Serratia marcescens 균주로부터 추출한 Prodigiosin의 흡수분광학적 연구 (Absorption Spectroscopic Studies of Prodigiosin Extracted from Serratia Marcescens Strain)

  • Serratia marcescens 2354(ATCC 25419) 균주로부터 추출한 붉은 색소는 prodigiosin (PG)이었고, 이를 methanol에 녹여 자외선 및 가시광선 흡수 스펙트라를 측정한 결과 537 nm의 최대흡수파장 (${\lambda}_{max}$)을 갖는 산성용액에서의 전형적인 PG의 흡수 스펙트라이었다. 또한 methanol 용액에서 PG의 농도를 $1.0{\times}10-5M$에서 $9.0{\times}10-5M$로 증가시키면, 537 nm의 흡수강도는 증가하고 467 nm의 흡수강도는 감소하였으며, 500 nm에서 isosbestic point가 관측되었다. 이러한 현상은 537 nm와 467 nm가 각각 산과 염기용액에서의 PG 흡수대이고, 500 nm의 isosbestic point 등을 고려하면 가역적 산-염기 평형반응에 의한 결과라고 볼 수 있다. 한편 pH, 4.75의 acetic acid 완충용액에서 PG의 농도를 $6.0{\times}10^{-4}M$에서 $1.0{\times}10^{-4}M$로 감소시키면 500 nm에서 ${\lambda}_{max}$를 가지는 새로운 흡수대가 나타난다. 이 흡수대는 pH 4.75의 수용액에서만 나타나는 것으로 같은 pH의 순수한 methanol 용액에서는 나타나지 않는다. 이는 PG 분자가 $H_2O$에 의해 ${\alpha}$-이성질체에서 ${\beta}$-이성질체로의 전환에 기인하는 것이다. 즉 PG의 색변화는 용액의 농도 및 용매의 특성에 의해서도 일어날 수 있음을 확인하였다.

노면표시 색상에 따른 최소재귀반사성능 연구 (Study on the Minimum Recursive Reflection Performance according to the Color of Road Surface)

  • 도로교통법 시행규칙과 단체표준에서 정하고 있는 8가지 색상에 대해 건조 시와 습윤 시의 최소재귀반사성능 기준 확보를 위한 현장실험을 실시하였으며, 백색 건조 시에는 260.8(mcd/㎡·lux), 습윤 시에는 154.6(mcd/㎡·lux)으로 결과가 산출되었다. 황색의 경우는 건조 시 백색의 반사성능 대비 황색, 가호1호에서는 67%, 가호2호에서는 63% 수준이다. 습윤 시는 각각 79%와 59% 수준으로 나타났다. 청색의 경우에는 백색대비 가호1호에서는 64% 수준이며, 가호2호에서는 72%이다. 습윤 시는 각각 63%와 72% 수준으로 나타났다. 습윤 시의 반사성능 변화 폭이 건조 시보다 높으며, 유리알이 없는 경우도 이전의 결과와 비슷하게 나타났다. 신규색상이 적색, 주황색, 분홍색, 연한 녹색, 녹색의 경우에도 백색 대비 50% 이상의 기준값을 가지는 것으로 분석되었으며 이를 토대로 도로교통법 시행규칙에 노면표시 색상에 따른 최소반사성능 기준 마련의 토대가 될 것으로 판단된다.

Development of Standard Operating Procedures (SOPs), Standardization, TLC and HPTLC Fingerprinting of a Polyherbal Unani Formulation

  • Background: Unani System of Medicine (USM) has its origin to Greece. To ensure and develop the quality, authenticity of Unani drugs, standardization on modern analytical parameter is essential requirement for drugs. Objectives: The aimed of the present study was to develop a standard profile of "Qurṣ-e-Mafasil" by systematic study through authenticated ingredients, pharmacognostic identification followed by physicochemical, TLC, HPTLC fingerprinting analysis as per standard protocol. Material and Methods: In this study three batches of "Qurṣ-e-Mafasil" QM were prepared by standard method as per UPI had been followed by organoleptic properties of formulation such as appearance, color, odor, taste. Powder Microscopy and physicochemical studies were carried out such as Uniformity of weight, Friability, Disintegration time, hardness, LOD, ash vales and extractive values in like aqueous, alcohol & hexane. Further qualitative tests such as Thin-Layer Chromatography (TLC), and High-Performance Thin Layer Chromatography (HPTLC) studies were also carried out to develop fingerprint pattern of the alcoholic solvent extract of QM. Phytochemical screening was carried out in different solvent extracts such as alcoholic, aqueous and chloroform extracts to detect the presence phytoconstituents in the formulation QM. Heavy metals, Microbial Load Contamination and pesticidal residues were also determined. Results: Qurṣ-e-Mafasil showed tablet-like appearance, light brown colour, mild pungent odour and acrid taste. Uniformity of weight (mg), friability (rpm), and hardness (kg/cm) and disintegration time was ranged between (500 to 503), (0.0340 to 0.038), (8.40 to 8.67) and (4-5 minutes) respectively for the three batches. Loss in weight on drying at 105℃ was ranged between (8.3425 to 8.7346). Extracted values were calculated in distilled water ranged between (30.9091 to 31.4358), hexane (1.1419 to 1.4281), and alcohol (3.3352 to 3.3962). The ash values recorded were ranged between (3.7336 to 3.8378), and acid insoluble ash (0.5859 to 0.6112).

해조의 육종을 위한 기초적 연구 3. 김 엽체에 미치는 식물호르몬의 생장효과 (Fundamental Studies for the Breeding of Marine Algae 3. Effects of Plant Hormones on the Growth of Porphyra-fronds)

  • 김엽체의 생장에 미치는 식물흐르몬의 영향을 조사하기 위해서 Gibberellin, IAA, Kinetin, NAA및 2.4-D 등을 농도별로 회석한 배지에서 김엽편을 32일간 배양하고 또 2차적으로 Kinetin의 농도별배지에서 수온을 $5^{\circ}C$$10^{\circ}C$로 구분하여 18일간 배양하였다. 그 결과를 요약하면 다음과 같다. (1) Gibberellin은 10.0mg/l 농도에서, 기타는 모두 5.0mg/1 농도의 배지에시 각각 최고생장효과가 나타났다. 간종량으로 본 효율치는 Kinetin이 $312.5\%$(엽면체:$345.3\%$)로 으뜸이며 2.4-D는 $257.5\% (236.1\%)$, Gibberellin $247.9\%(241.9\%)$, IAA, $166.7\%(147.6\%)$, NAA $141.7\% (167.7\%)$의 순이였다. (2) 특히 Kinetin 5.0mg/1농도에서 배양된 엽편은 그 색채가 진한 흑갈색으로 가장 좋았으며 세포마다 생기 있는 큰 색소체가 뚜렷하고 건전하여 뛰어난 효과가 나타났으나 기타는 색채가 담갈색 또는 황록갈색으로 변하고 사세포가 많이 발견되었으며 생세포도 원형질이 위축되었거나 큰 세포가 생겨 매우 불건전하였다.(3) 저온($5^{\circ}C$)과 고온($10^{\circ}C$)에서의 Kinetin의 생장효율은 저농도에서는 대차가 없었으나, 5.0mg/1의 고농도애시는 저온이 $366.7\%$, 고온이 $318.8\%$로서 $48\%$의 효율차가 생겼으며 세포의 건전성은 1차시험때와 마찬가지로 모두 좋았다.(4) 최고생장효과가 나타난 Kinetin 5.0mg/1농도에서는 엽편의 녹변에 지름 $25.0\sim27.5\mu$ 정도의 이상대형세포가 많이 발견되었으며 그 중에는 한 세포내에 수개의 색소체가 있는 것이 관찰되었다. 두께도 $41.0\sim42.0\mu$으로 비교적 두꺼웠다.(5) Kinetia 1.0mg/l농도의 배지에서 배양된 엽편중에는 조정기을 형성하고 있는 세포군이 관찰되었다.

HPLC-UVD를 이용한 폴리에틸렌, 폴리프로필렌 기구 및 용기·포장 유래 자외선흡수제 동시분석법 (Simultaneous Determination of UV Absorbers Migrated from Polyethylene and Polypropylene Food Packaging Materials into Food Simulants by HPLC-UVD)

  • 본 연구에서는 식품용 기구 및 용기 포장으로부터 식품유사용매로 이행되는 12종의 자외선흡수제(Uvinul 3000, Cyasorb UV 24, Uvinul 3040, Tinuvin 312 및 P, Seesorb 202, Chimassorb 81, Tinuvin 329, 234, 326, 328 및 327)의 분석법을 확립하였다. 식품유사용매 중 물, 4% 초산, 50% 에탄올의 경우 hydrophilic-lipophilic balance (HLB) 카트리지로 고체상 추출법(Solid Phase Extraction)을 이용하였고, n-헵탄의 경우 이소프로판올로 희석하여 HPLC-UVD(310 nm)로 자외선흡수제의 이행량을 분석하였다. 확립된 분석법으로 일회용기, 밀폐용기, 일회용백, 소스병, 물병, 도시락 등 국내 유통 폴리에틸렌(60건) 및 폴리프로필렌(140건) 재질의 식품용 기구 및 용기 포장 200건으로부터 식품유사용매로 이행되는 12종의 자외선흡수제 이행량 조사 결과, 물, 4% 초산, 50% 에탄올, n-헵탄 4가지 식품유사용매 모두에서 자외선흡수제는 검출되지 않았다. 따라서, 실제 가정에서 폴리에틸렌이나 폴리프로필렌 재질의 식품용 기구 및 용기 포장을 사용하는 환경에서는 자외선흡수제의 이행 정도는 안전한 수준인 것으로 판단된다. 또한, 자외선흡수제를 0.1 wt%, 0.5 wt%씩 첨가하여 제작한 시편에서의 이행정도는 0.02~0.5% 수준으로 나타나 통상적인 식품용 기구 및 용기 포장을 사용하는 조건에서 자외선흡수제의 이행량은 크지 않은 것으로 나타났고, 안전한 수준임을 확인할 수 있었다. 본 연구에서 확립된 12종 자외선흡수제의 식품유사용매로의 이행량 동시분석법은 폴리에틸렌 및 폴리프로필렌 식품용 기구 및 용기 포장의 안전관리를 위한 기초자료로 활용될 수 있을 것으로 기대된다.

  • In this study, the researcher studied the historical background, and the traditional culture about dress and ornament of Yunnan Province of China, The results of the study are as follows. 1. Dress and personal ornaments of the Va peoples vary with the locality. Their traditional dress and adornment is characterized by those in the Ximeng area. Men usually wear black or dark blue collarless jackets and black and dark blue loose and short bagged trousers with folding waist. Women usually wear close-fitting sleeveless pullover blouses with V-shaped necks and straight skirts with patterns of red and black cross stripes. 2. Jingpo men have changed to wear shirts with button down the front and trousers. They also entwine white turbans with red bobbles on both ends, and carry diagonally long knives, firelocks and red woolen figured satchels on their shoulders. Women usually wear black velvet blouses with silver bowl-shaped ornaments and chains around collars and on the fronts. They also wear red straight skirts with overlapped slit on the right, waistbands and waist hoops made of rattan and bamboo. 3. The Naxi nationality has a long history and excellent traditional culture. In modern times, women like to wear red, blue or purple laced blouses, long double-layered pleated skirts, waistbands and embroidered shoes. They wear their hair in buns with either hats or kerchiefs over them. While working or going out, they put on their "seven-star" capes made of sheepskin and embroidered with two big circles and seven small ones, while is a symbol of their frog totem. 4. The dress and adorment of the Jinuo people is simple, elegant and has its own unique characteristics. Men usually wear white buttonless shirts with round necks and an opening on the front, knee-length bagged trousers and legging. They wear cloth turbans, earrings and also put small bamboo or silver pipes in the holes of their earlobes. Women wear short buttonless blouses with round necks and seven coloured stripes and thin tight-fitting or embroidered triangular underwear. 5. The dress and ardorment of the Benglong (De' ang) nationality has its own strong national colour. Most of the men wear jackets with buttons arranged diagonally on the front, loose, short trousers and black or white turbans. Some young men like to wear eardrops and silver necklaces. Women's dress and adornment differs according to various branches. For example, the women of the Bielie and Liang branches have their hair shaved and wear black turbans. They use large square silver tablets as buttons and wear blue or black blouses with buttons down the front. 6. Oai men usually wear trousers, white or blue cloth turbans and round-necked shirts with buttons down the front or arranged diagonally on the front. Women usually wear long straight skirt and blouses. But dress and adornment varies in regions. 7. The Bai nationality dress and adornment has unique national style. The dress fabrics are mainly cotton cloth, silk and velvet. Men usually wear red velvet vests over white shirts with buttons down the front or black velvet vests over light blue shirts. They also wear white of blue turbans and carry satchels with beautiful embroidered designs over their shoulders. Women usually wear red velvet vests over white blouses, or black vests over light-coloured blouses.

경반층 토양인 장원통의 미세형태학적 및 광물학적 특성 (Micromorphological and Mineral Characteristics of the Jang-won Series which have Fragipan in the soil Profile)

  • 본 연구지역인 장원통은 자연 경반층을 함유하는 토양으로 명시되어 있다. 따라서, 장원통의 물리 화학적, 광물학적 그리고 미세형태학적 분석을 통해 경반층의 생성원인을 규명하고자 하였다. 토양의 층위별로 토양을 채취하여 물리화학적 광물학적 특성을 분석하였다. 토성분석은 피펫법으로 측정하였다. 점토광물은 $<2{\mu}m$ 이하로 분리한 후 정방위법으로 X-선 회절기를 이용하여 동정하였다. 미세형태학적 특성을 살펴보기 위해 불교란 시료를 채취하여 0.03mm 이하로 박편을 제작한 후 편광현미경을 이용하여 분석하였다. 장원통 시료에 대한 물리 화학적 특성을 분석한 결과, 토양조성은 모든 층위가 미사질 양토로 도시되었다. 점토의 함량이 약 19.3-20.6% 범위이고, 미사는 약 58.3-61.5%로 미사의 함량이 높게 분포하고 있으며 상부보다는 하부로 내려갈수록 미사의 함량이 더 높게 분포하였다. 표토의 pH는 4.9-4.8로 우리나라 밭 토양의 평균치 5.59 보다 낮게 분포 하고 있었다. 점토광물은 일라이트, 카올리나이트, 녹니석 그리고 질석이 동정되었다. 본 지역의 미세형태학적 특성을 관찰하기 위해 박편을 제작하여 편광현미경 하에서 박편을 관찰한 결과, 사장석과 석영반정이 주위에 미사크기의 각괴상 토양 입자가 우상-좌상방향으로 미약한 층을 이루고 있으나 기질과 경계가 명확하고 토양 기질 간에 미사 크기의 띠를 관찰할 수 있었다. 점토광물은 점토 집적층에서 공극을 채우는 형태로 존재하는 것이 아니라 일차광물에서 변질된 이차광물로 존재하고 있다. 따라서 장원통의 경반층은 점토집적 보다는 미사집적에 의한 영향이 더 크다고 볼 수 있다. 따라서 현재의 토양통 설명서에서 분류된 Typic Fragiochrepts의 해석이 올바르다고 판단된다.

  • 장군광산산 보울란저라이트는 침상이나 불규칙한 형태로 방연석, 차골석, 함은 사면동석과 밀접하게 공생하여 남광상에서 산출되며, 특히 남광상 A광체와 B광체 주변의 능망간석대에 그 산출이 뚜렷하다. 어떤 장소, 특히 남광상 B광체 상부에는 방연석, 황철석, 유비철석, 함망간 방해석, 석영 등과 공생하여 작은 정동중에 "머리털"이나 "깃털"모양의 보울 란저라이트 집합체를 형성하기도 한다. 이광물의 반사색은 녹회색을 띠고, 반사다색성은 차골석보다 강하고 이방성이 명료하다. 반사율은 공기중에서 파장이 560nm 일 때 $R_{max.}=42.3%$, $R_{min.}=35.7%$이고, 비커스경도 (VHN)는 50g의 하중에서 $146{\sim}173kg/mm^2$이다. 표준시약 (Short, 1941)에 의한 반응에서는 HN03에서는 즉시 흑색으로 변하지만, 그외의 시약과는 거의 반응하지 않는다. 8개 시료 23개 입자에 대하여 EPMA로 분석한 결과 $Pb_{56.1}Sb_{25.1}S_{18.5}$, Total 99.6 wt.%이고, S= 11로해 계산된 화학식은 $Pb_{5.16}Sb_{3.94}S_{11}$로서 거의 보울란저라이트의 이상적인 화학식 $Pb_5Sb_4S_{11}$을 만족하고 있다. X-선 회절분석에 의해 얻어진 X-선 회절패턴에 나타나는 주요한 회절선은 $3.73\;{\AA}\;(10)$, $3.22\;{\AA}\;(5)$, $3.03\;{\AA}\;(4)$, $2.82\;{\AA}\;(5)$로서 공간군 $C^5_{2h}-P2_{1/a}$ 과 잘 일치한다. 이상의 장군광산산 보울란저라이트는 그 산출상태와 광물의 공생관계로부터 열수성 연-아연-은광화작용의 최후기에 생성된 광물로 판단된다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm.
