Study on the Ontology Linking by Acupuncture Terms (혈성(효능), 치법 용어를 통한 온톨로지 연계 연구)

  • Objectives : The goal of this paper is to enhance the percentage of linkages between an acupuncture point ontology and an acupuncture treatment ontology. Methods : Terms of the effect from acupuncture points and terms of the treatment method from diseases are selected in the ontologies. We have disassembled the terms into minimum meaning units and then linked them again. Results : By linking terms of the effect and terms of the treatment method, the meaninful information on the acupuncture point and treatment ontology is increased. Conclusions : The links between points for acupuncture points and treatment methods for diseases could improve the information in the ontologies. This also enables the knowledge representation for the use of acupuncture points with accompanied symptoms, causes of disease, and treatment methods.

Studies on the Manufacturing of Ginseng Soft Drink II. Effect of pH and heat treatment on the stability of panaxadiol saponins (인삼청량음료 제조에 관한 연구 (제2보) pH 및 처리조건이 Panaxadiol Saponin의 안정성에 미치는 영향)

  • This investigation was carried out to study the influence of pH and heat treatment on the ginsenosides in the white ginseng extract. Changes in ginsenosides (Rb1, Rb2, ,Rc, Rd) and free sugar were measured by the peak area variation of HPLC chromatogram during 25 hours heat treatment at the various level of pH. It was found that :(1) The peak areas of Rb1. Rb2, Rc and Rd on the HPLC chromatogram were decreased remarkably below pH 4.0 and more decrease was found as the temperature and heating time increased. (2) Those of glucose and arabinose were increased remarkably. It is considrered that the increase of glucose and the formation of arabinose result from the hydrolysis of ginsenoside( Rb1, Rb2, Rc, Rd) linked with sugars.

Clinical Manifestations and Treatment in Korean Patients with X-Linked Agammaglobulinemia (성염색체 열성 범저감마글로불린혈증 환자의 임상 소견 및 치료)

  • Purpose: X-linked agammaglobulinemia (XLA) is a primary immunodeficiency caused by mutations in the Bruton's tyrosine kinase (Btk) gene. The aim of this study was to investigate the clinical manifestations, molecular features, and treatment status of XLA in Korean patients at Seoul National University Children's Hospital. Methods: Fourteen Korean boys with XLA showing serum agammaglobulinemia, non-detectable to less than 2% of peripheral B-cells, and mutation of the Btk gene were enrolled. We observed the clinical features, laboratory findings, status of treatment, and complications in these XLA patients. Results: All XLA patients had a history of recurrent bacterial infections before diagnosis, and 20% of them had a neutropenia. Of the XLA patients 35.7% had a family history of XLA and 75% of their mothers were carriers. Btk gene analysis showed variable gene mutations in Xq22 including 9 amino acid substitutions, 3 frameshifts, 1 premature stop codon, and 1 splice defect. After intravenous immunoglobulin replacement therapy, infection episodes decreased, but complications such as bronchiectasis and chronic sinusitis remained. Conclusions: In patients less than 4 years of age with recurrent infection, analysis of serum gamma globulin levels and the Btk gene are recommended for the early diagnosis of XLA and for the appropriate prevention of recurrent infection.

Lorenzo's Oil Treatment in X-linked Adrenoleukodystrophy (X 염색체 연관 부신백질이영양증에서 Lorenzo Oil의 투여효과)

  • Purpose : Though more than 15 years have passed after introduction of Lorenzo's oil therapy in X-linked adrenoleukodystrophy(ALD), its efficacy is still fully not known. In patients who already have neurologic symptoms, most reports indicate that the neurologic disability continues to increase. We evaluated its efficacy in X-linked ALD patients. Methods : Four patients who were treated with Lorenzo's oil for at least 12 months were enrolled from 1996 to 2003. During treatment, changes of neurologic symptoms, brain magnetic resonance imaging(MRI) findings, and serum saturated very long-chain fatty acid(C26:0) were assessed. Results : Two patients with childhood cerebral ALD had progression of neurologic symptoms and MRI lesions during treatment. One asymptomatic patient developed childhood cerebral ALD after six year treatment of Lorenzo's oil. One "Addison only" patient remained neurologically intact after three years. During Lorenzo's oil therapy, serum C26:0 levels which had increased at diagnosis decreased in all four patients. Conclusion : Treatment with Lorenzo's oil did not prevent disease progression in childhood cerebral ALD patients who were already symptomatic. Long term follow up will be needed in asymptomatic ALD including "Addison only" to reveal the efficacy of Lorenzo's oil.

Strength and durability characteristics of biopolymer-treated desert sand

  • Biopolymer treatment of geomaterials to develop sustainable geotechnical systems is an important step towards the reduction of global warming. The cutting edge technology of biopolymer treatment is not only environment friendly but also has widespread application. This paper presents the strength and slake durability characteristics of biopolymer-treated sand sampled from Al-Sharqia Desert in Oman. The specimens were prepared by mixing sand at various proportions by weight of xanthan gum biopolymer. To make a comparison with conventional methods of ground improvement, cement treated sand specimens were also prepared. To demonstrate the effects of wetting and drying, standard slake durability tests were also conducted on the specimens. According to the results of strength tests, xanthan gum treatment increased the unconfined strength of sand, similar to the strengthening effect of mixing cement in sand. The slake durability test results indicated that the resistance of biopolymer-treated sand to disintegration upon interaction with water is stronger than that of cement treated sand. The percentage of xanthan gum to treat sand is proposed as 2-3% for optimal performance in terms of strength and durability. SEM analysis of biopolymer-treated sand specimens also confirms that the sand particles are linked through the biopolymer, which has increased shear resistance and durability. Results of this study imply xanthan gum biopolymer treatment as an eco-friendly technique to improve the mechanical properties of desert sand. However, the strengthening effect due to the biopolymer treatment of sand can be weakened upon interaction with water.

Improvement of treatment efficiency for sanitary treatment facilities by process modifications (분뇨처리장의 공정개선에 의한 처리효율 향상에 관한 연구)

  • There is a need to improve the efficiency of the existing sanitary treatment facilities, because the effluent standard becomes more stricter and septic sludge increased. Thus, operating processes of sanitary treatment system in M city changed with installation of additional facilities. Process modifications were as follows: Dilution water was added to the next process after primary aeration tank. Some secondary sedimentation sludge was recycled to primary aerator so that most of the organics were stabilized in primary aeration tank under automatic control of dissolved oxygen. The line of effluent from dewatering process flowing to the activated sludge tank was changed to the primary aerator. The primary sedimentation sludge line was linked to a thickener. Polymer was added to the activated sludge tank. The effluent of primary aerator and aerobic digester was recycled from the 5th to the 1st sector. As consequencies of above process modifications, the improvement of removal efficiency was achieved as BOD 54%, COD 42%, SS 61%, T-N 39%, and T-P 12%, respectively.

  • XLH (X-linked hypophosphatemic rickets) is a form of rickets which is resistant to the usual dose of vitamin D and inherited in a X-linked dominant manner. It is also known as vitamin D-resistant rickets or familial hypophosphatemic rickets. Here we report a 6-year-and-6-month-old female patient of XLH who is diagnosed with in SNUB. She was referred from local clinic for impaction of maxillary left permanent incisor and its treatment. She presents bowing deformities of the legs, short stature, enlargement of wrist and ankles and spontaneous dental abscesses of clinically sound teeth delayed eruption, taurodontism, delayed apical closure, enlarged pulp chambers, and absent or poorly defined lamina dura. The purpose of this case is to review the literatures of XLH and report the dental and medical characteristics of this patient.

Oligomeric Characterization of GroESLx Chaperonin from Symbiotic X-Bacteric in Amoeba proteus

  • GroESLx proteins of symbiotic X-bacteria were overproduced in Escherichia coli and their structural characteristics were assayed after simple purification. The GroESx and GroELx were heat-stable at 8$0^{\circ}C$ and 5$0^{\circ}C$, respectively. After heat-treatment, GroESx was purified by DEAE Sephadex A-50 chromatography and GroELx was purified by step- and linear sucrose density gradient ultracentrifugation. Molecular masses of GroESx and GroELx were 50-80 kDa and 800 kDa, respectively, as estimated by sucrose density gradient ultracentrifugation. In chemical cross-linking analysis, subunits of GroESx were mostly cross-linked by incubation for 3 h in 0.4% glutaralde-hyde and GroESx was found to be composed of homo-heptamer subunits. Those of GroELx were cross-linked within 10 min in 0.3% glutaraldehyde and GroELx was in two stacks of homo-heptamer subunits. On the other hand, GroESx and GroELx proteins in a solution could not be cross-linked even after incubation for 3 h in 0.5% glutaraldehyde. GroELx was stable at 4-37$^{\circ}C$. In the presence of both GroESx and ATP, GroELx$_{14}$ was stable at 37$^{\circ}C$ but not at 4$^{\circ}C$ or 24$^{\circ}C$. Thus, we confirmed the oligomeric properties of GroESx$_{7}$ and GroELx$_{14}$ and their stability to heat and in the interaction with GroESx.x.

Simulation of 10-day Irrigation Water Quality Using SWAT-QUALKO2 Linkage Model (SWAT-QUALKO2 연계 모형을 이용한 관개기 순별 관개수질 모의)

  • The objectives of this study were to develop a linked watershed-waterbody modeling system and to assess the impacts of indirect wastewater reuse on irrigation water quality. The Osan stream watershed within Gyeonggi-do of South Korea was selected for this study. The linked modeling system was composed of the SWAT (Soil and water assessment tool) and QUALKO2 models. The SWAT model was calibrated and validated using the stream discharge and water quality data from 2010 to 2011. Runoff and non-point source pollutants from each subbasin and stream discharge from 1980 to 2009 were simulated by the SWAT model and applied to the QUALKO2 model. The QUALKO2 model was calibrated and validated under the conditions of low water and normal discharges, respectively. Finally, The 10-day irrigation water quality from April to September was simulated. The statistical measures of coefficient of determination ($R^2$), reliability index (RI), and efficiency index (EI) were used to evaluate the system performance. The $R^2$, RI and EI values ranged from 0.5 to 1.0, 1.03 to 1.92, and -35.03 to 0.95, respectively. The 10-day irrigation water quality showed the concentrations of BOD and coliform exceeded the water quality guidelines for wastewater reuse. The linked modeling system can be a useful tool to estimate non-point source pollutant loads in watershed and to control the water quality of effluent from a wastewater treatment plant and irrigation water in the downstream waterbody.

Integrin-linked Kinase Functions as a Tumor Promoter in Bladder Transitional Cell Carcinoma

  • The aim of this study was to elucidate the role of the integrin-linked kinase (ILK) gene in development of human bladder transitional cell carcinoma (BTCC). Expression of ILK protein and ILK mRNA in 56 cases of human BTCC tissue and in 30 cases of adjacent normal bladder tissue was detected by immunohistochemistry S-P and reverse transcription polymerase chain reaction (RT-PCR), respectively. Four specific miRNA RNAi vectors targeting human ILK were synthesized and transfected into BIU-87 cells by liposome to obtain stable expression cell strains. The influence of ILK on proliferation of BTCC was detected by MTT, FCM on athymic mouse tumorigenesis. The positive rate of ILK protein in BTCC tissue (53.6%) was much higher than adjacent normal bladder tissue (10.0%) (p<0.05). Similarly, expression of ILK mRNA in BTCC tissue ($0.540{\pm}0.083$) was significantly higher than in adjacent normal bladder tissue ($0.492{\pm}0.070$) (p<0.05). MTT showed that the proliferation ability of miRNA-ILK transfected group was clearly decreased (p<0.05), the cell cycle being arrested in G0/G1-S, an tumorigenesis in vivo was also significantly reduced (p<0.05). ILK gene transcription and protein expression may be involved in the development of BTCC, so that ILK might be the new marker for early diagnosis and the new target for gene treatment.
