT) is a microsomal enzyme that plays a central role in the metabolism of cellular glycerolipid. Recently, the generation of DGA T-deficient mice has provided a better understanding of triglyceride synthesis and its relationship to obesity. Therefore DGAT is an attractive target for treatments of triglyceride metabolism disorders, such as obesity or hypertriglyceridemia. (omitted)

  • Keloids are characterized by abnormal proliferation of fibroblasts and overproduction of collagen. Recently, it is reported that transforming growth factor beta (TGFb) and its signaling molecule, SMAD3 are related to the mitogenic effect of fibroblasts and a stimulatory factor for collagen synthesis. Cationic SLN was developed to improve the complex formation of DNA/SLN and enhance the uptake efficiency to cells. SLN was formulated by DC-Chol, DOPE, trimyristin as a solid core and other surfactant. The physical properties of the SLN and the ATS-SLN complex were characterized. (omitted)

  • All cells are equipped with intricate signaling networks to meet the energy demands and respond to the nutrient availability in the body. AMP-activated protein kinase (AMPK) is among the most potent regulators of cellular energy balance. Under ATP -deprived conditions, AMPK phosphorylates substrates and affects various biological processes, such as lipid/glucose metabolism and protein synthesis. These actions further affect the cell growth, death, and functions, altering the cellular outcomes in energy-restricted environments. AMPK plays vital roles in maintaining good health. AMPK dysfunction is observed in various chronic diseases, making it a promising target for preventing and alleviating such diseases. Herein, we highlight the different AMPK functions, especially in allergy, aging, and cancer, to facilitate the development of new therapeutic approaches in the future.
