Purpose: This study aimed to investigate the prognostic value of lymph node ratio (LNR) in patients with locally advanced gastric cancer who received neoadjuvant chemotherapy. Materials and Methods: We retrospectively enrolled gastric cancer patients treated with neoadjuvant chemotherapy and curative surgery at the First Affiliated Hospital of Zhejiang University from 2004 to 2015 as the study cohort. Patients with the same inclusion criteria treated in 2016-2017 were enrolled as the validation cohort. Kaplan-Meier curves were assessed using the log-rank test to analyze the differences in overall survival (OS). Multivariate survival analysis was performed using the Cox proportional hazards model. The areas under the receiver operating characteristic curve of ypN and LNR categories for predicting the actual 3-year OS were compared. Results: A total of 265 patients were included in the proposal cohort. The median number of retrieved lymph nodes (rLNs) was 32. The number of positive lymph nodes (pLNs) increased as rLN increased (P=0.037), but the LNR remained relatively constant (P=0.462). The LNR was categorized into 4 groups according to the prognosis: ypNr0, node-negative with rLN>25; ypNr1, node-negative with rLN≤25 or 00.3. In the validation cohort of 43 enrolled patients, there was a clear distinction in OS that significantly (P<0.001) varied depending on the LNR values and LNR was the only independent prognostic factor in multivariate analysis (P<0.001). Conclusions: LNR was an independent prognostic factor for survival of patients with gastric cancer after preoperative chemotherapy and might be an alternative predictor for ypN stage.
Objective: This study was performed to examine whether the porcine glutamic-oxaloacetic transaminase 1 (GOT1) gene has important functions in regulating adipocyte differentiation. Methods: Porcine GOT1 knockout and overexpression vectors were constructed and transfected into the mouse adipogenic 3T3-L1 cells. Lipid droplets levels were measured after 8 days of differentiation. The mechanisms through which GOT1 participated in lipid deposition were examined by measuring the expression of malate dehydrogenase 1 (MDH1) and malic enzyme (ME1) and the cellular nicotinamide adenine dinucleotide phosphate (NADPH) content. Results: GOT1 knockout significantly decreased lipid deposition in the 3T3-L1 cells (p<0.01), whereas GOT1 overexpression significantly increased lipid accumulation (p<0.01). At the same time, GOT1 knockout significantly decreased the NADPH content and the expression of MDH1 and ME1 in the 3T3-L1 cells. Overexpression of GOT1 significantly increased the NADPH content and the expression of MDH1 and ME1, suggesting that GOT1 regulated adipocyte differentiation by altering the NADPH content. Conclusion: The results preliminarily revealed the effector mechanisms of GOT1 in regulating adipose differentiation. Thus, a theoretical basis is provided for improving the quality of pork and studies on diseases associated with lipid metabolism.
In order to investigate the application of 3D base-seismic isolation system in nuclear power plants (NPPs), comprehensive analysis of constitution and design theory for 3-dimensional combined isolation bearing (3D-CIB) was presented and derived. Four different vertical stiffness of 3D-CIB was designed to isolate the nuclear island (NI) building. This paper aimed at investigating the isolation effectiveness of 3D-CIB through modal analysis and dynamic time-history analysis. Numerical results in terms of dynamic response of 3D-CIB, relative displacement response, acceleration and floor response spectra (FRS) of the superstructure were compared to validate the reliability of 3D-CIB in mitigating seismic response. The results showed that 3D-CIB can significantly attenuate the horizontal acceleration response, and a fair amount of the vertical acceleration response reduction of the upper structure was still observed. 3D-CIB plays a significant role in reducing the horizontal and vertical FRS, the vertical FRS basically do not vary with the floor height. The smaller the vertical stiffness of 3D-CIB is, the better the vertical isolation effectiveness is, whereas, it will increase the displacement and the rocking effect of superstructure. Although the advantage of 3D-CIB is that the vertical stiffness can be flexibly adjusted, it should be designed by properly accounting for the balance between the isolation effectiveness and displacement control including rocking effect. The results of this study can provide the technical basis and guidance for the application of 3D-CIB to engineering structure.
Unsaturated soil at high suctions is widespread. Many civil engineering projects are related to the hydro-mechanical behavior of unsaturated soils at high suctions, particularly in arid and semiarid areas. To investigate water retention behaviors of nine clayey soils (one is classified as fat clay and the others are classified as lean clay according to the unified soil classification system), the high suction (3.29-286.7 MPa) was imposed on the specimens at zero net stress by the vapor equilibrium technique. In this paper, the effect of void ratio on the water retention behavior at high suction was discussed in detail. Validation data showed that soil types, i.e., different mineralogical compositions, are critical in the soil water retention behavior at a high suction range. Second, the hysteresis behavior at a high suction range is mainly related to the clay content and the specific surface area. And the mechanism of water retention and hysteresis behavior at high suctions was discussed. Moreover, the maximum suction is not a unique value, and it is crucial to determine the maximum suction value accurately, especially for the shear strength prediction at high suctions. If the soil consists of hydrophilic minerals such as montmorillonite and illite, the maximum suction will be lower than 106 kPa. Finally, using the area of hysteresis to quantify the degree of hysteresis at a high suction range is proposed. There was a good correlation between the area of hydraulic hysteresis and the specific surface area.
This study aims to determine the effects of the experiences of ZEPETO platform users on their attitudes and purchase intentions. A survey on 10-20 Korean and Chinese virtual fashion consumers was conducted. Brand experiences were independent variables, and offline purchase was the dependent variable. The results showed that, first, among the four factors of the brand experience, Korea and China have the highest explanatory power. Second, the brand experience of Korean consumers significantly impacted their brand attitude. Additionally, cognitive and emotional experiences of the brand positively impacted their brand attitude. Third, the brand experience of Chinese consumers significantly affected their brand attitude, and among the sub-factors of brand experience, cognitive experience positively affected brand attitude. Fourth, the brand experience of Chinese consumers significantly influenced their offline purchase intention, and among the sub-factors of brand experience, cognitive experience positively influenced their offline purchase intention. Overall, an emotional branding strategy that provides brand and product information must be implemented for the Korean market encouraging consumers to develop a positive and emotional connection with the brand. The Chinese market would benefit from formulating a reliable branding strategy that easily provides a wide range of brand and product information focusing on cognitive experiences.
Buckling failure is a typical slope instability mode that should be paid more attention to. It is difficult to provide systematic guidance for the monitoring and management of such slopes due to unclear mechanism. Here we examine buckling failure as the potential instability mode for a slope above a railway tunnel in southwest China. A comprehensive model test system was developed that can be used to conduct buckling failure experiments. The displacement, stress, and strain of the slope were monitored to document the evolution of buckling failure during the experiment. Monitoring data reveal the deformation and stress characteristics of the slope with different slipping mass thicknesses and under different top loads. The test results show that the slipping mass is the main subject of the top load and is the key object of monitoring. Displacement and stress precede buckling failure, so maybe useful predictors of impending failure. However, the response of the stress variation is earlier than displacement variation during the failure process. It is also necessary to monitor the bedrock near the slip face because its stress evolution plays an important role in the early prediction of instability. The position near the slope foot is most prone to buckling failure, so it should be closely monitored.
Objective: With the rapid development of proteomics sequencing and RNA sequencing technology, multi-omics analysis has become a current research hotspot. Our previous study indicated that Xinjiang brown cattle have better meat quality than Kazakh cattle. In this study, Xinjiang brown cattle and Kazakh cattle were used as the research objects. Methods: Proteome sequencing and RNA sequencing technology were used to analyze the proteome and transcriptome of the longissimus dorsi muscle of the two breeds of adult steers (n = 3). Results: In this project, 22,677 transcripts and 1,874 proteins were identified through quantitative analysis of the transcriptome and proteome. By comparing the identified transcriptome and proteome, we found that 1,737 genes were identified at both the transcriptome and proteome levels. The results of the study revealed 12 differentially expressed genes and proteins: troponin I1, crystallin alpha B, cysteine, and glycine rich protein 3, phosphotriesterase-related, myosin-binding protein H, glutathione s-transferase mu 3, myosin light chain 3, nidogen 2, dihydropyrimidinase like 2, glutamate-oxaloacetic transaminase 1, receptor accessory protein 5, and aspartoacylase. We performed functional enrichment of these differentially expressed genes and proteins. The Kyoto encyclopedia of genes and genomes results showed that these differentially expressed genes and proteins are enriched in the fatty acid degradation and histidine metabolism signaling pathways. We performed parallel reaction monitoring (PRM) verification of the differentially expressed proteins, and the PRM results were consistent with the sequencing results. Conclusion: Our study provided and identified the differentially expressed genes and proteins. In addition, identifying functional genes and proteins with important breeding value will provide genetic resources and technical support for the breeding and industrialization of new genetically modified beef cattle breeds.
There are various technical problems need to be solved in the construction process of pre-setting an isolation wall into a double lane in the outburst prone mine. This study presents a methodology that pre-setting an isolation wall into a double lane without a coal pillar. This requires the excavation of two small section roadways to dig a wide section roadway, followed by construction of the separation wall. During this process the connecting lane is reserved. In order to ensure the stability of the separation wall, the required bearing capacity of the isolation wall is 4.66 MN/m and the deformation of the isolation wall is approximately 25 cm. To reduce the difficulty of implementing support the roadway is driven by 5 m/d. After the construction of the separation wall, the left side coal wall is brushed 1.5 m to make the width of the gas roadway reach 2.5 m and the roadway support utilizes anchor rod, ladder beam, anchor cable beam and net configuration. During construction, the concrete pump and removable self-propelled hydraulic wall mold are used to pump and pour the concrete of the isolation wall. In the process of mining, the stress distribution of coal body and isolation wall is detected and measured on site. The results demonstrate that the deformation of the surrounding rock of roadway and separation of roof in the roadway is small. The stress of the bolt and anchor cable is within equipment tolerance validating their selection. The roadway is well supported and the intended goal is achieved. The methodology can be used for reference for similar mine gas control.
Inevitable response loss under complex operational conditions significantly affects the integrity and quality of measured data, leading the structural health monitoring (SHM) ineffective. To remedy the impact of data loss, a common way is to transfer the recorded response of available measure point to where the data loss occurred by establishing the response mapping from measured data. However, the current research has yet addressed the structural condition changes afterward and response mapping learning from a small sample. So, this paper proposes a novel data driven structural response reconstruction method based on a sophisticated designed generating adversarial network (UAGAN). Advanced deep learning techniques including U-shaped dense blocks, self-attention and a customized loss function are specialized and embedded in UAGAN to improve the universal and representative features extraction and generalized responses mapping establishment. In numerical validation, UAGAN efficiently and accurately captures the distinguished features of structural response from only 40 training samples of the intact structure. Besides, the established response mapping is universal, which effectively reconstructs responses of the structure suffered up to 10% random stiffness reduction or structural damage. In the experimental validation, UAGAN is trained with ambient response and applied to reconstruct response measured under earthquake. The reconstruction losses of response in the time and frequency domains reached 16% and 17%, that is better than the previous research, demonstrating the leading performance of the sophisticated designed network. In addition, the identified modal parameters from reconstructed and the corresponding true responses are highly consistent indicates that the proposed UAGAN is very potential to be applied to practical civil engineering.
Guaranteeing the quality and integrity of structural health monitoring (SHM) data is very important for an effective assessment of structural condition. However, sensory system may malfunction due to sensor fault or harsh operational environment, resulting in multiple types of data anomaly existing in the measured data. Efficiently and automatically identifying anomalies from the vast amounts of measured data is significant for assessing the structural conditions and early warning for structural failure in SHM. The major challenges of current automated data anomaly detection methods are the imbalance of dataset categories. In terms of the feature of actual anomalous data, this paper proposes a data anomaly detection method based on data-level and deep learning technique for SHM of civil engineering structures. The proposed method consists of a data balancing phase to prepare a comprehensive training dataset based on data-level technique, and an anomaly detection phase based on a sophisticatedly designed network. The advanced densely connected convolutional network (DenseNet) and Transformer encoder are embedded in the specific network to facilitate extraction of both detail and global features of response data, and to establish the mapping between the highest level of abstractive features and data anomaly class. Numerical studies on a steel frame model are conducted to evaluate the performance and noise immunity of using the proposed network for data anomaly detection. The applicability of the proposed method for data anomaly classification is validated with the measured data of a practical supertall structure. The proposed method presents a remarkable performance on data anomaly detection, which reaches a 95.7% overall accuracy with practical engineering structural monitoring data, which demonstrates the effectiveness of data balancing and the robust classification capability of the proposed network.
