인삼 사포닌이 개 심실 형질막의 $K^+$-의존성 포스파타제 활성에 미치는 영향 (Effect of Ginseng Saponins on $K^+-Dependent$ Phosphatase Activity of Dog Cardiac Sarcolemma)

  • The effects of ginseng saponins, gypsophila saponin, sodium dodecyl sulfate(SDS), and Triton X-100 on membrane $K^+-dependent$ phosphatase activity which is lipid dependent and represents dephosphorylation step of the complete Na+, $K^+-ATPase$ reaction were investigated in this study to elucidate whether the effects of ginseng saponins are due to the detergent action, using sarcolemma enriched preparation isolated from dog ventricle. $Na^+$, $K^+-ATPase$ and $K^+-dependent$ phosphatase activities of cardiac sarcolemma were about $143\;{\mu}mol$ Pi/mg protein/hr and $34\;{\mu}mol$ p-nitrophenol/mg protein/hr, respectively. While ginseng saponins (triol>total>diol) inhibited $K^+-dependent$ phosphatase activity, gypsophila saponin, and low dose of SDS($0.4\;{\mu}g/{\mu}g$ protein), and Triton X-100 ($0.6\;{\mu}g/{\mu}g$ protein) increased the enzyme activity, indicating disruptive effect of detergents on membrane barriers. The activating effect of low doses of Triton X-100 on membrane $K^+-dependent$ phosphatase appeared at concentration decreasing light scattering. However, the inhibitory effect of ginseng saponin appeared before a decrease in light scattering. These results suggest that low concentrations of ginseng saponins inhibit the membrane $K^+-dependent$ phosphatase by interacting directly with enzyme before membrane disruption.

  • Isolated rat thoracic aorta which is pharmacologically precontracted by phenylephrine induces photorelaxation when exposed to long wave length UV-light. The aim of the present study was to characterize the mechanism of UV-light induced by photorelaxation in the rat aorta. 1. UV light relaxed both endothelium-intact and -denuded rat aortic rings contracted by phenylephrine. The magnitude of relaxation on UV light was dependent on the exposure time and slightly greatly in endothelium-denuded rings than in endothelium-intact preparations. 2. L-NAME (10 nM - 100 $\mu$M) but not D-NAME completely inhibited the photorelaxation in a concentration dependent manner. 3. The UV-induced relaxation was inhibited by methylene blue (1 - 100 uM), and verapamil (100 nM), and removal of extracellular $Ca^{2+}$. In contrast, UV-light induced photorelaxation was potentiated by $N^{w}$-nitro-L-arginine (L-NNA) treatment. These results suggest that UV light-induced photorelaxation may be due to nitric oxide from exogenously administered L-arginine as well as endogenous nitric oxide donors such as amino acid and arginine derivatives

  • We studied the chloroplast rearrangement, short-term regulation depending on the light conditions in plants, and the characteristic of photosynthic rate as affected by in Panax ginseng C.A. Meyer. The chloroplast rearrangement of ginseng mesophyll cell was induced with the irradiation of blue light (400~500 nm) and through this process the rate of leaf transmittance increased 5~7.5%. The time to reach the maximum value of photosynthesis was shorter above 20 minutes with the blue light irradiation than that of the red light.

저온처리한 오이의 자엽에서 광합성 활성의 광의존성 저해 (Light-Dependent Chilling Injury on the Photosynthetic Activities of Cucumber Cotyledons)

  • The photosynthetic activities in relation to oxygen evolution rates, quantum yield, CO2 uptake rates and room temperature chlorophyll fluorescence were investigated in cotyledons of cucumber seedlings exposed to low temperature (at 4$^{\circ}C$) for 24 h. Light-chilling caused more inhibition on light-saturated maximum oxygen evolution rates, quantum yield, and CO2 uptake rates than dark-chilling did in the cucumber plant. Light-chilling induced more marked increase in Fo and decrease in (Fv)m/Fm than dark-chilling did in the room temperature chlorophyll induction kinetics. The above results affected by chilling in the light are considered to be associated with the partial damage of the reaction center of PS II and the decreased photosynthetic activities. There occurred a large decrease in qQ with little change in qNP in the light-chilling plant. When light- and dark-chilled plants were recovered at room temperature for 24 h and their chlorophyll fluorescences were induced with light doubling technique, light-chilled plants showed more smaller magnitude and rate of fluorescence relaxation than dark-chilled plants. These suggest that light-chilling might cause some alterations in transthylakoid pH formation, and that photosynthetic apparatus of cucumber cotyledons is more susceptible to light-chilling. In the fast fluorescence induction kinetics, FR was decreased by 60% in the light-chilled plants with reference to $25^{\circ}C$ light-grown plants, while the dark-chilled plants showed a decreased rate of only 20% with reference to $25^{\circ}C$ dark-treated plants for 24 h, indicating that cucumber seedling is very sensitive to chilling stress. So, it is certain that chilling injury to the photosynthetic apparatus is strongly dependent on the presence of light in cucumber seedlings.

Numerical Study of Polarization-Dependent Emission Properties of Localized-Surface-Plasmon-Coupled Light Emitting Diodes with Ag/SiO2 Na

  • We study polarization-dependent spontaneous emission (SE) rate and light extraction efficiency (LEE) in localized-surface-plasmon (LSP)-coupled light emitting diodes (LEDs). The closely packed seven $Ag/SiO_2$ core-shell (CS) nanoparticles (NPs) lie on top of the GaN surface for LSP coupling with a radiated dipole. According to the dipole direction, both the SE rate and the LEE are significantly modified by the LSP effect at the $Ag/SiO_2$ CS NPs when the size of Ag, the thickness of $SiO_2$, and the position of the dipole source are varied. The enhancement of the SE rate is related to an induced dipole effect at the Ag, and the high LEE is caused by light scattering with an LSP mode at $Ag/SiO_2$ CS NPs. We suggest the optimum position of the quantum well (QW) in blue InGaN/GaN LEDs with $Ag/SiO_2$ CS NPs for practical application.

A Novel Phototransduction Pathway in the Pineal Gland and Retina

  • Light is a major environmental signal for entrainment of the circadian clock, but little is known about the phototransduction pathway triggered by light-activation of photoreceptive molecule(s) responsible for the phase shift of the clock in vertebrates. The chicken pineal gland and retina contain the autonomous circadian oscillators together with the photic entrainment pathway, and hence they provide useful experimental model for the clock system. We previously demonstrated the expression and light-dependent activation of rod-type transducin $\alpha$-subunit (Gtl$\alpha$) in the chicken pineal gland. It is unlikely, however, that the pineal Gt$_1$$\alpha$ plays a major role in the photic entrainment, because the light-induced phase shift is unaffected by bloking the signaling function of Gt$_1$$\alpha$. Here, we show the expression of G 11 $\alpha$, an $\alpha$-subunit of another heterotrimeric G-protein, in the chicken pineal gland and retina by cDNA cloning, Northern blot and Western blot analyses. GIl$\alpha$-immunoreactivity was colocalized with pinopsin in the chicken pineal cells and it was found predominantly at the outer segments of photoreceptor cells in the retinal sections, suggesting functional coupling of G11 $\alpha$ with opsins in the both the tissues. By coimmunoprecipitation experiments using the retina, we showed the light- and GTP-dependent interaction between rhodopsin and G11 $\alpha$. Upon ectopic expression of a Gq/ 11-coupled receptor in cultured pineal cells, pharmacological (non-photic) activation of endogenous G11 induced phase-dependent phase shifts of the melatonin rhythm in a manner very similar to the effect of light. These results suggested opsin-G11 pathway contributing to the photic entrainment of the circadian clock.

Continuous Viewing Angle Distribution Control of Liquid Crystal Displays Using Polarization-Dependent Prism Array Film Stacked on Directional Backlight Unit

  • We present a polarization-dependent prism array film for controlling the viewing angle distribution of liquid crystal (LC) display panels without loss of light efficiency. On a directional backlight unit, our polarization-dependent prism array film, made into a stacked bilayer with a well-aligned liquid crystalline reactive mesogen (RM) layer on the UV-imprinted prism structure, can continuously control the light refraction function of the prism array by electrically switching incident polarization states of a polarization-controlling layer prepared by a twisted nematic LC mode. The viewing angle control properties of the polarization-dependent prism array film are analyzed under different prism angle and refractive index conditions of the RM layer. A simple analytic model is also presented to describe the intermediate viewing angle distributions with continuously varying applied voltages and incident polarization states.

Developmental Changes in Photosynthetic Pigments and Chlorophyll Fluorescence in Etiolated Rice Seedlings During Greening

  • Developmental of photosynthetic pigments and changes in chlorophyll fluorescence of dark-grown rice seedlings were studied during greening. Light-illumination stimulated accumulations of total chlorophylls and carotenoids in leaves of etiolated seedlings, accompanied by a decrease in the ratio of chlorophyll a to chlorophyll b. When the composition of carotenoids was analyzed, violaxanthin level was shown to increase up to 24 h after the beginning of light illumination, followed by a subsequent decline. In contrast to this, zeaxanthin level increased consistently with progress of deetiolatin. The role of zeaxanthin is discussed in relation to chlorophyll fluorescence quenching. A study on chlorophyll fluorescence kinetics of the rice seedlings being deetiolated showed a time-dependent increase in Fv/Fm (yield of variable fluorescence/maximum yield of fluoresecnece) ratios, indicating that greening is responsible for the activation of photochemical reaction centers of the photosystem. When chlorophyll fluorescence quenching was examined, qNP (nonphotochemical quenching) and qE (energy-dependent quenching) exhibited a time-dependent decline with progress of greening. The presented results indicate that greening-induced development of the photosynthetic machinery is associated the conversion of the carotenoid violaxanthin to zeaxanthin, suggesting that zeaxanthin synthesized in the illuminated leaves may provide the protection from the damage when etiolated plants are exposed to light.

EXOSAT X-RAY LIGHT CURVES OF SS ARI

  • We construct the X-ray light curves of the W UMa type contact binary SS Arietis(HD12929) from the EXOSAT data in HEASARC data archive. The phase dependent X-ray light curves obtained by using the ephimeris of Kaluzny & Pojmanski of SS Ari is modulated on the orbital phase as in the case of other W UMa type binaries. Although a detailed analysis of these data is impossible because of the very low X-ray light curves in the context of the radiation in the corona region of W UMa type stars.

  • We proposed driving method with variable integration time for ambient light sensing. One operation period of the proposed driving method consists of several sub-integration periods with variable integration time which can enlarge dynamic range of ambient light sensing circuit. Temperature dependent characteristic of p-intrinsic-metal (p-i-m) diode can be compensated using the proposed driving method.
