A Study on the Depaminergic Control of T.S.H. Secretion (갑상선자극(甲狀腺刺戟) 호르몬 분비(分泌)에 대(對)한 Dopaminergic Contrl에 관(關)한 연구(硏究))

  • To elucidate the depaminergic control of T.S.H. secretion, we analized the pattern of T.S.H. secretion in seven normal controls and nine primary hypothyroid subjects, before and after single or combined administration of specific dopaminergic,receptor blocker, metoclopramide, and specific depaminergic receptor stimulant, bromergocryptine(CB-154). The results obtained were as follows: 1) There was a significant rise in T.S.H. levels after intra venous injection of metocloramide (10mg) in hypothyroid subjects. But there was no significant rise in T.S.H. levels in normal controls. The T.S.H. response to metoclopramide varied considerably, being large in mild cases and small in severely hypothyroid subjects. 2) There was a significant fall in T.S.H. levels after oral administration of bromergocryptine (2mg) in hypothyroid subjects, but there was no significant fall in T.S.H. levels in normal controls. 3) There was no significant fluctuation in T.S.H. levels after combined administration of both metoclopramide and bromergocrytine. 4) There was no significant fluctuation in T.S.H. levels after intravenous injection of normal saline(2ml) in both hypothyroid and normal subjects. 5) There was no significant change in serum $T_3\;and\;T_4$ after administration of metoclopramide and and bromergocryptine respectively and serially. These data support the fact that there is a dopaminergic control in the secretion of T.S.H. in the human.

Effects of Herbal Product (Miracle®) on the Performance, Nutrient Digestibility, Fecal Microflora and Immunoresponse in Weanling Pigs (생약제제(미라클®)첨가가 이유 자돈의 성장률, 영양소 이용율, 분내 미생물균총 및 면역기능에 미치는 영향)

  • An experiment was conducted to investigate the effects of dietary herbal product(Miracle$^{\circledR}$) supplementation on the performance, nutrient digestibility, fecal microflora and blood parameters in pigs. Forty eight cross bred(Y${\times}$L${\times}$D) weanling pigs (average initial body weight 7.84kg${\pm}$0.17, 28 d old) were used in 35 d feeding trial. Pigs were allotted to 16 raised floor cages(W 35.5cm${\times}$L45cm${\times}$H 55cm). Each treatment had four replications of 3 pigs each. Treatments were: control (T1), CTC 100ppm (T2), Miracle$^{\circledR}$ 0.15% (T3) and CTC 100ppm + Miracle$^{\circledR}$ 0.15% (T4). The herbal product supplementation(T3) tended to improve the weight gain, feed intake, feed/gain but there were no significant differences among treatments. Nutrient digestibility was significantly(P〈0.05) affected by dietary treatments. The pigs fed T3 diet showed the lowest digestibility of DM, crude protein, NFE, P, and amino acids among treatments. Serum cholesterol level of pigs fed T3 diet was lowest and significantly(P〈0.05) lower than those fed antibiotic supplemented diet(T2). Triglyceride and HDL level of the pigs fed T3 diet were also lower than those of others but the differences were not significant. The level of serum IgG was significantly(P〈0.05) different among treatments. The level of serum IgG was highest in T4, followed by T1, T2 and T3. Serum AST(Aspartate aminotransferase) level of the control(T1) was lower than other treatments. Levels of serum total protein, albumin and ALT(Alanine aminotransferase) were not significantly different among treatments. The colony forming units (CFU) of Lactobacillus, Cl. perfringens and E.coli in feces were not significantly different among treatments but those of Cl. perfringens of T2 (CTC 100ppm) showed the lowest count at both 3rd and 5th wk. It is concluded that herbal product Miracle$^{\circledR}$ does not significantly affect growth performance of weanling pigs but it influenced the digestibility, serum IgG, cholesterol and AST level. The combination of the herbal product Miracle$^{\circledR}$ with antibiotic(CTC) showed no synergistic effects.

A Comparative Study on the Etfects of 『Palmisoyo-San gamibang』, 『Gyebu-Tang』 and 『Boganyangje』 on the Thyrotoxicosis of Rats (갑상선중독유발 흰쥐에 미치는 팔미소요산가미방(八味消遙散加味方)과 계부탕(桂附湯)및 보간양제(補肝陽劑)의 영향 비교 연구)

  • In order to study comparatively the effects of "Palmisoyo-San gamibang(八味消遙散加味方)"(AP), "Gyebu-Tang(桂附湯)"(GT) and "Boganyangje(補肝陽劑)" (BG) on the thyrotoxicosis rats, we have made thyrotoxicosis rat model by administration sodium levothyroxine p.o. during 4 days(Control group), and have administered solid extract of AP(236.8mg/200g/day : Sample I group), GT(80mg/200g/day : Sample II group) and BG(70.4mg/200g/day : Sample III group) p.o. to thyrotoxicosis rats during 3 days from 3rd day. We measured the body weight(BW), body temperature(BT), levels of Serum $T_3{\cdot}T_4{\cdot}free\;T_3{\cdot}free\;T_4$ and TSH after administrating solid extract of AP, GT and BG. The results are summarized as follows: 1. Sample I group showed significantly high increasing rate compared with Control, Sample II and Sample III group, but it showed significantly lower levels of BT, $T_4$ and free $T_4$ than those of Control, Sample II and Sample III. group. 2. We could know that AP suppress the alteration of $T_4$ to $T_3$. 3. Sample II and Sample III group showed opposite results to Sample I group in the BW., BT, $T_4$ and free $T_4$ Especially Sample III was significantly higher than Control, Sample I and Sample II, so we could guess that BG stimulates the formation of thyroid hormones. According to the above experimental results, "Palmisoyo-San\;gamibang(八味消遙散加味方)" is assumed to have a curative effect against the thyrotoxicosis rat induced by sodium levothyroxine, and to suppress the alteration of $T_4$ to $T_3$. And we can suppose that "Boganyangje(補肝陽劑)" has a curative effect against the hypothyroidism.

Effects of Ca Intake Levels on Mg Utilization in Rats of Different Ages (칼슘의 섭취수준이 연령이 다른 흰쥐의 마그네슘 이용에 미치는 영향)

  • The purpose of this study was to investigate the effects of calcium intake levels on magnesium utilization in young and adult rats fed various levels of calcium(50, 100 or 200% of requirement) for 3 weeks. Feed intake, body weight gain, and feed efficiency ratios of the young group were significantly higher than those of the adult group. In calcium levels of tissues, serum calcium levels in young group were higher than in the adult group and kidney calcium levels in HCa(high-calcium group) were higher than in other groups. There was no significant difference in magnesium intake among age and diet groups. However, magnesium excretions through feces and urine in the adult group were higher than in the young group. Therefore, magnesium retention in the young group was higher than in the adult group. Especially in the young group, fecal magnesium excretion was increased with increasing levels of dietary calcium. Serum magnesium levels were lowest in YHCa(young 8t high-calcium) group, and there was no significant difference in liver and kidney levels among the calcium-diet groups. These results indicate that high calcium consumption promotes of fecal magnesium excretion and decreased magnesium retention rates in growing rats. Therefore, it is suggested that mineral interactions should be considered in the supplementation of dietary calcium intake for growth. (Korean J Nutrition 31(9) 1404-1410, 1998)

Assessment of Relationship between Wilms' Tumor Gene (WT1) Expression in Peripheral Blood of Acute Leukemia Patients and Serum IL-12 and C3 Levels

  • Background: Leukemia is a common cancer among children and adolescents. Wilms' tumor gene (WT1) is highly expressed in patients with acute leukemia. It is found as a tumor associated antigen (TAA) in various types of hematopoietic malignancies and can be employed as a useful marker for targeted immunotherapy and monitoring of minimal residual disease (MRD). In this regard, WT1 is a transcription factor that promotes gene activation or repression depending on cellular and promoter context. The purpose of this study was assessment of WT1 gene expression in patients with acute leukemia, measurement of IL-12 and C3 levels in serum and evaluation of the relationship between them. Materials and Methods: We evaluated the expression of WT1 mRNA using real-time quantitative RT-PCR and serum levels of IL-12 and C3 using ELISA and nephelometry in peripheral blood of 12 newly diagnosed patients with acute leukemia and 12 controls. Results: The results of our study showed that the average wT1 gene expression in patients was 7.7 times higher than in healthy controls (P <0.05). In addition, IL-12 (P = 0.003) and C3 (P <0.0001) were significantly decreased in the test group compared to controls. Conclusions: WT1 expression levels are significantly higher in patients compared with control subjects whereas serum levels of interleukin-12 and C3 are significantly lower in patients. Wt1 expression levels in patients are inversely related with serum levels of IL-12 and C3.

Mucosal Immunity Related to FOXP3+ Regulatory T Cells, Th17 Cells and Cytokines in Pediatric Inflammatory Bowel Disease

  • Background: We aimed to investigate mucosal immunity related to forkhead box P3 ($FOXP3^+$) regulatory T (Treg) cells, T helper 17 (Th17) cells and cytokines in pediatric inflammatory bowel disease (IBD). Methods: Mucosal tissues from terminal ileum and colon and serum samples were collected from twelve children with IBD and seven control children. Immunohistochemical staining was done using anti-human FOXP3 and anti-$ROR{\gamma}t$ antibodies. Serum levels of cytokines were analyzed using a multiplex assay covering interleukin $(IL)-1{\beta}$, IL-4, IL-6, IL-10, IL-17A/F, IL-21, IL-22, IL-23, IL-25, IL-31, IL-33, interferon $(IFN)-{\gamma}$, soluble CD40L, and tumor necrosis factor-${\alpha}$. Results: $FOXP3^+$ Treg cells in the lamina propria (LP) of terminal ileum of patients with Crohn's disease were significantly (P < 0.05) higher than those in the healthy controls. $ROR{\gamma}t^+$ T cells of terminal ileum tended to be higher in Crohn's disease than those in the control. In the multiplex assay, serum concentrations (pg/mL) of IL-4 ($9.6{\pm}1.5$ vs. $12.7{\pm}3.0$), IL-21 ($14.9{\pm}1.5$ vs. $26.4{\pm}9.1$), IL-33 ($14.3{\pm}0.9$ vs. $19.1{\pm}5.3$), and $IFN-{\gamma}$ ($15.2{\pm}5.9$ vs. $50.2{\pm}42.4$) were significantly lower in Crohn's disease than those in the control group. However, serum concentration of IL-6 ($119.1{\pm}79.6$ vs. $52.9{\pm}39.1$) was higher in Crohn's disease than that in the control. Serum concentrations of IL-17A ($64.2{\pm}17.2$ vs. $28.3{\pm}10.0$) and IL-22 ($37.5{\pm}8.8$ vs. $27.2{\pm}3.7$) were significantly higher in ulcerative colitis than those in Crohn's disease. Conclusion: Mucosal immunity analysis showed increased $FOXP3^+$ T reg cells in the LP with Crohn's disease while Th17 cell polarizing and signature cytokines were decreased in the serum samples of Crohn's disease but increased in ulcerative colitis.

The Effect of Samilshinkihwan on Immunosuppression Induced in Rats by Methotrexate (삼일신기환(三一腎氣丸)이 methotrexate로 유발(誘發)된 흰쥐의 면역기능저하(免疫機能低下)에 미치는 영향(影響))

  • Objective : To investigate the effect of Samilshinkihwan(SISKW) on white rats with deteriorated immunity caused by methotrexate. Methods : The test articles were dosed once a day for 14 days by gastric gavage at a dosage 1000, 500 and 250mg/kg/10ml of SISKW from 2 days after last MTX-dosing, and changes in body weight, spleen weight, total blood leukocyte numbers, total lymphocyte numbers, B and T lymphocyte ratio, CD3+CD4+, CD3+/CD8+ and CD4+/CD8+ T lymphocyte ratio in the blood and spleen were measured. In addition, the serum interleukin (IL)-2 levels and the productivity of IL-2 of splenic cells were also demonstrated in this study. Results : The changes on body weight increased significantly in the 1000mg/kgof SISKW group. The changes on the spleen weight, the total blood leukocytenumbers, the total lymphocyte numbers in the blood and spleen, the ratio of T-cell in the blood and spleen and the ratio of CD3+CD4+ T-cell in spleen increased significantly in all SISKW groups as compared with the control group. The ratio of CD4+/CD8+ T-cells in blood increased significantly. The serum IL-2 levels and productivity of IL-2 of splenic cells increased significantly in 1000 and 500mg/kg SISKW groups as compared with the control group. Conclusions : Samilshinkihwanhas an effect of increasing immune responses, especially on cellular immune responses, in white rats with deteriorated immunity caused by methotrexate.

Impact of dietary fiber intake on non-alcoholic fatty liver disease risk in Korean patients with obesity and type 2 diabetes mellitus

  • Purpose: Korean patients with type 2 diabetes mellitus (T2DM) and obesity are at a high risk of developing severe non-alcoholic fatty liver disease (NAFLD). This study examined the dietary intakes and compared the risks of NAFLD-related complications in Korean patients with T2DM and obesity. Methods: Data from the Korean National Diabetes Program cohort were used to study patients with T2DM. Two hundred and sixty-five obese patients with T2DM (body mass index ≥ 25 kg/m2) were classified into NAFLD and non-NAFLD groups. The nutrient intake was analyzed using a 24-hour dietary recall questionnaire. Anthropometric and biochemical data were also obtained. Statistical analyses were performed to determine the significant differences between the 2 groups. Results: The serum gamma-glutamyl transpeptidase levels in obese patients with T2DM and NAFLD were significantly higher than in obese T2DM patients without NAFLD (p < 0.05). The serum glucose and lipid profiles showed no significant differences between the NAFLD and non-NAFLD groups. The carbohydrate, protein, and fat levels also did not differ significantly. The results showed that the fiber intake of the NAFLD and non-NAFLD groups was 14.11 ± 3.86 g/100 kcal and 15.70 ± 4.56 g/1,000 kcal, respectively, showing that the dietary fiber intake of the non-NAFLD group was significantly higher (p < 0.05). A correlation was observed between total fiber intake and γ-glutamyl transpeptidase in either patient group. In addition, the odds ratio of developing NAFLD was 0.29× lower when the fiber was consumed at 125% of adequate intake. Conclusions: A higher dietary fiber intake may reduce the risk of NAFLD in obese patients with T2DM. The dietary intake of Korean obese patients with T2DM should include and be enriched in dietary fiber to aid in preventing and treating NAFLD.

Effect of Taraxacum Mongolicum Extract on Serum Heavy Metal Levels and Blood Parameters of Albino Rats Exposed to Lead and Mercury (포공영 추출물이 납 및 수은에 노출된 랫드 혈청의 중금속 및 혈액지표에 미치는 영향)

  • BACKGROUND: The purpose of this study was that serum metabolic and hematological variables were measured to investigate the detoxication effect of Taraxacum mongolicum extract on male albino rat exposed with lead and mercury. METHODS AND RESULTS: For this study, Pb and Hg-exposed( 50 ppm) albino rats was used, and the treatments were carried out in three doses of 100 mg/kg, 200 mg/kg and 500 mg/kg as dry weight of T. mongolicum extract. T. mongolicum extract could improved the body weight gain and feed efficiency ratio, except to food intake. The levels of biochemical factors elevated by Pb-Hg mix exposure, which are Bilirubin, Alkaline phosphatase(ALP), glutamic oxaloacetate transaminase(GOT), glutamic pyruvate transaminase (GPT), lactate dehydrogenase(LDH), ceatinine and blood urea nitrogen(BUN), were significantly reduced in all treated groups as compared to Pb-Hg mix exposure alone. T. mongolicum extract was shown to suppress the accumulation of Hg and Pb in serum by dose dependent manner. CONCLUSION: Therefore, this study suggest that T. mongolicum extract might have the potential effect to minimize the toxic effects of Pb and Hg.

Effect of Deer Antler on the Inhibitory Action of Obese Rats (녹용이 흰쥐의 비만억제에 미치는 영향)

  • Objective: We investigated the effects of deer antler extract (DAE) on the expression of the weight, leptin, and serum levels (lipid metabolism) of the obese SD rats induced by a $2\%$ cholesterol diet in vivo. Methods: In vivo, the weight, leptin and serum levels were measured after a period of 7 weeks for obese SD rats induced by a $2\%$ cholesterol diet and treated with DAE of three concentrations (0.938 g/kg/5ml, 1.875 g/kg/5ml, 3.75 g/kg/5ml). Results: 1. Body weight didn't change significantly. 2. Leptin levels significantly decreased (p<0.05). 3. Total cholesterol, triglyceride, LDH, sGOT & sGPT levels significantly decreased in the sample (p<0.05). 4. The HDL cholesterol levels significantly increased in the sample (p<0.05). 5. The average size of the epididymal fat cells significantly decreased as the concentration of deer antler extract increased (p<0.05). Conclusion: Deer antler effects inhibition of obese rats.
