Chemical Derivatization of Catecholamines for Gas Chromatography-Mass Spectrometry

  • GC/MS analysis of catecholamines (CAs) in biological sample may produce poor reproducible quantitaion when chemical derivatization is used as the technique to form a volatile derivative. Significant quantities of the side products can be formed from CAs with primary amine during the derivatization reaction under un-optimized conditions. We have tested various chemical derivatization techniques in an attempt to find an optimum derivatization method that will reduce side product formation, enable to separate several catecholamine derivatives in GC chromatogram, and obtain significant improvement of detection sensitivity in GC/MS analysis. Whereas several derivatization techniques such as trimethylsilylation (TMS), trifluoroacylation (TFA), and two step derivatization methods were active, selective derivatization to form O-TMS, N-heptafluorobutylacyl (HFBA) derivative using N-methyl-N-(trimethylsilyl)-trifluoroacetamide (MSTFA) and N-methyl-bis(heptafluorobutyramide) (MBHFBA) reagents was found to be the most effective method. Moreover, this derivative formed by selective derivatization could provide sufficient sensitivity and peak separation as well as produce higher mass ion as base peak to use selected ion in SIM mode. Calibration curves based on the use of an isotopically labeled internal standard show good linearity over the range assayed, 1 ~ 5000 ng/mL, with correlation coefficients of > 0.996. The detection limits of the method ranged from 0.2 to 5.0 ppb for the different CAs studied. The developed method will be applied to the analysis of various CAs in biological sample, combined with appropriate sample pretreatment.

  • A ISFET-based glucose sensor has inherent problems such as low sensitivity, drift effect and long response time. For that reason, a amperometric actuation technique was introduce to make a highly sensitivity of the ISFET glucose sensor with a Pt actuator, which electrolyzes $H_2O_2$, one of the by a by-products of the oxidation reaction of glucose. Moreover, a potential-step measurement method detecting response by only the electrolysis of $H_2O_2$ was developed for eliminating a drift problem. The operation characteristics of ISFET-based glucose sensor was improved by using the amperometric actuation and a measurement techniques. The fabricated ISFET glucose sensor is shown good operation such as characteristics(30mM PBS, about 26mV/decade) and linearity. A portable glucose meter with a highly resolution by using the fabricated ISFET-based glucose sensor with Pt actuation was developed and its characteristics investigated.

A study of Improvement on the Road Drainage Poor Site (도로배수 취약구간의 개선방안에 대한 연구)

  • This research aims to investigate the cause of the occurrence of a weak road drainage section scientifically and specifically through a site survey for a poorly drained section occurring due to rainfalls during road operation. This paper deeply reviewed the existing research results and current situation data on the poorly drained sections accumulated in Korea Expressway Corporation in order to investigate the cause of the occurrence of a weak road drainage section, and deeply verified and analyzed the weak sections for the road surface drainage facilities and the other road drainage facilities by visiting the expressway controlled by the 6 local headquarters and 33 branches of Korea Expressway Corporation. As a result of site surveys for the weak road drainage sections, i) in a road surface section, occurrence of ponding in the road shoulder pavement due to slope changes, bad collection of water in the collecting well at a median strip, shortage of road shoulder dike height, and inferior construction, etc. was analyzed to be the main cause of the occurrence of poorly drained sections, and ii) in a road neighborhood section, the occurrence of pavement height difference in a main road and shoulder section due to inferior ditches on a slope and the bad drain age at the inlet and outlet of a culvert due to soil deposits, debris, etc. were analyzed to be the main cause of the occurrence of weak sections. Proposed as a plan to improve the poorly drainage section of road were i)calculation of capacity through material changes at the ditch, enhancement of vertical sections and hydraulic analysis in terms of construction and other aspects, ii)derivation of a combined slope considering a slope and a vertical linearity and maintenance of proper distance between drainage structures in a vertical concave section in terms of geometrical structure, and iii)calculation of the drainage facility installation interval using a minutely rainfall intensity formula and a non-uniform flow analysis technique in terms of hydraulics and hydrologics and prompt removal of rainfalls from the road surface according to a linear drainage method.
