Primer RNA Synthesis by E. coli RNA Polymerase on the SSB-coated 229-nt ssi Signal of Lactococcal Plasmid pGKV21 (Lactococcal plasmid pGKV21의 SSB-coated 229-nt ssi signal 상에서 E. coli RNA polymerase에 의한 시발체 RNA 합성)

  • Plasmid pGKV21 contains a 229-nucleotide (nt) single-strand DNA initiation (ssi) signal. Using asymmetric PCR, we prepared a small single-stranded (ss) DNA fragment of the ssi signal and, using the 229-nt ssDNA fragment, determined the requirements of RNA polymerase for priming and DNA-protein interaction. The ssi fragment prepared was able to generate primer RNAs with almost the same efficiency as the $M13{\Delta}lac182/229$ phage DNA. However, the cssi (complementary strand of the ssi signal) fragment could not synthesize primer RNAs. This result suggests that the 229-nt ssi signal functions in a strand specific manner. Gel retardation and DNase I footprinting demonstrated that the synthesized ssi fragment could interact with both E. coli RNA polymerase and SSB protein to synthesize primer RNA. In Escherichia coli [pWVAp], an addition of rifampicin resulted in an accumulation of ssDNA, indicating that the host-encoded RNA polymerase is involved in the conversion of ssDNA to double-stranded plasmid DNA.

  • The phosphorylation of extracellular signal-regulated kinases (p-ERK) in the spinal cord of rats with acute monophasic experimental autoimmune encephalomyelitis (EAE) was studied using immunohistochemistry and treatment with inhibitor. P-ERK is constitutively expressed in glial cells in the normal spinal cord. In EAE, some inflammatory cells in the subarachnoid space were positive for p-ERK at the early stage, and its immunoreactivity declined when those cells infiltrated the parenchyma at the peak stage. In a blocking experiment using its inhibitor, the intravenous administration of PD98059 from day 7 to 13 post-immunization did not modulate EAE paralysis. Considering the results, we postulate that intravenous administration of PD98059 is not effective in ameliorating EAE paralysis, although many inflammatory cells express ERK in the subarachnoid space.

Andrographolide Promotes the Stemness of Epidermal Cells through the Extracellular Signal-regulated Kinase (ERK) Pathway (Andrographolide의 Extracellular Signal-regulated Kinase Pathway (ERK)를 통한 상피 세포 줄기세포능 향상)

  • Andrographolide, the main compound of Andrographis paniculata (A. paniculata), shows various biological properties including anti-viral, anti-inflammatory, anti-diabetic, and hepatoprotective effects. Our previous study has shown that A. paniculata extract exerts antiaging effects by activation of stemness in epidermal stem cells (EpSCs). In this study, we investigated the effect of andrographolide as a main compound of A. paniculata on EpSCs and its mechnism of action using several in vitro assays. Andrographolide increased the proliferation of EpSCs and induced cell cycle progression. Additionally, andrographolide increased VEGF production and the expression of stem cell markers integrin ${\beta}1$ and p63. Furthermore, phosphorylation levels of extracellular signal-regulated kinase 1/2 (ERK1/2), S6 ribosomal protein (S6RP) and Akt were increased by andrographolide. Taken together, these results indicate that andrographolide-induced proliferation of EpSCs is mediated by the ERK1/2, Akt-dependent pathway with increased production of VEGF and upregulated stemness through integrin ${\beta}1$ and p63.

Monitoring of Tool Life through AR Model and Correlation Dimension Analysis (시계열 모델과 상관차원 해석을 통한 공구수명의 감시)

  • Recently, monitoring of tool life is a matter of common interesting because tool life affects precision, productivity and cost in machining process. Especially flank wear has a direct effect on cutting mechanism, so the various pattern of cutting force is obtained experimentally according to variation of wear condition. By investigating cutting force signal, AR(Autoregressive) modeling and correlation dimension analysis is conducted in turning operation. In this modeling and analysis, we extract features through 6th AR model, correlation integral and normalized correlation integral. After the back-propagation model of the neural network is utilized to monitor tool life according to flank wear. As a result. a very reliable classification of tool life was obtained.

  • The experimental investigation is mainly focused to detect tool wear by cutting signal measurements in multi-insert face milling SS 41 and STS 304. This research have investigated the effects on the insert number, which has relationship with mean-cutting force. AE(acoustic emission) signal, tool life and surface roughness in machining SS41 and STS 304. The cutting force and AE signal are monitored to analyse the cutting process, The surface roughness of the specimens machined by TiN coated tool with the various insert numbers measured at various cutting speeds, feed rates and depths of cut, The width of flank wear is also observed.

Modulation of Cardiac ATP-Sensitive $K^+$ Channels Via Signal Transduction Mechanisms During Ischemic Preconditioning

  • In several species, a short period of ischemic preconditioning protects the heart by reducing the size of infarcts resulting from subsequent prolonged bouts of ischemia. The mechanism by which activation of ATP-sensitive $K^+$($K_ATP$) channels could provide the memory associated with ischemic preconditioning is still under debate. Several signal transduction pathways have been implicated in the mechanisms of protection induced by ischemic preconditioning. The exact receptor-coupled pathways involved in preconditioning remain to be identified. Likely extracellular agonists are those whose circulating levels increase under conditions that activate $K_ATP$ channels; these conditions include ischemia and ischemic preconditioning. Potential physiological agonists include the following: (1) nitric oxide; (2) catecholamine; (3) adenosine; (4) acetylcholine; (5) bradykinin and (6) prostacycline. The purpose of this review was to understand the mechanism by which biological signal transduction mechanism acts as a link in one or more known receptor-mediated pathways to increase $K_ATP$ channel activity during ischemic preconditioning.

  • In this paper we investigated the method of examining the accelerated life test on LED traffic signal module by the temperature. The longevity presumption of the LED type signal light by a general heat cycle test used and executed cycle when it was done to longevity by the heat cycle test number and the acceleration factor of a real system requirements of this heat cycle test. Therefore, it reports on the introduction of the acceleration type from which the LED traffic signal module is done here to clarify the above-mentioned acceleration factor with the object and the acceleration factor is requested the test atmosphere actually in the system requirements.

  • Heart related diseases mainly caused by heavy work load and increasing stress in human daily life. Therefore, researches on mobile healthcare monitoring for daily life has been carried out. Notably, wearable healthcare monitoring system which has least restriction has been tried to provide an emergency alert of abnormal heart rate. In this study, we developed chair type unconstrained BCG measurement system which able to perform continuous heart status monitoring at the office and daily life in the unconstrained way. Furthermore, adaptive threshold is used to detect the heart rate from BCG signals. The HRV(heart rate variability) is calculated from heart rate interval. ECG signal measured using conventional method and BCG signal measured using unconstraint system are carried out simultaneously for the purpose of performance evaluation. From the comparison result, BCG signal shows a similar heart beat characteristic as ECG signal. This proves the possibility of practical implementation of unconstraint healthcare monitoring system. In addition, medical examination like valsalva maneuver is performed to observe the changes in HRV due to stress. By performing valsalva maneuver, heart is said to be placed under an artificial physical stress condition. Under this artificial physical stress condition, the time and frequency domain of HRV parameters are evaluated.

  • E. intermedium Blocontrol activity of a P. chlororaphis rhizobacteium O6, depends to the synthesis of extracellular secondary metabolites and exoenzymes, thought to antagonize the pathogenicity of a variety of phytopathogenic fungi. The production of secondary metabolites and exoenzymes in O6, depends essentially on the GacS-mediated signal transduction pathway, which activates largely unknown signal transduction pathway. To exploit the GacS-mediated signal transdcution pathway involved in activation of ph genes that are necessary for biosynthesis of phenazine from P. chlororaphis O6, we cloned and sequenced the phz operon, rpoS gene encoding stationary specific sigma factor, ppx gene encoding polyphosphatase, and lon gene encoding ion protease. Expression of each gene in wild type and GacS mutant were analyzed by RT-PCR. Transcripts from rpoS, phzI enconing acylhomoserine lactone (AHL) synthase, and ph structural genes in the GacS mutant were reduced in each of these growth phases compared to the wild type. The GacS or Lon mutant was found to be deficient in the production of phenzines, exoenzymes, and the acylhomoserine lactone. These mutants were not complemented by ph operon and addition of exogenous AHL. These results indicate that the GacS global regulatory systems controls phenazine production at multiple levels. Future research will focus to identifying the GacS-mediated regulatory cascade involving in production of phenazine in P. chlororaphis.
