We suggest the fluorescence endoscope system that has light source apparatus providing selectable white or excitation light. White light source generates normal color images and is easily switched over to excitation light with the wide spectrum range from 380 nm to 580 nm. 5-ALA is deposited selectively in the abnormal tissue like cancer and causes fluorescence in the red spectrum range when excited by blue spectrum range. In addition, the others of excitation light make the color background image by reflected light to allow accurate orientation and visualization of the abnormal tissue and around. According to clinical studies, the fluorescence intensity contrast that defines the fluorescence intensity of lesion over the fluorescence intensity of around has more than 2 in tumour. Proposed system is useful and objective way in early diagnosis. Furthermore, it can be used in the biopsy for tumour classification at the highest fluorescence intensity point.

The light source was a Xenon lamp and filter wheel, composed of an optical output control through Iris and filters with several wavelength bands. It also makes the inducement of auto-fluorescence possible because it is designed to generate a wavelength band of 380-420nm, 430-480nm, and 480-560nm. The transmission part of the light source was developed to enhance the efficiency of light transmission. To evaluate this light source, the characteristics of light output and wavelength band were verified. To validate the capability of this device as PDD, the detection of auto-fluorescence using mouse models was performed.
Chlorophyll fluorescence has been researched for assessing fruit post-harvest quality and condition. The objective of this preliminary research was to investigate the potential of fluorescence spectroscopy for measuring apple fruit quality. Ultraviolet (UV) and blue light was used as an excitation source for inducing fluorescence in apples. Fluorescence spectra were measured from 'Golden Delicious' (GD) and 'Red Delicious' (RD) apples using a visible/near-infrared spectrometer after one, three, and five minutes of continuous UV/blue light illumination. Standard destructive tests were performed to measure fruit firmness, skin and flesh color, soluble solids and acid content from the apples. Calibration models for each of the three illumination time periods were developed to predict fruit quality indexes. The results showed that fluorescence emission decreased steadily during the first three minutes of UV/blue light illumination and was stable within five minutes. The differences were minimal in the model prediction results based on fluorescence data at one, three or five minutes of illumination. Overall, better predictions were obtained for apple skin chroma and hue and flesh hue with values for the correlation coefficient of validation between 0.80 and 0.90 for both GD and RD. Relatively poor predictions were obtained for fruit firmness, soluble solids content, titrational acid, and flesh chroma. This research has demonstrated that fluorescence spectroscopy is potentially useful for assessing selected quality attributes of apple fruit and further research is needed to improve fluorescence measurements so that better predictions of fruit quality can be achieved.
Photodynamic diagnosis(PDD) is a method to diagnose the possibility of cancer, both by the principle that if a photosensitizer is injected into an organic tissue, it is accumulated in the tissue of a malignant tumor selectively after a specific period, and by a comparison of the intensity of the fluorescence of normal tissue with abnormal tissue after investigating the excitation light of a tissue with accumulated photosensitizer. Since the selection of the wavelength band of excitation light has an interrelation with fluorescence generation according to the selection of a photosencitizer, it plays an important role in POD. Exposure of algae or plants to irradiance from above the light saturation point of photosynthesis is known as high light stress. This high light stress induces various responses including photoinhibition of the photosynthetic apparatus. The degree of photoinhibition could be clearly determined by measuring the parameters such as absorption and fluorescence of chromoproteins. In cyanobacteria and red algae, most of the photosystem (PS) II associated light harvesting is performed by a membrane attached complex called the phycobilisome (PBS). The effects of high intensity light (1000-4000 ${\mu}mol$ photons $m^{-2}s^{-1}$) on excitation energy transfer from PBSs to PS II in a cyanobacterium Spirulina platensis were studied by measuring room temperature PC fluorescence emission spectra. High light (3000 ${\mu}mol$ photons $m^{-2}s^{-1}$) stress had a significant effect on PC fluorescence emission spectra. On the other hand, light stress induced an increase in the ratio of PC fluorescence intensity of PBS indicating that light stress inhibits excitation energy transfer from PBS to PS II. The high light treatment to 3000 ${\mu}mol$ photons $m^{-2}s^{-1}$ caused disappearance of 31.5 kDa linker polypeptide which is known to link PC discs together. In addition we observed the similar decrease in the other polypeptide contents. Our data concludes that the Spirulina cells upon light treatment causes alterations in the phycobiliproteins (PBPs) and affects the energy transfer process within the PBSs.
The aim of the present study was to describe an safe and convenient method for the early detection of enamel caries using laser fluorescence. Fluorescence from natually carious lesion of human teeth illuminated by an argon laser(488nm) was observed and photographed using barrier filter. Intact enamel was found to fluorescence with a yellowish light. Whereas, incipient caries lesions in the enamel were dearly visible as dark areas in contrast to the fluorescence surroundings. For evaluation of accuracy of this method, lesion depth measured by the laser fluorescence in light microscope was compared with that polarizing microscope. The results from the present study can be summarized as follows : 1. Enamel caries of smooth surface was observed as pale white spot and undefined outline in ordinary light. Whereas, lesion was clearly visible as dark spot in laser fluorescence. 2. There was no difference between ordinary light view and laser fluorescence in occlusal surface and interproximal surface. 3. There was no significant difference between the lesion depth observed by laser fluorescence with light microscope and polarizing microscope. Apparent correlation exists between two groups.
A miniaturized fluorescence detection system based on total internal reflection (TIR) configuration, which is applicable to detecting the presence of biological materials labeled with fluorescence dye in micro total analysis systems (${\mu}TAS$), is proposed. In conventional fluorescence testing and analysis devices, interference between the excitation light beam and the emitted light from dyes is unavoidable. This paper presents a fluorescence detection system based on TIR configuration that allows the excitation light beam and the emitted light to be spatially perpendicular to each other so as to minimize the interference where fluorescence emission is detected at the orthogonal angle to the excitation beam. We achieved the limit of detection of about 5 nmol/L with a high linearity of 0.994 over a wide range of 6-FAM mol concentration, being comparable to that in earlier studies.
An apparatus for measuring oil oxidation was developed, which is capable of being mounted to mechanical devices for detecting power of fluorescent light reflected from oil in real time as an indication of the oil oxidation. This device has an advantage over conventional fluorescence spectrometers where the thin film is required for the measurement. Clean and used oil samples (mineral and synthetic oils) were tested by the developed apparatus that calculates a fluorescence quantum yield and a light absorption coefficient of the oil based upon the signals from the two light-receiving members and evaluates the degree of oil oxidation of test oils based on the fluorescence quantum yield. Results generally show that the developed device is able to effectively evaluate oil oxidation characteristics on-site in the field.
형광 이미징은 시간 분해능과 공간 해상도가 높기 때문에 기초연구에서 세포나 소동물 이미징에 널리 활용된다. 기존의 형광 이미징은 가시광선 영역의 광원을 활용하기 때문에 조직 내 광투과도가 낮고, 광원에 의한 광독성이 생길 수 있으며, 자가형광에 의한 간섭으로 검출 민감도가 떨어지는 한계가 있다. 이러한 점을 개선하기 위해 에너지가 낮은 장파장의 광원을 활용하고자 하며, 700~900 nm 영역을 활용하는 근적외선-I 형광 이미징이 개발되었고, 이미징 성능을 대폭 향상시키기 위해서 1000~1700 nm 영역의 장파장을 이용하는 근적외선-II 이미징이 연구자들의 관심을 받고 있다. 근적외선-II 영역은 광산란이 최소화되어 생체조직 내 투과도를 약 10 mm까지 향상시킬 수 있고, 생체조직의 자가형광도 최소화되어 고민감도와 고해상도의 형광 이미징이 가능하다. 본 총설에서는 다양한 근적외선-II 형광 이미징 탐침 중에서 광안정성이 뛰어나고 발광 파장 조절이 용이한 무기 나노입자 기반 탐침에 대해 살펴보았고, 그 중에서 단층 탄소 나노튜브와 양자점 및 란탄족 나노입자에 대해 중점적으로 기술하였다.
We fabricated photo-sensor for fluorescence detection in LOC. LOC is high throughput screening system. Our LOC screens biochemical reaction of protein using the immunoassay, and converts biochemical reaction into electrical signal using LIF(Laser Induced Fluorescence) detection method. Protein is labeled with rhodamine intercalating dye and finger PIN photodiode is used as photo-sensor We measured fluorescence emission of rhodamine dye and analyzed tendency of fluorescence detection, according to photo-sensor size, light intensity, and rhodamine concentration. Detection current was almost linearly proportional to two parameters, intensity and concentration, and was inversely proportional to photo-sensor size. Integrated LOC consists of optical-filter deposited photo-sensor and PDMS microchannel detected 50 (pg/${mu}ell$) rhodamine. For integrated LOC including light source, we used green LED as the light source and measured emitted fluorescence.
