Effect of Ketamine on the Oxidative Burst Activity of Canine Peripheral Blood Leukocytes In Vitro (In Vitro에서 개 말초혈액 백혈구의 순간산소과소비현상에 대한 케타민의 효과)

  • Ketamine, one of general anesthetics for human and veterinary use, is a non-competitive N-methyl-D-aspartate (NMDA) receptor antagonist which interferes with the action of excitatory amino acids. It has been reported to impair various leukocyte functions. In this study, the effect of ketamine on the oxidative burst activity (OBA) of canine peripheral blood leukocytes was examined. The OBA of canine peripheral blood phagocytes was analyzed by flow cytometry system. Ketamine at higher concentration such as $1,000{\mu}M$ exhibited a low viability of leukocytes. Thus, ketamine was used at concentration of 10 to $500{\mu}M$ showing no cytotoxic effect and high cell viability. The OBA of leukocytes in the presence or absence of latex beads was analyzed by addition of dihydrorhodamine 123. The direct treatment of ketamine revealed the inhibitory effect on the OBA of peripheral blood polymorphonuclear cells (PMN) and monocyte-rich cells but not peripheral blood mononuclear cells (PBMC) in the presence of latex beads. However, when latex beads were not added to PMN, its OBA was not inhibited by ketamine. The OBA of PMN and monocyte-rich cells but not PBMC in the presence of latex beads was also inhibited by culture supernatant from ketamine-treated- PBMC but not -PMN. But the OBA of PMN in the absence of latex beads was not inhibited by culture supernatant from PBMC treated with ketamine. Therefore, these results suggested that ketamine has the inhibitory effect on the OBA of canine peripheral blood phagocytes such as neutrophils and monocytes during phagocytic response.

A Characteristics of Phagocytic Plaque on Staphylococcus aureus Layer Formed by Leukocytes of the Alcoholics (Staphylococcus aureus를 이용한 주정중독환자 백혈구의 Phagocytic Plaque 형성력)

  • This study was conducted to develop a method for direct determination of phagocytic activities in human circulatic systems and to measure the phagocytic activities in human leukocytes from the alcoholics, since phagocytic activity was considered to be very important in human immune mechanism at early stage for the health care of the alcoholics. The subjects for this study were 130 among which 95 males and 3 females were diagnosed as alcoholism and 32 was healthy blood donors. A thin layer of heat-killed Staphylococcus aureus Cowan I was placed on a plastic dish and reacted with whole blood to measure the phagocytic plaque formation by human leukocytes. In order to determine the health conditions of the subjects, some clinical laboratory tests, such as white blood cell counts, hemoglobin contents (Hgb), mean corpuscular volume of red blood cells(MCV), serum electrophoresis, B and T-lymphocytes, T-lymphocyte subtypes and phytohemagglutination test were also implemented. Compared to the non-alcoholism, new and old alcoholic inpatients showed statistically significant differences on levels of Hgb and MCV (p<0.05), but showed that T and B-lymphocyte numbers decreased and Helper T cell/Suppressor T cell ratio ($1.6{\pm}0.8$%) increased. Compared to non-alcoholism, phagocytic plaque activities of leukocytes from alcoholic patients decreased significantly and an unusual pattern in phagocytic plaque was observed, showing a strange body and chain shaped phagocytosis. Based upon these results, it is concluded that a phagocytic-plaques of Staphylococcus aureus Cowan I by leukocytes was very simple and useful method for the early immunological determination of phagocytic activities in alcoholic patients without requiring any special equipments.

Hematological Studies in Reference to Immune Enhancement Mechanism of Formalin Applied to Formalin Inactivated Anthrax Vaccines in Rabbits (Formalin 처리(處理) 면역원(免疫原)에 대한 가토(家兎)의 혈액세포상(血液細胞像)에 관한 연구)

  • Hematological studies were conducted in reference to the immune enhancing mechanism of formalin applied to, as an inactivating agent, a formalin inactivaed anthrax vaccines in rabbits. Rabbits were inoculated two types of formalinized anthrax immunogens namely capsular and spore vaccines in addition of formalin saline as a control. From immune rabbits, peripheral blood was collected and subjected to count a total erythrocytes, leukocytes, and pyroninophilic lymphocytes. The experimental results were summarized as followings. At a level of 0.5M 0.5ml formalin with or without the addition to vaccine, a total leukocytes count was increased. Due to the increased lymphocytes, the ratio of neutrophil and lymphocyte was lowered within 4 to 12 hours of the postinoculation. Formalin saline, anthrax spore vaccine and capsular vaccine, without group difference, caused an increased level of pyroninophilic lymphocytes in peripheral blood. Throughout the studies, a possible role of immune enhancement by formlin was disscused and suggested.

Recent evidence has indicated that polymorphonuclear leukocytes (PMNs) have the ability to release IL-1 and $TNF-{\alpha}$ Calcium hydroxide is an effective medicament in root canal infections, reducing the microbial titre within the canal. It has been proposed that the therapeutic effect of $Ca(OH)_2$ may also be the result of direct inactivation of LPS.(omitted)

Levamisole Enhances the Natural Cytotoxic Cell Activity of Japanese Flounder (Paralichthys olivaceus) Head Kidney Leukocytes

  • Japanese flounder (Paralichthys olivaceus) head kidney (HK) leukocytes were incubated with $10^3$ to $10^{-3}$ ng levamisole/ml for 4, 24 or 48 h and then assayed for their natural cytotoxic activity against xenogeneic tumor cells. This activity was slightly increased after 24 h of incubation. In a second experiment, fish were fed 0, 75, 150 or 300 mg levamisole/kg diet for 10 consecutive days. The fish were then fed a commercial non-supplemented diet and sampled 0, 1, 2, 3, 4 or 6 weeks post-administration of levamisole. The cytotoxic activity was found to be increased with increasing levamisole dose and remained greatly enhanced until the end of the experiment. In conclusion, levamisole enhanced flounder natural cytotoxic cell activity both in vitro and in vivo and had a great lasting action when administered by feeding.

Efficacy of Intracytoplasmic Sperm Injection for Leukocytospermia

  • White blood cells (WBCs) are present in most human ejaculates, but abnormally high concentration of seminal leukocytes may reflect an underlying pathological condition. The World Health Organization (WHO) has defined leukocytospermia as status of more than $10^6$ WBC/mL of semen. The purpose of this study was firstly, to compare the outcomes between conventional IVF and ICSI in leukocytospermia, and secondly, to investigate whether ICSI may be an alternation treatment for patients with leukocytospermia. Total 346 cycles of conventional IVF and ICSI candidates underwent IVF cycles at Eulji Hospital Infertility Clinic. Semen Parameters including concentration, motility, morphology of spermatozoa and concentration of leukocytes were assessed from the raw ejaculates. There was no difference in sperm concentration, motility and morphology. The rates of fertilization and good embryo development from ICSI were significantly higher than those from conventional IVF in leukocytospermia (60.4% & 32.5%, respectively for ICSI group and 44.4% & 28.5%, respectively for IVF group, P<0.00l). The pregnancy rate after ICSI was also higher than that from conventional IVF (34.0% vs 29.1 %, P<0.05). These results indicate that the presence of seminal leukocyes (> 1$\times10^6$ WBC/mL of semen) is adversely related with fertilization, embryo development and pregnancy rate. Therfore the measurement of seminal leukocytes in routine semen analysis appears to be of prognostic value with regard to male fertilizing potential. In conclusion, it is suggested that ICSI is an alternative choice of treatment for patients with leukocytospermia.
