Construction of a High-Quality Yeast Two-Hybrid Library and Its Application in Identification of Interacting Proteins with Brn1 in Curvularia lunata

  • Curvularia lunata is an important maize foliar fungal pathogen that distributes widely in maize growing area in China, and several key pathogenic factors have been isolated. An yeast two-hybrid (Y2H) library is a very useful platform to further unravel novel pathogenic factors in C. lunata. To construct a high-quality full length-expression cDNA library from the C. lunata for application to pathogenesis-related protein-protein interaction screening, total RNA was extracted. The SMART (Switching Mechanism At 5' end of the RNA Transcript) technique was used for cDNA synthesis. Double-stranded cDNA was ligated into the pGADT7-Rec vector with Herring Testes Carrier DNA using homologous recombination method. The ligation mixture was transformed into competent yeast AH109 cells to construct the primary cDNA library. Eventually, a high qualitative library was successfully established according to an evaluation on quality. The transformation efficiency was about $6.39{\times}10^5$ transformants/$3{\mu}g$ pGADT7-Rec. The titer of the primary cDNA library was $2.5{\times}10^8cfu/mL$. The numbers for the cDNA library was $2.46{\times}10^5$. Randomly picked clones show that the recombination rate was 88.24%. Gel electrophoresis results indicated that the fragments ranged from 0.4 kb to 3.0 kb. Melanin synthesis protein Brn1 (1,3,8-hydroxynaphthalene reductase) was used as a "bait" to test the sufficiency of the Y2H library. As a result, a cDNA clone encoding VelB protein that was known to be involved in the regulation of diverse cellular processes, including control of secondary metabolism containing melanin and toxin production in many filamentous fungi was identified. Further study on the exact role of the VelB gene is underway.

Effects of Field-Grown Genetically Modified Zoysia Grass on Bacterial Community Structure

  • Herbicide-tolerant Zoysia grass has been previously developed through Agrobacterium-mediated transformation. We investigated the effects of genetically modified (GM) Zoysia grass and the associated herbicide application on bacterial community structure by using culture-independent approaches. To assess the possible horizontal gene transfer (HGT) of transgenic DNA to soil microorganisms, total soil DNAs were amplified by PCR with two primer sets for the bar and hpt genes, which were introduced into the GM Zoysia grass by a callus-type transformation. The transgenic genes were not detected from the total genomic DNAs extracted from 1.5 g of each rhizosphere soils of GM and non-GM Zoysia grasses. The structures and diversities of the bacterial communities in rhizosphere soils of GM and non-GM Zoysia grasses were investigated by constructing 16S rDNA clone libraries. Classifier, provided in the RDP II, assigned 100 clones in the 16S rRNA gene sequences library into 11 bacterial phyla. The most abundant phyla in both clone libraries were Acidobacteria and Proteobacteria. The bacterial diversity of the GM clone library was lower than that of the non- GM library. The former contained four phyla, whereas the latter had seven phyla. Phylogenetic trees were constructed to confirm these results. Phylogenetic analyses of the two clone libraries revealed considerable difference from each other. The significance of difference between clone libraries was examined with LIBSHUFF statistics. LIBSHUFF analysis revealed that the two clone libraries differed significantly (P<0.025), suggesting alterations in the composition of the microbial community associated with GM Zoysia grass.

연속적 전시공간 개념으로 본 서울대 뮤지엄 내 연결 공간의 역할과 특성에 관한 연구 (A Study on the Role and Characteristics of Middle Space in Museum of Seoul National University based on the Concept of Sequential Exhibition Space)

  • The purpose of this study is to analyze the spatial transformational patterns according to the spectator and the exhibition environment and to examine the role of 'the middle space' organically linking the exhibition space with MoA which is a representative example of 'sequential exhibition space'. The concept of sequential exhibition space in MoA is applied not only to building but also to extended continental concepts including neighborhood. MoA's exhibition space consists of the library area and the exhibition area. In particular, the spaces of library area perform a complex function as a quasi-exhibition space, and the exhibition spaces have variable spatial transformation through an associated structure of the library area and the educational area. The types of exhibition space those constitute the sequential exhibition space of MoA appear as 'Room type', 'Free plan type', and 'Gallery type'. Each exhibition space maintains independence, but it is changed according to the situation of exhibition. The flow is based on the spiral induction around the central void space, but with selective circulation considering the visitors. Therefore, through the transformation of the exhibition area and the division of the flow, the whole museum space appears as the area differentiation.

SWOT 분석을 통한 도서관의 경쟁력 제고 방안에 관한 연구 - A대학교 도서관의 사례를 중심으로 - (A Study on the Libraries' Competitiveness Reinforcement Measures through SWOT Analysis : Based on the Case Study of University A's Library)

  • 도서관이 보다 효율적으로 운영되기 위해서는 도서관 내부의 강 약점과 외부의 기회 위협 요인 등을 파악하여 미래의 발생가능성을 예측하고, 이에 대응하기 위한 최적의 전략방안을도출하여 활용할 수 있어야 한다. 이에 본 연구는 전략경쟁분석기법의 하나로 각광을 받고 있는 SWOT 분석을 통하여 A대학교 도서관의 전략경쟁을 분석하였다. 본 연구를 통하여 얻어진 결론은 다음과 같다. 첫째, A대학교 도서관의 강점은 도서관 직원의 전문성 체계적인 업무추진 계획 및 분장 디지털도서관 구축에 따른 고품질의 서비스 제공이다. 둘째, A대학교 도서관의 약점은 가용자원(인력, 예산, 공간)의 부족, 미래의 도서관 운영계획 부재, 능동적인 서비스 마인드의 부족이다. 셋째 A대학교 도서관의 기회는 디지털 정보환경에 따른 새로운 도약, 외부기관 및 도서관과의 교류협력 확대, 도서관 개방을 통한 지역거점 도서관으로의 위상 제고이다. 넷째, A대학교 도서관의 위협은 학교 당국의 도서관에 대한 인식 부족, 급변하는 정보환경에 따른 이용자의 다양한 정보요구, 강화된 저작권법에 따른 도서관 서비스의 제한이다. 다섯째, A대학교 도서관의 경쟁력 강화방안은 디지털 전문 인력의 확보를 통한 디지털 서비스의 강화, 외부기관 및 도서관과의 교류협력 강화를 통한 확장 서비스의 제공. 강화된 저작권법에 따른 차별화된 서비스의 개발이다.

SVE에서 절대위치를 갖는 오브젝트에 변형의 적용 (Applying Transformation to the Absolutely Positioned Object in SVE)

  • The Simple Virtual Environment (SVE) library is the application programming interface fur creating the virtual environment easily. Even though it has a lot of efficient features, applying transformation to absolutely positioned objects is difficult. In this paper, we designed a simple method with which it is possible to rotate the absolutely positioned object freely. To test the method, we designed a simple virtual environment. The environment is designed with people, street and building objects. To reduce time consumption of displaying high-detailed people models, we only used texture-based objects. The person objects are modeled as quadric-shape and textured with front-view images of people. To make more realistic environment, objects always change its orientation themselves following to the user location. The result and the testing environment will be demonstrated.

A recursive approach for mechanical system design sensitivity analysis

  • Recursive formulas have been effective in solving the equations of motion for large scale constratined mechanical sys-tems. However, derivation of the formulas has been limited to individual terms in the equations of motion, such as veloci-ty, acceleration. and generalized forces. The recursive formulas are generalized in this paper. The velocity transformation method is employed to transform the equations of motion from Cartesian to the joint spaces. Computational structure of the equations of motion in the joint space is carefully examined to classify all necessary computational operations into sev-eral categories. The generalized recursive formula for each category is then developed and applied whenever such a cate-gory of computation is encountered. Since the velocity transformation method yields the equations of motion in a compact form and computational efficiency is achieved by generalized recursive formulas, the proposed method is not only easy to implement but is also efficient. A library of generalized recursive formulas is developed to implement a dynamic analysis algorithm using backward difference.

용접판 구조물의 Feature 기반 모델링

  • Developed in this paper is a feature based modeling of welded plate construction(WPC) which is composed of flat or bended plates represented as reference plane with a constant thickness. First, the necessity and the characteristics of the feature based modeling of WPC as compared with the assemblies of mechanical parts are investigated. Second, feature library for the assemblied of WPC is shown which contains several types of features like joint feature, groove feature, materialfeature and precision feature. Third, the assembly procedures are presented whichmainly consist of both the assembly transformation and the correct assembly checking. Fourth, weld lines generated in the assembled WPC are defined so that those can be used in the process planning or the manufacturing stage. Finally, A prototype system isdeveloped by using a geometric modeling software Pro/Engineer, a graphic software GL(Graphic Library), and C language on a CAD workstation IRIS.

Cloning of Genomic DNAs of Trametes versicolor Acting as Autonomously Replicating Sequences in Saccharomyces cerevisiae

  • A genomic DNA library of the fungus Trametes versicolor was constructed in a yeast integration vector which contains the URA3 gene of the budding yeast Saccharomyces cerevisiae and the gene responsible for hygromycin B resistance, and fragments acting as autonomously replicating sequences (ARSes) in the budding yeast were identified from the genomic DNA library. Sixteen recombinant plasmids from the library transformed the budding yeast Saccharomyces cerevisiae to Ura+ at high frequencies. They were maintained stably under selective conditions, but were gradually lost from yeast cells at different rates under nonselective conditions, indicating that they contain eukaryotic origins of DNA replication and exist as extrachromosomal plasmids. Base sequences of four ARS DNAs among the 16 cloned fragments revealed that all or the four contain at least one 11 bp [(A/T)TTTA(T/C)(A/G)TTT(A/T)]consensus sequence of the budding yeast ARS.

네트웍환경에서 내용관리로의 패러다임 변화에 관한 연구 (A Study on the Paradigm Shift to Content Management in the Networked Environment)

  • 본고에서는 네트웍 환경에서 매체변화와 장서개발 이론의 향후관계에 대해서 연구하였다. 도서관에서, 취급하고 있는 정보가 인쇄매체에서 전자매체 중심으로 비중이 커짐에 따라서 장서개발전략이 물리적 대상이나, 배가, 보관등 형식 중심의 전통적인 장서관리 개념에서 벗어나서, 다양한 매체가 담고 있는 메시지인 '내용(content)' 을 관리하는 방향으로 전환되어야 할 필요성을 제시하였다. 도서관이 '내용'을 가지고 있는 '패키지(package)'로서 간주되어 왔던 기존의 기능으로는 이제 다양한 정보욕구를 가지고 도서관에 접근하는 이용자 중심의 정보제공을 충족시켜주기에는 한계가 있다. 따라서 미래의 장서개발은 기존의 장서 관리 (collection management)에서 매체가 담고 있는 내용중심의 내용관리(content management)로의 패러다임 전환이 필요함을 규명하였다.

Molecular Genetics of the Model Legume Medicago truncatula

  • Medicago truncatula is a diploid legume plant related to the forage crop alfalfa. Recently, it has been chosen as a model species for genomic studies due to its small genome, self-fertility, short generation time, and high transformation efficiency. M. truncatula engages in symbiosis with nitrogen-fixing soil bacterium Rhizobium meliloti. M. truncatula mutants that are defective in nodulation and developmental processes have been generated. Some of these mutants exhibited altered phenotypes in symbiotic responses such as root hair deformation, expression of nodulin genes, and calcium spiking. Thus, the genes controlling these traits are likely to encode functions that are required for Nod-factor signal transduction pathways. To facilitate genome analysis and map-based cloning of symbiotic genes, a bacterial artificial chromosome library was constructed. An efficient polymerase chain reaction-based screening of the library was devised to fasten physical mapping of specific genomic regions. As a genomics approach, comparative mapping revealed high levels of macro- and microsynteny between M. truncatula and other legume genomes. Expressed sequence tags and microarray profiles reflecting the genetic and biochemical events associated with the development and environmental interactions of M. truncatula are assembled in the databases. Together, these genomics programs will help enrich our understanding of the legume biology.
