  • A new phenolic compound and three known flavonoids isolated from the MeOH extracts of Lespedeza tomentosa. Based on spectral data, the isolated compounds were identified as methyl 4,5-dihydroxy-3-methoxy-2-(3-methylbut-2-en-1-yl)benzoate (1), 1-methoxylespeflorin G11 (2), farrerol (3) and 1-methoxylespeflorin I2 (4). Methyl 4,5-dihydroxy-3-methoxy-2-(3-methylbut-2-en-1-yl)benzoate (1) is newly isolated from plant source.

Breaking Physical Dormancy with Sulfuric Acid in Seeds of Lespedeza tomentosa (Thunb.) Siebold ex Maxim (황산처리를 이용한 개싸리 종자의 물리적 휴면 타파)

  • Lespedeza species are mainly used for wildlife food and cover and for erosion control. The germination of these species can be enhanced after a fire occurrence in forest, which is known as fire-activated seeds to germinate. While the heat treatment could break seed dormancy of Lespedeza, its germination rate was quite low. We investigated that chemical scarification could promote germination of L. tomentosa. Seeds were soaked in 100% sulfuric acid (H2SO4) for 0, 1, 3, 6, 12, 24, 48, 96, 192, and 384 min, and then washed in distilled water for 24 h. Very few seeds were germinated in control (H2SO4 for 0 min). More than 90% of seeds were germinated in H2SO4 for 24, 48, and 92 min. However, some damage was observed in roots and cotyledons of seedling dipped in H2SO4 for a long time. To search the optimal soaking time in H2SO4 without defects, seeds scarified in H2SO4 for 30, 60, 90, 120, 150, 180, and 300 min were sown the commercial soil medium. Seeds treated with H2SO4 for 90 min and 150 min emerged by about 92% and 84%, respectively. Therefore, H2SO4 treatment could break the seed dormancy of Lespedeza species, and especially in case of L. tomentosa the optimal treatment time in sulfuric acid was one to two hours. Germination of L. tomentosa began promptly following the scarification and was completed within about one month, indicating that seeds has no physiological dormancy, just has physical dormancy.

Hepatoprotective Effects of Plants Extracts from Baekdu Mountain on Tacrine-induced Cytotoxicity in HepG2 Cells (타크린으로 유발한 간 세포 독성에 대한 백두산 식물 추출물의 보호 효과)

  • The present study represents the screening of the protective effects of herbal methanolic extracts from Baekdu mountain against tacrine-induced cytotoxicity in HepG2 cells. Tacrine is an acetylcholinesterase inhibitor, and used for the treatment of Alzheimer's disease. However, administration of tacrine for the treatment of Alzheimer's disease results in a reversible hepatotoxicity in 30-50% of patients, consequently limiting clinical use. Therefore, studies for natural products with protective effect on the tacrine-induced hepatotoxicity would be valuable as providing potential therapeutic use. 109 plant sources were collected in Baekdu mountain, and extracted with methanol. These extracts had been screened the protective effects against tacrine-induced cytotoxicity in HepG2 cells at the 100 and 300 ${\mu}g/ml$. Of these, ten methanolic extracts, roots of Ampelosis japonica, aerial parts of Berberis amurensis, aerial parts of Sedum aizoon, aerial parts of Lespedeza tomentosa, aerial parts of Lespedeza juncea, aerial parts of Hypenricum ascyron, stem barks of Syringa reticulata, fruits of Gleditsia japonica, aerial parts of Chamaenerion angustifolium, branches of Ginkgo biloba, showed significant protective effects against tacrine-induced cytotoxicity in HepG2 cells.

Antioxidant and Nicotine Degradation Effects of Medicinal Herbs

  • Antioxidant activity and nicotine degradation activity (NDA) of 21 medicinal herbs were determined by using a 1,1-diphenol-2-picrylhydrazyl (DPPH) method and a PLC/PRF5 human liver cell line method, respectively, to develop an anti-smoking aid. The highest and lowest antioxidant activities represented by $IC_{50}$/ value were 30 $\mu$g/mL of Eugenia caryophyllus and 3,270 $\mu$g/mL of Panax ginseng C. A. Meyer, respectively. Antioxidant activity of Eugenia caryophyllus was equal to 38.0$\pm$1.2 mg VCEAC(vitamin C equivalent antioxidant capacity)/ g herb. The highest and lowest NDA values were 1.81 of Astrgalus membranaceus Bunge and 1.01 of Raphani seed and Lespedeza tomentosa Sieb, respectively. Eleven medicinal herbs with high antioxidant activity and/or NDA were selected to make an herbal tea. The herbal tea had high antioxidant activity (50 $\mu$g/mL $IC_{50}$/ and 22.4$\pm$1.4 mgVCEAC/g) and NDA (1.243). The medicinal herb tea could help smokers quitting smoking by degrading and exhausting nicotine accumulated in body and removing reactive oxygen species.

Vegetation Structure of Some Abandoned Coal Mine Lands in Mungyeong Area (경북(慶北) 문경지역 폐탄광지(廢炭鑛地)의 식생구조(植生構造)에 관한 연구(硏究))

  • Vegetation structure of Samchang, Bongmyeong, Kabjeong, Danbong coal mine and control site was investigated to provide basic information on vegetation rehabilitation of abandoned coal mine lands in Mungyeong. Gyeongbuk Province. The middle and lower layer consisted of 3 and 10 species for Samchang, 2 and 5 species for Bongmyeong, 7 and 11 species for Kabjeong, 9 and 12 species for Danbong coal mine, and 15 and 17 species appeared for control site, respectively. Importance value of Lespedeza cyrtobotrya in lower layer was the highest for in Samchang, and that of Rubus crataegifolius in other coal mine lands wast he highest. Importance value in herbaceous layer was the highest for Arundinella hirta in Samchang, for Corchoropsis tomentosa in Bongmyeong, for Miscanthus sinensis in Kabjeong, for Carex lanceolata and Artemisia princeps var. orientalis in Danbong coal mines, and Calamagrostis arundinacea in control site was the highest, respectively. Species diversity of middle and lower layer for four coal mine lands ranged from 0.201 to 0.666 and 0.612 to 0.895, respectively. Evenness of lower layer ranged from 0.683 to 0.875 for four coal mine lands, and it was 0.990 for control site.
