Amberlite에 고정화된 Lipase 제조 및 효소적 Interesterification을 이용한 반응 특성 연구 (Immobilization of Lipases on Amberlite and Their Interesterification Reaction Characteristics)

  • 본 연구는 미생물로부터 유래된 5종(AH, AK, AY, PS와 R)의 lipase들을 Amberlite XAD 7에 흡착법으로 고정화 시킨 후 각 immobilized lipase들의 특성을 알아보았다. 고정화 전과 후의 단백질 함량 및 각 free lipase들과 immobilized lipase들을 이용한 interesterification 반응물의 지방산과 TAG 조성을 분석하였다. 또한, immobilized lipase에 있어 중요한 요인인 reusability를 확인하였다. Free lipase의 단백질 함량은 2.22-11.41%로 AH가 가장 높았던 반면, immobilized lipase에서는 AH, PS와 AK가 mg protein/g support이 높았다. 한편, 반응 특성을 알아보기 위해 카놀라유, PEE와 StEE를 기질로 하여 batch type interesterification을 진행하였을 때, free lipase의 경우 free lipase R을 제외한 다른 free lipase들은 반응시간이 1시간에서 72시간으로 증가함에 따라 총 포화지방산 함량이 증가하였으며 그 중 free lipase AH가 반응성이 가장 높았다. 또한, RP-HPLC를 통해 free lipase AK 반응물을 분석한 결과, 반응시간이 길어질수록 카놀라유(0시간)에서 볼 수 있었던 57.49 area%의 ${\bigcirc}{\bigcirc}{\bigcirc}$가 6.53 area%로 감소하였다. 이는 각 free lipase들이 PEE와 StEE를 효소적 반응에 이용했기 때문이라고 판단된다. 한편, immobilized lipase AY와 R의 경우 반응시간이 1시간에서 48시간으로 증가하여도 카놀라유(0시간)의 총 포화지방산 함량과 큰 차이가 없었으나 immobilized AK의 경우 48시간에서 38.3 area%의 포화지방산 함량으로 가장 높은 활성을 보였다. 또한, 이를 사용하여 continuous type으로 반응하였을 때 유속이 느릴수록 효소와 기질 사이의 접촉 시간이 길어져 반응물의 총 포화지방산 함량이 증가함을 알 수 있었다. Reusability는 immobilized AK, AH와 PS 모두에서 두 번째 반복 반응을 하였을 때, 첫 번째 반응보다 총 포화지방산이 120-196.5% 증가하였다. 그러나 bounding protein 함량이 가장 높았던 immobilized AK는 support에 흡착되었던 free lipase AK의 탈착이 일어나 2번째 반응 후부터 활성이 감소한 반면, immobilized AH와 PS는 활성이 비교적 유지되었다.

폴리(에틸렌 테레프탈레이트)(PET) 올리고머의 합성과 생분해 거동 (Synthesis and Biodegradation Behavior of Poly(ethylene terephthalate) Oligomers)

  • Poly(ethylene terephthalate)(PET)의 올리고머인 oligo(ethylene terephthalate)(OET), oligo(ethylene succinate-co-terephthalate)(OEST) 및 oligo(butylene succinate-co-terephthalate)(OBST)를 각각 합성한 후, Pseudomonas cepacia 유래의 lipase PS를 사용하여, 올리고머의 분해실험을 실시, 분해시간에 의한 잔존중량 및 분해생성물의 분석에 의한 생분해기구를 해명함을 목적으로 하였다. OEST와 OBST의 합성에서는 배합 몰비에 대한 조성비의 조절이 가능한 올리고머를 얻을 수 있었으며, 조성비의 상이함에 의해 열적 성질이 현저한 차이가 있음을 확인하였다. 분해실험에서는 OEST, OBST 모두 lipase PS에 의해 분해가 발생되나, terephthalic acid의 조성이 높아질수록 분해능력이 저하됨을 확인하였고, lipase PS는 지방족에 가까운 조성을 가진 polyester를 쉽게 분해시킴을 확인하였다.

유기용매 내에서 중쇄지방질의 합성

  • Using 20 lipases from various microbial origins medium chain glycerides, namely, mono-, di-, and tri-carproyl glycerols from glycerol and acid were synthesized in isooctane. Enzyme reaction was performed at 0.35 M of capric acid, 0.025M of glycerol and the same mass of silica gel to remove water in 5ml of isooctane with 30mg of lyophilized lipase. Among 20 lipases, eleven lipases showed good synthetic activities, especially lipase from Pseudomonas aeruginosa (Lipase PS), Rhizomucor miehei origined lipase and Chromobacterium viscosum lipase (Lipase CV) showed good activities for production of tricaproylglycerol, while Lipase OF-360 (origined from Candida rugosa) and Lipase D (Rhizopus delemar) were good for production of dicaprolyglycerol. The lipases, especially Lipase PS, have high thermal stability at $ 60^{circ}C$, and optimum pH of lyophilization for dehydrating the lipase was pH 6.5.

방사선 조사선량에 따른 lipase의 interesterification 반응특성 변화 연구 (Studies of interesterification properties of irradiated lipases)

  • This study was conducted to prove the effect of irradiation on lipases (lipase AK, lipase AH, lipase PS-D, Lipozyme TLIM, Lipozyme RMIM and Novozyme SP435) which were used for interesterification reaction using batch type reactor. Through such interesterification, structured lipid (1(3)-palmitoyl-2-oleoyl-3(1)-stearoyl, POS) was synthesized by lipase treated with irradiation at different doses (0, 3, 7, 14, 29 and 59 kGy) using canola oil, palmitic ethyl ester (PEE) and stearic ethyl ester (StEE). After the reaction, fatty acid composition of triacylglycerol (TAG) in structured lipid was analyzed to compare the lipase activity. The results showed that activity of the irradiated lipase AH, PS-D and Novozyme SP435 with certain dose (3 kGy) were slightly improved. Such change of lipase activity suggested that irradiation might affect on the interesterification properties. Especially, Lipase AK, Lipozyme TLIM and Lipozyme RMIM after at 3 kGy irradiation showed that content of stearic acid ($C_{18:0}$) was increased while palmitic acid ($C_{16:0}$) decreased in the interesterified products.

Mechanism of Enzymatic Degradation of Poly(butylene succinate)

  • Poly(butylene succiate) (PBS), poly(butylene succinate-co-L-lactate) (PBSL), and poly(butylene succinate-co-6-hydroxycaproate) (PBSCL) polymers were degraded by lipase $PS^{(R)}$, and the enzymatic degradation mechanism of PBS was analyzed in detail. The enzymatic degradation of PBS gave 4-hydroxybutyl succinate (4HBS) as the main product. An exo-type hydrolysis mechanism was proposed based on this observation. The terminal chain of PBS had conformational similarity to ordinary tri- and diglycerides and could be incorporated as a substrate in the active site of this lipase. The surface adsorption of the lipase was much larger on PBS and its copolymer films than on the other polyester films because the lipase adhered quite strongly to the polymer terminal through a specific adsorption mechanism. Kinetic analysis showed that the total number of surface adsorption points per unit area of PBSL and PBSCL copolymers was larger than that of the PBS homopolymer.

Analysis of the Structure-stability Relationship of Cold-adapted Lipase PsLip1 from Homology Modeling

  • Two initial models of cold-adapted lipase PsLip1 have been constructed, based on homology with the bacterial lipases Chromobacterium viscosum (CvLip) and Pseudomonas cepacia (PcLip), whose X-ray structures have been solved and refined to high resolution. The mature polypeptide chains of these lipases have 84% similarity. The models of Mod1 and Mod2 have been compared with the tertiary structures of CvLip and PcLip, respectively, and analyzed in terms of stabilizing interactions. Several structural aspects that are believed to contribute to protein stability have been compared: the number of conserved salt bridges, aromatic interactions, hydrogen bonds, helix capping, and disulfide bridges. The 3-dimensional structural model of PsLip1 has been constructed in order to elucidate the structural reasons for the decreased thermostability of the enzyme in comparison with its mesophilic counterparts.

Biochemical Properties and Substrate Specificity of Lipase from Staphylococcus aureus B56

  • A lipase of Staphylococcus aureus B56 was purified from a culture supernatant and its molecular weight was estimated to be 45 kDa by SDS-PAGE. The optimum temperature and pH for the hydrolysis of olive oil was $42^{\circ}C$ and pH 8-8.5, respectively. The enzyme was stable up to $55^{\circ}C$ in the presence of $Ca^++$ at pHs 5-11. The lipase gene was cloned using the PCR amplification method. The sequence analysis showed an open reading frame of 2,076 bp, which encoded a preproenzyme of 691 amino acids. The preproenzyme was composed of a signal sequence (37 aa), propeptide (255 aa), and mature enzyme (399 aa). Based on a sequence comparison, lipase B56 constituted of a separate subgroup among the staphylococcal lipase groups, such as S. aureus PS54 and S. haemolyticus L62 lipases, and was distinct from other lipases in their optimum pH and substrate specificity.

급성췌장염 유발된 흰쥐에 대한 패장의 억제 효과 (Inhibitory Effect of Acute Pancreatitis in Rats by Patrinia Scabiosaefolia)

  • Objectives : Patrinia scabiosaefolia (PS) has long been as a remedy for treating infectious diseases in Korea. In the present experiments, the author examined the effects of PS on the cholecystokinin-octapeptide (CCK)-induced pancreatitis (AP) in rats. Methods : Male Wister rats weighing 200 to 250 g were divided into two group. Normal untreated group, in treatment with PS group; PS was administered orally, followed by $75{\mu}g/kg$ CCK subcutaneously three times, after 1, 3 and 5 h. This whole procedure was repeated for 5 days. In treatment with saline group, the protocol was the same as in treatment group with PS. The author determined the pancreatic weight/body weight ratio, the levels of pancreatic heat shock proteins(HSP)60, HSP72 and the secretion of pro-inflammatory cytokines. Results and Conclusion : PS was significantly decreased the pancreatic weight/body weight ratio in CCK-induced AP. PS increased HSP60 and HSP72 compared with CCK-induced AP. Additionally, the secretion of tumor necrosis factor(TNF)-${\alpha}$, interleukin(IL)-$1{\beta}$ and IL-6 the levels of amylase and lipase were lower than that of saline. These results suggest that PS may has a inhibitory effect against CCK-induced AP.

효소적 공법을 이용한 기능성 modified-butterfat의 향기성분 특성 분석 (Flavor Characteristic of Functional Modified-butterfat Synthesized by Lipase-catalyzed Interesterification)

  • 효소적 공법을 이용하여 합성한 기능성 modified-butterfat들(MF668과 MF866)의 향기성분 분석을 전자코와 SPME-GC/MS를 가지고 수행하였다. 전자코 분석결과, 기질로 사용된 ABF와 FSO, 그리고 생성물인 MF668과 MF866은 제1주성분 값의 95.16%기여율로 서로 다른 향기패턴을 갖음을 확인하였다. 신속하며 간편한 SPME-GC/MS분석결과, MF668과 MF866에서는 기질에서보다 더 다양한 aldehyde류, ketone류, acid류 등이 검출되었다. 이는 반응과정 중 생성될 수 있는 산화물로 추후 항산화제 연구가 필요함을 입증하였다.
