Korean Curcuma longa L. induces lipolysis and regulates leptin in adipocyte cells and rats

  • BACKGROUND/OBJECTIVES: Turmeric (Curcuma longa L.) has been reported to have many biological functions including anti-obesity. Leptin, peptide hormone produced by adipocytes and its concentration is increased in proportion to the amount of the adipocytes. In the present study, we examined the effects of Korean turmeric on the regulation of adiposity and leptin levels in 3T3-L1 adipocytes and rats fed a high-fat and high-cholesterol diet. MATERIALS/METHODS: Leptin secretion, free fatty acid and glycerol contents in 3T3-L1 adipocytes were measured after incubation of cells with turmeric for 24 hours. Rats were divided into four experimental groups: a normal diet group (N), a high-fat and high-cholesterol diet group (HF), a high-fat and high-cholesterol diet group supplemented with 2.5% turmeric extracts (TPA group) and a high-fat and high-cholesterol diet group supplemented with 5% turmeric extracts (TPB group). Serum samples were used for the measurement of leptin concentration. RESULTS: Contents of free fatty acid and glycerol showed concentration dependent increase in response to turmeric extracts. Effects of turmeric extracts on reduction of lipid accumulation in 3T3-L1 cells were examined by Oil Red O staining. Treatment with turmeric extracts resulted in increased expression levels of adipose triglyceride lipase and hormone-sensitive lipase mRNA. The concentration of leptin from 3T3-L1 adipocytes was significantly decreased by turmeric. Proportional abdominal and epididymal fats weights of the turmeric 5% supplemented group, TPB has significantly decreased compared to the HF group. The serum levels of leptin in the TPA and TPB groups were significantly lower than those of the HF group. CONCLUSIONS: Based on these results, we suggested that Korean turmeric may contribute to the decreasing of body fat and regulating leptin secretion.

The Effects of Vitamin D Supplementation to Peak-producing Hens Fed Diets Differing in Fat Source and Level on Laying Performance, Metabolic Profile, and Egg Quality

  • This experiment was designed to examine the effects of supplemental vitamin D on laying performance, metabolic profile and egg quality of hens fed diets containing different fat sources and levels. Lohman strains (n = 480) were assigned to one of 10 diets: basal diet (BD), BD plus 2.5 and 5.0% sunflower oil (SO) or tallow (T) at vitamin D provided $1{\times}$ and $3{\times}$ of the current recommendation. The experiment lasted from week 30 to 44 of age. Each diet was tested in 12 replicate cages of 4 hens. Production, metabolism, and egg quality data were subjected to three-way ANOVA. Both fats decreased feed intake (FI) as compared to BD. Increasing SO and T levels linearly decreased and quadratically increased FI, respectively. The dietary factors did not affect egg production (EP) and egg weight. Vitamin D supplementation increased and decreased EP when diets contained SO and T, respectively. Feed conversion efficiency (FCE) for hens fed SO was lower than for hens fed T. However, increasing T level improved FCE, whereas increasing SO level worsened FCR. Vitamin D supplementation increased serum vitamin D and glucose concentrations. Vitamin D supplementation also caused a decrease and an increase in serum vitamin D concentration when diets contained SO and T, respectively. Serum glucose concentration for hens fed SO was lower than hens fed T. Increasing fat level linearly increased serum triglyceride and VLDL concentrations, regardless of the fat type. Increasing SO level linearly decreased serum cholesterol concentration. Vitamin D supplementation did not alter lipid metabolites. The dietary factors did not affect serum total protein, Ca, and P concentrations. As compared with BD, feeding SO decreased dry tibia and ash weights more than feeding T. Vitamin D supplementation tended to increase dry tibia weight and decrease tibia ash weight. Eggshell strength and thickness, yolk and albumen indexes, and Haugh unit were not responsive to the dietary factors. Eggshell strength quadratically increased with increasing T level. Yolk color for hens fed SO was lower than for hens fed T. The dietary factors did not affect most of yolk fatty acids. Increasing SO level quadratically decreased yolk $C_{18:2}$ concentration. Vitamin D supplementation increased and decreased yolk $C_{18:2}$ concentration when diets contained SO and T, respectively. In conclusion, increasing fat level improved laying performance without altering metabolic profile and egg quality. Vitamin D supplementation had minor alteration effects on laying performance, metabolic profile, and egg quality in response to fat feeding.

Expression of Fas/FasL in CD8+ T and CD3+ Foxp3+ Treg Cells - Relationship with Apoptosis of Circulating CD8+ T Cells in Hepatocellular Carcinoma Patients

  • Aims: Dysfunction of the host immune system in cancer patients can be due to a number of factors, including lymphocyte apoptosis. Several studies showed that $Foxp3^+T$ cells take part in inducing this process by expressing FasL in tumor patients. However, the relationship between apoptosis, $CD8^+T$ cells and $Foxp3^+T$ cells in HCC patients is still unclear. The present study was designed to investigate the correlation between apoptosis levels and Fas/FasL expression in $CD8^+T$ lymphocytes and $Foxp3^+T$ cells in patients with HCC. Methods: $CD8^+T$ cells and $CD3^+Foxp3^+T$ cells were tested from peripheral blood of HCC patients and normal controls and subjected to multicolor flow cytometry. The expression of an apoptosis marker (annexin V) and the death receptor Fas in $CD8^+T$ cells and FasL in $CD3^+Foxp3^+T$ cells were evaluated. Serum TGF-${\beta}1$ levels in patients with HCC were measured by enzyme-linked immunosorbent assay. The relationship between apoptosis and Fas expression, as well as FasL expression in $CD3^+Foxp3^+T$ cells was then evaluated. Results: The frequency of $CD8^+T$ cells binding annexin V and Fas expression in $CD8^+T$ cells, were all higher in HCC patients than normal controls and the proportion of apoptotic $CD8^+T$ cells correlated with their Fas expression. Serum TGF-${\beta}1$ levels correlated inversely with $CD3^+Foxp3^+T$ cells. Conclusions: Fas/FasL interactions might lead to excessive turnover of $CD8^+T$ cells and reduce anti-tumor immune responses in patients with HCC. Further investigations of apoptosis induction in $Fas^+CD8^+T$ cells in vitro are required.

Study of Thyroid-Related Hormones During Second Trimester of Pregnancy

  • A study during second trimester of pregnancy was conducted among 100 women from a population with mild to moderate deficient iodine intake (median 7 ${\mu}g$/L)to monitor the changes in serum free thyroxin ($FT_4$), triiodothyionine ($T_3$) and thyrtropin (TSH). Thyroid related hormones were assessed using radioimmunoassay (RIA) and immunoradiometricassay (IRMA). As compared with non pregnant women, mean $FT_4$ levels decreased, $T_3$ levels increased and TSH levels were slightly depressed in pregnant women. Mean $FT_4$ concentration reached its highest level in the $4^{th}$ month and linearly decreased until the $6^{th}$ month. Mean $T_3$ level remained almost constant during $2^{nd}$ trimester of pregnancy. Mean TSH level remained unaltered during the $4^{th}$ and $5^{th}$ months and was slightly lower than that for the control group. However, these differences were statistically insignificant At the end of the second trimester, hypothyroxinemia and $T_3$ levels greater than the upper limit of the laboratory reference range were found in $34\%$ and $38\%$ of women, respectively. These changes in thyroid hormones reflect typical adaptations of the thyroid gland during pregnancy to the limited availability of iodine.

A Study on Serum Vitamin E Levels and Deficiency in Cattle (소의 혈청 비타민 E 농도와 결핍증에 관한 연구)

  • Serum $\alpha$-tocopherol was measured in cattle to evaluate normal range and to investigate the difference of $\alpha$-tocopherol levels between healthy cattle and diseased cattle. Seventy two heads of 1 year old beef Cattle have 429.9$\pm$77.2 $\mu\textrm{g}$/100ml of Serum $\alpha$-tocopherol. The Serum $\alpha$-tocopherol values of calves with diarrhea(8 heads), pneumonia(6 heads) and piroplasmosis(4 heads) were 87.1$\pm$19.2, 126.3$\pm$45.7 and 106.3$\pm$30.9$\mu\textrm{g}$/100ml, respectively. But that of calves in good health (S heads) was 357.t$\pm$68.4 $\mu\textrm{g}$/100m1. And the values of diseased calves are significantly lower an that of calves in good health(p<0.05). Seasonaly, serum $\alpha$-tocopherol levels of dairy Holstein cows were 529.9$\pm$ 120.3(March), 540.2$\pm$127.2(June), 566.9$\pm$149.5(September) and 550,0$\pm$ 125.4(December)$\mu\textrm{g}$/100m1, respectively. The values on autumn was the highest than that of othor seasons. Serum H-tocopherol level of cows with retained placenta was 262.2$\pm$40.6$\mu\textrm{g}$/100m1. And the level of retained placenta was significantly lower than that of healthy cattle regardless seasonal variation(p<0.05).

Effects of Smoking, Drinking, Exercise on Body Composition, Nutrient Intakes and Serum Lipids in Male High School Students (흡연, 음주, 운동이 남자 고등학생의 체성분, 영양소 섭취 및 혈청 지질 양상에 미치는 영향)

  • This study was done to investigate effects of smoking, drinking and exercise on body composition, dietary intakes, serum lipids of male high school students in Seoul. Body Composition was analyzed by the Bioelectrical Impedance Fatness Analyzer. Dietary survey was conducted by 24-hour recall method and nutrient intakes were analyzed by the Computer Aided Nutritional analysis program for professional (CAN-Pro). All data were statistically analyzed by SAS PC package program. Mean and standard error were examined for each items. The significant difference was examined by student's t-test. Body composition data showed that protein mass, mineral mass and lean body mass of regular exercisers were significantly higher (p < 0.05) than those of irregular exercisers. Energy and protein intakes of male high school students were 1868 kcal/day (69.2% RDA), 68 g/day (91.3% RDA), respectively. Ca, Fe and Vitamin B$_2$ intakes were lower than 2000 RDA. Analysis of serum lipids showed that serum levels of HDL-cholesterol of drinkers were significantly higher (p < 0.05) than those of nondrinkers. Overall results imply that smoking and alcohol drinking of adolescent males may not much influence on serum lipids levels because pack-years of smoking or the amount of alcohol drinking was not serious enough to be harmful to health.

Effects of Boyangmaksung-bang (BYMSB) on the Membranous Nephropathy Induced by Cationic Bovine Serum Albumin in Mouse Model (보양막성방(補陽膜性方)이 Cationic Boivine Serum Albumin 투여로 유발된 Mouse의 Membranous Nephropathy에 미치는 영향)

  • Objective: The purpose of experimental study was to prove the effects of Boyangmakseong-bang (BYMSB) treatment on cBSA-induced in a MN mouse model. Methods: We divided mice into 4 groups. The Normal group had no treatment. We used cBSA and induced MN mouse model to the other 3 groups. The Control group was treated with cBSA (9mg/kg i.p) only. The second group, named 'BY-250', was treated with cBSA (9mg/kg i.p) and BYMSB extract (250mg/kg, p.o). The third group, named 'BY-500', was treated with cBSA (9mg/kg i.p) and BYMSB extract (500mg/kg, p.o). After cBSA and BYMSB extract treatment for 4 weeks, the increase in percentage of body weight, proteinuria, serum albumin, total cholesterol, creatinine and BUN of all groups were measured. The CD3+, CD19+, CD4+, CD8+ cell levels of spleen of all groups were analyzed. IgA, IgG, IgM, IL-$1{\beta}$, TNF-${\alpha}$, IL-6 and IFN-${\gamma}$ levels of all groups were gauged. H&E staining, immunofluorescence staining and electron microscopy of kidney were observed. Results: BYMSB showed significant decrease in the 24hrs proteinuria, serum total cholesterol, serum IgG levels and BUN levels, and showed significant increase in the serum albumin levels compared with the control group. BYMSB showed increase in the increasing percentage of body weight and IFN-${\gamma}$ levels compared with the control. BYMSB showed decrease in the CD3+ T cells, CD4+ Th cells, IL-$1{\beta}$, TNF-${\alpha}$ and IL-6 levels, but did not show significant change compared with the control. BYMSB showed considerable decrease in the thickening of the GBM on H&E staining, deposition of IgG on immunofluorescence staining and deposition of electron-density on electron microscopy of kidney compared with the control. Conclusions: According to the above results, it is suggested that BYMSB decreases the symptoms of MN induced by cBSA in a mouse model. Therefore BYMSB seems to be applicable to MN in clinical practice.

Effect of Feeding Ammoniated Wheat Straw Treated with Hydrochloric Acid on Blood Biochemical Profile in Growing Male Buffalo (Bubalus bubalis) Calves

  • An experiment was conducted to study the effect of feeding ammoniated wheat straw treated with HCl on blood biochemical profiles in growing male buffalo (Bubalus bubalis) calves. Twenty-four growing male buffalo calves (one year of age, 88.54${\pm}$3.81 kg average body weight) were divided into three groups in a completely randomized design on the basis of their body weight. Animals in all the three groups were fed on concentrate mixture. In addition, they were offered wheat straw, ammoniated wheat straw (4% urea at 50% moisture level) and HCI treated ammoniated wheat straw (4% urea at 50% moisture level and HCl added to trap 30% of the NH3 evolved) in groups I, II and III, respectively for a period of 180 days, as per Kearl (1982) for body weight gain of 500 g/d. In all diets, concentrate:roughage ratio was fixed at 50:50 and were made isonitrogenous by adjusting CP levels of conc. mixtures. Blood was collected from jugular vein of each buffalo calf at the beginning and subsequently at two months interval of experimental feeding. Due to urea-ammoniation, the CP content of wheat straw increased from 2.90 to 6.96% and addition of HCl along with urea further increased the CP content to 10.09%. In all the three groups, the mean values of plasma glucose (mg %) and serum globulin (g %), showed a decreasing trend, while the mean value of serum TP (g %), serum A:G ratio, serum urea (mg %), serum creatinine (mg %), serum ALP (KA units), SGOT (units/ml.), SGPT (units/ml), serum T$_3$ and T$_4$ (ng/ml) showed an increasing trend with the advancement of feeding period. The cumulative period mean values of serum TP (6.15 to 6.20 g %), serum albumin (3.07 to 3.18, g %), serum globulin (2.98 to 3.09, g %), serum A:G ratio (1.03 to 1.10), serum ALP (23.15 to 23.63, KA units), serum T$_3$ (1.20 to 1.23 ng/ml) and serum T$_4$ (21.33 to 21.88 ng/ml) were comparable among the groups. The cumulative period mean plasma glucose (mg %) in group III (57.28) was similar to groups I (55.31) and II (59.41), however, the cumulative period mean plasma glucose in group II was significantly (p<0.01) higher than group I. The cumulative period mean serum urea (mg %) in group III (47.34) was significantly (p<0.001) higher than group I (38.38) and II (42.24), which were statistically alike. However, the cumulative period mean serum creatinine values (mg %) in groups II (1.43) and III (1.52) were similar and were significantly (p<0.01) higher than group I (1.24). The cumulative period mean SGOT (units/ml) in groups I, II and III was 91.71, 96.04 and 96.64, respectively. Similarly the cumulative period mean SGPT (units/ml) was 19.00, 19.93 and 20.01 in groups I, II and III, respectively. The cumulative period mean values of SGOT (p<0.05) and SGPT (p<0.001) in groups II and III were similar and were significantly higher than group I. The cumulative period mean serum T$_3$ and T$_4$ values in groups I (1.21 and 21.81), II (1.23 and 21.42) and III (1.20 and 21.33) were comparable. From the present study it may be concluded that feeding of AWS treated with and without HCI to growing male buffalo calves for 180 days had no significant adverse effect on blood biochemical profile.

Effect of Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparat On Prevention of Osteoporosis In Ovariectomized Rats (생하수오(生何首烏)와 제하수오(製何首烏)가 난소적출(卵巢摘出)로 유발된 흰쥐의 골다공증(骨多孔症) 예방효과에 미치는 영향)

  • Objectives : The present study has been undertaken to investigate the effects of Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparat on prevention of osteoporosis in ovariectomized rats. Method : In this experiment, the rats were ovariectomized. Rats were administered by 2 kinds of medicine, Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparat. Results : The levels of spinal bone mineral density was significantly increased in comparison with OVX group at 6 weeks in Polygoni Multiflori Radix Preparat group. The levels of serum osteoclacin and calcium didn't show significant changes in comparison with OVX group in Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparat group. The levels of serum ALP showed significant decrease in comparison with OVX group at 3 weeks in Polygoni Multiflori Radix group, and showed significant decrease in comparison with OVX group at 6 weeks in Polygoni Multiflori Radix Preparat group. The levels of serum phosphorus showed significant decrease in comparison with OVX group at 3, 6 weeks in Polygoni Multiflori Radix, and showed significant decrease in comparison with OVX group at 6 weeks in Polygoni Multiflori Radix Preparat group. The levels of urine calcium and phosphorus didn't show significant changes in comparison with OVX group in Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparat group. The levels of femoral and fibula-tibial calcium showed significant increase in comparison with OVX group in Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparat group. The levels of femoral phosphorus showed significant increase in comparison with OVX group in Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparat group. The levels of femoral and fibula-tibial ash weight showed significant increase in comparison with OVX group in Polygoni Multiflori Radix Preparat group. And the levels of fibula-tibial ash weight showed significant increase in comparison with OVX group in Polygoni Multiflori Radix group. The levels of body weight and uterus weight didn't show significant changes in comparison with OVX group in Polygoni Multiflon Radix and Polygoni Multiflori Radix Preparat group. Conclusion : Reviewing these experimetal results, it appears that Polygoni Multiflori Radix and Polygoni Multiflori Radix Preparat have efficacy on prevention of osteoporosis. In particular Polygoni Multiflori Radix Preparat showed good efficacy in comparison with Polygoni Multiflori Radix Preparat.

Effects of Supplementing Jeju Native Chicken Meat Fed a Tangerine By-product on Lipid Metabolism, Protein Levels and Enzyme Activities in White Rats (감귤 부산물을 급여한 제주도 토종 닭고기가 흰쥐의 지질대사, 단백질 농도 및 효소 활성에 미치는 영향)

  • In this experiment, three different diets were produced to investigate the effects of Jeju native chicken meat, from chickens that were fed a tangerine by-product, on physiological activities in rats. The first diet did not contain any chicken (TS), the second diet contained 10% chicken that had not been fed the tangerine by-product (T0), and the third diet contained 10% that had been fed the by-products (T1). These diets were provided to 11-week-old male rats for four weeks. Weight gain, feed intake, feed efficiency, liver, kidney, and epididymis fat weights were not significantly different among the TS, T0, and T1 groups. Total lipid, triglycerides, and cholesterol levels in the liver were significantly lower in T0 and T1 than in TS (p<0.05). And total lipid, phospholipid, triglycerides, total cholesterol, HDL-cholesterol, DL+VLDL-cholesterol, HDL-cholesterol/total cholesterol, and atherogenic index levels in the blood serum were similar between T0 and T1, which did not present any significant differences. The feed containing the tangerine by-product did not cause any statistically significant differences in serum protein, glucose, or hemoglobin. Finally, T0 and T1 showed similar trends in terms of $\gamma$-GTP, ALT, AST, and ALP activities, which again did not present any statistically significant differences.
