The Influence of Saturated Fats, ${\alpha}-linolenic$ Acid, EPA and DHA on the Lipid Hydroperoxide Level and Fatty Acid Composition in Liver Microsomes and in Plasma Lipid of Rabbits

  • To investigate the influence of saturated fats, ${\alpha}-linolenic$ acid, EPA and DHA on the lipid hydroperoxide concentration and fatty acid composition in liver microsomes and in plasma lipid of rabbits, the animals were fed on the perilla oil rich ${\alpha}-linolenic$ acid or sardine oil rich EPA and DHA diet for four weeks Were examined. The fatty acid composition of plasma lipid and liver microsomes of rabbits fed on the perilla oil diet was an accumulation of arachidonic acid(AA) 20:4 n-6, eicosapentaenoic acid(EPA) 20:5 n-3, and docosahexaenoic acid(DHA) 22:6 n-3, The fatty acid composition of plasma lipid and liver microsomes of rabbits fed on the sardine oil was an accumulation of ${\alpha}-linolenic$ acid(LNA) 18:3 n-3, and arachidonic acid(AA) 20:4. The p/s ratio of rabbits fed on the perilla oil diet changed from 7.4 to 2.27 for plasma lipid and 2.47 for liver microsomes. The concentration of lipid hydroperoxide was 3.48 nmol MDA/ml and 4.35 nmol MDA/ml for plasma lipid and liver microsomes, respectively, in perilla oil diet. The lipid hydroperoxide liver was 4.22 nmol MDA/ml and 67 nmol MDA/ml for plasma lipid and liver microsornes in sardine oil diet.

미역의 부위별 중성 및 극성 지방질의 지방산 조성 (Fatty Acid Compositions of Nutural Lipids and Polar Lipids in the Parts of Miyeok (Undaria pinnatifida))

  • 미역 (Undaria pinnatifida)의 엽상부, 줄기 및 포자엽 부위별 지방산을 분석하여 n-3 폴리엔산, n-6 폴리엔산. ${\gamma}$-linolenic acid (18:3, n-6) 및 $\alpha$-linolenic acid (18:3, n-3)의 영양학적 의의를 규명코자 하였다. 총지방질 함량은 엽상부 3.37%, 포자엽 4.59% 및 줄기 2.50%였으나, 지방산 조성을 볼 때 엽상부는 총 폴리엔산 44.18% 중 $\alpha$-linolenic acid (18:3, n-3), stearidonic acid(18:4, n-3), eicosapentaenoic acid (20:5, n-3) 및 docosahexaenoic acid (22:6, n-3)와 같은 n-3계열의 폴리엔산은 29.69%였으나 ${\gamma}$-linol enic acid (18:3, n-6) 0.90%로 매우 낮았다. 총 폴리엔산과n-3계열의 폴리엔산 조성비율을 볼 때 엽상부는 44.18% 및 29.69%였으나. 포자엽은 23.63% 및 5.58%였고 줄기는 26.66% 및 7.37%였다. 한편 중성 지방질, 당지방질 및 인지방질의 n-3 폴리엔산의 함량을 볼 때 엽상부는 22.40%, 35.82% 및 3.53%로 현저하게 높았으나 줄기는 5.26%, 9.51% 및 2.37%였고 포자엽은 4.49%, 8.01% 및 2.14%로 가장 낮았다. 따라서 미역 엽상부의 총 폴리에산과 n-3 폴리엔산은 기능성 식품자원으로 활용이 가능한 것으로 시사된다.

  • In order to study the influence of culture conditions on cell growth and lipid formation by Mucor sp, various carbon and nitrogen sources initial pH and C./N ratio of medium were investigated. Glucose was found to be suitable carbon source in terms of lipid yield and ${\gamma}$-linolenic acid(GLA) content. When NH4Cl and (NH4)2SO4 were used as nitrogen source lipid content was high(19-21%) but GLA content was low(15-17%) On the other hand when NaNO3 and KNO3 were used lipid content was low(about 13%) but GLA content was high(22-23%). The highest production of lipid was obtained at a C/N ratio of 40 using glucose and (NH4)2SO4 as carbon and nitrogen source respectively. it was found that lipid yield was high at pH4.6 Also this fungus did not grow at 35$^{\circ}C$ and lipid yield was highr at 15$^{\circ}C$ than $25^{\circ}C$.

감마 리놀렌산의 혈액 지질 대사 (Effect of Dietary ${\gamma}$-Linolenic Acid on Plasma Lipid Metabolism in Rats)

  • The purpose of this study was to determine the effect of dietary ${\gamma}$-linolenic acid on plasma lipid metabolism and anti thrombotic activity in male Sprague Dwaley Strain rats. Rats weighing an average of $100{\sim}120g$ were fed a experimental diets containing 5% lard (saturated fatty acids), corn oil(linoleic acid), evening promise oil(EPO, 9% ${\gamma}$-linolenic acid) or borage oil(BO, 24% ${\gamma}$-linolenic acid) for 3Odays, respectively. Though there were no significant difference in the food intake among the groups, the body weight gain of the BO group was significantly lower than that of other group. The spleen weight of the lard group was significantly lower than that of other group. The bleeding time of the BO group was significantly longer than that of other group. The blood clotting time was significantly tended to long in EPO and BO groups compared with lard group. The plasma triacylglyceride and total cholesterol concentration were high in order of lard, com oil, EPO and BO, groups and there were significant differences among the groups. The plasma HDL-C concentrations were high in order of BO, EPO, com oil and lard groups and there were significant differences among the groups. The plasma LDL-C concentrations were significantly the highest in lard group, but the lowest in BO group. These data indicate that ${\gamma}$-linolenic acid has a antithrombotic activity, and decrease the plasma triacylglyceride, total cholesterol and LDL-C concentrations in rats.

싸리종자(Lespedeza bicolor)의 지질성분(脂質成分)에 관한 연구(硏究) (Studies on the Lipid Composition of Bush Clover (Lespedeza bicolor) Seed)

  • 싸리종자의 지질을 chlorform: methanol(2:1, v/v)로 추출한 후 중성지질, 당지질, 인지질로 분획한 다음 TLC와 GC로 분리, 정량하여 다음과 같은 결과를 얻었다. 1. 싸리종자의 일반성분은 수분이 11.60%, 조단백이 20.10%, 조지방이 15.25%, 당질이 48.92%, 회분이 4.13%로 나타났다. 2. 싸리종자의 지질은 중성지질이 71.75%, 당지질이 23.26%, 인지질이 4.99%였다. 3. 중성지질 중에는 triglyceride 61.9%, free fatty acid 22.04%가 주성분을 이루었고 그 외에 free sterol, esterified sterol, diglyceride, monoglyceride 지질 성분이 존재하였다. 4. 당지질 중에는 monogalactosyl diglyceride 38.19%의 함량이 가장 높았고 esterified steryl glycoside, steryl glycoside, digalactosyl diglyceride, cerebroside 등의 지질 성분이 비슷한 함량으로 존재하였다. 5. 인지질 중에는 phosphatidyl choline 36.46%, phosphatidyl inositol 21.52%, phosphatidyl ethanolamine 17.29%가 주성분이었고 lysophosphatidyl choline, phosphatidyl serine과 1가지 미확인 인지질 성분이 존재하였다. 6. 전지질의 주요 지방산은 linoleic acid 36.83%, linolenic acid 19.65%, oleic acid 11.25, palmitic acid 9.61%였고 중성지질의 주요 지방산은 linoleic acid 55.36%, linolenic acid 19.40%, oleic acid 12.47%, palmitic acid 9.81%였다. 당지질의 주요 지방산은 linoleic acid 53.26%, linolenic acid 18.67%, palmitic acid 12.20%, oleic acid 11.39%였다. 인지질의 주요 지방산은 linoleic acid 53.86%, palmitic acid 27.83%, linolenic acid 9.51%, stearic acid 5.00%로 전지질, 중성지질, 당지질의 지방산 조성 pattern과는 차이가 있었다.

국내산 및 중국산 참기름과 변조 참기름의 지방산 조성에 관한 연구 (Comparative Studies on the Fatty Acid Composition of Korean and Chinese Sesame Oils and Adulterated Sesame Oils with Commercial Edible Oils)

  • 경남 도내에서 유통 중인 국내산 참기름과 중국산 참기름 각각 30건과 콩기름 및 옥수수기름 등의 식용유지를 첨가하여 변조한 참기를 12건에 대한 지방산 함량을 GC에 의한 정량법으로 분석하여 다음과 같은 결과를 얻었다. 국내산 참기름의 주요 지방산 평균 함량은 palmitic acid 8.26$\pm$0.04%, stearic acid 4.57$\pm$0.05%, oleic acid 39.25$\pm$0.05%, linoleic acid 46.82$\pm$0.07, linolenic acid 0.52$\pm$0.02%, arachidic acid 0.58$\pm$0.03% 등으로 나타났으며, 중국산 참기름은 palmitic acid 9.24$\pm$0.03%, stearic acid 5.11$\pm$0.04%, oleic acid 38.65$\pm$0.04%, linoleic acid 46.02$\pm$0.06%, linolenic acid 0.42$\pm$0.04%, arachidic acid 0.56%$\pm$0.04% 등으로 각각 나타났다. 국내산 참기름은 포화 지방산인 palmitic acid와 stearic acid가 중국산 참기름보다 적게 함유되어 있었고 불포화 지방산인 linolenic acid는 반대로 많이 함유되어 있었다. 국내산 참기름에 콩기름을 0%, 3%, 9%씩 첨가했을 때 불포화지방산인 linolenic acid가 각각 0.52$\pm$0.02%, 0.73$\pm$0.05%, 1.25$\pm$0.04%로 현저히 증가되었으며, 옥수수기름을 0%, 5%, 9%씩 첨가했을 때는 불포화지방산인 linoleic acid의 함량이 각각 46.82$\pm$0.07%, 50.22$\pm$0.06%, 51.14$\pm$0.05%로 증가됨이 확인되었다. 참기름의 산지별 확인과 식용유지를 첨가한 변조참기름에 대해서는 각각의 지방산 함량에서 특이성을 보이고 있으나 일부지역에서 구입한 국내산과 중국산 참기름에서 유사한 결과치를 보여 앞으로 국내에서 유통되는 참깨나 참기름에 대하여 더 많은 연구가 필요할 것으로 사료된다.

Escherichia coli와 Bacillus subtilis의 당지질 생합성과 지방산 조성에 미치는 여러가지 금속화합물의 영향 (The Effect of Metal Compounds on Phospholipid Biosynthesis and Fatty Acid Composition in Escherichia coli and Bacillus subtilis)

  • The biosynthesis of galactolipid and galactose and their composition of fatty acid in E. coli and B. subtilis treated ] with copper chloride (10 ppm), nickel chloride (50 ppm), manganese chloride (100 ppm) during the culture were analyzed. The contents of MGDG, DGDG and total lipids in treatment with metal compounds were lower to compared with the control. In E. coli, the major fatty acid unitized for biosyntheis of MGDG were palimitic acid (ave. 36.87%) and linolenic acid (ave. 14.79%) in control. In MGDG, the major fatty acids were utilized for palmitic acid (ave. 20.00%) and myristic acid (ave. 7.32%) in treatment with copper chloride, lauric acid (ave. 11.71%) and linolenic acid (ave. 11.06%) in manganese chloride treatment. And in nickel chloride treatment, it was palmitic acid (ave. 36.16%) and oleic acid (ave. 6.43%) were use in MGDG formation. In DGDG, in copper chloride treatment, it was lauric acid (ave. 19.41%) and oleic acid (ave. 9.95%) in biosynthesis of galactolipid. and in treatment with nickel chloride linolenic acid (ave. 15.39%) and linoleic acid (ave. 13.51%), in manganese chloride treatment palmitic acid (ave. 29.76%) and palmitoleic acid (ave. 11.35%) were used in DGDG formation. In B. subtilis, the major fatty acids utilized for biosynthesis of galactolipid was palmitic acid (ave. 30.86%) and linolenic acid (ave. 8.36%) in control. Otherwise, in MGDG, the major fatty acids were utilized for palmitic acid (ave. 28.92%) and stearic acid (ave. 13.25%) in treatment with copper chloride, and palmitic acid (ave. 15.73%) and lauric acid (ave. 11.88%) in manganese chloride treatment. It was continned that nickel chloride treatment was palmitic acid (ave. 35.16%) and palmitoleic acid (ave. 12.47%). The major fatty acids in DGDG were utilized for palmitic acid(ave. 34.19%) and linoleic acid (ave. 17.45%) in copper chloride treatment, and lauric acid (ave. 11.16%) and myrisitic acid (ave. 8.65%) in manganese chloride treatment. In treatment with nickel chloride, it was palmitoleic acid (ave. 10.30%) and myristic acid (ave. 7.81%) were used galactolipid formation.

감마-리놀렌산을 함유하는 식이지방이 흰쥐의 혈전작용 및 혈액지질 수준에 미치는 영향 (Effect of Dietary Oil Containing $\gamma$-Linolenic Acid on the Plasma Lipid Levels and Thrombotic Activity in Rats)

  • This study was carried out to determine the effect of dietary ${\gamma}$-linolenic acid on decreasing the plasma lipid levels and the thrombotic activity in rats. Sprague-Dawley male rats (B.W 120 g) were fed a experimental diet containing 5% lard (46.05% saturated fatty acids) , corn oil (51.36% linoleic acid) , evening primrose oil (EPO,72.80% linoleic acid and 9.16% ${\gamma}$-linolenic acid) or borage oil (BO,40.29% linoleic acid and 24.25% ${\gamma}$-liolenic acid) for 30 days. Although there were no significant differences in the food intake among the groups, the body weight gain of the BO group was significantly lower than that of the other groups. The bleeding time of the BO group was significantly longer than that of the other groups. There were significantly differences in the whole blood clotting time among the groups except for the EPO and corn oil groups, where the whole blood clotting time of the BO group was the highest among the groups, and that of the lard group was the lowest. The plasma triacyglyceride (TAG) , total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) concentrations were the lowest in the BO group, but highest in the lard group, and there were significant differences among the groups. The plasma HDL-C concentrations were in the following order: BO, EPO, corn oil and lard groups and there were significant differences among the groups. The excretions of fecal neutial steroids and acidic steroids of the BO group were the highest among the groups, and there were significant differences compared to the other groups. The results suggest that dietary EPO and BO containing ${\gamma}$-linolenic acid has an antithrombotic activity, and inhibits the increasing of plasma TAG, TC and LDL-C concentrations compared to lard, which contains saturated fatty acids, or corn oil, which contains linoleic acid.

국내 약용자원식물의 지방산 조성과 ${\gamma}-linolenic$ acid 탐색 (Screening of ${\gamma}-linolenic$ Acid Resources and Fatty Acid Composition in Korean Native Medicinal Plants Resources)

  • 우리나라에서 생산되고 있는 60여종의 약용식물의 지방산 조성을 조사하여 식물체의 부위 와 종류에 따른 지방산 조성의 특성을 관찰한 결과 아래와 같은 결과얻었다. 1. 달맞이꽃종자, 컴프리잎, 지치뿌리에서 각각 총지방산함량의 9.1, 3,5, 7,2%의 GLA가 검출되었으며 달맞이 꽃종자가 컴프리잎이나 지치뿌리보다 GLA의 절대량이 많았다. 2. 들깨종자, 참깨종자, 호박종자등의 지방산이 양(量)과 불포화도면에서 좋은 자원임이 확인되었다. 3. 식물체의 잎, 줄기, 뿌리에서의 지방산조성은 종자의 경우에 비하여 18 : 3의 분포 비율이 현저히 높게 나타났으며 18 : 0의 비율은 비교적 같게 나타났다.

Elicitor에 의한 baccatin III의 세포내 생합성 조절연구 (Elicitors for the Regulation of baccatin III Biosynthesis in Plant Cell Culture System)

  • The yeast extract, coconut water, safflower seed oil, arachidonic acid, linolenic acid, jasmonic acid and methyl jasmonate were added to Gamborg's B$_{5}$ medium. The changes on productivity of baccatin III were estimated every 30 minutes and the results were compared using the selected high yielding cell culture system of Taxus cuspidata. In most cases, the peaks of baccatin III productivity occured at 90~120 min after addition of elicitors. Among the compound elicitors, safflower seed oil showed the highest productivity of baccatin III. Also arachidonic acid and linolenic acid increased the baccatin III production.
