Determination of Pedo-Transfer Function Using the Relation Between Soil Particle Distribution, Organic Matter and Water Movement in Soil Originated from Limestone (석회암 유래 토양에서의 물의 이동특성과 토양 입자 및 유기물과의 관계에 따른 Pedo-Transfer Function의 결정)

  • Soils originated from limestone, located at the southern part of Kangwon province and Jecheon, Danyang of Chungbuk province are mainly composed of fine texture, have different properties from soils originated from granite and granite gneiss, especially for water movement. This study was conducted for making PTF(Pedo-Transfer Function) for Kfs(field saturaton hydraulic conductivity) estimation, and for investigating the relation between soil particle distribution and the infiltration and percolation rate in soils originated from limestone. Soils used for the experiment were 6 soils of Gwarim, Mosan, Jangseong, Maji, Anmi and Pyongan series. Infiltration and percolation rate for the soil were measured by a disc tension infiltrometer and a Guelph permeameter, respectively. The particle size distribution and organic matter content of the soils were analyzed. Kfs was not related with sand, silt, clay, and organic mattrer (OM) content because of forest soils which contained high gravel, pebble, and cobble content, and O layer with high OM content. After Mosan soil series and O layer of Gwarim series were excluded for the data analysis, Kfs was explained as a linear function with sand and clay content and a exponential function with OM content. As a result, the PTF equation was obtained as Kfs=-4.20558+0.479706*(S)+0.023187*exp(1.829*OM) ($R^2=0.6558^{*}$).

EFFICIENCY OF ENERGY TRANSFER BY A POPULATION OF THE FARMED PACIFIC OYSTER, CRASSOSTREA GIGAS IN GEOJE-HANSAN BAY (거제${\cdot}$한산만 양식굴 Crassostrea gigas의 에너지 전환 효율)

  • The efficiency of energy transfer by a population of the farmed pacific oyster, Crassostrea gigas was studied during culture period of 10 months July 1979-April 1980, in Geoje-Hansan Bay near Chungmu City. Energy use by the farmed oyster population was calculated from estimates of half-a-month unit age specific natural mortality rate and data on growth, gonad output, shell organic matter production and respiration. Total mortality during the culture period was estimated approximate $36\%$ from data on survivor individual number per cluster. Growth may be dual consisted of a curved line during the first half culture period (July-November) and a linear line in the later half period (December-April). The first half growth was approximated by the von Bertalanffy growth model; shell height, $SH=6.33\;(1-e^{0.2421(t+0.54)})$, where t is age in half-a-month unit. In the later half growth period shell height was related to t by SH=4.44+0.14t. Dry meat weight (DW) was related to shell height by log $DW=-2.2907+2.589{\cdot}log\;SH,\;(2, and/or log $DW=-5.8153+7.208{\cdot}log\;SH,\;(5. Size specific gonad output (G) as calculated by condition index of before and after the spawning season, was related to shell height by $G=0.0145+(3.95\times10^{-3}{\times}SH^{2.9861})$. Shell organic matter production (SO) was related to shell height by log $SO=-3.1884+2.527{\cdot}1og\;SH$. Size and temperature specific respiration rate (R) as determined in biotron system with controlled temperature, was related to dry meat weight and temperature (T) by log $R=(0.386T-0.5381)+(0.6409-0.0083T){\cdot}log\;DW$. The energy used in metabolism was calculated from size, temperature specific respiration and data on body composition. The calorie contents of oyster meat were estimated by bomb calorimetry based on nitrogen correction. The assimilation efficiency of the oyster estimated directly by a insoluble crude silicate method gave $55.5\%$. From the information presently available by other workers, the assimilation efficiency ranges between $40\%\;and\;70\%$. Twenty seven point four percent of the filtered food material expressed by energy value for oyster population was estimated to have been rejected as pseudofaeces : $17.2\%$ was passed as faeces; $35.04\%$ was respired and lost as heat; $0.38\%$ was bounded up in shell organics; $2.74\%$ was released as gonad output, $2.06\%$ was fell as meat reducing by mortality. The remaining $15.28\%$ was used as meat production. The net efficiency of energy transfer from assimilation to meat production (yield/assimilation) of a farm population of the oyster was estimated to be $28\%$ during culture period July 1979-April 1980. The gross efficiency of energy transfer from ingestion to meat production (yield/food filtered) is probably between $11\%\;and\;20\%$.

Effect of Production In Vitro Embryo using Boar Frozen Semen (돼지 동결 정액을 이용한 체외 수정란 생산 효율)

  • This study was carried out to investigate the effective genetic resources preservation system using the frozen boar semen. The porcine oocytes were matured for 44 hours in NCSU-23 medium with or without 10% Porcine Follicle Fluid (PFF), 0.5 ${\mu}g/ml$ porcine FSH, 0.5 ${\mu}g/ml$ equine LH, 1.0 ${\mu}g/ml$ 17 $\beta$-estradiol ($E_2$) and 10 ng/ml Epidermal Growth Factor (EGF) under mineral oil at $38.5^{\circ}C$ in humidified atmosphere of 5% $CO_2$ in air. After 44 h of culture, the oocytes were inseminated with frozen-thawed semen and fresh semen prepared with mTBM medium for 6 h. Later, set of 50 presumptive zygotes were transferred into 4-well dish (500 ${\mu}l$) of IVC medium. for embryos freezing, slow-freezing and vitrification methods were used as a cryopreservation. Differences among treatments were analyzed using General Linear Model Procedure by SAS Package (version 6.12) differences were considered significant when p<0.05. Following IVF and IVC, the rates of cleavage and blastocysts formation were significantly higher (p<0.05) in hormone supplemented group than that of hormone-free group (25.7 vs, 12.1). The development rates to cleavage and blastocysts were significantly higher in PZM-5 group than NCSU-23 group (60.3%, 46.6% vs 27.4%, 11.1%). Further improvement was achieved when PZM-5 was supplemented with FBS. Cleavage rates was significantly higher in fresh semen source group than frozen semen (66.7% vs 43.7%). However in blastocysts rates was similar two groups. Post-thaw survival rates of embryos were 1.2% and 2.2% in slow-frezing and vitrification groups, respectively. The results of our study suggest that it is still possible to improve the culture conditions and boar semen cryopreservation for enhance reproductive technology and animal genetic resources conservation.

Preparation and Luminescence Optimization of CeO2:Er/Yb Phosphor Prepared by Spray Pyrolysis (분무열분해법으로 CeO2:Er/Yb 형광체 제조 및 발광특성 최적화)

  • Submicron-sized $CeO_2:Er^{3+}/Yb^{3+}$ upconversion phosphor particles were synthesized by spray pyrolysis, and their luminescent properties were characterized by changing the concentration of $Er^{3+}$ and $Yb^{3+}$. $CeO_2:Er^{3+}/Yb^{3+}$ showed an intense green and red emission due to the $^4S_{3/2}$ or $^2H_{11/2}{\rightarrow}^4I_{15/2}$ and $^4F_{9/2}{\rightarrow}^4I_{15/2}$ transition of $Er^{3+}$ ions, respectively. In terms of the emission intensity, the optimal concentrations of Er and Yb were 1.0 % and 2.0%, respectively, and the concentration quenching was found to occur via the dipole-dipole interaction. Upconversion mechanism was discussed by using the dependency of emission intensities on pumping powers and considering the dominant depletion processes of intermediate energy levels for the red and green emission with changing the $Er^{3+}$ concentration. An energy transfer from $Yb^{3+}$ to $Er^{3+}$ in $CeO_2$ host was mainly involved in ground-state absorption (GSA), and non-radiative relaxation from $^4I_{11/2}$ to $^4I_{13/2}$ of $Er^{3+}$ was accelerated by the $Yb^{3+}$ co-doping. As a result, the $Yb^{3+}$ co-doping led to greatly enhance the upconversion intensity with increasing ratios of the red to green emission. Finally, it is revealed that the upconversion emission is achieved by two photon processes in which the linear decay dominates the depletion of intermediate energy levels for green and red emissions for $CeO_2:Er^{3+}/Yb^{3+}$ phosphor.

Horizontal 2-D Finite Element Model for Analysis of Mixing Transport of Heat Pollutant (열오염 혼합 거동 해석을 위한 수평 2차원 유한요소모형)

  • A numerical model has been developed by employing a finite element method to simulate the depth-averaged 2-D dispersion of the heat pollutant, which is an important pollutant material in natural streams. Among the finite element methods, the Streamline Upwind/Petrov Galerkin (SUPG) method was applied. Also both linear and quadratic elements can be applied so that irregular river boundaries can be easily represented. To show the movement of heat pollutants, the reaction term describing heat transfer was represented as an equation in which sink/source term is proportional to the difference between the equilibrium temperature and water surface temperature. The equation was expressed so that the water surface temperature changes according to the temperature transfer coefficient and the equilibrium temperature. For the calibration of the model developed, analytic and numerical results from a case of rectangular channel with full width continuous injection have been compared in a steady state. The comparisons showed that the numerical results were in good agreement with analytical solutions. The application site was selected from the downstream of Paldang dam to Jamsil submerged weir, and overall length of this site is about 22.5 km. The change of water temperature caused by the discharge from the Guri sewage treatment plant has been simulated, and results were similar to the observed data. Overall it is concluded that the developed model can represent the water temperature changes due to heat transport accurately. But the verification using observed data will further enhance the validity of the model.

  • Cryopreservation of boar semen is continually researched in reproductive technologies and genetic resource banking in breed conservation. For evaluating the boar semen quality, sperm motility (MOT) is an important parameter because the movement of spermatozoa indicates active metabolism, membrane integrity and fertilizing capacity. Various researches have been trying to improve the quality of semen post-thawed in boar. Recently, polymorphism (g.358A>T) of cluster-of-differentiation antigen 9 (CD9) gene reported to be significant association with MOT. Also, CD9 gene was expressed in the male germ line stem cells is crucial for sperm-egg fusion, and was therefore selected as candidate gene for boar semen. This study was conducted to evaluate the pig SNP (g.358A>T) of CD9 gene as a positional controlling for semen parameters of post-thawed boar semen. To results, the g.358A>T SNP of the CD9 gene was significantly associated with the traits such as MOT, curve linear velocity, straight line velocity, average path velocity and amplitude of lateral head displacement. Particularly, the g.358A>T SNP significantly has the highest association with MOT and animals with AA genotype (p<0.001). Therefore, we suggest that the g.358A>T in the intron 6 region of the porcine CD9 may be used as a molecular marker for Duroc boar Post-thawed semen quality, although its functional effect was not defined yet.

Mass Transfer during Salting and Desalting Processes of Chinese Cabbage (배추의 염절임 및 탈염 공정중 물질이동)

  • The diffusion phenomena of water, solid and reducing sugar in Chinese cabbage during salting (5$0^{\circ}C$, 25% salt solution) and desalting (5$0^{\circ}C$, distilled water) were investigated. Water loss and solid gain during salting were rapid in the first 6hrs and then almost leveled off. After 24hrs of salting, water loss and solid gain in 100g of initial wet Chinese cabbage were 33.35g and 6.26g respectively. Moisture content was changed from 94.29% to 83.11% during 24hrs of salting. The reducing sugar concentration was also changed from 29.2 mg/$m\ell$ to 6.5mg/$m\ell$, which was linearized as a function of the square root of salting time and showing that Y=30.1841-5.0269√t. After 24hrs salting, water gain and solid loss during desalting were rapid in the first 4hrs and then increased linearly. After 12hrs of desalting, the water gain and solid loss in 100g of initial wet Chinese cabbage were 20.82g and 9.14g respectively. The amount of solid loss after 12hrs desalting was higher than that of solid gain after 24hrs salting due to the diffusion of solute presented initially in the Chinese cabbage during salting and desalting. The concentration of salt in Chinese cabbage after 12hrs desalting was 2.98% which was a suitable salt concentration for the preparation of Kimchi. At this time, the concentration of reducing sugar was only 1.6mg/$m\ell$. The linear regression equation of reducing sugar concentration during desalting was Y=6.7854-1.5992√t.

A Study on Dynamic Behaviour of Cable-Stayed Bridge by Vehicle Load (차량하중에 의한 사장교의 동적거동에 관한 연구)

  • This paper is considered on the dynamic behavior and the dynamic impact coefficient on the cable-stayed bridge under the vehicle load. The method of static analysis, that is, the transfer matrix method is used to get influence values about displacements, section forces of girder and cable forces. Gotten influence values were used as basic data to analyse dynamic behavior. This paper used the transfer matrix method because it is relatively simpler than the finite element method, and calculating speed of computer is very fast and the precision of computation is high. In the process of dynamic analysis, the uncoupled equation of motion is derived from simultaneous equation of the motion of cable-stayed bridge and vehicle travelling by using mode shape, which was borne from system of undamped free vibration. The solution of the uncoupled equation of motion, that is, time history of response of deflections, velocity and acceleration on reference coordinate system, is found by Newmark-${\beta}$ method, a kind of direct integral method. After the time history of dynamic response was gotten, and it was transfered to the time history of dynamic response of cable-stayed bridge by linear transformation of coordinates. As a result of this numerical analysis, effect of dynamic behavior for cable-stayed bridge under the vehicle load has varied depending on parameter of design, that is, the ratio of span, the ratio of main span length, tower height, the flexural rigidity of longitudinal girder, the flexural rigidity of tower, and the cable stiffness, investigated. Very good agreements with the existing solution in the literature are shown for the uncracked plate as well as the cracked plate.

Analysis of the Bearing Capacity of Drilled Shafts Compared with Driven Piles (항타말뚝과 비교한 현장타설말뚝의 지지력분석)

  • In this study an iterative procedure for the analysis of drilled shafts was proposed on the basis of the load transfer mechanism. Special attention was given to the estimation of bearing capacity of drilled shafts which was compared with driven piles, and then with the results of pile load test. The load displacement at the pile head was calculated by load than sfer curves (t -z curves, q-z curves) by using Vljayvergiya, Castelli and hi -linear models. Bab ed on the analytical results, it is found that the behavior of drilled shafts is different from that of driven piles the smaller the skin friction mobilized at the pile-boil interface, the smaller the development of the bearing capacity. Hence the greater pile head movement is required to mobilize the same mainitride of bearing capacity. This trend is more noticeable in sand than in clay. It is also found that as the length-todiameter ratios increase, the dirtference of ultimate bearing capacity between drilled shafts and driven piles is becoming lass ger in sand, but it is minor in clay.

Development of Sperm MTT Assay for Its Application in Boar Semen

  • The MTT assay is one of superior evaluation methods widely used to analyze the viability of metabolically active cell. It can be used to determine the percentage of viable sperm through measurement of the reduction of MTT granules at mitochondria in sperm tail. The purpose of this study is to determine the optimal condition of a simple and easy MTT assay to validate boar sperm viability and compare the accuracy of this test with microscopic examination. The MTT reduction rate for sperm viability were analyzed in microtiter plates (96 well) from 1 hr to 5 hr incubation periods at $37^{\circ}C$ using spectrophotometer (microplate reader) at 550 nm wavelength. The remainder of semen sample was simultaneously examined to compare the correlation of accuracy between MTT assay and other sperm parameters. Those sperm parameters were included the motility, survival rates, membrane integrity, mitochondria activity and acrosome integrity. The OD values of MTT assay (MTT reduction rates) did not greatly change at 1 hr to 5 hr incubation periods in different proportion of live and freeze-killed sperms (dead sperm). The MTT reduction rates or survival rates were decreased according to the different concentration of live and dead sperm. The linear regression at 1 hr and 4 hr incubation periods in sperm MTT assay was y=291.55x-72.176 and y= 180.64x-44.569, respectively. There are high correlation between 1 hr and 4 hr incubation periods (p<0.001). The results of MTT assay and other sperm parameters has a positive correlation (p<0.01 or 0.05). The correlation coefficients for MTT assay was 0.88115 for motility, 0.89868 for survival rates, 0.91722 for membrane integrity and 0.77372 for acrosome integrity, respectively. In conclusion, the MTT assay can be used as a reliable and efficient evaluation method for boar sperm viability. It can be use practical means to evaluate the quality of boar sperm by a fast, inexpensive and easy method.
